Natural rubber (NR), derived from Hevea brasiliensis, has properties for biomedical applications. Several studies indicate that these properties can be amplified when we associate another bioproduct. However, there are no studies of aging aspects of this biomaterial regarding changes in functionality, structure and composition. The objective was to evaluate the aging process of natural rubber membranes - copaiba (NRC) subjected to controlled conditions of time, light and presence of oxygen. The NRC was prepared and stored in the presence or absence of light and vacuum, for periods of 30, 60 and 90 days. Subsequently, the membranes were characterized through the techniques of wettability, infrared spectroscopy, thermal analysis, scanning microscopy and antioxidant activity. The wettability analysis, showed that NRC membranes both in the zero time and in the aging time were hydrophilic. Through thermogravimetric analysis and differential exploratory analysis the membranes remained thermally stable. The scanning electronic microscopy, indicated no morphological alterations during the observed period. After 90 days, the packaged membranes showed satisfactory antioxidant activity. Our results suggest that the membranes were resistant to the storage period, since they maintained their chemical, thermal, morphological and antioxidant characteristics. Hence, it corroborates to use of membranes as a possible curative for biomedical applications.

Candida albicans is responsible for most of the nosocomial infections that affect immunocompromised individuals. We investigated the application of eosin in photodynamic inactivation as a strategy in the inhibition of the growth of C. albicans and the morphological variation and growth dynamics in light of fractal theory. The damage caused to fungal structures alters the roughness of the colony, and these changes were described by parameters that were defined by mathematical models. Proliferation of the fungi should be inhibited in the center of the colonies and the analysis of the edges gives an indication about the dynamics of growth and cell reproduction.

Livestock production is considered an important factor influencing global warming; however, climate change has also been considered one of the major factors affecting livestock production and reproduction. The present study was carried out in the Southern region of Brazil and aimed to assess the seasonal influence of climatic factors and external morphology of the animals on the semen quality. Three ejaculates were obtained from each bull (Crioulo Lageano and Angus) in summer and winter seasons. Motile cells, sperm vigor, sperm concentration, sperm membrane status, and sperm abnormalities were analyzed in fresh and post-thawed semen. In addition, the sperm kinetics using CASA was assessed on post-thawed semen. Environmental data such as air temperature (AT), relative humidity (RH), mean radiant temperature (MRT) and temperature-humidity index (THI) were measured. Furthermore, body and testicular metrics, as well as hair coat characteristics, were recorded in each season on all bulls. Environmental traits differed significantly between seasons. Results of the PLS regression and Spearman's correlations confirmed a negative relationship between climatic variables and sperm traits in both seasons. Semen quality between breeds and seasons was similar, satisfactory, and heat stress was not sufficient to impair spermatozoa viability. External morphological traits were considered non-important in the final model. The findings of this study suggest that AT, MRT, and THI were the most important factors that could affect seminal characteristics of bulls raised in a subtropical climate.

Despite their importance as biological control agents, zoophytophagous dicyphine mirids can produce economically important damage. We evaluated the phytophagy and potential impact on tomato plants of Dicyphus cerastii and Nesidiocoris tenuis. We developed a study in three parts: (i) a semi-field trial to characterize the type of plant damage produced by these species on caged tomato plants; (ii) a laboratory experiment to assess the effect of fruit ripeness, mirid age, and prey availability on feeding injuries on fruit; and (iii) a laboratory assay to compare the position of both species on either fruit or plants, over time. Both species produced plant damage, however, although both species produced scar punctures on leaves and necrotic patches on petioles, only N. tenuis produced necrotic rings. Both species caused flower abortion at a similar level. Overall, N. tenuis females produced more damage to tomato fruit than D. cerastii. There was an increased frequency of D. cerastii females found on the plants over time, which did not happen with N. tenuis. Our results suggested that, although D. cerastii caused less damage to fruit than N. tenuis, it still fed on them and could cause floral abortion, which requires field evaluation and caution in its use in biological control strategies.

RESUMO

The current pandemic called COVID-19 caused by the SARS-CoV-2 virus brought the need for the search for fast alternatives to both control and fight the SARS-CoV-2 infection. Therefore, a race for a vaccine against COVID-19 took place, and some vaccines have been approved for emergency use in several countries in a record time. Ongoing prophylactic research has sought faster, safer, and precise alternatives by redirecting knowledge of other vaccines, and/or the development of new strategies using available tools, mainly in the areas of genomics and bioinformatics. The current review highlights the development of synthetic antigen vaccines, focusing on the usage of bioinformatics tools for the selection and construction of antigens on the different vaccine constructions under development, as well as strategies to optimize vaccines for COVID-19.

