Tadalafil, a potent phosphodiesterase inhibitor 5 (PDE-5), is commonly used for the management of erectile dysfunction. However, its therapeutic potential extends beyond this indication. This study aimed to investigate the impact of tadalafil on the recovery of testicular parenchyma in male Wistar rats exposed to testicular thermal stress. Fifty-four Wistar rats were subjected to testicular thermal stress and randomly assigned to receive either tadalafil treatment (TAD) or no treatment (control). TAD was administered intraperitoneally at a dose of either 0.9 mg/kg or 1.8 mg/kg. Biometric parameters, histopathological assessment of the testis, serum testosterone levels, oxidative stress, and interleukin levels were evaluated on days 7, 15, and 30 after thermal shock. The animals were euthanized at the end of each experimental period, and samples were collected. TAD treatment maintained testicular weight and reduced the testicular degenerative process up to day 7 post-injury. However, despite TAD therapy, serum testosterone levels were decreased in the treated groups at days 7 and 15 post-thermal stress. TAD also decreased TNF-α and NO levels at different doses but had no effect on IL-6. The treatment with TAD after heat shock demonstrated anti-inflammatory and antioxidant properties but did not prevent the aggravation of testicular lesions in subsequent periods, even with the systematic reduction in TNF-α and NO levels. Therefore, this selective PDE-5 inhibitor, at the dosages used, did not have a positive impact on testosterone levels during the post-thermal stress period, which could compromise the resumption of the spermatogenic process.

Food deprivation has been associated with the development of metabolic pathologies. Few studies have explored the repercussions of a partial food deprivation following the reestablishment of an ad libitum diet. This study investigates the impact of a partial food deprivation (an 8-hour food intake restriction coupled with a 4-hour feeding window during the active phase) and the subsequent return to ad libitum feeding on the glycemic curve, food intake, and locomotor behavior. Wistar rats aged 45 days were subjected to 6 weeks of a partial food deprivation followed by 6 weeks of ad libitum feeding. Body weight, visceral fat, food intake, circadian glycemia, oral glucose tolerance, and locomotor activity were evaluated. It was found that the partial food deprivation resulted in the reduction of both the body weight and food intake; however, it increased visceral fat by 60%. Circadian glycemic values were altered at all intervals during the light phase, and glucose sensitivity improved at 60 minutes in the oral glucose tolerance test (OGTT). In the food-deprived group, the locomotor activity rhythm was reduced, with an observed delay in the peak of activity, reduction in total activity, and a decrease in the rhythmicity percentage. After the reestablishment of the ad libitum feeding, there was recovery of body weight, no difference in visceral fat, normalization of the food intake pattern, circadian glycemia, and oral glucose tolerance. Additionally, the return to ad libitum feeding restored locomotor activity, although the duration required for its complete recovery warrants further investigation. In conclusion, partial food deprivation induces physio-metabolic changes in rats, most of which are reversed after reestablishing ad libitum feeding.

This study aimed to evaluate metal exposure through the concentration of essential and toxic micro-minerals in biological samples of Phrynops geoffroanus from an anthropized river. The work was carried out in four areas with different flow characteristics and uses of the river, where individuals of both sexes were captured during the dry and rainy seasons. The elements Al, Cd, Co, Cr, Cu, Fe, Mn, Mo, Ni, Pb, and Zn were quantified in samples of serum (168), muscle (62), liver (61), and kidney (61) by inductively coupled plasma optical emission spectrometry. The concentration of the elements varied according to the sample type, being higher in the liver and the kidney. In the serum, many elements were below the limit of quantification, but it was possible to determine Al, Cu, Fe, Mn, Pb, and Zn. The liver showed high levels of Cu, Fe, Pb, and Zn, and muscle for Fe, Ni, Pb, and Zn, with most of the elements accumulated in the kidney (Al, Cd, Co, Cr, Mn, Mo, and Ni) relative to other tissues. There was no significant difference between the sexes in the accumulation of elements. Between seasons, Cu was higher in serum and Mn in muscle and liver in the dry period, while in the kidney, almost all the elements were higher in the rainy period. The concentrations of the elements in the samples indicated a high degree of environmental contamination, representing risk in the use of the river and consumption of food from local fisheries.

