RESUMO

The COVID-19 pandemic has been a major health concern in Bangladesh until very recently. Although the Bangladesh government has employed various infection control strategies, more targeted Non-Pharmaceutical interventions (NPIs), including school closure, mask-wearing, hand washing, and social distancing have gained special attention. Despite significant long-term adverse effects of school closures, authorities have opted to keep schools closed to curb the spread of COVID-19 infection. However, there is limited knowledge about the impact of reopening schools alongside other NPI measures on the course of the epidemic. In this study, we implemented a mathematical modeling framework developed by the CoMo Consortium to explore the impact of NPIs on the dynamics of the COVID-19 outbreak and deaths for Bangladesh. For robustness, the results of prediction models are then validated through model calibration with incidence and mortality data and using external sources. Hypothetical projections are made under alternative NPIs where we compare the impact of current NPIs with school closures versus enhanced NPIs with school openings. Results suggest that enhanced NPIs with schools opened may have lower COVID-19 related prevalence and deaths. This finding indicates that enhanced NPIs and school openings may mitigate the long-term negative impacts of COVID-19 in low- and middle-income countries. Potential shortcomings and ways to improve the research are also discussed.

In different regions of the world, cowpea (Vigna unguiculata (L.) Walp.) is an important vegetable and an excellent source of protein. It lessens the malnutrition of the underprivileged in developing nations and has some positive effects on health, such as a reduction in the prevalence of cancer and cardiovascular disease. However, occasionally, certain biotic and abiotic stresses caused a sharp fall in cowpea yield. Major RNA interference (RNAi) genes like Dicer-like (DCL), Argonaute (AGO), and RNA-dependent RNA polymerase (RDR) are essential for the synthesis of their associated factors like domain, small RNAs (sRNAs), transcription factors, micro-RNAs, and cis-acting factors that shield plants from biotic and abiotic stresses. In this study, applying BLASTP search and phylogenetic tree analysis with reference to the Arabidopsis RNAi (AtRNAi) genes, we discovered 28 VuRNAi genes, including 7 VuDCL, 14 VuAGO, and 7 VuRDR genes in cowpea. We looked at the domains, motifs, gene structures, chromosomal locations, subcellular locations, gene ontology (GO) terms, and regulatory factors (transcription factors, micro-RNAs, and cis-acting elements (CAEs)) to characterize the VuRNAi genes and proteins in cowpea in response to stresses. Predicted VuDCL1, VuDCL2(a, b), VuAGO7, VuAGO10, and VuRDR6 genes might have an impact on cowpea growth, development of the vegetative and flowering stages, and antiviral defense. The VuRNAi gene regulatory features miR395 and miR396 might contribute to grain quality improvement, immunity boosting, and pathogen infection resistance under salinity and drought conditions. Predicted CAEs from the VuRNAi genes might play a role in plant growth and development, improving grain quality and production and protecting plants from biotic and abiotic stresses. Therefore, our study provides crucial information about the functional roles of VuRNAi genes and their associated components, which would aid in the development of future cowpeas that are more resilient to biotic and abiotic stress. To combat with energy crisis considering clean energy, oxygen evolution reaction (OER) is crucial to implement electrolytic hydrogen fuel production in real life. Here, straightforward chemical synthesis pathways are followed to prepare cobalt tetraoxide nanoparticles (Co3 O4 NPs) in an alkaline OER process using poly[(2-methacryloyloxy)ethyl]trimethylammonium chloride (Co3 O4 NPs@PMTC) as support to prevent aggregation. In material characterization, the X-ray diffraction (XRD) pattern confirms the crystallinity of the synthesized Co3 O4 NPs@PMTC, and Raman spectroscopy indicates that the Co3 O4 NPs contain cubic close-packed oxides. The morphological analysis reveals the wrinkle-like disruption which is distributed evenly owing to the folded nanosheet arrays. Energy-dispersive X-ray spectroscopy indicates the presence of a significant number of cobalt atoms in the Co3 O4 NPs, and elemental mapping analysis demonstrates the composition of the NPs. At a current density of 10 mA cm-2 , oxygen is emitted at 1.67 V delivering an overpotential of 440 mV. This unique structure of Co3 O4 NPs@PMTC provides beneficial functions that are responsible for a large number of active sites and the rapid release of oxygen gas with long-term stability. Through kinetic study, we found a Tafel slope of 48.9 mV dec-1 which proves the catalytic behavior of Co3 O4 NPs@PMTC is promising toward the OER process.

