Designing Pt-skin on the catalyst surface is critical to developing efficient and stable electrocatalysts toward oxygen reduction reaction (ORR) in proton exchange membrane fuel cells. In this paper, an acidic reductant is proposed to synchronously manipulate in-situ growth of Pt-skin on the surface of alloyed Pt-Cu nanospheres (PtCuNSs) by a facile one-pot synthesis in an aqueous solution. Ascorbic acid can create a Pt-skin of three atomic layers to make the typical PtCu-alloy@Pt-skin core/shell nanostructure rather than the uniform alloys generated by using alkaline reductants. Surfactant as soft-template can make the alloyed PtCuNSs with a three-dimensional porous network structure. Multiple characterizations of XRD, XPS and XAFS are used to confirm Pt-alloying with Cu and formation of core/shell structure of such a catalyst. This PtCuNSs/C exhibits a half-wave potential of 0.913 V (vs. RHE), with mass activity and specific activity about 3.5 and 6.4 times higher than those of Pt/C, respectively. Fuel cell tests verify the excellent activity of PtCuNSs/C catalyst with a maximum power density of about 1.2 W cm-2. Moreover, this catalyst shows excellent stability, achieving a long-term operation of 40,000 cycles. Furthermore, theoretical calculations reveal the enhancement effect of characteristic PtCu-alloy@Pt-skin nanostructure on both catalytic ORR activity and stability.

Clinical updates suggest conserving metastatic sentinel lymph nodes (SLNs) of breast cancer (BC) patients during surgery; however, the immunoadjuvant potential of this strategy is unknown. Here we leverage an immune-fueling flex-patch to animate metastatic SLNs with personalized antitumor immunity. The flex-patch is implanted on the postoperative wound and spatiotemporally releases immunotherapeutic anti-PD-1 antibodies (aPD-1) and adjuvants (magnesium iron-layered double hydroxide, LDH) into the SLN. Genes associated with citric acid cycle and oxidative phosphorylation are enriched in activated CD8+ T cells (CTLs) from metastatic SLNs. Delivered aPD-1 and LDH confer CTLs with upregulated glycolytic activity, promoting CTL activation and cytotoxic killing via metal cation-mediated shaping. Ultimately, CTLs in patch-driven metastatic SLNs could long-termly maintain tumor antigen-specific memory, protecting against high-incidence BC recurrence in female mice. This study indicates a clinical value of metastatic SLN in immunoadjuvant therapy.

Organ morphogenesis needs orchestration of a series of cellular events, including cell division, cell shape change, cell rearrangement and cell death. Cytokinesis, the final step of cell division, is involved in the control of organ size, shape and function. Mechanistically, it is unclear how the molecules involved in cytokinesis regulate organ size and shape. Here, we demonstrate that the centralspindlin complex coordinates cell division and epithelial morphogenesis by regulating cytokinesis. Loss of the centralspindlin components CYK-4 and ZEN-4 disrupts cell division, resulting in altered cell arrangement and malformation of the Caenorhabditis elegans spermatheca. Further investigation revealed that most spermathecal cells undergo nuclear division without completion of cytokinesis. Germline mutant-based analyses suggest that CYK-4 regulates cytokinesis of spermathecal cells in a GTPase activator activity-independent manner. Spermathecal morphology defects can be enhanced by double knockdown of rho-1 and cyk-4, and partially suppressed by double knockdown of cdc-42 and cyk-4. Thus, the centralspindlin components CYK-4 and ZEN-4, together with RHO-1 and CDC-42, are central players of a signaling network that guides spermathecal morphogenesis by enabling completion of cytokinesis.

Using in vivo multiphoton fluorescent dosimetry, we demonstrate that the clearance dynamics of Indocyanine Green (ICG) in the blood can quickly reveal liver function reserve. In normal rats, the ICG retention rate was below 10% at the 15-minute post-administration; While in the rat with severe hepatocellular carcinoma (HCC), the 15-minute retention rate is over 40% due to poor liver metabolism. With a 785 nm CW laser, the fluorescence dosimeter can evaluate the liver function reserve at a 1/10 clinical dosage of ICG without any blood sampling. In the future, this low-dosage ICG 15-minute retention dosimetry can be applied for the preoperative assessment of hepatectomy or timely perioperative examination.

Developing a facile and eco-friendly method for the large-scale synthesis of highly active and stable catalysts toward oxygen reduction reaction (ORR) is very important for the practical application of proton exchange membrane fuel cells (PEMFCs). In this paper, a mild aqueous-solution route has been successfully developed for the gram-scale synthesis of three-dimensional porous Pt nanospheres (Pt-NSs) that are composed of network-structured nanodendrites and/or oval multipods. In comparison with the commercial Pt/C catalyst, X-ray photoelectron spectroscopy (XPS) demonstrates the dominant metallic-state of Pt and electrochemical impedance spectroscopy (EIS) indicates the substantial improvement of conductivity for the Pt-NSs/C catalyst. The surfactant-induced porous network nanostructure improves both the catalytic ORR activity and durability. The optimal Pt-NSs/C catalyst exhibits a half-wave potential of 0.898 V (vs. RHE), leading to the mass activity of 0.18 A mgPt -1 and specific activity of 0.68 mA cm-2 which are respectively 1.9 and 5.7 times greater than those of Pt/C. Moreover, the highly-active Pt-NSs/C catalyst shows a superior stability with the tenable morphology and the retained 78% of initial mass activity rather than the severe Pt aggregation and the only 58% retention of the commercial Pt/C catalyst after 10000 cycles.

