AbstractHibernators save energy during winter by expressing torpor bouts characterized by strongly reduced body temperature and metabolic rate. Polyunsaturated fatty acids (PUFAs), specifically n-6 PUFAs, are known to positively affect hibernation performance and thereby energy savings predominantly in fat-storing hibernators. Accordingly, hibernators usually retain PUFAs and mobilize monounsaturated fatty acids (MUFAs) or saturated fatty acids (SFAs) during hibernation. In food-storing common hamsters (Cricetus cricetus), however, we previously found that PUFA proportions in white adipose tissue (WAT) decreased during winter, indicating that individuals did mobilize PUFAs. To further investigate these patterns, we analyzed PUFA changes in WAT during hibernation as well as hibernation performance in free-ranging and captive common hamsters with lower prehibernation PUFA proportions compared to those in the previous study. Under controlled conditions, total PUFAs, n-6 PUFAs, and SFAs increased while n-3 PUFAs and MUFAs decreased during hibernation. Higher prehibernation n-6 PUFA proportions resulted in fewer torpor bouts and less time spent in torpor. In free-ranging hamsters, n-6 PUFAs increased while n-3 PUFAs and SFAs decreased during winter. Prehibernation n-6 PUFA proportions, however, did not affect hibernation performance. In summary, these results indicate that the mobilization or retention of n-6 PUFAs during hibernation could depend on their availability in WAT or in the diet before the onset of the hibernation period.

Common hamsters (Cricetus cricetus) are hibernators that rely both on body fat reserves and food stores for the winter period. They face an ongoing population decline in most parts of their distribution and recently were classified as critically endangered. Knowledge on individual body fat proportions in this species is of particular interest for conservation, because it could contribute to better understand the high plasticity in overwintering strategies, overwinter mortality rates, individual variations in reproductive output, and give information on the animals' health state. To calculate body fat proportions, we validated a method that can be applied in the field without the use of anesthesia. To develop this method, we first analyzed the body fat in carcasses of common hamsters using Soxhlet extractions and measured four morphometric parameters (body mass, head length, tibia length, foot length). The morphometric measurements were then integrated in a linear regression model to predict body fat proportions based on the measured values. The morphometric variables yielded an explained variance (adjusted R 2) of 96.42% and body fat proportions were predicted with a mean absolute error of 1.27 ± 0.11% from measured values. We applied the model to predict body fat for available field data, which consistently produced reliable values. By measuring the four morphometric parameters and following the provided instructions, body fat proportions can be reliably and noninvasively estimated in captive or free-ranging common hamsters. Furthermore, the method could be applicable to other rodents after species-specific validation.

Dietary intake of polyunsaturated fatty acids (PUFAs) is crucial for neuronal functions, can positively affect cognition, and reduce glucocorticoid (e.g. corticosterone) concentrations in response to stress. We investigated the effects of walnut oil high in PUFAs on spatial cognition and fecal corticosterone metabolite (FCM) concentrations under non-stressed conditions in rats. Unexpectedly, PUFA-supplemented rats had higher FCM concentrations and elevated concentrations generally impaired learning in the subsequent T-maze task. Statistically adjusting for individual FCM concentrations, however, revealed that learning performance was improved in PUFA-supplemented rats. The results suggest that glucocorticoids can modulate the effects of PUFAs on spatial learning under normal (non-stressed) conditions and call for consideration of basal physiological conditions in spatial learning tasks.

