RESUMO

The usage of 0.6-0.8 mg of lysenile in the course of 6 months gives a possibility to stimulate pregnancy in 70% of patients and gain pregnancy in 20% of patients. In order to exceed the percentage of impregnantion, the course of medicine can be enlarged up to 15 months.

RESUMO

The usage of metergoline in patients with infertility and different forms (hinds) of hyperprolactinaemia was studied for 90 days at a dose of 12 mg. It gives a possibility to stimulate ovulation in 64% of patients and gain pregnancy in 8% of patients. In order to exceed the percentage of inpregnantion the course of medicine can be enlarged up to 6 months.

RESUMO

The study of hormonal balance in the presence of pregnancy and labor in females with a history of infertility treated with parlodel failed to reveal any negative effect of elevated prolactin levels on the fetoplacental system. No differences in the course of pregnancy and labor were revealed as compared to the untreated age-matched. Weight and height figures of neonates born to parlodel-treated mothers were similar to those from the control group. There was no evidence of malformations or developmental defects in the studied group of neonates.

A study was made of function of the hypothalamohypophyseal system in women with infertility combined with hyperprolactinemia. Changes in the levels of prolactin, LH, FSH and TSH in response to the administration of releasing hormones (gonado- and thyroliberin) and metoclopramide (dopamine antagonist) were determined in 28 women with hyperprolactinemic amenorrhea and in 5 women with a normal menstrual cycle. Resistance of lactotrophs to functional tests was revealed. It grew with an increase in a prolactin level. On the contrary, a response of gonado- and thyrotrophs to functional tests grew with an increase in a degree of hyperprolactinemia. The authors arrived at a conclusion that stable hyperprolactinemia resulted from disorder of the receptor apparatus of lactotrophs with the resultant nonsusceptibility of the latter to an inhibitory effect of dopamine to prolactin secretion. Proceeding from their own and literature data, the authors propose for discussion a scheme of fertility disturbance in stable hyperprolactinemia.

RESUMO

The response of water-electrolyte metabolism regulating hormones to neurosurgical stress was studied in 54 patients with tumors of the chiasma-sellar area of the brain. Three types of responses of hormones (the activity of renin, aldosterone, cortisol, and vasopressin) were revealed. Type I was characterised by a sharp activation of the hormones studied, type II by an absence of a response, type III by activation of renin and aldosterone and a decrease in vasopressin activity. Responses of types II and III were attended by deep disorders of water-electrolyte balance. The work substantiates the necessity of taking into account the state of the humoral mechanisms of regulation in conducting intensive therapy in neurosurgical patients.

RESUMO

The content and metabolism of cyclic nucleotides in the aorta as well as contractility were studied in rats given different doses of indometacin. High doses of the drug (5 mg/kg) favored an abrupt decrease in cAMP and cGMP levels in the aorta and did not essentially affect vascular response to electric stimulation. Low doses of indometacin (2 mg/kg), particularly when coupled with salt load, resulted in the decreased cAMP/cGMP ratio and in the potentiation of vasoconstrictor responses. It has been demonstrated that the reduced proportion of cAMP in cyclic nucleotide metabolism is related to the activation effect of indometacin on cAMP-phosphodiesterase activity. The role of the prostaglandin-cyclic nucleotide system in the vascular tone control is discussed.

RESUMO

Taurine produced no effect on the cyclic nucleotides level in the heart of intact rats but sharply inhibited the cAMP and cGMP level elevation in the rat heart occuring in stress. After atropine pretreatment of the animals no effect of taurine on the heart cGMP level was observed; its effect on the cAMP level was significantly inhibited against the background of partial beta-adrenoreceptors block. It is suggested that taurine is a nonspecific regulator of the myocardial cells sensitivity to the biologically active drugs.

