BACKGROUND: Analyte-triggered semiconductor quantum dots (QDs) modulation in the presence of non-consistently responsive fluorescent species represents a challenging analytical issue in concrete multi-way data handling. QDs with heterogeneous sizes and/or uneven distribution of functional moieties on their surfaces exhibit significant fluctuations in the fluorescent response components, known as chemical rank, across different excitation/emission modes. This phenomenon may lead to a substantial deviation from the proportionality prescribed by Beer-Lambert law. Nonetheless, even in the presence of such deviation, a multi-way model may be successfully selected after determining a proper chemical rank in a QDs system. RESULTS: We show that in a valid PARAllel FACtor (PARAFAC) model under properly determined chemical rank, meaningfully resolved pure spectral profiles can be reached for each fluorescent responsive constituent in the original excitation-emission fluorescence matrix (EEFM) measurements. This was thoroughly illustrated by applying PARAFAC trilinear decomposition of a three-way data array of two distinct datasets acquired from semiconductor QDs sensing systems with low-rank trilinear assumption. The first dataset, presented here for the first time, comprises EEFM measurements of the ligand-driven quenching of thiomalic acid (TMA)-capped AgInS2 (AIS) QDs by vomitoxin. The second dataset, employed for illustrative purposes, comprises EEFM measurements of the quenching, via cation bridging, of glutathione (GSH)-capped CdTe QDs by Pb(II). The results of this study enabled the determination of vomitoxin at a ppb level in real samples of fish feeds, showcasing the efficacy of the PARAFAC model in resolving spectral signatures (loadings) and pure concentration profiles (scores). SIGNIFICANCE: PARAFAC under a properly examined chemical rank can be easily adapted for retrieval the underlying Beer-Lambert law of the original EEFM measurements with a low-rank trilinear structure through the chemically meaningful information either when (i) no deviation of Beer-Lambert law was observed as deeply discussed in connection with the dataset acquired from vomitoxin-driven molecular sensing through TMA-capped AIS QDs, or when (ii) substantial deviations of the Beer-Lambert law are evident, as discussed in connection with the dataset collected from sensing ionic species through Pb(II) bridging of GSH-capped CdTe QDs.

According to the World Health Organization, 17 million people die every year of cardiovascular diseases (CVD), particularly heart attacks and strokes. More than half of all deaths across the Europe are caused by CVDs. 80 % of premature deaths from these causes could be avoided by controlling the main risk factors: tobacco, unhealthy diet and physical inactivity. Local authorities provide sport and recreation facilities to their communities. They also have a central role in forging partnerships with public and private sector to enhance the local sport delivery system. The present paper consists of an empirical exercise about the statistical relationship between the financial effort of Portuguese municipalities in the implementation of sports policies and the prevalence of CVDs. The estimation of a Spatial Autocorrelation Model confirms the strong spatial dependence between neighbouring municipalities and validates the existence of a positive impact of local sports policies on mortality rate from cardiovascular diseases. The results raise the need to reinforce local policies to promote active living based on holistic approaches to combating heart disease and promoting healthy lifestyles in partnerships with health professionals, educators, and community leaders.

