RESUMO

The volume of urine of high power field of analyzer Iris IQ 200 TM was identified according standard of stabilizing erythrocytes and amounted up to 0.18 mkl. The amount of urine corpuscles detected by analyzer in this volume corresponds to amount of urine corpuscles under microscopy of sediment often times concentrated urine at thickness of preparation in 0.1 mm, under microscope with objective x40 and ocular 10/18. The count of corpuscles by analyzer in 1 mkl of urine is the most objective and convenient technique for quantitative evaluation of urine corpuscles.

RESUMO

The authors have studied the species-specific composition of major periodontopathogenic bacteria (P. gingivalis, P. intermedia, B. forsythus, A. actinomycetemcomitans, and T. denticola), by applying the Russian reagent kit developed and made by HPF "GENETECH", and some fungi, microorganisms, and viruses among the Moscow Region inhabitants with chronic generalized periodontitis in 2006-2008. The studies have shown it possible to use molecular genetic methods for diagnosis, drug therapy monitoring, and epidemiological studies in periodontology and implantology.

Since the express-diagnostics of mycoses in immune-deficit patients still remains an acute problem, we developed an effective test system (Kan-Am) to detect DNA Candida albicans, which is a leader in the list of causative agents of candidosis. A comparison study of three PCR-systems used to detect a broad spectrum of fungoid pathogens was carried out, and a universal system (FungAm), which ensures the detection of DNAs of above 78 strains of 25 types of pathogenic fungi, was selected. The results of clinical testing of the species-specific and universal PCR-systems are well confirmed by the culture method, and they are indicative of the efficacy of applying them for the diagnostics of mycoses in neonatology. The use of the mentioned systems is a promising factor for the express-diagnostics of mycoses in immunodeficiency patients. The high sensitivity of the method makes it possible to detect 10 to 100 cells of a causative agent in 100 mcl of the examined biological material, which is compatible with the culture method. A kit of dry reagents (IonoMix) designed for an accelerated sample preparation and isolation, from them, of DNAs on the basis of Chelex-100 and of proteinase K was worked out; the kit is portable and meant for a long-term storing.

RESUMO

A simple method for detecting pathogenic microorganisms in clinical samples has been developed. This method is based on identification of specific nucleotide sequences by Pt[(dien)Cl] Cl-labelled DNA probes and simultaneous immunochemical control of total DNA content in each clinical sample. Such control is provided by a simple semiquantitative enzyme-linked immunoassay using antibodies to denatured DNA; its implementation requires the same procedures, materials and solutions used in the hybridization analysis. The information about the DNA content in the sample is necessary for adequate interpretation of results of hybridization analysis. Pt[(dien)Cl]Cl for the probe and affinity antibodies to DNA-Pt[(dien)Cl] Cl and rabbit immunoglobulin antibodies conjugated to phosphatase for DNA hybrid detection were used. The results of hybridization analysis show a good correlation with those of the PCR-test. The method is simple, relatively inexpensive and fit for population monitoring.

The paper describes a sensitive latex hybridization assay (LHA) method applied for indirect detection of biotinylated nucleic acid hybrids immobilized on a synthetic membrane. The biotinylated hybrids were visualized by means of latex particles containing the fluorescent dye pyronine G and coated with streptavidin; 1.6 and 0.3 pg of lambda-phage DNA was detected by dot blot hybridizations on nylon membrane and polyethyleneimine-cellophane, respectively. The assay sensitivity was increased by three orders of magnitude over that with fluorescently labeled probes due to encapsulation of the fluorescent dye in polymer particles. LHA is simple (single-stage detection procedure), fast, and more sensitive than any of the other nonradioactive hybridization methods.

RESUMO

A new reagent, polyphotobiotin (PPhB), for labelling DNA probes has been obtained using low molecule oligoethylenimine (M 600 Da). Photoreactive group of PPhB is 4-azido-salicylic acid. The procedure is simple and quick, the reaction time being about 20 min. PPhB-labelled DNA has the same efficiency in the hybridisation analysis as DNA labelled with Bio-4-dUTP via PCR.

RESUMO

Electron structure of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) and some of its analogs--the substrates of monoaminooxidase--substituted by phenyl cycles was studied by semiempiric quantum-chemical CNDOR, MINDOB methods. The relationship between the obtained electron and conformation parameters (orientation of the phenyl ring in particular) and biological activity of the compounds under consideration is discussed. A comparative analysis of the distribution pattern of the electron density for the MPTP molecule calculated by the above methods showed a good agreement between the results obtained.

Method of the latex hybridization analysis has been developed: after hybridization of NA-target with biotinylated probe visualization of hybrids was carried out using latex particles, containing fluorescent dye pyronin G and coated with streptavidin. Due to encapsulation of the fluorescent dye in polymer particles sensitivity of the analysis was increased by several orders of magnitude in comparison with methods, using fluorescently labelled probes. Possessing a number of advantages, the method yields to none of any other methods of NA hybridization analysis in sensitivity.

RESUMO

The electronic and conformational properties of some acyclonucleosides of guanine and hypoxanthine were studied by the modified intermediate neglect of differential overlap (MINDO/3) molecular orbital method. There is a significant linear relationship between the ionization potentials of these compounds and their inhibitory effect on purine nucleoside phosphorylase, a key enzyme in purine metabolism.

RESUMO

A two-step chemical method of introduction of nonradioactive labels in DNA was proposed. At first step DNA is modified by succinic dihydrazide at pH 5.0 and 95 degrees C, or at pH 4.5 and 37 degrees C in presence of sodium bisulfite. Then FITC or biotin are joined to the hydrazide groups. DNA modified in this way were shown to be effective hybridisation probes.
