RESUMO

Lanthanide metal-organic frameworks (Ln-MOFs) broaden the optical sensing applications of lanthanide ions due to the antenna effect between organic ligands and metals. However, the sensitization ability of the ligand to metal ions is limited, and maximizing the sensitization of the electrochemiluminescence behavior of Eu3+ is still a challenge for the application of Ln-MOFs. Therefore, under the guidance of the "cascade sensitization mechanism" based on the antenna effect sensitizing the electrochemiluminescence of bimetallic Ln-MOFs, we proposed Eu/Tb-MOFs with high luminescence intensity as a signal probe. According to the antenna effect, the conjugated structure and high extinction coefficient of the benzene ring of 2-amino terephthalic acid (NH2-BDC) can enhance the ECL luminescence intensity of Eu/Tb-MOFs. Tb3+ can act as an energy bridge between NH2-BDC and Eu3+, buffering the energy gap. The bimetallic sensitization is formed between Tb3+ and Eu3+, which can inhibit the reverse internal flow of energy and ensure the high luminous efficiency of Eu3+. In addition, the nanosphere mixed valence Fe3O4 as a co-reactant accelerator promotes the formation of transient free radical SO4•- through the valence change of Fe2+/Fe3+. The ECL immunosensor constructed by luminophores Eu/Tb-MOFs and nanosphere Fe3O4 provided a new explanation for the ECL self-luminous of Eu/Tb-MOFs.

Polystyrene microplastics (PM) is a pressing global environmental concern, posing substantial risks to aquatic ecosystems. Microalgal astaxanthin (MA), a heme pigment, safeguards cells against oxidative damage induced by free radicals, which contributes to various health conditions, including aging, inflammation and chronic diseases. Herein, we investigated the potential of MA in ameliorating the immunotoxicity of PM on carp (Cyprinus carpio L.) based on head kidney lymphocytes treated with PM (250 µM) and/or MA (100 µM). Firstly, CCK8 results showed that PM resulted in excessive death of head kidney lymphocytes. Secondly, head kidney lymphocytes treated with PM had a higher proportion of necroptosis, and the levels of necroptosis-related genes in head kidney lymphocytes were increased. Thirdly, the relative red fluorescence intensity of JC-1 and MitoSox showed decreased mitochondrial membrane potential and increased mtROS in head kidney lymphocytes treated with PM. MitoTracker® Green FM fluorescence analysis revealed enhanced mitochondrial Ca2+ levels in PM-treated lymphocytes, corroborating the association between PM exposure and elevated intracellular Ca2+ dynamics. PM exposure resulted in upregulation of calcium homeostasis-related gene (Orail, CAMKIIδ and SLC8A1) in lymphocytes. Subsequent investigations revealed that PM exposure reduced miR-25-5p expression while increasing levels of MCU, MICU1, and MCUR1. Notably, these effects were counteracted by treatment with MA. Furthermore, PM led to the elevated secretion of inflammatory factors (IFN-γ, IL-1ß, IL-2 and TNF-α), thereby inducing immune dysfunction in head kidney lymphocytes. Encouragingly, MA treatment effectively mitigated the immunotoxic effects induced by PM, demonstrating its potential in ameliorating necroptosis, mitochondrial dysfunction and immune impairment via regulating the miR-25-5p/MCU axis in lymphocytes. This study sheds light on safeguarding farmed fish against agrobiological threats posed by PM, highlighting the valuable applications of MA in aquaculture.

The highly water-soluble nematicide fosthiazate is anticipated to undergo microencapsulation in order to enhance its retention around plant roots and mitigate leaching into groundwater. However, the underlying mechanism governing the influence of hydrophilicity of the microcapsule (MC) core on the evolution of the microcapsule shell remains unclear, posing challenges for encapsulating water-soluble core materials. This study elucidates the microlevel formation mechanism of microcapsules by investigating the impact of interfacial mass transfer on shell formation and proposes a method for regulating the structure of shells. The study reveals that enhancing the hydrophilicity of the core enhances the shuttle effect between the oil and aqueous phase, expands the region of polymerization reactions, and forms a loose and thick shell. The thickness of the microcapsule shell prepared using solvent oil 150# (MCs-SOL) measures only 264 nm, while that of the microcapsules prepared using propylene glycol diacetate and solvent oil 150# at a ratio of 2:1 (MCs-P2S1) is 5.2 times greater. The enhanced compactness of the shell reduced the release rate of microcapsules and the leaching distance of fosthiazate in soil, thereby mitigating the risk of leaching loss and facilitating the distribution of active ingredients within crop roots. The MCs-SOL had a limited leaching distance measurement of 8 cm and exhibited a satisfactory efficacy of 87.3% in controlling root galling nematodes. The thickness and compactness of the MCs shell can be regulated by manipulating the interfacial shuttle effect, providing a promising approach to enhancing utilization efficiency while mitigating potential environmental risks.