Animal tuberculosis (TB) is often maintained by multi-host communities, including livestock and wildlife. Quantitative studies of such communities require estimating the true prevalence of TB, correcting the apparent prevalence by the diagnostic sensitivity (Se) and specificity (Sp) of the test. The goal of this study was to lay the foundations for estimating the true prevalence of TB in wild ungulate populations (wild boar and two cervids: red deer and fallow deer). We used Bayesian latent class models to assess the Se and Sp of gross pathology, IS6110 real-time PCR in tissues, bacteriological culture, and P22 indirect ELISA. We analyzed 308 harvested wild ungulates (211 wild boar and 97 cervids: 92 red deer and 5 fallow deer). The Se of bacteriological culture (80.4%, CI95 61.0-96.3%) and gross pathology (87.9%, CI95 69.5-99.9%) was reasonably good in wild boar. These tests showed lower Se in cervids: 60.2% (CI95 38.3-82.3%) for bacteriological culture and 81.5% (CI95 63.6-96.2%) for gross pathology. The Se of the real-time PCR was low (50.7% in wild boar and 53.0% in cervids). These tests showed Sp between 95.2 and 99.1% in both taxa. The P22 ELISA performed reasonably well in wild boar (Se = 71.9%, CI95 59.2-83.4%; Sp = 98.8%, CI95 96.9-99.9%) but lacked Sp in cervids (Se = 77.1%, CI95 62.9-89.7%; Sp = 74.5%, CI95 65.7-83.3%). The real-time PCR in wild boar and cervids and bacteriological culture in cervids tended to show higher Se in low-prevalence populations, possibly due to a higher proportion of early-stage TB lesions. In cervids, the parallel interpretation of gross pathology and bacteriological culture significantly improved the diagnostic performance (Se = 93.1%, CI95 84.7-98.9%; Sp = 92.9%, CI95 86.0-98.3%). Our results allow the estimation of true prevalence from the results of a single diagnostic test applied to harvested wild boar, red deer, and fallow deer, paving the way for more precise quantitative ecological studies of the multi-host TB maintenance community.

Agricultural pests can cause direct damage to crops, including chlorosis, loss of vigor, defoliation, and wilting. In addition, they can also indirectly damage plants, such as by transmitting pathogenic micro-organisms while feeding on plant tissues, affecting the productivity and quality of crops and interfering with agricultural production. Among the known arthropod pests, mites are highly prevalent in global agriculture, particularly those from the Tetranychidae family. The two-spotted spider mite, Tetranychus urticae, is especially notorious, infesting about 1600 plant species and causing significant agricultural losses. Despite its impact on agriculture, the virome of T. urticae is poorly characterized in the literature. This lack of knowledge is concerning, as these mites could potentially transmit plant-infecting viral pathogens, compromising food security and complicating integrated pest management efforts. Our study aimed to characterize the virome of the mite T. urticae by taking advantage of publicly available RNA deep sequencing libraries. A total of 30 libraries were selected, covering a wide range of geographic and sampling conditions. The library selection step included selecting 1 control library from each project in the NCBI SRA database (16 in total), in addition to the 14 unique libraries from a project containing field-collected mites. The analysis was conducted using an integrated de novo virus discovery bioinformatics pipeline developed by our group. This approach revealed 20 viral sequences, including 11 related to new viruses. Through phylogenetic analysis, eight of these were classified into the Nodaviridae, Kitaviridae, Phenuiviridae, Rhabdoviridae, Birnaviridae, and Qinviridae viral families, while three were characterized only at the order level within Picornavirales and Reovirales. The remaining nine viral sequences showed high similarity at the nucleotide level with known viral species, likely representing new strains of previously characterized viruses. Notably, these include the known Bean common mosaic virus (BCMV) and Phaseolus vulgaris alphaendornavirus 1, both of which have significant impacts on bean agriculture. Altogether, our results expand the virome associated with the ubiquitous mite pest T. urticae and highlight its potential role as a transmitter of important plant pathogens. Our data emphasize the importance of continuous virus surveillance for help in the preparedness of future emerging threats.

Biochar is an effective material for enhancing soil ecosystem services. However, the specific impacts of biochar on microbial indicators, particularly in degraded soils, remain poorly understood. This study aimed to evaluate the effects of biochar produced from cashew residues and sewage sludge, in a highly degraded soil, on microbial indicators. We analyzed soil chemical composition and microbial biomass C and N, enzyme activity, and stoichiometry. Cashew biochar increased soil respiration, indicating a higher availability of C to microorganisms compared to sewage sludge biochar and a better adaptation of soil microbial communities to C-rich organic material obtained from a native plant. Although the biochar differentially impacted microbial biomass C, both significantly increased N in the microbial biomass. Arylsulphatase activity did not respond to biochar application, while ß-glucosidase, urease, and phosphatases showed significant changes with biochar treatments. Importantly, stoichiometry and vector analysis revealed that both types of biochar increased P limitation for soil microbes. Conversely, both biochar alleviated C and N limitations for the soil microbes. Thus, biochar applications in highly degraded soils should be supplemented with external P sources to maintain soil functions, mainly for cashew residues. Our results provide evidence that biochar can restore soil biological properties and enhance the availability of C and N to microorganisms. These findings have significant implications for restoration practices in degraded lands of semiarid regions.