Adsorption-mediated methods for environmental pollution control are suitable as they are cost-effective and easy to use. Porous materials can play an important role in adsorption studies. Herein, we synthesized a p-n heterostructure of phosphorene and metal oxide using a simple hydrothermal approach. The synthesized material is porous in nature, with a surface area of 127.44 m2/g and pore volume of about 1.73 nm with appreciable thermal stability. As the material is microporous, we used it for the adsorption of CO2 gas and dye. For CO2 adsorption, we determined the CO2 gas uptake according to the mass balance principle of the ideal gas equation, and it was found to be about 21.478 mol/kg. We have also studied different isotherm models to check the adsorption phenomena. Moreover, for dye adsorption, we have chosen the xanthene-derived rose bengal (RB) dye, which shows a removal percentage of about 92.02%. In the case of dye adsorption, the material shows good reusability and significant adsorption up to five cycles.

Background: In South Asia, women often experience intimate partner violence (IPV) and have limited access to maternal health services (MHS). However, the effects of IPV on antenatal care (ANC) visits remain unclear. This study aimed to examine the impact of IPV of different forms and severities on ANC visits in South Asia. Methods: This cross-sectional study used the latest available data from demographic and health surveys conducted in Bangladesh, India, Afghanistan, Nepal, Maldives, and Pakistan. The study sampled 4467 women who had given birth within the past 12 months and were interviewed for IPV. IPV was measured by binary variables indicating the presence of physical violence (PV), categorised into less severe (LSPV) and severe physical violence (SPV), emotional violence (EV), and sexual violence (SV). ANC utilization was measured using binary variables indicating whether respondents had any, at least four, or at least eight ANC visits, as recommended by World Health Organization (WHO). Logistic regressions adjusted for survey weights were used to assess associations between ANC utilization and exposure to IPV during pregnancy and lifetime. Results: The prevalence of LSPV, SPV, EV, and SV during pregnancy were 14.5%, 4.4%, 11.6%, and 4.1%. LSPV experience during pregnancy was associated with decreased likelihoods of at least four ANC visits (odds ratio (OR) = 0.55; 95% confidence interval (CI) = 0.40-0.76) and eight ANC visits (OR = 0.53; 95% CI = 0.31-0.90). Results of lifetime exposure to IPV followed similar patterns. Lifetime exposure to LSPV was associated with decreased likelihoods of at least four ANC visits (OR = 0.55; 95% CI = 0.41-0.74) and eight ANC visits (OR = 0.47; 95% CI = 0.29-0.77). Conclusion: This study highlights the negativities of LSPV on the frequency of women seeking ANC visits. Policies are necessary to identify women at risk of the often-overlooked LSPV early and provide protective interventions to promote maternal health in South Asia.

Monitoring therapeutic drugs and their elimination is crucial because they may cause severe side effects on the human body. Methotrexate (MTX) is a widely used anti-cancer drug, which is highly expensive, and the detection of unwanted overdoses of MTX using traditional procedures is time-consuming and involves complex instrumentation. In this work, we have developed a nanocomposite material using phosphorene, cystine, and gold (Ph-Cys-Au) that shows excellent optical properties. This nanocomposite can be used as an optical sensing platform for the detection of MTX in the range 0-260 µM. The synthesized sensing platform is very sensitive, selective, and cost-effective for the detection of MTX. Ph-Cys-Au can effectively detect MTX in aqueous media with a limit of detection (LOD) of about 0.0266 nM (for a linear range of 0-140 µM) and 0.0077 nM (for a linear range of 160-260 µM). The nanocomposite is equally selective for real samples, such as human blood serum (HBS) and artificial urine (AU) with a LOD of 0.0914 nM and 0.0734 nM, respectively. We have also determined the limit of quantification (LOQ); the LOQ values for the aqueous media were 0.0807 nM (for a linear range of 0-140 µM) and 0.0234 nM (for a linear range of 160-260 µM), whereas, the values for HBS and AU were around 0.2771 nM and 0.2226 nM, respectively. Moreover, the nanocomposite also provides a feasible platform for cytotoxicity screening in cancerous cells (Caco-2 cell lines) and non-cancerous cells (L-929 cell lines).

