RESUMO

Cytokinin oxidase/dehydrogenase (CKX) is the key enzyme that has been observed to catalyze irreversible inactivation of cytokinins and thus modulate cytokinin levels in plants. CKX gene family is known to have few members which are, expanded in the genome mainly due to duplication events. A total of nine MiCKXs were identified in Morus indica cv K2 with almost similar gene structures and conserved motifs and domains. The cis-elements along with expression analysis of these MiCKXs revealed their contrasting and specific role in plant development across different developmental stages. The localization of these enzymes in ER and Golgi bodies signifies their functional specification and property of getting modified post-translationally to carry out their activities. The overexpression of MiCKX4, an ortholog of AtCKX4, displayed longer primary root and higher number of lateral roots. Under ABA stress also the transgenic lines showed higher number of lateral roots and tolerance against drought stress as compared to wild-type plants. In this study, the CKX gene family members were analyzed bioinformatically for their roles under abiotic stresses.

Unravelling the perplexing nature of negative differential resistance (NDR) in 2D transition metal dichalcogenide (2D TMD) devices, especially regarding intrinsic properties, is hindered by experiments conducted in ambient environments. A thorough investigation is essential for unveiling the actual mechanism. In this study, we provide compelling evidence of the NDR effect with a remarkably high peak-to-valley current ratio and proton-diffused superionic conductivity in quantum-confined water molecules anchored to a thin film of 2D TMDs. Our investigation underscores the crucial role of ambient moisture for this robust NDR effect independent of underlying materials used. The bonding of water molecules to the existing sulfur defect sites on 2D TMD nanoflakes facilitates the formation of bridges between two planar metal electrodes, thus enabling superionic in-plane protonic conduction. During electrolysis of chemisorbed water, protons are liberated at the anode and migrate toward the cathode during bias voltage sweeping. Nevertheless, proton diffusion encounters increasing impedance beyond a certain applied bias, thereby restricting current flow even with higher biasing voltages, which is attributed to the interfacial Schottky energy barrier influenced by the Fermi level pinning effect. Our DFT simulations corroborate this mechanism, revealing minimal intermolecular interaction of H+ ions compared to OH- ions at distinct atomic sites on 2D TMD nanoflakes.

RESUMO

A previously identified wheat drought stress responsive Universal stress protein, TaUSP_3B-1 has been found to work in an auxin dependent manner in the plant root tissues in the differentiation zone. We also found a novel interacting partner, TaGolS, which physically interacts with TaUSP_3B-1 and colocalizes in the endoplasmic reticulum. TaGolS is a key enzyme in the RFO (Raffinose oligosaccharides) biosynthesis which is well reported to provide tolerance under water deficit conditions. TaUSP_3B-1 overexpression lines showed an early flowering phenotype under drought stress which might be attributed to the increased levels of AtTPPB and AtTPS transcripts under drought stress. Moreover, at the cellular levels ER stress induced TaUSP_3B-1 transcription and provides tolerance in both adaptive and acute ER stress via less ROS accumulation in the overexpression lines. TaUSP_3B-1 overexpression plants had increased silique numbers and a denser root architecture as compared to the WT plants under drought stress.

RESUMO

Molecular hydrogen (H2) production by the electrochemical hydrogen evolution reaction (HER) is being actively explored for non-precious metal-based electrocatalysts that are earth-abundant and low cost like MoS2. Although it is acid-stable, its applicability is limited by catalytically inactive basal planes, poor electrical transport and inefficient charge transfer at the interface. Therefore, the present work examines its bilayer van der Waals heterostructure (vdW HTS). The second constituent monolayer boron phosphide (BP) is advantageous as an electrode material owing to its chemical stability in both oxygen and water environments. Here, we have performed first-principles based calculations under the framework of density functional theory (DFT) for the HER in an electrochemical double layer model with the BP monolayer, MoS2/BP and MoSSe/BP vdW HTSs. The climbing image nudged elastic band method (CI-NEB) has been employed to determine the minimum energy pathways for Tafel and Heyrovsky reactions. The calculations reveal that the Tafel reaction shows no reaction barrier. Thereafter, for the Heyrovsky reaction, we obtained a low reaction barrier in the vdW HTSs as compared to that in the BP monolayer. Subsequently, we have observed no significant difference in the reaction profile of MoS2/BP and MoSSe/BP vdW HTSs in the case of 2 × 2 supercell configuration. However, in the case of 3 × 3 and 4 × 4 configurations, MoSSe/BP shows a feasible Heyrovsky reaction with no reaction barrier. The coverages with 1/4H+ concentration (conc.) deduced high coverage with low conc. and low coverage with high conc. to be apt for the HER via the Heyrovsky reaction path. Finally, on observing the activation barrier of the Heyrovsky pathway along with that of second H adsorption at the surface, the Heyrovsky path is expected to be favoured.

RESUMO

Phonon scattering at grain boundaries (GBs) is significant in controlling the nanoscale device thermal conductivity. However, GBs could also act as waveguides for selected modes. To measure localized GB phonon modes, milli-electron volt (meV) energy resolution is needed with subnanometer spatial resolution. Using monochromated electron energy loss spectroscopy (EELS) in the scanning transmission electron microscope (STEM) we have mapped the 60 meV optic mode across GBs in silicon at atomic resolution and compared it to calculated phonon densities of states (DOS). The intensity is strongly reduced at GBs characterized by the presence of 5- and 7-fold rings where bond angles differ from the bulk. The excellent agreement between theory and experiment strongly supports the existence of localized phonon modes and thus of GBs acting as waveguides.

KEY MESSAGE: TaUSPs are localized in Endoplasmic reticulum and form homo and hetero dimers within themselves. They play significant role in multiple abiotic stress responses in yeast heterologous system and in plants. Universal Stress Proteins are stress responsive proteins present in a variety of life forms ranging from bacteria to multicellular plants and animals. In this study we have identified 85 TaUSP genes in the wheat genome and have characterised their abiotic stress responsive members in yeast under different stress conditions. Localization and Y2H studies suggest that wheat, USP proteins are localized in the ER complex, and extensively crosstalk amongst themselves through forming hetero and homodimers. Expression analysis of these TaUSP genes suggests their role in adaptation to multiple abiotic stresses. TaUSP_5D-1 was found to have some DNA binding activity in yeast. Certain abiotic stress responsive TaUSP genes are found to impart tolerance to temperature stress, oxidative stress, ER stress (DTT treatment) and LiCl2 stress in the yeast heterologous system. TaUSP_5D-1 overexpression in A. thaliana imparts drought tolerance via better lateral root network in transgenic lines. The TaUSP represents an important repertoire of genes for engineering abiotic stress responsiveness in crop plants.