The aim of this study was to evaluate the effect of the replacement of soybean meal by increasing levels of sugarcane yeast and urea on the energy, enzymatic, protein, mineral, and hormonal profiles of buffalo heifers. Twenty Murrah heifers with an average body weight of 168 ± 2.0 kg and 1 year old were used in a completely randomized design. The treatments consisted of the replacement of soybean meal by increasing levels of sugarcane yeast and urea (0, 33, 67, and 100% dry matter). The animals were weighed at the beginning and end of the experimental period, and blood was collected on the final day. The replacement of soybean meal with sugarcane yeast influenced the performance and metabolic dynamics, with total weight gain (P = 0.005) and average daily gain (P = 0.015), presenting, for both variables, a quadratic effect. Considering the biochemical profile, there was an influence on the serum concentration of triglycerides (P = 0.055), the serum activities of aspartate aminotransferase, gamma-glutamyl transferase, and alkaline phosphatase (P < 0.05), in addition to the serum concentration of phosphorus (P = 0.007) and potassium (P = 0.053), which showed positive linear effects. The hormonal profile was not influenced by diets (P > 0.05). The elements copper, iron, and zinc were able to be quantified at all levels of sugarcane yeast and urea in the diets offered. The use of sugarcane yeast with urea promotes adequate metabolic response of buffalo heifers and can replace up to 100% of soybean meal in the concentrate.

Goat farming is concentrated in semi-arid and tropical regions in Brazil. From 2006 to 2017, the number of goats sold in the country increased by 65.7%. The dairy products from these animals present higher digestibility, high vitamin A and B content, hypoallergenicity, and less lactose compared to dairy products from cows, in addition to having a higher sales value. Since corn and soybean meal generate an expense for feed management, crude glycerin, originating from the manufacture of biodiesel, has been studied as an energy substitute. However, this product contains heavy metals, posing risks to animal and human health. Few data are available on trace elements in biological samples and products derived from goats' milk with the dietary introduction of glycerin. The objective was to quantify aluminum (Al), cadmium (Cd), cobalt (Co), chromium (Cr), copper (Cu), iron (Fe), manganese (Mn), molybdenum (Mo), nickel (Ni), lead (Pb), and zinc (Zn) in the serum, urine, milk, and artisanal "coalho" cheese of dairy goats fed different levels of crude glycerin in feed management. In total, 16 Saanen goats were selected, lactating females, that were distributed in four Latin squares and randomly treated with different levels of crude glycerin (0, 5, 10, and 15%). After the end of each experiment cycle, serum, urine, milk, and artisanal "coalho" cheese samples were collected at the four moments. The samples were submitted to digestion assisted by microwave radiation. The multi-element analysis was carried out using inductively coupled plasma optical emission spectrometry (ICP OES). There was no influence of crude glycerin levels replacing corn in serum, milk, urine, and artisanal "coalho" cheese. The serum concentration of the metals Cu, Fe, Zn, and Mn; urine concentrations of Cu, Zn, Mn, Mo, and Cr; and milk and artisanal "coalho" cheese concentrations of Cu, Zn, and Mn remained within the standards found in the literature. The inclusion of 5 to 15% of crude glycerin, derived from cotton oil, in the diets of dairy goats in partial replacement of corn, does not alter the concentration of essential and toxic metals in serum, urine, milk, and artisanal "coalho" cheese and does not, therefore, present a risk factor for intoxication.

The effects of systemic insulin administration at different concentrations on the testicular tissue of diabetic adult rats, induced by streptozotocin, are evaluated by the morphological analysis of spermatogenic process. Twenty-four adult male rats were divided into 1) Control Group: they received citrate buffer, by intraperitoneal injection; 2) Diabetic Group: induced by intraperitoneal injection of streptozotocin (60 mg. kg-1 of body weight); 3) Insulin 50%: induced diabetes treated with half of standard dosage of insulin; 4) Insulin 100%: induced diabetes treated with standard dose of insulin. After eight weeks, animals were weighted and anesthetized; testicles were removed and processed in resin. Body and testicular weight of diabetic rats decreased when compared to that of control. Parameters increased with insulin therapy. Testosterone levels were low in diabetic animals but rates recovered after insulin therapy. Nuclear diameter and volume of Leydig cells decreased in diabetic rats although they significantly increased after insulin therapy. Results showed that the administration of insulin in diabetic rats promoted a protective effect of testicular parenchyma, enhancing efficient recovery on testosterone levels and increase in daily sperm production.

The aim of this study was to determine the serum levels of folic acid, cobalamin, and indicators of energy metabolism during the peripartum period of ewes submitted to different diets and with or without pregnancy toxemia. Forty Santa Inês ewes were divided into three groups, a group of 20 animals fed only roughage-diet with low energy density (G1), a group of 10 ewes submitted to diet containing roughage and concentrate-diet with high energy density (G2), and a group of 10 ewes with diagnosis of pregnancy toxemia (PT) (G3). In the ewes of groups G1 and G2, blood samples were collected -30, -15, -7 days pre-partum, on the day of partum, and 7, 15, and 30 days postpartum; blood samples from G3 animals were collected during the veterinary clinic care. Significant variations were observed in the plasma concentration of glucose and serum folic acid, cobalamin, fructosamine, NEFA, and ß-hydroxybutyrate in the groups fed diets with different energy levels, with and without PT (P < 0.05). A positive correlation of folic acid and cobalamin was observed with plasma glucose and serum NEFA concentrations, in addition to negative correlation of glucose with NEFA and ß-hydroxybutyrate and positive correlation between the latter biomarkers of energy metabolism. The ewes diagnosed with PT showed higher levels of folic acid, NEFA, and ß-hydroxybutyrate (P < 0.05). Ewes fed diet containing roughage and concentrate present higher serum levels of folic acid, cobalamin, and glucose, in the pre-partum period and in the lactation period. The results obtained contribute to better understanding of the application of blood concentration tests for folic acid and cobalamin in the energy metabolism and in PT in ewes.