Background and Aim: Informal prescribers (IPs) significantly contribute to the development of antimicrobial resistance and in disseminating pathogens from poultry to humans and other animals through the food chain, posing a serious global health threat. Therefore, this study aimed to assess whether the knowledge of IPs has an impact on their attitude and practice toward antimicrobial use, antibiotic residues, and antimicrobial resistance. Materials and Methods: In this cross-sectional study, we conducted a pre-tested and questionnaire-based survey to investigate the knowledge, attitude, and practice of IPs in selected parts of the Mymensingh division, Bangladesh. Then, we used the linear regression model test with R-squared (R2) to measure the association between the study variables. Results: Our investigation revealed that 70% of the IPs knew about antibiotics and 75% had good knowledge about antibiotic resistance, whereas only 50% were aware of withdrawal periods. Informal prescribers also displayed good attitudes toward the use and sale of antibiotics with withdrawal periods and completion of medication (50%). Analysis of their practice on the sale and prescription of antibiotics showed that 70% and 30% of IPs use antibiotics against bacterial infections and other conditions, respectively. Most of them do not consult a veterinarian before selling or prescribing antibiotics, although 80% claim to do so. This is because 75% of IPs gave other options regarding their consultations. However, 95% of IPs uses antibiotics only for therapeutic purposes. Furthermore, only 10% sell antibiotics based on a veterinarian's recommendation. Approximately 45% of IPs use single antibiotics at a time, while the rest use multiple antibiotics, individually or combined. Approximately 15% use antibiotics monthly, while 85% use them whenever the need arises. The knowledge and attitude of IPs are significantly affected by their age (p ≤ 0.025). The district of domicile also impacted their knowledge. Surprisingly, IPs from Jamalpur had significantly better knowledge compared to those from Mymensingh and Sherpur (p ≤ 0.01). The attitude of IPs from Jamalpur and Netrokona also differed significantly (p ≤ 0.001) from that of Mymensingh and Sherpur. The knowledge of IPs influenced their attitude up to 80.5% (r2 = 0.628) and their practice up to 75.4% (r2 = 0.545). Conclusion: The knowledge of IPs greatly influenced their attitude and practice, while sociodemographics also influenced their knowledge and attitude toward antimicrobial use, antibiotic residues, and antimicrobial resistance.

BACKGROUND: Medicinal plants or their products are popular substitutes for antibiotics or other synthetic growth promoters for their beneficial effects and cost-effectiveness. Clove (Cv) and tulsi (Ts) are rich in phytochemicals and are used to augment the growth performance of poultry. OBJECTIVES: The current investigation was carried out to evaluate the effects of Cv and Ts supplementation in drinking water on the morphologic and morphometric adaptations in the lymphoid organs. METHODS: The experiment was conducted on 60 broiler chicks, divided into 4 homogenous groups (5 pens/group and 3 broilers/pen) named T0 (control), T1 (0.5% Cv and 2% Ts), T2 (1% Cv and 3% Ts) and T3 (1.5% Cv and 4% Ts). The treatments were administered from days 8 to 28. Lymphoid organs (thymus, spleen and bursa of Fabricius) were collected on days 14, 21 and 28 after sacrificing five broilers from each group. RESULTS: The treatment groups had higher thymus and spleen weights on day 21, whereas the weights of the bursa of Fabricius were higher on days 14, 21 and 28. In the histomorphologic study, no noticeable difference in the histoarchitectures of the lymphoid organs was noticed. However, substantial differences in the cortico-medullary ratio and the number of follicles in the thymus and bursa of Fabricius, respectively, were found in the histomorphometric investigation. CONCLUSION: The inclusion of Cv and Ts in drinking water augments the weight of lymphoid organs with histomorphometric adaptations that might improve the immune status in broilers.

