RESUMO

Glass serves as the solid support for a variety of array types; however, the chemical nature of glass makes it unsuitable for high-affinity binding to most biomolecules. In this chapter, we describe the activation and surface coating of glass with silane, a wide-ranging group of molecules that can covalently attach to the surface of glass and modify it with a variety of functional groups.

BACKGROUND: Neonatal and post-weaning colibacillosis caused by enterotoxigenic E. coli is responsible for substantial economic losses encountered by the pork industry. Intestinal colonization of young piglets by E. coli depends on the efficiency of bacterial attachment to host gastrointestinal epithelium that is mediated by fimbriae. We tested the effect of porcine individual milk fat globule membrane (MFGM) proteins on F4ac positive E. coli attachment to porcine enterocytes in vitro. RESULTS: Butyrophilin, lactadherin and fatty acid binding protein inhibited fimbriae-dependent adherence of E. coli to enterocytes in vitro, while xanthine dehydrogenase did not. The inhibiting activity was dose-dependent for all three proteins, but the inhibiting efficiency was different. CONCLUSIONS: The results indicate that MFGM proteins may interfere with attachment of E. coli to porcine neonatal intestinal mucosa.

Last October's Healthcare Estates 2013 conference saw one of the first day's 'Engineering' sessions debate the topic, 'Why do so many buildings disappoint their owners and occupants?' Much of the discussion centered on the problems caused by 'inadequate management of the commissioning process'. A roundtable debate jointly staged recently in London by IHEEM and the B&ES, the leading U.K. trade association for building services engineering contractors, took the debate forward. As HEJ editor, Jonathan Baillie reports, the discussions confirmed that one of the key contributors to poor commissioning is a failure to involve specialist building services contractors sufficiently early. It was also agreed that finding a really effective 'client-side' project manager, with the panoply of skills and experience the role requires, can be 'a tough ask'. In this issue of HEJ we report on the debate's first 'half'; in June's edition, we will cover 'part two' of a lively, forthright, and positive debate.