This study aimed to evaluate the effects of four varieties of cactus pear resistant to carmine cochineal as exclusive roughage for lambs on the biochemical, histopathological, and histomorphometric parameters of kidneys. Twenty-four castrated male crossbred lambs at eight months of age and an initial body weight of 21.0 ± 0.5 kg were distributed in a completely randomized design, with four treatments and six repetitions. The experimental treatments consisted of four diets containing Miúda cactus pear, IPA-Sertânia cactus pear, IPA-F21 cactus pear, or Orelha de Elefante Mexicana (OEM) cactus pear as the only roughage. Blood samples were collected every two weeks (14 d, 28 d, 42 d, and 56 d) to quantify serum urea and creatinine levels. After 72 days of the introduction of the tested diets, the animals were slaughtered and fragments of the kidneys were collected for histological analysis. The serum urea level was higher in animals fed a diet based on the Miúda variety (49.38 mg dL-1), and the serum creatinine levels were lower in the last two collections (P = 0.009). The most frequent histopathological findings in the kidneys were calcification, congestion, glomerular atrophy, presence of luminal cellular debris, and nephrosis, regardless of the cactus pear variety. The Miúda cactus pear and OEM cactus pear varieties caused more severe damage to the nephron components, while the varieties IPA F-21 and IPA-Sertânia caused less significant injuries. The use of IPA-Sertânia and IPA F-21 cactus pear varieties is suggested in lamb's diets, due to the lower impact on the renal parenchyma. However, there was no expressive impairment of renal function, and there was no difference between the cactus pear varieties tested in this study on the weight gain of the animals, and they can all be used to feed feedlot sheep.

Two experiments were performed to evaluate the effects of partial replacement of Tifton hay by Miúda cactus cladodes (Nopalea) or Orelha de Elefante Mexicana (O.E.M.) cactus cladodes (Opuntia) on serum and liver concentrations of iron (Fe), copper (Cu), zinc (Zn), and manganese (Mn) in feedlot small ruminants. The experiment 1 (Exp. 1) lasted 86 days, and thirty-six male Santa Inês lambs (22.0 ± 2.9 kg initial body weight-IBW) were used. In experiment 2 (Exp. 2), lasting 100 days, thirty-six male goats of undefined breed (19.0 ± 2.8 kg IBW) were used. In both studies, the animals were distributed in a completely randomized design, with three treatments (diet control-without cactus; and diets with hay partially replaced by Miúda or O.E.M. cactus cladodes) and twelve replicates. Blood samples were collected to quantify serum micromineral levels, and at the end of the feedlot periods, sheep and goats were slaughtered and liver fragments were collected for micromineral dosage. In Exp. 1, lambs fed Nopalea cactus cladodes diet showed greater hepatic Cu accumulation (441.87 mg/kg DM; P = 0.027). The diets containing cactus cladodes tended to increase serum levels of Cu (P = 0.075) and Zn (P = 0.058). In Exp. 2, goats fed diet containing Opuntia cactus cladodes presented higher serum levels of Fe (P = 0.024) and Cu (P = 0.012). In conclusion, the partial replacement of Tifton hay by cactus cladodes increases hepatic concentrations of Cu in lambs and serum levels of Fe and Cu in goats.

Type 2 diabetes mellitus (T2D) during pregnancy is characterized by high levels of reactive oxygen species and pro-inflammatory factors in the placenta. Once these reactive species reach the foetus, they trigger physiological adaptations that allow the foetus to survive, but programme the organism to develop metabolic disorders in adulthood. The male reproductive system is highly susceptible to foetal programming. This study aimed to investigate the effects of intrauterine exposure to T2D on testicular histomorphometry and redox homeostasis of adult rats and evaluate the effects of maternal treatment with metformin and pentoxifylline. Female rats were induced to T2D, then treated with metformin and pentoxifylline, or co-treated with both drugs. The females were mated, the male offspring were sacrificed on postnatal day 90, and the testicles were collected for analysis. Metformin protected the tubular compartment, with the maintenance of the Sertoli cell population and daily sperm production. Pentoxifylline attenuated the effects of diabetes on Leydig cells, in addition to stimulating testosterone production and lowering lipid peroxidation. Intrauterine exposure to T2D results in important testicular alterations that compromise gonadal function, and the co-treatment with metformin and pentoxifylline may represent a promising therapy that attenuates these effects by combining the positive influences in both the tubular and interstitial compartments of the testicular parenchyma.