The reaction of cyclic nucleotides in blood to an orthostatic position, furosemid administration, and submaximal bicycle ergometry load was studied in 20 healthy individuals and in 50 patients with hypertensive disease. A special group was composed of 25 patients the disease in whom was marked by crises. It is shown that a walk of one hour and intravenous infusion of 40 mg of furosemid caused an increase in the blood cAMP but did not change the level of cGMP. During a bicycle ergometry load the levels of cGMP and cGMP increased to an equal measure and returned to their initial values 30 min after its cessation. In patients with arterial hypertension the cGMP system becomes most functionally mobile, whereas the cAMP level, if it increases, does so at a later term after the effect of the stimulus begins. The increased cGMP level is maintained for quite a lengthy period of time after the load is discontinued. In patients suffering from hypertensive disease with crises the cAMP/cGMP ratio, which is reduced at rest also, diminishes still more in an orthostatic position and particularly during a crisis. The role of changes in the metabolism of cyclic nucleotides and their sensitivity to regulatory factors in the pathogenesis of arterial hypertension is discussed.

The content of cyclic nucleotides (cAMP and cGMP) in the blood plasma, urine and tissues, and also morphological changes of the vascular renal bed were studied in rats with arterial hypertension induced by chronic inhibition of prostaglandin synthesis. A considerable thickening of the wall of the interlobular and arcuate arteries with marked lumen narrowing occurred mainly on account of hypertrophy and the swelling of smooth muscle cells. At the same time there was a marked increase in the cGMP concentration, a decrease of cAMP level, and a reduction of the cAMP/cGMP coefficient in the biological fluids. It is suggested that the changed cyclic nucleotides metabolism is associated with organic and functional changes of the peripheral vascular bed underlying an increase of the total vascular resistance in arterial hypertension.

Administration of hydrocortisone into healthy rabbits activated adenilate cyclase and phosphodiesterase in liver tissue; activity of the enzymes was normalized within 5 days after the treatment. The hormone, administered into animals with experimental cholesterol-induced atherosclerosis, caused an activation of adenilate cyclase and inhibition of phosphodiesterase; due to the phenomenon more distinct and long-term increase in cAMP concentration was observed in kidney, liver and fatty tissues. Concentration of cAMP exceeded considerably its initial content in the tissues within 5 days after the hydrocortisone administration. Hydrocortisone inhibited the adenilate cyclase system activity in adrenal cortex of experimental and control animals at early period of the experiment. In health rabbits content of cAMP was increased in adrenal cortex within 5 days after the hormone administration. As the similar effect was not found in animals with experimental atherosclerosis these data suggest that the hypophysis-adrenal cortex system under the experimental conditions studied was inhibited.

Chronic administration of small doses of prostaglandin-synthesis inhibitor indometacin against the background of a salt load or unilateral nephrectomy induces the development of arterial hypertension in rats. Arterial pressure increases (two fold on the average) on the 6th week of the experiment in 60-80% of the animals. Arterial hypertension developing against the background of a salt load is marked by retention of sodium in the organism and increase the intravascular volume, while that developing in unilateral nephrectomy--by increased sodium excretion and reduced intravascular volume. Depressed activity of the renin-angiotensin-aldosterone system and conspicious changes in the renal vascular channel are noted in both forms of arterial hypertension. It is assumed that disorders in the metabolism of cyclic nucleotides underlie the changes occurring in the renal vessels due to the effect of indometacin. Similar generalized changes in the peripheral vascular channel on the whole may be the cause of the increased vascular resistance and one of the causative factors of the hypertension development.

Content of 3',5'-cycloAMP was estimated in rabbit blood plasma and urine; activities of adenylcyclase and phosphodiesterase were determined in liver tissue, adipose tissue and adrenal glands within 5, 24 and 48 hrs after hydrocortisone administration. Concentration of 3',5'-cycloAMP was found to be increased 4-6-fold in plasma and urine after the hormone administration. Cortisol was shown to increase also the activity of adenylcyclase and phosphodiesterase in liver tissue within 5 hrs and in adipose tissue--within 24 hrs and 48 hrs after the administration. In adrenal glands hydrocortisone caused an inhibitory effect on the adenylcyclase activity at early steps of the experiment.