The role of Clove (Cv) and Tulsi (Ts) supplementation on broiler growth performance and gut health as an alternative to antibiotic growth promoters has already been established. Therefore, the objectives of this study were to investigate the role of Cv and Ts on the serum biochemical profile and meat quality traits in broilers. A total of sixty (60) one-day-old commercial broiler chicks were randomly allotted into four homogenous groups (15 birds per group). They were then fed Cv powder and Ts extract from day (d) 8 to d 28 with drinking water i.e. zero level of Cv or Ts (T0), 0.5% Cv + 2% Ts (T1), 1.0% Cv + 3% Ts (T2), 1.5% Cv + 4% Ts (T3). Blood and meat samples were collected on d 14, 21, and 28 to analyze the serum-biochemical profile and meat quality. Supplementation of Cv and Ts improved serum-biochemical profile by reducing total cholesterol and low-density lipoprotein levels in broilers. However, they did not affect the serum protein levels while the 0.5-1% Cv and 2-3% Ts supplemented groups had higher glucose levels on d 21. Production of breast and thigh meat increased with increased size, and density of myofibers while fed 0.5-1% Cv and 2-3% Ts. On the contrary, 1.5% Cv powder and 4% Ts extract supplementation improved thigh meat color, breast and thigh meat pH as well as the water-holding capacity. The current study findings suggest that Cv and Ts can be used in combination to improve broiler health, production, and meat quality.

BACKGROUND: Thymus, bursa of Fabricius and spleen are the major lymphoid organs of avian species that plays a crucial role in their immunity. Though glucocorticoids are reportedly used as growth promoters, they also suppress the immune system. OBJECTIVES: The objective of this study was to investigate the morphologic and morphometric adaptations in the lymphoid organs as well as the mortality rate in broilers in response to long-term treatment with dexamethasone (DEX). METHODS: A total of 80 one-day-old broiler chicks (Cobb 500) were randomly divided into four homogenous groups (control - C and treatment groups - T1, T2, and T3). The treatment groups received DEX at the rate of 3, 5, and 7 mg/kg commercial feed. Samples, i.e. thymus, bursa of Fabricius, and spleen, were collected on 7, 14, 21, and 28 days of the experiment. Relative weight of the organs was calculated on each sampling day. The tissues were then processed and stained with haematoxylin and eosin stain for morphological and morphometric study. RESULTS: The relative weight of lymphoid organs was found substantially (p < 0.05) less in the DEX-treated groups. Significant (p < 0.05) reduction in lobular size and the cortical-medullary ratio was observed in the thymus of the DEX-treated broilers. Follicular atrophy and massive depletion of lymphocytes were evident in the bursa of Fabricius. The mortality rate was also increased which was largely dependent on the dose and duration of DEX treatment. CONCLUSIONS: The study results indicate that DEX treatment can alter the morphology and morphometry of lymphoid organs which might result in severe immunosuppression and increased mortality rate in broilers.

Ovarian cancer represents a major health concern for the female population: there is no obvious cause, it is frequently misdiagnosed, and it is characterized by a poor prognosis. Additionally, patients are inclined to recurrences because of metastasis and poor treatment tolerance. Combining innovative therapeutic techniques with established approaches can aid in improving treatment outcomes. Because of their multi-target actions, long application history, and widespread availability, natural compounds have particular advantages in this connection. Thus, effective therapeutic alternatives with improved patient tolerance hopefully can be identified within the world of natural and nature-derived products. Moreover, natural compounds are generally perceived to have more limited adverse effects on healthy cells or tissues, suggesting their potential role as valid treatment alternatives. In general, the anticancer mechanisms of such molecules are connected to the reduction of cell proliferation and metastasis, autophagy stimulation and improved response to chemotherapeutics. This review aims at discussing the mechanistic insights and possible targets of natural compounds against ovarian cancer, from the perspective of medicinal chemists. In addition, an overview of the pharmacology of natural products studied to date for their potential application towards ovarian cancer models is presented. The chemical aspects as well as available bioactivity data are discussed and commented on, with particular attention to the underlying molecular mechanism(s).

