RESUMO

Candida albicans is an opportunistic fungal human pathogen that has been causing an increasing number of deaths each year. Due to the widespread use of broad-spectrum antibiotics and immunosuppressants, C. albicans resistance to these therapies has increased. Thus, natural plant inhibitors are being investigated for treating C. albicans infections. Schinifoline is a 4-quinolinone alkaloid with antibacterial, insecticidal, antitumor, and other biological activities. Here, we explored the effects of schinifoline on C. albicans in C. elegans and extracted RNA from uninfected C. elegans, C. elegans infected with C. albicans, and C. elegans infected with C. albicans and treated with 100 mg/l schinifoline. Our results showed that there were significant differences among the three groups. The GO and KEGG pathway analysis suggested that the pathogenicity of C. albicans to C. elegans was caused by abnormal protein function. Schinifoline regulates lysosomal pathway related genes that accelerate the metabolism and degradation of abnormal proteins, thereby inhibiting the negative effects of C. albicans in vivo. These findings advance our understanding of the molecular mechanisms underlying schinifoline inhibition of C. albicans.

RESUMO

OBJECTIVE: To study the constituents isolated from Juncus effuses Linn.. METHODS: The compounds were isolated through chromatographic approaches, such as Silica gel. CC, Sephadex LH-20 and were identified by a combination of spectroscopic methods (UV, MS, 1H-NMR, 13C-NMR and NOE). RESULTS: 8 kinds of compounds were identified which included four flavonoids, luteolin (I), eriodictyol (II), 2', 5', 5, 7-tetrahydroxyflavone (III), glucoluteolin (IV); two aromatic compounds, methyl p-hydroxybenzoate (V), vanillic acid (VI); a fatty acid, hexadecanoic acid (I) and an alkane, n-Tetradecane (VIII). CONCLUSION: Compound II, III, V, VI, VII, VIII were firstly isolated from this plant.

OBJECTIVE: To compare the quality of cultivated Chinese yam (Dioscorea polystachya Turcz.) from Yunnan (Kunming yam) and Henan (Tiegum yam) by determining the content of dry substance in fresh sample and the contents of water extracts, ethanol extracts, starch, protein, crude-fat and polysaccharide in dry sample. METHOD: The above-mentioned indexes were determined by methods of air desiccation, cold-immersing, hot-immersing, iodine-blue coloration, half Micro-Kjeldahl, Soxhlet extraction and spectrophotometric method respectively. RESULT: In fresh sample, the content of dry substance of Kunming yam is a little lower than Tiegun yam; in dry sample, all indexes of Kunming yam are higher than or approach to those of Tiegun yam except the content of crude-fat. Especially the content of polysaccharide in Kunming yam is 64% higher than that of Tiegun yam. CONCLUSION: Kunming yam, with high quality, high yield and good taste, deserves paying close attention as a good variety.