Dietary intake of polyunsaturated fatty acids (PUFAs) or saturated fatty acids (SFAs) differently modulates neurophysiological and behavioral functions in response to altered hypothalamic-pituitary-adrenal (HPA)-axis activity and an individual's development. In this context, an individual's social environment, including social interactions and social hierarchies, is closely related to hormone concentrations and possibly interacts with dietary fatty acid effects. We investigated if dietary supplementation with walnut oil (high in PUFAs) and coconut fat (high in SFAs), compared to a control group, affects body mass gain, cortisol and testosterone concentrations, plasma fatty acids, and social behavior in male domestic guinea pigs from adolescence to adulthood. For analyses of cortisol and testosterone concentrations, social interactions were included as covariates in order to consider effects of social behavior on hormone concentrations. Our results revealed that SFAs increased escalated conflicts like fights and stimulated cortisol and testosterone concentrations, which limited body mass gain and first-year survival. PUFAs did not remarkably affect social behavior and hormone concentrations, but enabled the strongest body mass gain, which probably resulted from an energetic advantage. Neither sociopositive nor agonistic behaviors explained age-specific differences in hormone concentrations between groups. However, a high number of subdominant individuals and lower testosterone concentrations were related to increased cortisol concentrations in adult PUFA males. Our findings demonstrate the importance of dietary fatty acids regarding behavioral and endocrine developmental processes and adaptations to the social environment by modulating HPA-axis function and body homeostasis.

Animals in urban environments face challenging situations and have to cope with human activities. This study investigated the ecology and behaviour of a population of European hamsters (Cricetus cricetus) living in the city centre of Vienna (Austria). We recorded the surface activities of 35 hamsters in May 2015. Each focal animal was observed for 15 minutes, and a total of 66 focal samples were analysable. As a prey species in an environment teeming with human activities, we predicted a high level of vigilance by the hamsters. The results show that while animals dedicated a lot of time to vigilance, most of their time was spent foraging. The study also explores whether the frequency of vigilance behaviours differ between males and females. We found that vigilance behaviours were expressed in a different manner by males and females. Finally, we investigated the distribution of the burrows on green spaces depending on proximity to trees and on noise levels. We found a biased distribution of burrows, with a spatial preference for location protected by the vegetation and distant to noise sources. Although burrows were located preferentially under vegetation cover, levels of noise did not determine their positions. Moreover, this species does not respond to disturbances like daily urban noises, probably due to habituation. The common hamster is an endangered species; our results lead to a greater knowledge of its behaviour in a persistent urban population.

Hibernation is characterized by reduced metabolism and body temperature during torpor bouts. Energy reserves available during winter play an important role for hibernation and some species respond to high energy reserves with reduced torpor expression. Common hamsters are food-storing hibernators and females hibernate for shorter periods than males, probably related to larger food stores. In this study, we provided free-ranging common hamsters with sunflower seeds shortly before winter and recorded body temperature using subcutaneously implanted data loggers. We compared hibernation patterns and body mass changes between individuals with and without food supplements and analysed reproductive onset in females. Supplemented males delayed hibernation onset, hibernated for much shorter periods, and emerged in spring with higher body mass than unsupplemented ones. Additional food did not affect hibernation performance in females, but supplemented females emerged earlier and preceded those without food supplements in reproductive onset. Thus, males and females differently responded to food supplementation: access to energy-rich food stores enabled males to shorten the hibernation period and emerge in better body condition, probably enhancing mating opportunities and reproductive success. Females did not alter hibernation patterns, but started to reproduce earlier than unsupplemented individuals, enabling reproductive benefits by an extended breeding period.

Reproductive functions in female mammals can be significantly affected by the actions of dietary polyunsaturated fatty acids (PUFAs) on steroid hormone secretion rates. Nevertheless, the effects of plasma free PUFAs on the oestrous cycle have seldom been considered. Therefore, in the present study, the diet of domestic guinea pigs was supplemented with high concentrations of different PUFAs and the effects of altered plasma PUFA patterns on steroid hormone concentrations, measured non-invasively, and body mass during oestrus and dioestrus were analysed. The oestrous cycle was characterised by increased oestrogen and cortisol concentrations in oestrus, corroborated by lowest bodyweight, whereas progesterone concentrations were highest in dioestrus. Plasma concentrations of the long-chain PUFAs docosahexaenoic acid (DHA; 22:6 ω3) and arachidonic acid (AA; 20:5 ω6) affected steroid hormone concentrations differently in oestrus and dioestrus. DHA positively affected oestrogen and progesterone concentrations and diminished cortisol concentrations only in oestrus. In contrast, AA negatively affected oestrogen and stimulated cortisol concentrations in oestrus and reduced progesterone concentrations in general. These findings imply selective and opposite contributions of DHA and AA to ovarian functions during different stages of the oestrous cycle, indicating a high biological relevance of plasma free PUFAs in female reproductive function.