This study aimed to determine the mineral profile of hair samples of free-ranging capybaras (Hydrochoerus hydrochaeris) in remnants of the Atlantic Forest located in the Northeast of Brazil, and to evaluate the effects of origin, gender and age of the animals on their mineral accumulation in hair. Twenty hair samples from animals of different areas, genders and ages were prepared using microwave-assisted digestion, employing 5 mL of HNO3 and 2.5 mL of H2SO4 for 100 mg of sample. Digestion efficiency was assessed by residual carbon content. The digested samples were analyzed by inductively coupled plasma optical emission spectrometry (ICP OES). Principal component analysis (PCA) was performed considering the composition data of 11 elements (Al, Ca, Cd, Co, Cr, Cu, Fe, K, Mn, Mo, and Na). The digestion method applied was efficient and the most abundant elements with their respective concentration ranges in mg kg-1 were Al (396-2746), Ca (36-3420), Fe (476-51180), K (115-4843), and Na (72-473). ANOVA and PCA differentiated among the capybaras regarding age (adults) and origin (A3), both associated with higher concentrations of Al, Cd, Fe, and K. Although a higher metal bioaccumulation was observed in adult animals, it is important to highlight that this result could have been affected by diet and physiological parameters. The results suggest that A3 was the most anthropized remnant area due to agricultural and urban influences. Multi-elemental determination in hair can be used as a non-invasive method to assess heavy metal contamination in capybaras.

The aim of this study was to evaluate the effect of green tea extract (GTE) on the spermatic parameters of Wistar rats, submitted or not to testicular heat shock (HS). For this, 48 animals were treated according to the experimental groups (G1: not exposed to HS and untreated; G2: exposed to HS and untreated; G3: not exposed to HS and treated with GTE; G4: exposed to HS and treated with GTE). Subgroups of rats were euthanized on days 15, 30, and 60 to recover the spermatozoa. The total motility (TM), vigor, spermatic morphology and concentration, mitochondrial membrane potential, plasma membrane integrity, and acrosome integrity (ACi) were analyzed. The TM was higher in G1 and G3 than in G2 and G4 on day 30, and higher in G4 on day 60. The overall means of TM and vigor were higher in G1 and G3 than in G2 and G4, as well as TM on day 60. For the morphology, G2 and G4 were lower than G1 and G3 on day 15, and G4 was lower than G1 and G3 on day 30. Moreover, in G1 and G3 morphology was higher on days 15 and 30, and in G4 it was lower on day 30, with the overall means being higher in G1 and G3 than in G2 and G4, as well as on days 15 and 60 compared to day 30. The overall mean of ACi, on day 30, was lower than on days 15 and 60 for all the groups. Therefore, HS is shown to be widely deleterious to the gametes, and the daily administration of 100 mg/kg green tea extract does not improve the spermatic parameters of Wistar rats, submitted or not to testicular HS, although it leads to better recovery of spermatic motility and morphology at 60 days.

Bisphenol A (BPA) is a plasticizer and a risk when it interacts with organisms, and can cause changes in the development and reproduction of them. This study aimed to evaluate the effects of BPA, by acute and chronic toxicity tests with neonates and adults of Pomacea lineata. Adults and neonates were divided into groups exposed to BPA (1-20mg/L), or 17ß-estradiol (1mg/L) and control in the acute and chronic toxicity tests. Behavior, heart rate, reproduction and hemolymph biochemical analysis were measured. In the acute toxicity test, the 96-h LC50 with adults was 11.09 and with neonates was 3.14mg/L. In this test, it was observed lethargic behavior and an increase of 77.6% of aspartate aminotransferase in the adults' hemolymph (p<0.05); and neonates' heart rate decreased 72.7% (p<0.05). In the chronic toxicity test, it was observed behaviors associated with reproduction, as Copulate, in the groups exposed to BPA. The results that were found in this study proved that BPA is a potentially toxic agent to Pomacea lineata according to biological parameters evaluated. These data contribute to the understanding of BPA toxic effects' in the aquatic invertebrates.