The search for new materials that can be applied in the treatment of injured human tissues has led to the development of new dressings. Membranes have potential as dressing materials because they can be fitted to and interact with the tissue surface. In this study, we analyze the morphological properties and wettability of latex membranes, along with the incorporation of the photosensitizer methylene blue, in the context of the utility of the membranes in curative applications involving photodynamic therapy (PDT). It was observed that deposition of the photosensitizer into latex membranes increased both the surface roughness and wettability. Antifungal testing indicated that antimicrobial PDT assisted by the latex membranes incorporating methylene blue effectively inactivated Candida albicans.

The aim of this study was to use estrus synchronization protocols to favor fixed-time artificial insemination and consequently fixed-time embryo collection, and increase embryo production using eCG, in gits. In a cross over design, nine Piau breed gilts were subjected to 18 days of oral progesterone; P4 group did not receive any further; GnRH group received 25µg of GnRH 104 hours after the final application of P4; and eCG+GnRH group received 1000IU of eCG 24 hours after the final P4 in addition to GnRH for subsequent embryo collection, that was performed six days after first AI, by laparotomy. Artificial insemination was performed after 12 and 24 hours of estrus in P4 group, and 128 and 144 hours in GnRH and eCG+GnRH groups. The number of CL (8.6±3.9; 8.3±2.1; 26.7±15.0) and anovulatory follicles (4.3±3.7; 3.9±3.9; 17.2±9.5) was higher in the eCG+GnRH gilts (P<0.05). However, the use of 1000 IU of eCG reduced (P<0.05) the number of total structures (5.2±3.6; 5.1±3.1; 1.7±2.7), viable embryos (5.0±3.5; 4.8±3.3; 0.4±0.7), freezable embryos (3.6±3.4; 3.3±3.8; 0.1±0.3) and recovery rate (63.7±38.9; 58.6±24.7; 5.38±9.5). P4 and GnRH protocols were effective in the production and recovery of embryos. However, the use of 1000 IU of eCG, 24 hours after P4, was not effective in promoting the production of embryos, although the animals had superovulated.

This study was designed to evaluate the seasonal expression of seminal plasma proteins from two bovine breeds adapted to a subtropical climate and their associations with post-thawing sperm and environmental characteristics. Semen samples were obtained three times in summer and three times in winter from four Crioulo Lageano and four Angus bulls. Seminal plasma was obtained by centrifugation, and the other portion of the semen was cryopreserved. Seminal plasma proteins were identified by 2D-nanoUPLC-MSE. Post-thawing assessments of sperm kinetics, morphology and membrane integrity were performed. Environmental data such as air temperature, air humidity and black globe temperature (BGT) were recorded, and the temperature-humidity index (THI) was calculated in summer and winter. Results showed that the climate varied significantly between seasons. Although no statistical differences were observed in semen quality between breeds, the protein profiles varied within and between seasons. We suggest that the most critical proteins in summer affecting sperm characteristics were TIMP-2, DNase, Clusterin, CFAH and GPx6. TIMP-2 and DNase showed a higher abundance in Crioulo Lageano in comparison with Angus, while Clusterin, CFAH and GPx6 presented a lower abundance. To the best of our knowledge, this is the first report of a recently evolved type of glutathione peroxidase, GPx6, in seminal plasma of bovines. In winter, five proteins were considered to be more critical: BSP1, BSP3, CCL2, Sulfhydryl oxidase and TIMP-2. BSP1 and TIMP-2 showed a lower abundance while BSP3, CCL2 and Sulfhydryl oxidase presented a higher abundance in this season in Crioulo Lageano in comparison with Angus.