Crop landcover datasets are crucial for modern agriculture, aiding farmers, researchers, policymakers, and stakeholders. These databases offer extensive insights into crop distribution, facilitating informed decision-making for sustainable practices, particularly under a changing climate. Moreover, these datasets drive research, fostering collaborations and innovation for resilient agriculture. In Portugal, the COS dataset is vital, offering insights into agrarian landscapes and supporting sustainable practices. However, in recent versions, since 2007, information on permanent crops has been aggregated, necessitating complementary datasets and tools. The current paper addresses this gap by providing an open-source dataset focusing on perennial crops in mainland Portugal. Based on the 2019 agricultural census from the Portuguese Statistical Institute (INE), this dataset contributes to the spatial understanding of permanent crop distribution, being freely available for researchers, farmers and policymakers. The dataset includes a selection of perennial crops commonly cultivated in Portugal, such as Prunus dulcis (Almond), Malus domestica (Apple), Castanea sativa (Chestnut), Ceratonia siliqua (Carob), Prunus avium (Sweet Cherry), Vitis vinifera (Grapevine), Olea europaea (Olive), Citrus limon (Lemon), Citrus sinensis (Sweet Orange), Juglans regia (Walnut), Citrus reticulata (Mandarin), Prunus persica (Peach), Pyrus communis (Pear), and Prunus domestica (Plum). Further information regarding the Administrative Units of each crop is also available. This comprehensive list provides a detailed overview of the types of permanent crops included in the dataset, offering valuable insights into the Portuguese agricultural landscape.

Objectives: The association between ultra-processed foods (UPF) consumption and cognitive performance needs to be better characterized in adolescents, especially in low-income settings, where the cost of human capital is high. This study investigated the association between cognitive performance and UPF in adolescents from the countryside of the Brazilian Northeast. Methods: Adolescents (15-18 years old) from three public high schools were included. Food intake was assessed using three 24-hour dietary recalls. The classification of foods as UPF was determined according to the Nova classification. Cognitive performance was evaluated using the Non-Verbal General Intelligence Test. Results: 116 adolescents were included, of which 50 (43.1%) showed low cognitive performance. The average energy intake was 1973.5 kcal, with 24.2% coming from UPF. Participants with low cognitive performance consumed 26.5% (95% CI: [22.2; 30.7]%) of daily energy intake from UPF compared to 22.5% ([18.8; 26.2]%) of those with medium-high cognitive performance (P = 0.17), without differences in energy and macronutrient intake. Conclusion: Despite similar UPF consumption compared to the Brazilian average, no association was found between UPF consumption and cognitive performance in this low-income adolescent sample.

BACKGROUND: The Golden Syrian hamster (Mesocricetus auratus), Ferrets (Mustela putorius furo), and macaques have been described as useful laboratory animals naturally susceptible to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. OBJECTIVES: To study the mechanism of lung injury, we describe the histopathological features of SARS-CoV-2 infection in Golden Syrian hamsters inoculated intranasally with the A.2 Brazilian strain. METHODS: Hamsters were intranasally inoculated with the A.2 variant and euthanised at 3-, 5-, 10- and 15-days post-inoculation. The physical examination and body weight were recorded daily. Neutralising antibodies and viral RNA load of the respiratory tract were assessed during necropsies. FINDINGS: The coronavirus disease 2019 (COVID-19) model presented body weight loss, high levels of respiratory viral RNA load, severe segmentary pneumonitis, and bronchial fistula besides lymphatic trapping and infiltration, like the human SARS-COV-2 pathogenesis. The presence of subepithelial lymphoeosinophilic infiltrate was highlighted in our results; it contributed to the detachment of SARS-CoV-2 nucleocapsid-positive epithelial cells resulting in the infectious cell plugs. MAIN CONCLUSIONS: The SARS-CoV-2 caused segmentary pneumonia and vascular damage. In our comprehension, the infectious cell plugs, as being aspirated from the upper respiratory tract into the terminal bronchial lumen, work as a "Trojan horse", thus contributing to the dissemination of the SARS-CoV-2 infection into specific regions of the deep lung parenchyma.