Abstract The transplacental transmission is the primary route of Neospora caninum infection in bovine herds around the world. This study aimed to determine the frequency of transplacental transmission of the parasite in dairy cattle of Agreste region of Pernambuco through serological tests (IFAT and ELISA). Three hundred sixteen serum samples from cows and heifers and their offspring were analyzed. The transplacental transmission rate was 72.22% (13/18) for cows and 69.23% (9/13) for heifers by IFAT. ELISA test showed transplacental transmission rate of 43.58% (17/39) for cows and 50% (9/18) for heifers. The transplacental transmission rates were similar, in both groups in test, but a higher seropositivity was found in cows by IFAT. Data were statistically analyzed using the chi-square and Fisher’s exact test. A significant relationship of dependence between seropositivity of mothers and their offspring was found. The more frequent IFAT antibody titers and ELISA levels for N. caninum were, respectively, 200 and between four (cows) and five (heifers and offspring). In the Spearman correlation, no association was found between the magnitude of antibody titers for N. caninum between mothers and their offspring. The kappa test showed an index of 0.35, indicating a mild correlation between the serological tests used. The study suggests that cows and heifers are the main transmitters of N. caninum in the studied region and that vertical transmission is the major form of transmission in dairy herds of the Agreste region of Pernambuco.
Resumo A transmissão transplacentária é a principal via de infecção do Neospora caninum nos rebanhos bovinos em todo o mundo. O presente estudo teve como objetivo determinar a frequência da transmissão transplacentária do parasita em bovinos leiteiros do Agreste Pernambucano, por meio de testes sorológicos (RIFI e ELISA). Foram analisadas 316 amostras de soro de fêmeas bovinas (vacas e novilhas) e de suas crias. A taxa de transmissão transplacentária pela RIFI foi de 72,22% (13/18) para vacas e 69,23% (9/13) para as novilhas. O ELISA teste mostrou taxa de transmissão transplacentária de 43,58% (17/39) para as vacas e 50% (9/18) para as novilhas. As taxas de transmissão transplacentária foram similares para os dois testes em geral, porém uma maior soropositividade foi encontrada nas vacas pela RIFI. Os dados foram estatisticamente analisados pelo teste de qui-quadrado e teste exato de Fischer. Foi encontrada uma relação significativa de dependência entre a soropositividade das mães e de suas crias. Os títulos de anticorpos anti- N. caninum foi de 200 na RIFI e posicionados entre o nível quatro (vacas) e cinco (novilhas e bezerros) pelo ELISA. Pela correlação de Spearman, não foi observada associação entre a magnitude de títulos de anticorpos anti- N. caninum de fêmeas com o de suas crias. O teste de concordância kappa revelou um índice de 0,35, indicando uma concordância leve entre os testes sorológicos utilizados. O estudo sugere que vacas e novilhas são as principais transmissoras do N. caninum na região estudada, sendo a transmissão vertical, a principal forma de transmissão do agente em rebanhos leiteiros do Agreste de Pernambuco.

The transplacental transmission is the primary route of Neospora caninum infection in bovine herds around the world. This study aimed to determine the frequency of transplacental transmission of the parasite in dairy cattle of Agreste region of Pernambuco through serological tests (IFAT and ELISA). Three hundred sixteen serum samples from cows and heifers and their offspring were analyzed. The transplacental transmission rate was 72.22% (13/18) for cows and 69.23% (9/13) for heifers by IFAT. ELISA test showed transplacental transmission rate of 43.58% (17/39) for cows and 50% (9/18) for heifers. The transplacental transmission rates were similar, in both groups in test, but a higher seropositivity was found in cows by IFAT. Data were statistically analyzed using the chi-square and Fisher's exact test. A significant relationship of dependence between seropositivity of mothers and their offspring was found. The more frequent IFAT antibody titers and ELISA levels for N. caninum were, respectively, 200 and between four (cows) and five (heifers and offspring). In the Spearman correlation, no association was found between the magnitude of antibody titers for N. caninum between mothers and their offspring. The kappa test showed an index of 0.35, indicating a mild correlation between the serological tests used. The study suggests that cows and heifers are the main transmitters of N. caninum in the studied region and that vertical transmission is the major form of transmission in dairy herds of the Agreste region of Pernambuco.

(AU)O presente estudo teve por objetivo avaliar a influência da administração de propileno glicol e cobalto associado à vitamina B12 sobre o perfil metabólico e a atividade enzimática de ovelhas da raça Santa Inês no período do periparto. Foram utilizadas 18 ovelhas prenhes, pesando em torno de 40kg. Aproximadamente 30 dias antes da data prevista para o parto foram separadas de maneira aleatória em três grupos e administrados os suplementos conforme a seguir: (G1/n=6) grupo que recebeu propileno glicol (30mL por via oral diariamente); (G2/n=6) grupo que recebeu cobalto (1mg de cloreto de cobalto a 1%, via oral diariamente) associado a vitamina B12 (2mg via intramuscular, semanalmente) e (G3/n=6) grupo controle. As amostras de sangue das ovelhas para avaliação do perfil metabólico e enzimático (glicose, β-hidroxibutirato-BHB, NEFA, proteína total, albumina, uréia, creatinina, AST, GGT, FA e CK) foram colhidas 30 dias antes da data prevista para o parto, uma semana antes (ante-parto), no parto, às 24h, 72h, 5 dias, 15 dias e 30 dias após o parto. Não foi observado cetonúria nos momentos que antecederam ao parto. A administração dos suplementos não influenciou sobre o perfil metabólico, protéico e energético, assim como não houve comprometimento hepático das ovelhas no período do periparto.(AU)
The aim of this study was to evaluate the influence of the administration of propylene glycol and cobalt associated with vitamin B12 on the metabolic profile and enzymatic activity of Santa Inês ewes in the peripartum period. A total of 18 pregnant ewes, weighing around 40kg were used. Approximately 30 days before the expected date of delivery were randomly separated into three groups and administered supplements as follows: (G1/n = 6) group received propylene glycol (30mL orally daily); (G2/n = 6) group receiving cobalt (1mg cobalt chloride 1%, orally daily) associated with vitamin B12 (2mg intramuscular weekly) and (G3/n = 6) control group. Blood samples from ewes to evaluate the enzymatic and metabolic profile (glucose, β-hydroxybutyrate, BHB, NEFA, total protein, albumin, urea, creatinine, AST, GGT, ALP and CK) were taken 30 days before the date set for delivery, one week before (ante partum), delivery at 24h, 72h, 5 days, 15 days and 30 days after delivery. ketonuria was not observed in pre partum. The administration of supplements had no effect on the metabolic profile, protein and energy, and no liver disorders was observed in peripartum.(AU)