RESUMO: Este estudo foi desenvolvido para avaliar a expressão sazonal de proteínas plasmáticas seminais de duas raças bovinas adaptadas ao clima subtropical e suas associações com espermatozóides pós-descongelamento e características ambientais. Amostras de sêmen foram obtidas três vezes no verão e três no inverno de quatro touros Crioulo Lageano e quatro Angus. O plasma seminal foi obtido por centrifugação e outra porção do sêmen foi criopreservada. As proteínas plasmáticas seminais foram identificadas por 2D-nanoUPLC-MSE. Foram realizadas avaliações pós-descongelamento da cinética espermática, morfologia e integridade da membrana. Dados ambientais como temperatura do ar, umidade do ar e temperatura do globo negro (BGT) foram registrados, e o índice temperatura-umidade (THI) foi calculado no verão e no inverno. Os resultados mostraram que o clima variou significativamente entre as estações. Embora não tenham sido observadas diferenças estatísticas na qualidade do sêmen entre as raças, os perfis proteicos variaram dentro e entre as estações. Sugerimos que as proteínas mais críticas no verão que afetam as características espermáticas foram TIMP-2, DNase, Clusterin, CFAH e GPx6. TIMP-2 e DNase apresentaram maior abundância em Crioulo Lageano em comparação com Angus, enquanto Clusterin, CFAH e GPx6 apresentaram menor abundância. Até onde sabemos, este é o primeiro relato de um tipo recentemente desenvolvido de glutationa peroxidase, GPx6, no plasma seminal de bovinos. No inverno, cinco proteínas foram consideradas mais críticas: BSP1, BSP3, CCL2, sulfidril oxidase e TIMP-2. BSP1 e TIMP-2 apresentaram menor abundância, enquanto BSP3, CCL2 e Sulfidril oxidase apresentaram maior abundância nesta temporada em Crioulo Lageano em comparação com Angus.

The present study aimed to investigate the effects of a combination of progesterone with different doses of E-17ß on following end points: (1) ovarian follicular dynamics and emergence of a new follicular wave, and (2) superovulatory response and embryo yield. In Experiment 1, 28 ewes were randomly divided into four groups (n = 7) to receive either 2.0 mg, 1.0 mg, 0.5 mg or none E-17ß one day after insertion of a progesterone device. The different doses of estradiol similarly delayed the moment of follicular emergence (overall mean = 3.1 ± 1.0 days vs. control group = 0.86 ± 1.0 days; P < 0.01), but the emergence of the new wave showed greater synchronization with the 0.5 mg dosage of E-17ß. In Experiment 2, sixty-two donor ewes received an internal progesterone release device (day -1) for 7 d and 1 d after the insertion of this device (day 0) were allocated randomly to receive 0.5 mg of E-17ß or only the vehicle (control group). Superstimulation was initiated on day 3 with the administration of 133 mg of pFSH in eight decreasing doses. Contrary to expectations, the protocol with the administration of 0.5 mg E-17ß did not improve the percentage of donors with > 2 CL, the number of CL and the production of embryos (P > 0.05). It was concluded that the combination of progesterone and 0.5 mg E-17ß was more efficient in synchronizing the emergence of the new follicular wave, however this approach seems to be unnecessary in ewe's superovulation programs.

The placenta works as a selective barrier, protecting the fetus from potential infections that may affect the maternal organism during pregnancy. In this review, we will discuss several challenging infections that are common within Latin American countries and that may affect the maternal-fetal interface and pose risks to fetal development. Specifically, we will focus on emerging infectious diseases including the arboviruses, malaria, leishmaniasis, and the bacterial foodborne disease caused by Shiga toxin-producing Escherichia coli. We will also highlight some topics of interest currently being studied by research groups that comprise an international effort aimed at filling the knowledge gaps in this field. These topics address the relationship between exposure to microorganisms and placental abnormalities, congenital anomalies, and complications of pregnancy. Introduction: The placenta is a temporary and unique organ that allows for the physical connection between a mother and fetus; this organ regulates the transport of gases and nutrients mediating the elimination of waste products contained in the fetal circulation. The placenta performs metabolic and excretion functions, on the basis of multiple enzymatic systems responsible for the oxidation, reduction, hydrolysis, and conjugation of xenobiotics. These mechanisms give the placenta a protective role that limits the fetal exposure to harmful compounds. During pregnancy, some diseases require uninterrupted treatment even if it is detrimental to the fetus. Drugs and other xenobiotics alter gene expression in the placenta with repercussions for the fetus and mother's well-being.Areas covered: This review provides a brief description of the human placental structure and function, the main drug and xenobiotic transporters and metabolizing enzymes, placenta-metabolized substrates, and alterations in gene expression that the exposure to xenobiotics may cause.Expert opinion: Research should be focused on the identification and validation of biological markers for the assessment of the harmful effects of some drugs in pregnancy, including the evaluation of polymorphisms and methylation patterns in chorionic villous samples and/or amniotic fluid.

