RESUMO

The common features of the pathogenesis of chronic lymphocytic leukemia (CLL) and chronic glomerulonephritis (CGN) have been shown as a result of a comprehensive study of the functional properties of B cells. Both CLL and CGN are accompanied by the appearance and accumulation of different types of activated B cells characterized by high spontaneous proliferation and/or Ig secretion; or sensitivity to T cell growth and differentiation factors; or capacity for Ig secretion; in mitomycin C treated B cell cultures (i.e. without previous proliferation). The present data may be useful in the integral evaluation of B cell function in health and in immunopathologies (lymphoproliferative and autoimmune/immune complex diseases).

RESUMO

The data presented indicate that B cells from patients with chronic lymphocytic leukemia (CLL) may produce suppressive factors capable of inhibiting T and B cell proliferation and Ig secretion. Two factors with the molecular weight 65-75 KDa (SBF1) and 15-25 KDa (SBF2) have been characterized. SBFs are primarily produced by B cells with a low flow density. There is a reverse dependence between the capacity of leukemic B cells for spontaneous Ig secretion and their ability for the production of suppressive factors. The autocrine and paracrine pathways of B cell regulation in the pathogenesis of chronic lymphocytic leukemia are discussed.

RESUMO

The influence of the previous cultivation of human peripheral blood mononuclear cells in suspension with mitogen on the consequent PHA-stimulated response in suspension and agar cultures was studied. It has been established that proliferative response of mononuclear cells in suspension with mitogen is not changed, but the intensity of T-cell colony formation in two-layer agar systems in increased after preincubation mainly at the expense of increasing the number of type II colonies and clusters. It is concluded that the free cell-cell contacts are needed to activate T-lymphocyte colony precursors.

Altogether 67 patients with chronic glomerulonephritis (CGN) were examined for the content of thymus-dependent precursor lymphocytes (auto-RFC, pre-T-cells). The patients suffering from CGN were characterized by the high content of the young post-thymic T-cells (auto-RFC) and high affinity T lymphocytes carrying a receptor for the Fc fragment of IgM as compared to normal donors. In patients with different clinical patterns of CGN, an analysis was made of the content of precursor thymus-dependent lymphocytes. A relationship was discovered between the content of pre-T-cells and the activity of the nephritic process. The activity of the pathological process was associated with the high content of precursor lymphocytes. The inactive course of nephritis was characterized by a well-defined reduction of the number of precursor cells. This might be regarded as depletion of the amount of precursor lymphocytes.

RESUMO

The results of the study of natural cytotoxic activity of human peripheral blood lymphocytes are presented. Two types of natural killer cells isolated in discontinuous percoll density gradient have been shown to exist. Lymphocytes with low floating density lyse NK-sensitive target cells and leukemic B-lymphocytes, increase the lytic activity with respect to blasts of K-562 line under the effect of alpha-interferon. Lymphocytes isolated in percoll heavy fractions chiefly lyse leukemic B cells of patients by B-variant CLL and are not sensitive to the alpha-interferon effect.

RESUMO

The role of T- and B-cell cooperative interaction in the regulation of natural killer (NK) activity in human peripheral blood lymphocytes was studied. It has been shown that preincubation of normal donor mononuclear cells (MNC) for 48 h is followed by the loss of NK activity, while the incubation of the isolated T- and non-T-cell subsets does not lead to an analogous fall in the killer lymphocyte function. NK activity of MNC and isolated lymphocyte subsets in normal donors is shown to exceed that of CLL patients. The absence of preincubation effect on NK activity level of MNC, T- and non-T-cells in CLL patients has been also found. The findings obtained suggest that as a result of T- and B-cell interaction during preincubation differentiation of young T lymphocytes with NK cellular properties takes place. It is followed by the loss in NK activity. B-cell defect in CLL patients might cause the absence of preincubation effect on NK activity of T cells.

RESUMO

The proliferative response of human lymphocytes was studied in one-way and two-way mixed lymphocyte cultures (MLC). The maximal proliferation was shown to be attained in a two-way MLC containing unequal quantities of the cells from two paired donors. It was also demonstrated in a one-way MLC that the cells of one of the two paired donors are more effective responders. The most powerful proliferation in the MLC of these donors was observed in the cultures containing the excess of more effective cells. Thymosine increased the response of human lymphocytes in the two-way MLC. In vitro cell preincubation reduced the response in the cultures with a high proliferative response in the control. It has been thus demonstrated that lymphocytes from paired donors possess different functional activity in the MLC.