Currently, there is a pressing need to develop an agrochemical-loaded system that is both uncomplicated and efficient, thereby enhancing the adhesion of agrochemical to leaf surfaces and optimizing their insecticidal efficacy, while concurrently mitigating environmental risks. The flexible eugenol-loaded particles were synthesized via a one-step polyurethane self-assembly reaction, utilizing polyethylene glycol (PEG) as the soft segment and 4,4-diphenylmethane diisocyanate (MDI) as the hard segment. The increase in the length of the soft segment enhances the flexibility of the particles, thereby improving the contact area and adhesion with the foliar surface. When flexible particles are applied on the foliar surface, they can achieve satisfactory resistance to rainfall erosion. When the PEG molecular weight is 800, the residual concentration of eugenol can still reach 42.11% after 6 washes. The carrier protects the active ingredients and improves the resistance to ultraviolet irradiation. After 5 h of ultraviolet irradiation, the concentration of eugenol remained at 59.03% when PEG with a molecular weight of 200 was employed. Greenhouse experiments showed that the flexible transformation of particles greatly enhanced the application effect of spray on the foliar surface of particles. After undergoing three washes, the mortality of the particles can be enhanced by 5.4-8.4 times compared to that of emulsion concentrate (EC) sample. The enhancement of leaf retention performance reduces environmental risks caused by pesticide loss. Meanwhile, the controlled release of particles also reduces the acute toxicity to zebrafish. The toxicity selection pressure of the EUG@P800-Ps sample is 10.6 times that of the EC sample. In conclusion, the preparation process of the system is simple, and the flexible transformation is an effective strategy to improve the foliar application effect of spray and improve the environmental safety.

Detection of arterial stiffness is an important method to predict the occurrence of hypertension complications and to screen patients with high cardiovascular risk. In order to predict the damage of AASI to the renal function of patients with essential hypertension, the prediction of AASI based on stepwise Regression equation scanning for renal function damage in patients with essential hypertension is proposed. Measure the 24 h ambulatory blood pressure of the selected subjects, establish a linear Regression equation scanning, and calculate the slope of the straight line, and finally, the slope is AASI. According to the quartiles, AASI is divided into four parts: group I < 0.53 (n = 49); 0.53 ≤ group II < 0.60 (n = 51); 0.60 ≤ group III < 0.69 (n = 48); group IV ≥ 0.69 (n = 44). Experiment result shows the following: with the increase of AASI, cystatin (CysC) also increased significantly, while CysC-eGFR decreased significantly (P < 0.05). Compared with groups I, II, and III, Scr and CysC in group IV increased (P < 0.05), and Ccr, CysC-eGFR, and (CKD-EPI)-eGFR all decreased (P < 0.05). AASI is positively correlated with CysC performance, and the correlation coefficient r is 0.637. It is negatively correlated with Ccr performance, and r is -0.361. It is negatively correlated with CysC-eGFR, and r is -0.698. And it is negatively correlated with (CKD-EPI)-eGFR, and r is -0.331. Age and 24 h PP also showed an increasing trend with the increase of AASI, and it suggests that age may be an influencing factor that promotes kidney damage caused by hypertension; it also suggests that AASI can be used as a new indicator of arterial compliance; AASI is linearly related to various indicators of renal damage and can be used as a predictive indicator of renal damage caused by essential hypertension; cystatin C and the estimated glomerular filtration rate CysC-eGFR based on cystatin C are better than other indicators reflecting glomerular filtration rate, more sensitively assess the degree of early renal damage. Obesity may also be a factor that promotes kidney damage caused by hypertension.

Echinoderms exhibit several fascinating evolutionary innovations that are rarely seen in the animal kingdom, but how these animals attained such features is not well understood. Here we report the sequencing and analysis of the genome and extensive transcriptomes of the sea cucumber Apostichopus japonicus, a species from a special echinoderm group with extraordinary potential for saponin synthesis, aestivation and organ regeneration. The sea cucumber does not possess a reorganized Hox cluster as previously assumed for all echinoderms, and the spatial expression of Hox7 and Hox11/13b potentially guides the embryo-to-larva axial transformation. Contrary to the typical production of lanosterol in animal cholesterol synthesis, the oxidosqualene cyclase of sea cucumber produces parkeol for saponin synthesis and has "plant-like" motifs suggestive of convergent evolution. The transcriptional factors Klf2 and Egr1 are identified as key regulators of aestivation, probably exerting their effects through a clock gene-controlled process. Intestinal hypometabolism during aestivation is driven by the DNA hypermethylation of various metabolic gene pathways, whereas the transcriptional network of intestine regeneration involves diverse signaling pathways, including Wnt, Hippo and FGF. Decoding the sea cucumber genome provides a new avenue for an in-depth understanding of the extraordinary features of sea cucumbers and other echinoderms.