The present study aimed to evaluate the anti-staphylococcal, antibiofilm, cytotoxicity and trypanocidal activity, mechanisms of parasite death and immunomodulatory effect of CrataBL encapsulated into liposomes (CrataBL-Lipo). CrataBL-Lipo were prepared by the freeze-thaw technique and characterized. Anti-staphylococcal and antibiofilm activities of CrataBL and CrataBL-Lipo were evaluated against standard and clinical strains of Staphylococcus aureus susceptible and resistant. Thus, broth microdilution method was performed to determine the Minimum Inhibitory Concentration (MIC). Antibiofilm activity at subinhibitory concentrations was evaluated using the crystal violet staining method. Cytotoxicity of CrataBL-Lipo was verified in L929 fibroblasts and J774A.1 macrophages by determining the inhibitory concentration necessary to kill 50 % of cells (IC50). Trypanocidal activities of CrataBL-Lipo was evaluated in Trypanosoma cruzi and the efficacy was expressed as the concentration necessary to kill 50 % of parasites (EC50). The mechanisms of parasite death and immunomodulatory effect of CrataBL-Lipo were evaluated using flow cytometry analysis. CrataBL-Lipo presented Ø of 101.9 ± 1.3 nm (PDI = 0.245), ζ of +33.8 ± 1.3 mV and %EE = 80 ± 0.84 %. CrataBL-Lipo presented anti-staphylococcal activity (MIC = 0.56 mg/mL to 0.72 mg/mL). CrataBL-Lipo inhibited 45.4 %-75.6 % of biofilm formation. No cytotoxicity of CrataBL-Lipo was found (IC50 > 100 mg/L). CrataBL-Lipo presented EC50 of 1.1 mg/L, presenting autophagy, apoptosis and necrosis as death profile. In addition, CrataBL-Lipo reduced the production of IL-10 and TNF-α levels, causing an immunomodulatory effect. CrataBL-Lipo has a therapeutic potential for the treatment of staphylococcal infections and Chagas disease exhibiting a high degree of selectivity for the microorganism, and immunomodulatory properties.

OBJECTIVE: To describe a protocol to achieve consensus on valid and important indicators to assess primary health care (PHC) quality regarding all clinical contexts of PHC in European health systems. STUDY DESIGN: Qualitative study using the Delphi technique to gain consensus among European panels of experts comprising a heterogeneous professional background. METHODS: Potential candidate indicators were extracted and translated according to a set of informative elements (i.e., name, description, formula, unit of analysis, and sources). This list was then independently reviewed, and duplicates were removed totaling 1726 indicators. To guarantee a good response rate, indicators were distributed across 57 Delphi panels organized by clinical context. Each panel is a Delphi process, assessing between 23 to 33 indicators. Experts' opinions on the validity and importance of the extracted indicators will be obtained through two rounds of online questionnaires, using a 9-point Likert scale and free-text boxes. To prevent biased responses, participation will be anonymous to other participants and to the team administrating panels. Consensus will be considered if at least 70% of ratings (≥7 assuming 10 participants) lie within the 7-9 range and less than 15% of ratings (<2 assuming 10 participants) are in the 1-3 range. Analysis of results will be streamlined and generalizable across panels using scripts. CONCLUSIONS: This protocol will contribute to improve the quality of PHC in Europe by achieving a consensual and concise list of PHC quality indicators retrieved from the scientific literature that fit current clinical guidelines and populations' needs in countries from the European region according to the World Health Organization.