Spiders are important natural enemies of rice pests. Studying the effects of herbivore-induced rice volatiles on spider attraction and predation ability may lead to safer methods for pest prevention and control. In this study, four-arm olfactometer, predation ability experiment, and field trapping experiment were used to evaluate the effects of herbivore-induced rice volatiles on Pirata subpiraticus Bösenberg et Strand (Araneae: Lycosidae) and Pardosa pseudoannulata Bösenberg et Strand (Araneae: Lycosidae). The 0.5 µg/µL linalool concentration was attractive, and also shortened the predation latency in male P. subpiraticus and female P. pseudoannulata. The 0.5 µg/µL linalool concentration increased the daily predation capacity of female P. pseudoannulata. Male P. pseudoannulata were attracted to 1.0 g/L methyl salicylate, which also shortened their predation latency. In field experiments, methyl salicylate and linalool were effective for trapping spiders. Herbivore-induced rice volatiles attract rice field spiders and affect their predatory ability. These results suggest that herbivore-induced rice volatiles can be used to attract spiders and provide improved control of rice pests.

Pluripotent stem cells (PSCs) can be maintained in a continuum of cellular states with distinct features. Exogenous lipid supplements can relieve the dependence on de novo lipogenesis and shift global metabolism. However, it is largely unexplored how specific lipid components regulate metabolism and subsequently the pluripotency state. In this study, we report that the metabolic landscape of human PSCs (hPSCs) is shifted by signaling lipid lysophosphatidic acid (LPA), which naturally exists. LPA leads to a distinctive transcriptome profile that is not associated with de novo lipogenesis. Although exogenous lipids such as cholesterol, common free fatty acids, and LPA can affect cellular metabolism, they are not necessary for maintaining primed pluripotency. Instead, LPA induces distinct and reversible phenotypes in cell cycle, morphology, and mitochondria. This study reveals a distinct primed state that could be used to alter cell physiology in hPSCs for basic research and stem cell applications.

Highly sensitive and selectivite detection of copper ions (Cu2+) and hydrogen sulfide (H2S) have become important research topics due to the potential harmful impacts of these chemicals to human health and the environment. In this study, we report the synthesis of a dual-functional peptide-based probe L (FITC-AhxSerSerHis), designed to mimic a copper-sulfur metalloprotein, and capable of continuous detection of Cu2+ and S2- based on colorimetric and fluorescent methods. The new probe L displayed excellent "turn off" fluorescence response and good selectivity for Cu2+ ions via a modification of the tripeptide and fluorescein isothiocyanate group, and produced an obvious color change visible to the naked eye. Furthermore, as an excitable probe, the L-Cu complex could continuously detect S2- with high selectivity and sensitivity in 100% aqueous buffered solutions. The detection limits for fluorescence titration measurements, calculated using the equation 3σ/k, were 76.7 nM (Cu2+) and 27.2 nM (S2-), which were well below U.S. EPA safety levels. In addition, L could be cycled to alternately detect Cu2+ and S2-, thereby making it a promising reversible probe. Moreover, L was successfully applied to monitoring Cu2+ and S2- in live RKO cells through fluorescence imaging, exhibiting low cytotoxicity and good cell permeability.

BACKGROUND: NAD kinases (NADKs) are the only known enzymes that directly phosphorylate NAD(H) to generate NADP(H) in different subcellular compartments. They participate in multiple life activities, such as modulating the NADP/NAD ratio, maintaining the intracellular redox balance and responding to environmental stresses. However, the functions of individual NADK in plants are still under investigation. Here, a rice NADK, namely, OsNADK1, was identified, and its functions in plant growth regulation and stress tolerance were analysed by employing a series of transgenic plant lines. RESULTS: OsNADK1 is a cytosol-localized NADK in rice. It was expressed in all rice tissues examined, and its transcriptional expression could be stimulated by a number of environmental stress treatments. Compared with wild-type (WT) rice, the mutant plant osnadk1 in which OsNADK1 was knocked out was a dwarf at the heading stage and had decreased NADP(H)/NAD(H), ascorbic acid (ASA)/dehydroascorbate (DHA) and reduced glutathione (GSH)/oxidized glutathione (GSSG) ratios, which led to increased oxidation states in the rice cells and sensitivity to drought. Moreover, certain stress-related genes showed differential expression patterns in osnadk1 under both normal growth and drought-stress conditions compared with WT. Among these genes, OsDREB1B and several WRKY family transcription factors, e.g., OsWRKY21 and OsWRKY42, showed correlated co-expression patterns with OsNADK1 in osnadk1 and the plants overexpressing or underexpressing OsNADK1, implying roles for these transcription factors in OsNADK1-mediated processes. In addition, overexpression of OsNADK1 enhanced the drought tolerance of rice plants, whereas loss of function of the gene reduced the tolerance. Furthermore, the proline content was dramatically increased in the leaves of the OsNADK1-overexpressing lines under drought conditions. CONCLUSIONS: Altogether, the results suggest that an OsNADK1-mediated intracellular redox balance is involved in the tolerance of rice plants to drought.