RESUMO

Here, we propose Uanauna gen. nov. (Lampyridae: Photinini), a new monotypic genus of fireflies endemic to the Brazilian Atlantic Forest. Uanauna angaporan sp. nov. occurs in Submontane and Montane forest habitats at the Serra do Mar mountain range, in the Southern Fluminense region (Rio de Janeiro State, Southeastern Brazil). The new species was recorded both on the mainland, at the municipality of Itaguaí and Rio de Janeiro, and at the continental island of Ilha Grande, in Angra dos Reis, at the Rio de Janeiro State. An occurrence map and illustrations of diagnostic features are given for the new taxa, as well as a key to Lucidotina genera in Brazil.

Candida albicans is responsible for the majority of nosocomial infections affecting immunocompromised patients. Systemic antifungals may promote microbial resistance, which has led to the search for alternative treatments, such as photothermal therapy (PTT). PTT assumes that the interaction of electromagnetic radiation with a photothermal agent generates heat that can lead to the destruction of tumor cells and the death of microorganisms. Carbon nanotubes (CNTs) have the potential for applications in biomedical systems, including acting as controlled deliverers of drugs, biosensors and scaffolds for tissue engineering and regenerative medicine. Furthermore, the absorption of radiation by CNTs in the infrared region induces an increase in temperature, which makes CNTs candidates for photothermal agents. In this work, the photothermal inactivation of C. albicans was evaluated by multiple wall CNTs associated with laser radiation in the near-infrared region. The mechanisms that are involved in inactivation were evaluated through cell susceptibility studies and an analysis of microscopic images that are associated with mathematical models and fractal concepts. The results indicate that direct contact between the cells and CNTs without irradiation does not lead to cell death, whereas the laser-mediated process is effective in inactivation. The application of the laws of scale and fractal concepts indicate that in the control groups, there are two distinct regimes that are delimited by the mean diameter of the microorganisms, as described by the Eden model and by the quasi-Euclidean surface. For the irradiated groups, the surfaces present only one regime described by Kardar-Parisi-Zhang, KPZ. The analysis of the fractality of the system by mathematical models can help in the identification of new strategies for the inactivation of microorganisms.

Our aim was to evaluate effects of natural rubber membrane (NR) associated with aqueous propolis extract (P) on the wound healing in rats burn model. The membranes were characterized for wettability and pore presence after the membrane preparation by casting the liquid solution in Petri dishes, at different temperatures (27 and 60 °C). Male Wistar rats were divided into treatment groups (n = 5 per group: control without treatment, silver sulfadiazine, NR, NRP) initiated immediately after second-degree burn and maintained during 10 days. The NRP membranes presented hydrophilic and porous characteristics. Macroscopic analysis from lesions showed that all groups presented crust formation. At the end of the experiment, the lesions treated with NRP membranes prepared at 60 °C had a higher regression percentage indicating a faster wound healing. The biopsies showed that the treatment with the NRP membranes induced a wound healing with collagen production, angiogenesis, reepithelization, and a small number of inflammatory cells, characterized by the crust detachment and the epithelium formation. In conclusion, NRP membranes are promising as a dressing for the burns treatment since were able to accelerate the healing process and tissue repair without the curative switch.

Successful placentation is a key event for fetal development, which commences following embryo implantation into the uterine wall, eliciting decidualization, placentation, and remodeling of blood vessels to provide physiological exchange between embryo-fetus and mother. Several signaling pathways are recruited to modulate such important processes and specific proteins that regulate placental function are a target for the glycosylation with O-linked ß-N-acetylglucosamine (O-GlcNAc), or O-GlcNAcylation. This is a reversible post-translational modification on nuclear and cytoplasmic proteins, mainly controlled by O-GlcNAc transferase (OGT) and O-GlcNAcase (OGA). O-GlcNAcylation has been implicated as a modulator of proteins, both in physiological and pathological conditions and, more recently, O-GlcNAc has also been shown to be an important modulator in placental tissue. In this mini-review, the interplay between O-GlcNAcylation of proteins and placental function will be addressed, discussing the possible implications of this post-translational modification through placental development and pregnancy.

The use of layer-by-layer (LbL) deposition technique allows materials, such as drugs, to be self-assembled in multilayers with other electrolytes by combining their properties in a nanostructured system. Triclosan (TCS) is commonly used as a drug because of its bactericidal action, while erythrosine (ERY) has been used as a photosensitizer in photodynamic therapies because of its high light absorptivity in the visible region of the electromagnetic spectrum. The major advantage of investigating systems immobilized in LbL films is the benefit of characterizing the interaction through available substances in solid state techniques. It was possible to immobilize in LbL films, ERY, and ERY + TCS. The results show that the growth of the films was linear, indicating the deposition of the same amount of material from the first bilayer without substrate interference. The release analysis showed slow kinetics, which occurred more rapidly for ERY LbL films, probably due to apparent activation energy, which were higher for films with TCS. The combination of TCS, ERY, and laser light (532 nm) for photodynamic inactivation of the fungus Candida albicans was analyzed, and the results were promising for future studies in applications, such as coating surfaces of dental implants.