Bangladesh struggles with undernutrition in women and young children. Nutrition-sensitive agriculture programmes can help address rural undernutrition. However, questions remain on the costs of multisectoral programmes. This study estimates the economic costs of the Targeting and Re-aligning Agriculture to Improve Nutrition (TRAIN) programme, which integrated nutrition behaviour change and agricultural extension with a credit platform to support women's income generation. We used the Strengthening Economic Evaluation for Multisectoral Strategies for Nutrition (SEEMS-Nutrition) approach. The approach aligns costs with a multisectoral nutrition typology, identifying inputs and costs along programme impact pathways. We measure and allocate costs for activities and inputs, combining expenditures and micro-costing. Quantitative and qualitative data were collected retrospectively from implementers and beneficiaries. Expenditure data and economic costs were combined to calculate incremental economic costs. The intervention was designed around a randomised control trial. Incremental costs are presented by treatment arm. The total incremental cost was $795,040.34 for a 3.5-year period. The annual incremental costs per household were US$65.37 (Arm 2), USD$114.15 (Arm 3) and $157.11 (Arm 4). Total costs were led by nutrition counselling (37%), agriculture extension (12%), supervision (12%), training (12%), monitoring and evaluation (9%) and community events (5%). Total input costs were led by personnel (68%), travel (12%) and supplies (7%). This study presents the total incremental costs of an agriculture-nutrition intervention implemented through a microcredit platform. Costs per household compare favourably with similar interventions. Our results illustrate the value of a standardised costing approach for comparison with other multisectoral nutrition interventions.

In an epoch of the growing risk of antibiotic resistance, there is a dire need to establish an effective novel feeding practice for broiler nutrition as an alternative to antibiotics. Hence, the aim of the current study was to evaluate the impact of clove powder and tulsi extract on the growth performance, gut morphologic and morphometric indices, and cecal microbial status of broiler, as an alternative to antibiotic growth promoters (AGPs). Sixty day-old chicks of Cobb-500 strain were randomly divided into 4 groups, each having 15 birds. Chicks of the control group (T0) were fed commercial broiler feed with no additional supplementation. The treatment groups were offered commercial broiler feed and received clove powder and tulsi extract with drinking water at the rate of 0.5% + 2% (T1), 1% + 3% (T2), and 1.5% + 4% (T3), respectively. Results showed a nonlinear relationship with the dosage of clove and tulsi. All the growth parameters substantially (P < 0.05) improved in T2 while T1 and T3 showed no significant improvement compared to T0. The final body weight was significantly (P < 0.05) higher in T2. Giblet and offal weights showed no noticeable differences except in the intestine and heart where intestine weight markedly (P < 0.05) decreased in T3 and heart weight significantly (P < 0.05) increased in T1 and T2. Clove and tulsi supplementation substantially improved the villus height and villus surface area of the small intestine in T2 while the large intestine remained mostly unaffected by the treatment. Cecal microbial status significantly improved in all the treatment groups having increased (P < 0.05) Lactobacillus spp. count and decreased (P < 0.05) E. coli count compared to T0. Based on the aforementioned findings, it can be concluded that the combination of clove and tulsi can improve the growth performance and gut health of broilers which is largely dose-dependent and might be supplied as a potential alternative to AGPs.

G-protein-coupled receptors (GPCRs) are activated by manifold neurotransmitters, and their activation, in turn, evokes slow synaptic transmission. They are profoundly related to numerous psychiatric and neurological disorders such as schizophrenia and Parkinson's disease. The significant malady indications for GPCR modulators demonstrate a change towards obesity, diabetes, and Alzheimer's disease, while other central nervous system disorders persist highly represented. GPR52, GPR6, and GPR8 are recognised as orphan GPCRs, co-exist either with both the dopamine D2 and D1 receptors in neurons of the basal ganglia or with the dopamine D2 receptor alone, and recommend that between these orphan receptors, GPR52 has the maximum potential of being a therapeutic psychiatric receptor. Genetically modified creature models and molecular biological investigations have suggested that these improved GPCRs could be potential therapeutic psychiatric receptors. In this perspective, the role of molecular targets in GPCR-mediated signalling has been discussed that would be novel drug design and discovery options for a scientist to elaborate previous knowledge with modern techniques.