Obsessive-compulsive disorder is a psychiatric disorder characterized by intrusive thoughts and repetitive behaviors. There are two prominent features: Harm Avoidance (HA) and Incompleteness (INC). Previous resting-state studies reported abnormally elevated connectivity between prefrontal cortical (PFC) and subcortical regions (thalamus, striatum) in OCD participants. Yet, little is known about the white matter (WM) structural abnormalities in these connections. Using brain parcellation and segmentation, whole brain tractography, and Neurite Orientation Dispersion and Density Imaging (NODDI), we aimed to characterize WM structural abnormalities in OCD vs. healthy controls and determine the extent to which NODDI indices of these connections were associated with subthreshold-threshold HA, INC and overall OCD symptom severity across all participants. Four PFC regions were segmented: ventral medial (vmPFC), ventrolateral (vlPFC), dorsomedial (dmPFC), and dorsolateral (dlPFC). NODDI Neurite Density (NDI) and Orientation Dispersion (ODI) indices of WM structure were extracted from connections between these PFC regions and the thalamus (42 OCD, 44 healthy controls, mean age[SD] = 23.65[4.25]y, 63.9% female) and striatum (38 OCD, 41 healthy controls, mean age[SD] = 23.59[4.27]y, 64.5% female). Multivariate analyses of covariance revealed no between-group differences in these indices. Multivariate regression models revealed that greater NDI in vmPFC-thalamus, greater NDI and ODI in vmPFC-striatum, and greater NDI in dmPFC-thalamus connections were associated with greater INC severity (Q ≤ 0.032). These findings highlight the utility of NODDI in the examination of WM structure in OCD, provide valuable insights into specific WM alterations underlying dimensional INC, and can facilitate the development of customized treatments for OCD individuals with treatment-resistant symptoms.

OBJECTIVES: Sleep and internalizing problems escalate during adolescence and can negatively impact long-term health. However, the directionality of this risk-relationship remains poorly understood within a developmental context. The current study aimed to determine the directionality of this relationship in adolescents with no history of psychiatric disorder and whether sex at birth played a role in this relationship. METHODS: We used data from the Adolescent Brain Cognitive Development, an ongoing multisite longitudinal US study, that covered four waves (W1:9-11 years; W2:10-12 years; W3:11-13 years; W4:12-14 years). Analyses included 3,128 youth (50.99%girls) with no past or current psychiatric disorders at W1. The Sleep Disturbance Scale for Children and the Child Behavior Checklist were used to measure sleep and internalizing problems. Cross-lagged panel models were used to evaluate the cross-lagged relationships across waves. RESULTS: The sleep-internalizing cross-lagged relationship was unidirectional, with medium-large effect sizes: greater total sleep problems were associated with more severe internalizing problems at later waves (W2âW3, coefficient = 0.052, p = .021; W3âW4, coefficient = 0.091, p < .001), with problems in initiating and maintaining sleep predicting internalizing problems early on. Girls showed greater sleep-internalizing risk than boys. CONCLUSIONS: Sleep-internalizing relationships change across adolescence, becoming significant and more specific from early to mid-adolescence. Sleep interventions delivered in early adolescence, to girls in particular, may have a positive short and long-term impact on internalizing outcomes.

OBJECTIVES: To analyze the immediate impact of vocal demand, probability of dysphonia, and performance aspects in amateur protestant singers. METHODS: Cross-sectional, analytical study, with 76 amateur singers from a protestant church, 60 women and 16 men. All participants responded to a sociodemographic and performance questionnaire, the Evaluation of the Ability to Sing Easily for Brazil (EASE-BR), and the Dysphonia Screening Tool (DST-Br). The data were analyzed descriptively and inferentially, considering a significance level of 5%. RESULTS: The singers reported good voice status after a performance, with the possibility of singing again if necessary. Most individuals had a low probability of dysphonia, showing a lower mean score compared to those with a moderate probability. There was an association between "trying to sing louder than others" and "vocal self-assessment" with the immediate impact of vocal demand and dysphonia screening. Implementing vocal rest was significantly associated with EASE-BR, and trying to sing louder than the instruments and being able to hear yourself well while singing and water intake were associated with the total DST-Br score. CONCLUSIONS: The immediate impact of vocal demand after performance on this population did not compromise their ability to sing easily. Most singers had a low probability of dysphonia. "Trying to sing louder than the instruments" and "ability to hear oneself well while singing and water intake" were associated with dysphonia screening.