Hibernating animals can adjust torpor expression according to available energy reserves. Besides the quantity, the quality of energy reserves could play an important role for overwintering strategies. Common hamsters are food-storing hibernators and show high individual variation in hibernation performance, which might be related to the quality of food hoards in the hibernacula. In this study, we tested the effects of food stores high in fat content, particularly polyunsaturated fatty acids (PUFAs), on hibernation patterns under laboratory conditions. Control animals received standard rodent pellets only, while in the other group pellets were supplemented with sunflower seeds. We recorded body temperature during winter using subcutaneously implanted data loggers, documented total food consumption during winter, and analysed PUFA proportions in white adipose tissue (WAT) before and after the winter period. About half of the individuals in both groups hibernated and torpor expression did not differ between these animals. Among the high-fat group, however, individuals with high sunflower seeds intake strongly reduced the time spent in deep torpor. PUFA proportions in WAT decreased during winter in both groups and this decline was positively related to the time an individual spent in deep torpor. Sunflower seeds intake dampened the PUFA decline resulting in higher PUFA levels in animals of the high-fat group after winter. In conclusion, our results showed that common hamsters adjusted torpor expression and food intake in relation to the total energy of food reserves, underlining the importance of food hoard quality on hibernation performance.

Hibernating species significantly reduce energy expenditure during winter by entering torpor. Nevertheless, the various benefits of hibernation might be counteracted by negative effects of torpor such as immune depression, oxidative stress, or neuronal impairment. Considering these trade-offs, adequate energy reserves could allow animals to reduce the time spent in torpor or the extent of metabolic depression. Common hamsters use food stores during hibernation and previously documented high individual variations in body temperature patterns during winter could, therefore, be related to differences in external energy reserves. In this study, we manipulated the availability of food stores under laboratory conditions to investigate potential effects on hibernation patterns. Female hamsters were kept in artificial burrows in climate chambers and subcutaneous temperature was recorded using implanted data loggers. One group had access to large food stores, whereas another group received daily food portions which were removed on the next day if not consumed. Almost all hamsters without access to food stores hibernated, while less than half of the individuals with food stores entered deep torpor. Individuals without food hoards additionally expressed more short torpor bouts and exhibited lower minimum subcutaneous temperatures during torpor than those with food stores. Thus, individuals confronted with lacking food reserves were more likely to hibernate and additionally saved energy by entering short torpor bouts more frequently and remaining at lower subcutaneous temperature both during torpor and euthermic periods. In conclusion, our results demonstrate that food store availability affects torpor expression and also highlight variation in torpor patterns and energy-saving strategies in common hamsters.

BACKGROUND: Dietary saturated (SFAs) and polyunsaturated (PUFAs) fatty acids can highly affect reproductive functions by providing additional energy, modulating the biochemical properties of tissues, and hormone secretions. In precocial mammals such as domestic guinea pigs the offspring is born highly developed. Gestation might be the most critical reproductive period in this species and dietary fatty acids may profoundly influence the gestational effort. We therefore determined the hormonal status at conception, the reproductive success, and body mass changes during gestation in guinea pigs maintained on diets high in PUFAs or SFAs, or a control diet. RESULTS: The diets significantly affected the females' plasma fatty acid status at conception, while cortisol and estrogen levels did not differ among groups. SFA females exhibited a significantly lower body mass and litter size, while the individual birth mass of pups did not differ among groups and a general higher pup mortality rate in larger litters was diminished by PUFAs and SFAs. The gestational effort, determined by a mother's body mass gain during gestation, increased with total litter mass, whereas this increase was lowest in SFA and highest in PUFA individuals. The mother's body mass after parturition did not differ among groups and was positively affected by the total litter mass in PUFA females. CONCLUSIONS: While SFAs reduce the litter size, but also the gestational effort as a consequence, PUFA supplementation may contribute to an adjustment of energy accumulations to the total litter mass, which may both favor a mother's body condition at parturition and perhaps increase the offspring survival at birth.