Objective: This study sought to determine the effectiveness of the cryodehydration technique in preserving the morphologic and morphometric attributes of the anatomical specimens of goats. Materials and methods: Different anatomical parts of a goat, i.e., heart, lungs, spleen, liver, kidney, and musculoskeletal specimens, were collected and fixed in 15% formalin. Later on, the fixed specimens were cryodehydrated by fast freezing (burning process) and repeated freezing-thawing sessions, followed by wood glue coating. Finally, the macroscopic (i.e., weight, color, texture, odor, and durability) and microscopic characteristics (by routine hematoxylin and eosin staining) of the cryodehydrated specimens were studied. Results: The resultant specimens produced excellent color and texture and were lightweight (60%-80% weight loss), soft, dry, odorless, durable, and easy to handle. The histoarchitectural details of the heart and skeletal muscle were well preserved, while some distinctive alterations were observed in the parenchymatous organs, i.e., breach in cellular integrity, loss of cell cytoplasm, loss of cytoplasmic and nuclear clarity, increased sinusoidal space, dilatation of the renal tubules, and reduction in glomerular size. Nevertheless, the basic histoarchitecture of each specimen was yet to be distinctly identifiable. Conclusion: The current study findings suggest that the cryodehydration technique can preserve gross anatomical features as well as histoarchitectural details and can be an effective tool for facilitating veterinary education and research.

Objective: The study aimed to develop and assess an Android app designed for farmers with a low educational status that can formulate a least-cost ration. Materials and Methods: First, a computer-android-based app named BLRI FeedMaster was developed to guide users in formulating a balanced ration at the least cost. A survey was conducted on 30 livestock officers and 18 farmers with 50 cattle to evaluate its efficacy at the field level. The study outcomes were milk yield, feeding cost, milk composition, time, and cost for management before and after using the BLRI FeedMaster app. Descriptive statistics and paired sample t-tests were used to analyze the data. Results: After adopting the BLRI FeedMaster app, a significant increase was observed in daily average milk yield (9.39 ± 0.32 l from 8.37 ± 0.36 l), while a considerable decrease was observed in daily average feed quantity (4.88 ± 0.15 kg from 5.60 ± 0.17 kg) and feed cost (BDT 28.00 ± 0.50 from BDT 29.75 ± 0.49). Besides, the number of visits, time, and cost for seeking professional services regarding feed, health care, and other information was significantly minimized. The number of visits decreased to 0.36 ± 013 from 3.07 ± 0.38, and the consumed time was reduced from 270 ± 34.30 to 235.71 ± 59.42 min (p < 0.05) after adopting the app. Conclusion: Hence, this app was very beneficial for farmers with a low economic and educational background and may ultimately help farmers with profitable animal farming and sustainable production in the least developed countries like Bangladesh.

Black Bengal goat (BBG) is the most adaptable, widely distributed, and prominent goat breed in Bangladesh, well known in the world for its high prolificacy, low demand of feed, tolerance to harsh weather conditions, and disease resistance with remarkably good quality red meat and skin. A large number of indiscriminate research reports on BBG have been published; however, the review on the productive and reproductive performances with different physiological features of BBG in Bangladesh is scarce. This review was conducted to investigate and summarize the available research reports on BBGs to highlight the gaps and provide coherent recommendations for the future research plan for sustainable BBG production in Bangladesh. It covers research works in morphometric features, feeding and nutrition, reproduction, diseases and health management, husbandry practices, and production performances of BBG under local conditions. Due to the contemporary increased demand for animal protein (meat and milk), the scope of small ruminants, especially goat farming, increases with other large ruminants farming. The key constraints of BBG production in Bangladesh include higher disease prevalence with low or no management practices, kid mortality, inadequate feeds and fodder supply, and poor marketing channel with some other stumpy genetic potentialities (slower body weight gain, low milk production) of this goat. Future research would be required to assess the contribution of BBG to household economies and food securities throughout the year and evaluate the constraints, adaptation and extension of artificial insemination (AI), and genetic improvement of economically important traits using molecular techniques and the selective breeding program.