In this work, the photo-, electro-, and photo-electro-oxidation of chloramphenicol was investigated. The photo-experiments were carried out with different irradiation sources (an ultraviolet and a simulated solar source) using self-doped titanium nanotubes (SDTNT), a very promising and innovative material that deserves further investigations in the degradation of different pollutants. The photo-electrooxidation (j = 15 mA cm-2) under simulated solar irradiation presented the best efficiency, with ca. 100% degradation and kinetic constant of k = 0.04427 min-1. The FTIR analysis demonstrated a structural modification of the standard molecule occurred for all conditions used, suggesting a modification in functional groups responsible for the biological activity. Furthermore, the TOC analysis showed a significant mineralization of the pollutant (66% from the initial concentration). In addition, both photo-electrooxidation approaches have demonstrated a positive value of S, where the simulated solar irradiation reached the highest value S = 0.6960. The experimental results pointed out evidence that the methodology employed herein for chloramphenicol degradation is greatly interesting and the photo-electrooxidation under simulated solar irradiation is a promising approach for this purpose.

The objective was to identify through integrative review the approach on maternity and motherhood in street situation. This is an integrative review performed in the databases PubMed, BIREME, Medline, and SciELO search portal, in Portuguese, English and Spanish. The survey was conducted between October and December 2021, following the recommendations of the Prisma flowchart and support of the Mendeley software, submitted to content analysis by Minayo. Eighteen articles were selected. The reading allowed the identification of two thematic categories: "Maternity and mothering in the streets" and "Main challenges experienced in maternity and mothering in street situation". Address about motherhood and mothering as a field of knowledge production creates a tension of the social constructions that do not have effecting rights for lack of programs and forceful intersectoral policies. In addition to the stigmatization for use of psychoactive substances, as well as the patriarchal structure attributed to motherhood. The responsibility for care entails several issues that cross the motherhood and mothering in the streets, namely: concern with the health of children, the fear of losing custody of their loved ones.
Objetivou-se identificar através da revisão integrativa a abordagem sobre a maternidade e maternagem em situação de rua. Trata-se de uma revisão integrativa realizada nas bases de dados PubMed, BIREME, Medline, e portal de buscas SciELO, em português, inglês e espanhol. A pesquisa foi realizada entre os meses de outubro a dezembro de 2021, seguindo às recomendações do fluxograma Prisma e suporte do software Mendeley, submetidos à análise de conteúdo de Minayo. Foram selecionados 18 artigos. A partir da leitura foi possível identificar duas categorias temáticas: "Maternidade e maternagem nas ruas" e "Principais desafios vivenciados na maternidade e maternagem em situação de rua". Abordar sobre a maternidade e maternagem enquanto um campo de produção de saberes cria-se um tensionamento das construções sociais que não possuem efetivação de direitos por falta de programas e políticas intersetoriais contundentes. Além da estigmatização por uso de substâncias psicoativas como também a estrutura patriarcal atribuída à maternidade. A responsabilidade pelo cuidado acarreta diversas questões que atravessam a maternidade e maternagem nas ruas, são elas: preocupação com a saúde dos filhos, a criação da prole, bem como, o temor pela perda da custódia de seus entes.

The Brazilian western Amazon is experiencing its largest laboratory-confirmed Oropouche virus (OROV) outbreak, with more than 6,300 reported cases between 2022 and 2024. In this study, we sequenced and analyzed 382 OROV genomes from human samples collected in Amazonas, Acre, Rondônia and Roraima states, between August 2022 and February 2024, to uncover the origin and genetic evolution of OROV in the current outbreak. Genomic analyses revealed that the upsurge of OROV cases in the Brazilian Amazon coincides with spread of a novel reassortant lineage containing the M segment of viruses detected in the eastern Amazon region (2009-2018) and the L and S segments of viruses detected in Peru, Colombia and Ecuador (2008-2021). The novel reassortant likely emerged in the Amazonas state between 2010 and 2014 and spread through long-range dispersion events during the second half of the 2010s. Phylodynamics reconstructions showed that the current OROV spread was driven mainly by short-range (< 2 km) movements consistent with the flight range of vectors. Nevertheless, a substantial proportion (22%) of long-range (>10 km) OROV migrations were also detected, consistent with viral dispersion by humans. Our data provide a view of the unprecedented spread and evolution of OROV in the Brazilian western Amazon region.