In order to understand the effect of the non-coal heating and the traditional coal-fired heating on the indoor environment of the rural houses, the humidity environment and indoor air quality in several households were investigated during the heating period in Beichen District and Wuqing District of Tianjin, China. The results showed that the indoor average temperature for the heating by the electricity and the natural gas was higher than that by the traditional coal fire. The indoor relative humidity for the heating by the electricity and the natural gas was lower than that by the traditional coal fire. The indoor air quality (IAQ) for the heating by the electricity and the natural gas was better than that by the traditional coal fire. For traditional coal-fire heating households, the indoor pollutant emission (CO emission) by using the clean coal was lower than that by using the raw coal. The indoor ventilation rate which was an important parameter for the indoor air quality was generally poor in winter. The total volatile organic compounds (TVOC) emission in the indoors of the coal-fired heating households was generally higher than that of the non-coaled heating households.

Isolength restriction site-associated DNA (isoRAD) sequencing is a very simple but powerful approach that was originally developed for genome-wide genotyping at minimal labor and cost, and it has recently extended its applicability to allow quantification of DNA methylation levels. The isoRAD method is distinct from other genotyping-by-sequencing (GBS) methods because of its use of special restriction enzymes to produce isolength tags (32-36 bp), and sequencing of these uniform tags can bring many benefits. However, the relatively short tags produced by the original protocol are mostly suited to single-end (SE) sequencing (36-50 bp), and therefore they cannot efficiently match the gradually increased sequencing capacity of next-generation sequencing (NGS) platforms. To address this issue, we describe an advanced protocol that allows the preparation of five concatenated isoRAD tags for Illumina paired-end (PE) sequencing (100-150 bp). The configuration of the five concatenated tags is highly flexible, and can be defined by users to work with a desired combination of samples and/or restriction enzymes to suit specific research purposes. In comparison with the original protocol, the advanced protocol has an additional digestion and ligation step, and library preparation can be completed in ∼8 h.

BACKGROUND: The most common treatment for old calcaneal fractures accompanied by subtalar joint injury is the use of subtalar in situ arthrodesis and subtalar distraction bone-block arthrodesis or osteotomy. This article describes the introduction of a novel surgical treatment, gradual subtalar distraction with external fixation and restoration of the calcaneal height, and presents an assessment of its efficacy. METHODS: The protruding lateral calcaneus and the articular surfaces and subchondral bone of the posterior facet of the subtalar joint were surgically removed. An external fixator, attached with 2 pins in the subcutaneous tibia and 2 pins in the posterolateral calcaneus, was used to fix the subtalar joint for 7 to 10 days followed by gradual subtalar distraction at 1 mm/d. The lengthening procedure was stopped when the calcaneal height was restored according to radiography. The external fixator was removed after bone fusion. Seven cases of old calcaneal fractures accompanied by severe subtalar joint injury (8 feet) were treated using this method. Average follow-up was 14.3 months (range, 7-36 months). RESULTS: In all 7 cases (1 case of both feet), the postoperative wound healed primarily. The calcaneal heights of all 8 feet were partially restored. Subtalar joint bone fusion was completed within 4 to 6 months after the operation. The average preoperative American Orthopedic Foot & Ankle Society (AOFAS) hindfoot score was 25.3, and the average postoperative AOFAS score was 76.3. CONCLUSION: Subtalar distraction osteogenesis with external fixation was a novel and effective method for the treatment of old calcaneal fractures accompanied by severe subtalar joint injury in this small group of patients. OBJECTIVE: To compare the outcome of damage control in patients with severe polytrauma in normal time and in the earthquake of 2008 Sichuan China. METHODS: In the retrospective study, 26 cases with severity polytrauma in peacetime and 24 cases with severe polytrauma who have mainly sustained in the earthquake of 2008 Sichuan China were involovd. The patients' demographics, the methods and effects of damage control were evaluated. RESULTS: In the earthquake group, 24 cases succeeded in the rescuing life; among whom, 3 patients needed the further debridement and dermoplasty. In the normal time group, 26 cases succeeded in the rescuing life; among whom, 19 patients had returned to their former work and 3 patients needed the further treatment. There was no difference between two groups in therapeutic effects (P > 0.05). CONCLUSION: Both in earthquake and in normal time, treating the severity polytrauma by damage control orthopedics can acquire satisfied effect.