RESUMO

Luciuranus Silveira, Khattar Mermudes, 2016 is a firefly genus whose species bear an intricate, species-specific lock-and-key mechanism of reproductive isolation. Here we propose four new species, Luciuranus magnoculus sp. nov., L. desideratus sp. nov., L. takiyae sp. nov. and L. carioca sp. nov., and provide illustrations of their diagnostic features and an updated key to species. As previously reported for their congenerics, each of the four new species have stereotypical morphology of both male and female terminalia, and are regarded as prima facie endemics of single massifs of the Serra da Mantiqueira and Serra do Mar, in the Brazilian Atlantic Rainforest.

RESUMO

A new species of Tricorythodes Ulmer (Ephemeroptera: Leptohyphidae) is described and illustrated based on nymphs and adults from the Doce River and surrounding areas in southeastern Brazil. Tricorythodes tragoedia sp. nov. is related to T. arequita Traver, T. mirca Molineri and T. sallesi Dias, Cabette & De Sousa, but its nymphs can be distinguished from these species by having a three-segmented maxillary palp with apical seta and one pair of submarginal denticles on the tarsal claws. Nymphs were collected from small to large rivers at altitudes of less than 100 m where they inhabit submersed substrates in areas with slow current. Nymphs and adults were found throughout the year.

Two Gram-negative, heterotrophic, aerobic, prosthecated, marine bacteria, designated strains MCS23T and MCS27T, were isolated from seawater samples. NaCl was required for growth. The major polar lipid detected in strain MCS27T was phosphatidylglycerol, whereas those detected in MCS23T were phosphatidylglycerol, sulfoquinovosyl diacylglycerol and 1,2-diacyl-3-α-d-glucuronopyranosyl-sn-glycerol taurineamide. The most abundant cellular fatty acids were C18 : 1ω7 and C16 : 0, hydroxyl-fatty acids were 3-OH C12 : 0 in both strains and 3-OH C11 : 0 in MCS23T. Strains MCS23T and MCS27T had DNA G+C contents of 57.0 and 55.0 mol%, respectively. The two strains shared 99.3 % 16S rRNA gene sequence similarity; levels of similarity with the type strains of species of the genus Henriciella were 99.4-97.8 % but DNA-DNA hybridizations were 53 % or lower. Besides their 16S rRNA gene sequences, the novel strains can be differentiated from other species of the genus Henriciella by cell morphology, lipid and fatty acid patterns and enzyme activities. The data obtained led to the identification of two novel species, for which the names Henriciella barbarensis sp. nov. (type strain MCS23T=LMG 28705T=CCUG 66934T) and Henriciella algicola sp. nov. (type strain MCS27T=LMG 29152T=CCUG 67844T) are proposed. As these two novel species are the first prosthecate species in the genus Henriciella, an emended genus description is also provided.

Candida albicans is responsible for many of the infections affecting immunocompromised individuals. Although most C. albicans are susceptible to antifungal drugs, uncontrolled use of these drugs has promoted the development of resistance to current antifungals. The clinical implication of resistant strains has led to the search for safer and more effective drugs as well as alternative approaches, such as controlled drug release using liposomes and photodynamic inactivation (PDI), to eliminate pathogens by combining light and photosensitizers. In this study, we used layer-by-layer (LBL) assembly to immobilize triclosan and acridine orange encapsulated in liposomes and investigated the possibility of controlled release using light. Experiments were carried out to examine the susceptibility of C. albicans to PDI. The effects of laser irradiation were investigated by fluorescence microscopy, atomic force microscopy, and release kinetics. Liposomes were successfully prepared and immobilized using the self-assembly LBL technique. Triclosan was released more quickly when the LBL film was irradiated. The release rate was approximately 40% higher in irradiated films (fluence of 15J/cm2) than in non-irradiated films. The results of the susceptibility experiments and surface morphological analysis indicated that C. albicans cell death is caused by photodynamic inactivation. Liposomes containing triclosan and acridine orange may be useful for inactivating C. albicans using light. Our results lay the foundation for the development of new clinical strategies to control resistant strains.