The influence of inorganic mercury (Hg(II)) exposure on photosynthetic microorganisms and their pigments remains understudied. Here, we employed resonance Raman (RR) spectroscopy to investigate the responses of two freshwater phytoplankton species, the green alga Chlamydomonas reinhardtii and the diatom Cyclotella meneghiniana to Hg(II) exposure. We selectively recorded the spectral RR signature of carotenoids in intact cells exposed to concentrations of 10 nM and 100 nM of Hg(II), representative for contaminated environment and unexposed control cells. A two-hour exposure of C. reinhardtii resulted in a slight decrease in lutein and ß-carotene levels, while total carotenoids RR band broadening, as revealed by the FWHM of the υ1(C=C) stretching mode from averaged RR spectra, suggested conformational changes in pigments. Higher Hg(II) concentration induced more pronounced conformational changes. Similarly, a two-hour exposure of C. meneghiniana resulted in slight decreased level of the fucoxanthin, while diadinoxanthin showed an opposite trend compared to control: when fucoxanthin decreased, diadinoxanthin increased under 10 nM Hg (II) exposure. At higher concentrations, the decrease in fucoxanthin was less pronounced, accompanied by a broadening of the band area, (with FHHM increased), indicating possible conformer occurrence in response to Hg-induced stress. The changes in the main carotenoid species of the two algae are species-specific, Hg(II) concentration-specific, and dependent on exposure time. The calculated spectral differences in absorbances from UV-VIS spectra of methanol extracts from each group supported the main findings obtained by RR, though with caution due to the selective extraction efficiency of the respective carotenoids. This study highlighted for a first time the capability of single-cell RR spectroscopy as a valuable tool for toxicity assessment and for comprehending early-stage alterations in carotenoid metabolism due to toxic metal exposure in vivo.

Theobroma cacao plantations are of significant economic importance worldwide, primarily for chocolate production. During the harvest and processing of cocoa beans, they are subjected to fermentation either by microorganisms present in the environment (spontaneous fermentation) or the addition of starter cultures, with different strains directly contributing distinct flavor and color characteristics to the beans. In addition to fungi and bacteria, viruses are ubiquitous and can affect the quality of the fermentation process by infecting fermenting organisms, destabilizing microbial diversity, and consequently affecting fermentation quality. Therefore, in this study, we explored publicly available metatranscriptomic libraries of cocoa bean fermentation in Limon Province, Costa Rica, looking for viruses associated with fermenting microorganisms. Libraries were derived from the same sample at different time points: 7, 20, and 68 h of fermentation, corresponding to yeast- and lactic acid bacteria-driven phases. Using a comprehensive pipeline, we identified 68 viral sequences that could be assigned to 62 new viral species and 6 known viruses distributed among at least nine families, with particular abundance of elements from the Lenarviricota phylum. Interestingly, 44 of these sequences were specifically associated with ssRNA phages (Fiersviridae) and mostly fungi-infecting viral families (Botourmiaviridae, Narnaviridae, and Mitoviridae). Of note, viruses from those families show a complex evolutionary relationship, transitioning from infecting bacteria to infecting fungi. We also identified 10 and 3 viruses classified within the Totiviridae and Nodaviridae families, respectively. The quantification of the virus-derived RNAs shows a general pattern of decline, similar to the dynamic profile of some microorganism genera during the fermentation process. Unexpectedly, we identified narnavirus-related elements that showed similarity to segmented viral species. By exploring the molecular characteristics of these viral sequences and applying Hidden Markov Models, we were capable of associating these additional segments with a specific taxon. In summary, our study elucidates the complex virome associated with the microbial consortia engaged in cocoa bean fermentation that could contribute to organism/strain selection, altering metabolite production and, consequently, affecting the sensory characteristics of cocoa beans.