O trabalho objetivou conhecer os teores de Cu, Mo, Fe e Zn em soro e fígado de ovinos e caprinos criados na região semiárida do estado de Pernambuco e abatidos nas épocas da chuva e seca, e estabelecer se a carência de Cu é causada por deficiência primária de Cu ou secundária à ingestão de quantidades excessivas de Fe ou Mo. Amostras de soro e fígado de 141 ovinos e 141 caprinos foram submetidas à digestão úmida em ácido nítrico-perclórico e, posteriormente, analisados em espectrofotômetro de absorção atômica indutivamente acoplado (ICPOES). A concentração sérica de Cu em caprinos teve uma média de 9,85±2,71µmol/L e em caprinos de 11,37±2,57µmol/L, enquanto que a concentração hepática média foi de 158,45±83,05mg/kg para ovinos e 152,46±79,58mg/kg para caprinos. Os teores séricos de Fe foram de 35,58± 14,89µmol/L em ovinos e de 25,06±8,10µmol/L em caprinos e as concentrações no fígado foram de 156,10±55,99mg/ kg em ovinos e 210,53±121,99mg/kg em caprinos. As concentrações médias séricas de Mo foram de 0,28±0,11µmol/ L em caprinos e 0,31±0,16µmol/L em ovinos, enquanto que no fígado sua concentração foi de 6,53±4,13mg/kg e 8,10± 4,01mg/kg, respectivamente. A concentração sérica de Zn foi de 11,9±6,07µmol/L em ovinos e 11,79±7,42µmol/ L em caprinos e a concentração no fígado foi de 126,43 ±51,50mg/kg e 132,91±55,28mg/kg em ovinos e caprinos, respectivamente. Verificou-se variação na concentração destes minerais considerando os fatores de variação, como o período sazonal, espécie e sexo. Baseado nos valores observados, considerados marginais, e na ocorrência de surtos de ataxia enzoótica em caprinos e ovinos na região, recomenda-se a suplementação com Cu em animais a campo, tanto na seca quanto na chuva. Considerando as concentrações séricas e hepáticas de Fe e Mo, sugerese que as concentrações marginais de Cu não estejam diretamente relacionadas com o excesso destes minerais. The aim of this research was to determine serum and liver concentrations of Cu, Mo, Fe and Zn in sheep and goats slaughtered in the semiarid region of the state of Pernambuco, northeastern Brazil, during the rainy and dry seasons, and to establish if Cu deficiency which occurs in the region is primary or secondary to high levels of Mo and/or Fe. Serum and liver samples from 141 goats and 141 sheep were submitted to wet digestion in nitric-perchloric acid and analyzed by coupled plasma atomic absorption spectrophotometry (ICPOES). The mean serum concentrations of Cu were 9.85±2.71µmol/L in sheep and 11.37±2.57µmol/L in goats. Mean Cu liver concentrations were 158.45±83.05mg/kg in sheep and 152.46±79.58mg/ kg in goats. Considering that serum and hepatic concentrations of Fe and Mo are within normal ranges or are marginal, it is suggested that the low Cu concentrations are not related with high Fe or Mo ingestion. Para testar a eficiência de vários tratamentos de intoxicação por amônia em bovinos, foram utilizados 25 garrotes que receberam cloreto de amônio por infusão intravenosa (iv) até o surgimento de quadro convulsivo. Em seguida, os animais foram alocados em um dos cinco grupos experimentais e tratados da seguinte forma: 1) controle: infusão (iv) de 300mL de solução salina isotônica (SSI), no decorrer de 4h; 2) infusão (iv) de 30mL kg-1 PV de SSI no decorrer de 4h e administração de 4L de água intraruminal por meio de sonda esofágica (ASE); 3) mesmo tratamento do grupo 2 e dose única (iv) de furosemida (2mg kg-1 PV) (F); 4) injeção (iv) de 5mL kg-1 PV de solução salina hipertônica (SSH) 7,2 por cento nos primeiros 30min, seguida de 20mL kg-1 PV de SSI e 4L de ASE; 5) mesmo tratamento do grupo 4 e dose única de F. No decorrer de 4h após a convulsão, foram determinados os teores plasmáticos de amônia e glicose, ureia, creatinina, potássio e sódio séricos, volume e gravidade específica da urina, e excreção urinária de amônio e ureia. No momento da convulsão, os teores de amônia plasmáticos foram muito altos e idênticos em todos os tratamentos, mas no 120°min, nos grupos tratados com associação de SSH+SSI+ASE (grupos 4 e 5), houve diminuição desse metabólito. O uso de furosemida (grupos 3 e 5) não aumentou a excreção total de urina. A terapia com associação de SSH+SSI+ASE aumentou ainda o volume urinário e a excreção percentual urinária de ureia e amônia durante o período crítico da 1ª hora de tratamento, mas o uso de SSI+ASE (grupos 2 e 3) teve resultados intermediários. A eficiência do tratamento com SSH+SSI+ASE ou SSI+ASE foi superior ao grupo controle. Embora com efeito menor que o observado com SSH+SSI+ASE, a SSI+ASE promoveu melhora no quadro clínico geral e, ao término do experimento, promoveu também uma adequada desintoxicação da amônia.
To test the efficiency of some treatments for ammonia poisoning in cattle, 25 steers were used. Ammonium chloride solution was infused intravenously (iv) in each steer until the onset of convulsive episode. Thereafter, the animals were distributed in one of the five different groups, as follows: 1) control: infusion (iv) of 300mL isotonic saline solution (ISS) throughout the following 4h; 2) infusion (iv) of ISS 30mL kg-1 BW throughout the following 4h and administration of 4L water (W) through stomach tube; 3) the same as group 2, plus a single dose (iv) 2g furosemide kg-1 BW (F); 4) injection of 5mL kg-1 BW hypertonic saline solution (HSS) (7.2 percent) (iv) throughout the first 30min, followed by 20mL kg-1 BW of ISS and 4L W.; 5) the same as group 4 and a single dose of F. For the next 4h after the convulsion, plasma concentration of ammonia and glucose, serum urea, creatinine, sodium and potassium, urine total volume and specific gravity, and urinary excretion of ammonium and urea were followed. At the convulsion, the ammonia blood levels were high and similar among the groups, but at the 120th min the animals treated with HSS+ISS+W (groups 4 and 5) had lower values than the control. Furosemide (groups 3 and 5) did not prevent an increase in the total excretion of urine. The therapy with HSS+ISS+W still increased the urinary volume and the total urinary excretion of urea and ammonium during the critical 1st h of treatment, while the use of ISS+W had intermediate results. The efficiency of the treatment with HSS+ISS+W or ISS+W was better than the control group. Although with lower efficiency as seen in the treatment with HSS+ISS+W, the ISS+W caused an improvement in the clinical picture and at the end of the experiment generated an adequate ammonia detoxification.