OBJECTIVE: To discuss damage control orthopaedics in 53 cases of severe polytrauma who have mainly sustained orthopaedic trauma. METHODS: The data of 53 cases of severe polytrauma who had mainly sustained orthopaedic trauma were retrospectively analyzed. And the methods and timing of damage control orthopaedics were discussed in this study. RESULTS: We succeeded in rescuing the lives of all the 53 patients, and 38 patients returned to their former work. CONCLUSIONS: Injury Severity Score (ISS(90)) should be 17 in severe polytrauma patients, but in severe polytrauma patients who have mainly sustained orthopaedic trauma, the ISS(90) of bone and joint injuries should be 16. We recommend that primary minimally-invasive external fracture stabilization should be made for extremities and pelvis in these patients to avoid additional surgical trauma and that definitive secondary fracture care should be performed after medical stabilization for these patients in intensive care unit (ICU).

OBJECTIVE: To evaluate the effect of testis homotransplantation in the treatment of androgen deficiency and infertility. METHODS: We retrospectively analyzed 12 cases of testis homotransplantation. RESULTS: Surgical success was achieved in 11 cases, all with a significantly increased level of serum testosterone, and markedly improved secondary sex characteristics and sexual function. CONCLUSION: Testis homotransplantation is highly effective for the treatment of androgen deficiency in males, but has little effect on spermatogenesis.

OBJECTIVE: To evaluate the application of injury severity score ( ISS) to multiple injuries headed by spinal cord injury. METHODS: The data of 55 cases (40 males and 15 females, aged 17-69 years, mean equal to 41 years) of multiple injuries headed by spinal cord injury treated in our hospital from January 2000 to December 2004 were reviewed and analyzed with ISS (Version of AIS-2005) to explore their relationship. RESULTS: The ISS values increased with the number of injured regions, so did the complications. The recovery rate was negatively related to ISS values. During the period of immunity observation, the patients with ISS larger than 25 could undergo planned operations safely. CONCLUSIONS: Treatment for multiple injuries headed by spinal cord injury closely depends on the general and local conditions of the patients. ISS may provide useful data for the choice of treatment methods.

OBJECTIVE: To explore the operative indications and operative methods of primary total hip arthroplasty for acetabular fracture and to observe the clinical curative effect. METHODS: We retrospectively summarized and analyzed the traumatic conditions, fracture types, complications, operative time, operative techniques, and short term curative effect of 11 patients (10 males and 1 female, with a mean age of 42.4 years) with acetabular fracture who underwent primary total hip arthroplasty. RESULTS: The patients were followed up for 6-45 months (mean=28 months). Their average Harris score of postoperative hip joint was 78. CONCLUSION: Under strict mastery of indications, patients with acetabular fracture may undergo primary total hip arthroplasty, but stable acetabular components should be made.

OBJECTIVE: To analyze retrospectively the surgical treatment of 21 cases of infected arterial injuries of the major limb hospitalized from 1989 to 2003 in our department. METHODS: After a radical debridement and drainage, an autologous vessel bypass reconstruction, simple vessel ligation or amputation were carried out respectively according to each patient's condition. In order to make a better milieu for the grafts, local musculo-cutaneous flaps were translocated to cover the wounds in 9 cases. RESULTS: 1 case (4.8%) died during the treatment. 5 cases (5/21, 23.8%) underwent amputations. Blood supply reconstruction was successful in 15 cases (15/21, 71.4%). 93.3% (14/15) of the cases were successful in getting an unobstructed vessel at early stage. Thrombosis occurred in 1 case (1/15, 6.7%) and a sound result was obtained after a second operation. CONCLUSIONS: Radical debridement combined with bypass vessel reconstruction with autologous vein and with musculo-cutaneous flap translocation is an effective treatment for infective limb vascular injuries.

OBJECTIVE: To explore an early stage repair method for soft tissue defect of limbs of modern firearm wound, and to improve treating result. METHODS: Defects of the hind limbs of dogs were repaired with skin, muscle and myocutaneous flaps. RESULTS: Wounds healed within 2 weeks in the experimental group except one that healed in 3 weeks because of infection. Limb function was close to normal. The treatment result was better in the experimental group than the control. CONCLUSIONS: Skin, muscle and myocutaneous flaps can cover soft tissue defect at an early stage, prevent and reduce infection, promote the healing and recovery of combined injury, reduce the time of treatment and disability rate.