BACKGROUND: Monitoring training load and competition load is crucial for evaluating and improving athlete performance. This study proposes an applied approach to characterize and classify the training task specificity in relation to competition in a top-level rink hockey team, considering external and internal load from training tasks and competition. HYPOTHESIS: Training tasks and game demands have significant dose-response differences, and exercises can be classified successfully based on their physiological and biomechanical demands. STUDY DESIGN: Cross-sectional study. LEVEL OF EVIDENCE: Level 5. METHODS: Ten elite-level male rink hockey players participated in this study. Players were monitored on 6 different task categories during 8 training sessions and 2 official games. A linear mixed model with random intercepts was used to compare training tasks and competition load, accounting for individual repeated measures. A 2-step cluster analysis was performed to classify the training tasks and games based on physiological and biomechanical load, employing log-likelihood as the distance measure and Schwartz's Bayesian criterion. RESULTS: Average heartrate , maximum heartrate, and high-speed skating (18.1-30 km/h) were the best physiological load predictors, while the most effective biomechanical load predictors were impacts [8-10] g(n), decelerations [-10 to -3]m/s²(n), and accelerations [3-10]m/s²(n). Different physiological and biomechanical responses were verified between training tasks and match demands. A 4-quadrant efforts assessment for each task category revealed that training tasks used by the team in the analysis presented lower biomechanical and physiological load demands than competition. CONCLUSION: Training tasks failed to adequately replicate the specific demands of competition, especially regarding high mechanical stress, such as the absence of high-intensity impacts and decelerations. CLINICAL RELEVANCE: This method of classification of training tasks may allow coaches to understand further the specificity and contribution of each task to competition demands, consequently improving the capacity of load management and the preparedness and readiness of players for competition.

Public Health Consortiums (PHC) in Brazil represent a strategy to enhance regionalization in the Public Health Care System (SUS in Portuguese) and State/interstate/intermunicipal cooperation. The establishment of regional polyclinics aims to improve access to services with greater technological concentration and closer to users' homes. This study specifically aims to analyze the process of creating PHC and regional polyclinics in Bahia State, based on documental analysis, identifying aspects related to entering the political agenda that facilitate and hinder, the role of state administration, specificities of the composition of these PHC and regional polyclinics. A documental analysis was carried out of the minutes of the Comissão Intergestores Bipartite (CIB-BA) [Bipartite Administrative Commission] from 2015 to 2018 and of constitutive documents of the consortiums established in this state. The Ceará State experience inspired the creation of consortiums in Bahia, especially the structuring of polyclinics. Municipal administrators demonstrated a favorable position regarding the potential of consortiums with cooperation among municipalities. The role of the state government proved to be a facilitating condition, whereas the maintenance of the municipal counterpart in finance constituted a hindering element. The consortiums contributed to the regionalization of health in the state as well as expansion of access to specialized care.
Os consórcios públicos de saúde (CPS) representam uma estratégia para potencializar a regionalização no SUS e a cooperação interfederativa. A implantação de policlínicas regionais visa melhorar o acesso a serviços de maior densidade tecnológica, mais próximos ao município de origem dos usuários. Este estudo objetiva analisar o processo de implantação dos consórcios públicos de saúde e das policlínicas regionais na Bahia, com base em análise documental, identificando aspectos facilitadores e dificultadores relativos à entrada na agenda política, ao papel da gestão estadual e às especificidades da composição dos CPS e das policlínicas regionais no estado da Bahia. Realizou-se análise documental das atas da Comissão Intergestores Bipartite dos anos de 2015 a 2018 e de documentos constitutivos dos consórcios implantados na Bahia. A experiência do Ceará inspirou a criação dos consórcios na Bahia, sobretudo na estruturação das policlínicas. Verificou-se posicionamento favorável dos gestores municipais acerca da potencialidade do consórcio na cooperação entre municípios. A atuação do governo do estado revelou-se condição facilitadora, e a manutenção da contrapartida municipal no financiamento, como elemento dificultador. Os consórcios contribuíram para a regionalização da saúde no estado e a ampliação do acesso à atenção especializada.