Este estudo teve por objetivo avaliar o proteinograma e os teores de cobre, ferro e zinco no soro sangüíneo de ovelhas com mastite induzida por cepa de campo de Staphylococcus aureus. Foram utilizadas 10 ovelhas da raça Santa Inês, primíparas, recém-paridas, com aproximadamente dois anos de idade e bom estado nutricional. Inoculou-se na metade direita da glândula mamária 1,0x10(4) UFC/mL da bactéria, enquanto que a metade esquerda serviu como controle. Os animais foram acompanhados diariamente e a partir do diagnóstico clínico de mastite, procedeu-se colheita do material para realização do proteinograma sérico em gel de poliacrilamida contendo dodecil sulfato de sódio (SDS-PAGE) e para determinação do teor plasmático de fibrinogênio e das concentrações séricas de cobre, ferro e zinco em 16 momentos a saber: antes da inoculação (controle) e 12h, 24h, 36h, 48h, 60h, 72h, 84h, 96h, 108h, 120h, 132h, 168h, 180h, 288h e 336h após a inoculação (p.i.). Todas as ovelhas apresentaram quadro clínico de mastite, com perda da funcionalidade da glândula mamária. O proteinograma permitiu a identificação de 23 proteínas, cujos pesos moleculares (PM) variaram de 26.000 a 185.000 dáltons (Da), incluindo proteínas de fase aguda, IgG e IgA. Notou-se aumento significativo nas concentrações de haptoglobina e ceruloplasmina, assim como de IgG e IgA. Não se constatou alteração nos teores de antitripisina e de glicoproteína ácida .Verificou-se diminuição nos teores de ferro e zinco e elevação na concentração de cobre. Constatou-se correlação positiva entre o teor plasmático de fibrinogênio e as concentrações séricas de ceruloplasmina (r=0,74), a haptoglobina (r=0,62) e IgA(r=0,62). Estes resultados mostram a importância das proteínas de fase aguda ceruloplasmina e haptoglobina como indicadores auxiliares da infecção intramamária de ovelhas, assim como ratifica a relevância do fibrinogênio como marcador inflamatório em razão de sua alta correlação com as proteínas...
The aim of the present study was to evaluate the effect of Staphylococcus aureus experimentally induced mastitis on proteinogram and serum concentrations of cupper, iron and zinc levels of Santa Ines primiparous ewes . The right mammary gland of ten healthy ewes was inoculated with 1,0x10(4) UFC/mL of S. aureus. Clinical examination and determination of serum concentrations of proteins by electrophoresis in polyacrylamide gel (SDS-PAGE), cupper, iron and zinc, as well plasma level of fibrinogen were measured before the inoculation (control) and 12h, 24h, 36h, 48h, 60h, 72h, 84h, 96h, 108h, 120h, 132h, 168h, 180h, 288h and 336h after bacteria inoculation. All animals experimentally infected presented clinical mastitis and subsequent loss of mammary gland function. The electrophoretogram allowed the identification of 23 proteins with molecular weights (MW) ranged from 26.000 to 185.000 daltons (Da) including acute-phase proteins, IgG and IgA. A significant increase (P<0,05) in haptoglobin, ceruloplasmin, IgG and IgA concentrations was observed. Antitrypsin and acid glicoprotein concentrations did not alter. The levels of iron and zinc decreased and the cupper concentration increased . A positive correlation between plasma fibrinogen and serum ceruloplasmin (r=0.74), haptoglobin (r=0.62) and IgA (r=0.62) was also identified. Results showed the importance of ceruloplasmin and haptoglobin as acute-phase proteins in ewes with intramammary infections and confirms fibrinogen as an inflammatory marker because its high correlation with specific proteins. The alterations in the serum levels of Cu, Fe and Zn suggest the action of inflammatory mediators triggered by S. aureus.