In this study, we investigated the timing and duration of hibernation as well as body temperature patterns in free-ranging common hamsters (Cricetus cricetus) with regard to sex and age differences. Body temperature was recorded using subcutaneously implanted data loggers. The results demonstrate that although immergence and vernal emergence sequences of sex and age groups resembled those of most hibernators, particularly adult females delayed hibernation onset until up to early January. Thus, in contrast to other hibernators, female common hamsters hibernated for shorter periods than males and correspondingly spent less time in torpor. These sex differences were absent in juvenile hamsters. The period between the termination of hibernation and vernal emergence varied among individuals but did not differ between the sex and age groups. This period of preemergence euthermy was related to emergence body mass: individuals that terminated hibernation earlier in spring and had longer euthermic phases prior to emergence started the active season in a better condition. In addition, males with longer periods of preemergence euthermy had larger testes at emergence. In conclusion, females have to rely on sufficient food stores but may adjust the use of torpor in relation to the available external energy reserves, whereas males show a more pronounced energy-saving strategy by hibernating for longer periods. Nonetheless, food caches seem to be important for both males and females as indicated by the euthermic preemergence phase and the fact that some individuals, mainly yearlings, emerged with a higher body mass than shortly before immergence in autumn.

In seasonally breeding animals, timing of reproduction in females can influence offspring development and survival. Temporal and energetic constraints are often more pronounced in juveniles born late in the season, and could activate the stress axis. Common hamsters (Cricetus cricetus) are hibernating rodents, and adult females produce up to three litters during the active season. Birth dates range from May to September, and shortly after natal emergence pups are weaned and start to disperse. In this study, we used faecal cortisol metabolites (FCM) as a non-invasive measure of recent glucocorticoid exposure. We compared FCM levels between early- (June/July) and late- (August/September) born juvenile Common hamsters during their first weeks after natal emergence. We further compared FCM levels in juveniles born in the study area and individuals that immigrated either early or late in the season. Additionally, we investigated potential effects of human disturbances on cortisol secretion patterns. FCM levels in late-born juveniles increased during the first weeks post-emergence and were significantly higher than those in early-born individuals during the post-weaning period. Late-immigrating juveniles had significantly higher FCM levels than early immigrants and residents during the same time period. Individuals that inhabited areas frequently used by humans had higher FCM levels than those in low-impact areas. These results indicate that the seasonal timing of birth and dispersal affect cortisol secretion patterns in juvenile Common hamsters. As all juveniles immerged into their hibernacula during early October, we assume that late-born and late-immigrated individuals have less time to prepare for hibernation. This could elevate energetic demands and increase exposure to predators and/or humans during food caching. These factors might lead to increased adrenal activity and thus, elevated FCM concentrations in these individuals. Potential consequences of these time constraints and associated physiological effects could be reflected in the lower overwinter survival rates of juveniles born late in the season.

In this study, we investigated endocrine factors and behaviour in free-living Common hamsters (Cricetus cricetus) during reproductive and non-reproductive periods of the annual cycle. We applied a non-invasive method to gain information on seasonal changes in adrenocortical activity in male and female hamsters by analysing faecal glucocorticoid metabolite concentrations (FCM). In addition, plasma progesterone concentrations were monitored in females throughout the non-hibernation season. The animals were live-trapped from spring emergence until the onset of hibernation in autumn. Reproductive status was determined at capture and blood and faecal samples were collected. During behavioural observations, agonistic and sexual interactions were recorded. FCM concentrations were significantly higher in males than in females during the reproductive period. In males, a pronounced increase in FCM during the reproductive period coincided with high frequencies of intrasexual aggression. In females, FCM levels remained relatively constant. Aggressive behaviour in females increased during the reproductive period, but was much less frequent than in males. Females, which successfully raised a second litter after a postpartum oestrus and concurrent lactation and gestation had lower FCM levels than individuals, which lost their second litter after parturition. As expected, plasma progesterone concentrations were low before and after the reproductive period. During gestation, levels peaked and remained elevated during lactation. The results of this field study provide insight in critical periods associated with reproduction in male and female Common hamsters.
