RESUMO

In China, the re-emerging pseudorabies virus (PRV) variant has caused large-scale outbreaks of pseudorabies in swine herds with classical PRV vaccine immunization since late 2011. Here, a recombinant PRV with TK/gI/gE/11k/28k deletion was constructed based on variant HN1201 strain isolated in 2012, by the bacterial artificial chromosome infectious clones. Compared with the parental virus, the recombinant PRV rHN1201TK-/gE-/gI-/11k-/28k- showed a similar virus grown curve and exhibited smaller plaques. The vaccination of rHN1201TK-/gE-/gI-/11k-/28k- could elicit an earlier and higher level of gB antibody, and the neutralizing antibodies elicited by rHN1201TK-/gE-/gI-/11k-/28k- were effective against both PRV classical and variant strains. Clinically, the body temperature of the pigs immunized with rHN1201TK-/gE-/gI-/11k-/28k- was significantly lower than that of the classical PRV vaccine immunized pigs, and the recombinant PRV could provide effective protection against the challenge with the PRV variant. These results imply that the rHN1201TK-/gE-/gI-/11k-/28k- could be a promising vaccine candidate for the prevention of the current epidemic of pseudorabies in China.

Inflammation reaction plays an important role in the pathogenesis of ankle fracture. The aim of the present study was to investigate the effect of RvD1 on the inflammatory response and underlying molecular mechanisms in MG-63 cells. Reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and ELISA were used to determine the mRNA and protein expression of cytokines extracted from peripheral blood in children with or without ankle fracture, respectively. MG-63 cells were pre-treated with/without RvD1 and stimulated with 1 µg/ml LPS. The cell viability was detected by MTT assay. The production of cytokines from MG-63 cells was assessed by RT-qPCR and western blot, respectively. The expression of p-p38, NF-κB (p50) and cyclooxygenase-2 (COX-2) mRNA and protein were detected by western blot and/or RT-qPCR. The levels of NLRP3, associated recruitment domain (ASC), cleaved caspase1, caspase-1 were measured by RT-qPCR and/or western blot. The levels of interleukin (IL)-1ß, IL-6 and tumor necrosis factor (TNF-α) mRNA and protein were up-regulated in children with ankle fracture compared with healthy children. RvD1 treatment did not induce cytotoxicity in MG-63 cells, but it can inhibit LPS induced MG-63 cell proliferation inhibition. RvD1 was able to dose-dependently reverse LPS induced up-regulation of TNF-α, IL-1ß, IL-6 mRNA and protein expression. Furthermore, the LPS induced up-regulation of p-p38, NF-κB (p50), and NLRP3, ASC, cleaved caspase-1/caspase-1, and COX-2 was dose-dependently reversed by RvD1. In conclusion, The present study demonstrated that RvD1 inhibited inflammation though inhibiting MAPKp38/NF-κB pathway and NLRP3 inflammasome expression in MG-63 cells, indicating that it may be an effective drug for the treatment of ankle fracture.

Inclusion body hepatitis and hepatitis-hydropericardium syndrome caused by high-pathogenic fowl adenovirus serotype 4 has recently plagued Chinese poultry industry and caused huge economic losses since 2013. So far, there is no commercial vaccine available to control this disease. In this study, we reported the development of both embryo-adapted and cell-culture derived inactivated FAdV-4 vaccines and evaluated their efficacies in chicken. Compared to embryo-adapted vaccine, cell-culture derived vaccine induced significantly earlier and higher serological response measured by AGP and ELISA. After virus challenge, chicken immunized with cell-culture derived vaccine did not showed any gross and histopathological lesions, whereas inclusion body hepatitis was observed in the liver of chicken vaccinated with embryo-adapted vaccine. No mortality was observed in both the vaccinated groups. The above results suggested that cell-culture derived FAdV-4 inactivated vaccine could be a better vaccine candidate than embryo-adapted vaccine to control FADV-4 infections in China.