G protein-coupled receptors (GPCRs) are intricately involved in the conversion of extracellular feedback to intracellular responses. These specialized receptors possess a crucial role in neurological and psychiatric disorders. Most nonsensory GPCRs are active in almost 90% of complex brain functions. At the time of receptor phosphorylation, a GPCR pathway is essentially activated through a G protein signaling mechanism via a G protein-coupled receptor kinase (GRK). Dopamine, an important neurotransmitter, is primarily involved in the pathophysiology of several CNS disorders; for instance, bipolar disorder, schizophrenia, Parkinson's disease, and ADHD. Since dopamine, acetylcholine, and glutamate are potent neuropharmacological targets, dopamine itself has potential therapeutic effects in several CNS disorders. GPCRs essentially regulate brain functions by modulating downstream signaling pathways. GPR6, GPR52, and GPR8 are termed orphan GPCRs because they colocalize with dopamine D1 and D2 receptors in neurons of the basal ganglia, either alone or with both receptors. Among the orphan GPCRs, the GPR52 is recognized for being an effective psychiatric receptor. Various antipsychotics like aripiprazole and quetiapine mainly target GPCRs to exert their actions. One of the most important parts of signal transduction is the regulation of G protein signaling (RGS). These substances inhibit the activation of the G protein that initiates GPCR signaling. Developing a combination of RGS inhibitors with GPCR agonists may prove to have promising therapeutic potential. Indeed, several recent studies have suggested that GPCRs represent potentially valuable therapeutic targets for various psychiatric disorders. Molecular biology and genetically modified animal model studies recommend that these enriched GPCRs may also act as potential therapeutic psychoreceptors. Neurotransmitter and neuropeptide GPCR malfunction in the frontal cortex and limbic-related regions, including the hippocampus, hypothalamus, and brainstem, is likely responsible for the complex clinical picture that includes cognitive, perceptual, emotional, and motor symptoms. G protein and GPCR-mediated signaling play a critical role in developing new treatment options for mental health issues, and this study is aimed at offering a thorough picture of that involvement. For patients who are resistant to current therapies, the development of new drugs that target GPCR signaling cascades remains an interesting possibility. These discoveries might serve as a fresh foundation for the creation of creative methods for pharmacologically useful modulation of GPCR function.

This study explores the impact of dexamethasone (DEX) on the serum glucose profile, morphological attributes of the liver, and the expression of hepatic glucocorticoid receptors (GRs) in the broiler. To conduct this study, four homogenous groups of day-old chicks (DOCs) were used (n = 20 chicks per group); one control group (C), and three treatment groups (T1, T2, and T3). All the groups were maintained on a commercial feed with which 3, 5, and 7 mg DEX (per kg feed) were supplied to the T1, T2, and T3 groups, respectively. Samples were collected on days 7, 14, 21, and 28 of the experiment. The serum glucose profile was measured by spectrophotometry. The livers' morphometric attributes were recorded before being processed and stained with hematoxylin and eosin. The expression pattern of hepatic GR proteins was evaluated by immunohistochemical staining. DEX residue was detected in liver tissue using thin-layer chromatography. Increased serum glucose level was observed in the DEX groups. Fatty liver, hepatic congestion, and decreased morphometric attributes were the critical findings in the DEX groups. Congestion of the central veins, sinusoids, and accumulation of lipid droplets was also observed in the DEX groups. GR proteins were mostly localized in the central vein and cytoplasm of the hepatocytes, the expression of which was found to be upregulated with the increased dose of DEX. The residue of DEX was detected in the liver tissues in the higher dose groups. The findings imply that DEX can substantially alter the blood glucose profile and liver morphology.

Growth promoters are added with broiler feed to boost the overall feed efficiency and growth rate. The current study investigated the effect of dexamethasone (DEX)-a commonly used growth promoter-on the broiler growth rate, meat quality, and muscle biology. Four homogenous groups (20 chicks/group) of broiler one-day-old chicks were fed commercial broiler feed where the treatment groups received 3, 5, and 7 mg/kg of DEX with their diet for 28 d. Feed consumption and body weight were monitored on a daily basis. Muscle samples were collected on 7, 14, 21, and 28 d of the experiment to investigate meat quality and muscular biology. The residue of DEX in meat was detected using thin-layer chromatography. We observed that DEX had substantially decreased (P < 0.05) feed intake, feed efficiency, and overall weight gain in the broiler. While the weight of breast and thigh meat was decreased, the relative meat weight (meat/body weight) was increased significantly in chicks fed DEX. Simultaneously, body fat decreased while the percentage of fat increased significantly (P < 0.05) in the DEX groups. Contrariwise, DEX improved the investigated meat quality parameters with the potential threat of accumulation of DEX residue in the meat at a high dose (7 mg/kg). We also observed that DEX significantly increased the number of myofibers and decreased the cross-sectional area of myofibers. Based on these findings, we conclude that DEX reduces feed intake, feed efficiency, and growth rate, but might improve meat quality with a potential risk of residual DEX accumulation if fed at a high dose.