Blood samples collected from 14 healthy, four month-old, male goats of mixed breed, weighting from 30 to 45 kg, were analyzed in order to evaluate the effect of refrigeration on blood gas analysis. Blood samples to be used in the blood gas analysis were collected in duplicates, using disposable needles and plastic syringes containing around 1,000 IU of sodium heparin. Unpreserved samples were kept at room temperature, between 23 and 25 ºC, and those to be kept in refrigeration temperatures were placed in styrofoam coolers containing three liters of cold water and three kilograms of ice, in order to keep temperatures between zero and 4 ºC. Blood gas analyses were carried out immediately after collection and after 1, 2, 3, 4, 5, 6, 8, 10, 12 and 24 hours. Storage at room temperature affected significantly the group of variables studied, except blood concentration of HCO3. When samples were kept under refrigeration, partial pressure of O2, CO2 and SO2 were significantly affected. The significant variation in mean values of these variables when compared with initial mean values was greater in samples kept at room temperature. Results for pH, HCO3 - and ABE in the refrigerated samples were stable for up to 24 hours of blood collection. It was concluded, therefore, that blood gas analysis of caprine venous blood may be safely carried out in up to six hours of blood collection, provided that samples are kept under adequate refrigeration.
Objetivando-se avaliar o efeito da refrigeração sobre o exame hemogasométrico, foram utilizados 14 caprinos machos, hígidos, mestiços, com cerca de quatro meses de idade e peso variando entre 30 e 45 kg. As amostras de sangue destinadas ao exame hemogasométrico foram coletadas em duplicata, utilizando-se agulhas descartáveis acopladas a seringas plásticas contendo cerca de 1000 UI de heparina sódica. As amostras não conservadas foram mantidas a temperatura ambiente, entre 23 e 25 ºC e aquelas destinadas à refrigeração foram acondicionadas em isopor contendo três litros de água gelada e três quilos de gelo, mantendo-se assim uma temperatura entre zero e 4 ºC. As análises hemogasométricas foram determinadas imediatamente após coleta e 1, 2, 3, 4, 5, 6, 8, 10, 12 e 24 horas após. Verificou-se efeito significativo da temperatura ambiente sobre o conjunto de variáveis, com exceção da concentração sanguínea de HCO3. Quanto às amostras mantidas sob refrigeração, verificou-se efeito significativo sobre a tensão parcial de O2 e CO2 e SO2; sendo que a variação significativa dos valores médios destas variáveis com os valores médios iniciais foram mais tardias em relação às amostras mantidas a temperatura ambiente. Os valores de pH, HCO3 e ABE das amostras refrigeradas mantiveramse estáveis até 24 horas após a colheita de sangue. Conclui-se, portanto, o exame hemogasométrico de sangue venoso de caprinos pode ser efetivado com segurança até seis horas após sua colheita, desde que mantidos sob refrigeração adequada.
