RESUMO

Mercury (Hg), especially methylmercury (MeHg), which is highly neurotoxic, is a global pollutant that can affect human health because of its accumulation in aquatic products. Poyang Lake, an inland lake in China, has been significantly affected by human activity, yet there is limited understanding of local mercury contamination and potential exposure pathways to humans. In this study, we explored the risks of mercury exposure by sampling sediments, plants, and aquatic organisms in the lake and surrounding areas and analyzing total Hg (THg) and MeHg levels. Sediment sampling was conducted at the main lake, rivers, rice paddies, and fishponds. Two dominant species of plants and 15 species of aquatic organisms were sampled and analyzed. We assessed the characteristics of mercury in sediments using the geo-accumulation index (Igeo), mercury exposure using the biomagnification factor (BMF) and biota sediment accumulation factor (BSAF), and risks using thresholds for adverse effects. The highest THg concentrations (137.04 ± 44.3 ng g-1 dw) were detected in the main lake sediments, whereas the highest MeHg concentrations (0.47 ± 0.6 ng g-1 dw) were detected in fishpond sediments. Mercury accumulation in the main lake sediments could be assessed as contaminated (Igeo > 0: 81.6%). Yellow catfish had the highest mercury concentration (THg 770.69 ± 199.7 ng g-1 dw; MeHg 741.93 ± 168.8 ng g-1 dw). Piscivores were adversely affected by carnivorous fish (50.8%), but all fish concentrations did not exceed the food safety standards recommend by China and the WHO. The mercury exposure results revealed significant Hg biomagnification and enrichment (BMF >1: 94.55%; BSAFmax = 1218). Long-term monitoring of aquatic organisms is warranted.

8-hydroxy-2'-deoxyguanosine (8-OHdG) is a sensitive and stable biomarker for evaluating DNA oxidative damage. A rapid and sensitive colloidal gold immunochromatographic strip was developed for 8-OHdG detection by a competitive method. The sample pad (glass cellulose film), bonding pad (glass cellulose film), nitrocellulose film and absorbent pad were pasted on the polyvinyl chloride (PVC) base plate to construct the test strip. Colloidal gold (AuNPs) was prepared by the reduction of chloroauric acid with sodium citrate. 8-OHdG antibody (Ab) was coated on the outer layer of AuNPs to form Ab@AuNPs as a probe. Bovine serum albumin (BSA) and 8-OHdG were conjugated with carbodiimide hydrochloride to prepare an artificial antigen, which was used as the coating antigen of detection line. Goat anti mouse polyclonal antibody IgG was used as the coating antibody of control line. The experimental parameters were optimized including the type of nitrocellulose membrane, the formula of loading solution, and the spraying amount of gold labeled antibody. The results showed that the appropriate nitrocellulose membrane was CN 95. The optimal loading solution included BSA (1%), Tween-20 (3%), sucrose (3%) and NaCl (0.9%). The optimal spraying amount of gold labeled antibody was 4 µL. 8-OHdG can be detected by the strip under visible light, and the level of 8-OHdG in urine can be preliminarily determined by comparing the color intensity of T line and C line. The 8-OHdG concentration in urine was further calculated by the gray value of T line and the threshold of detection was 2.55 µg/L. This colloidal gold immunochromatographic strip is simple, rapid and specific for detecting 8-OHdG in human urine to preliminarily evaluate the human status.

Using total counts in simultaneous annual surveys, we monitored the population size and spatial distribution of oriental white storks (Ciconia boyciana) wintering in Poyang Lake between 1998 and 2011. Results showed that Poyang Lake wetland is an important wintering ground for oriental white storks, with an annual average population number of 2 305±326. The population sizes in 2004, 2005, 2010, and 2011 were higher than the highest-ever estimate of its global population. In 2005, we recorded 3 789 individuals, which was the maximum population number within the period of 1998-2011. The storks inhabited 52 lakes, with the greatest distance between these lakes being 180.3 km. The storks presented a clustered distribution pattern in the Poyang Lake wetland, irrespective of the number of individuals or occurrence frequencies. Shahu, Dahuchi, Banghu, and Hanchihu were most frequently used lakes and had the largest annual average numbers of storks. There was a significant positive correlation between occurrence frequency and annual average number of storks in the lakes. Most of the lakes important for storks were covered by existing nature reserves, though some lakes outside the reserves were also frequently used. About 64.9%±5.5% of the storks were found in nature reserves. In addition, the storks more frequently used and clumped in significantly larger flocks in lakes within nature reserves than lakes outside.

Using simultaneous land surveys, we monitored the population size and spatial distribution of wintering Siberian cranes at 64 lakes around Poyang Lake between 1998 and 2010. The results showed that 46 lakes were inhabited by wintering cranes, and in 25 of those, the number of wintering cranes accounted for more than 1% of the Siberian cranes' global population. The lakes where over 40.0% of the global population, e.g. 1 280 individuals, included Dachahu Lake in Jiujiang region, and Banghu Lake and Candouhu Lake in the Poyang Lake Nature Reserve. The average yearly population of the wintering Siberian crane in the Poyang lakes was 3 108±849, with the maximum of 4 004 individuals in winter 2002. On the whole, there was no drastic fluctuation, but population numbers have shown considerable fluctuation since 2003. We also found the Poyang Lake Nature Reserve was the major wintering area of the Siberian crane, with over 60% of Siberian cranes wintering in the reserve since 2002 (except in 2006). Most of the inhabited lakes are covered in existing nature reserves, though some lakes outside the reserve were also considerably used by Siberian cranes.
