Pectin was extracted from red dragon fruit (Hylocereus polyrhizus) peel using two different extraction methods: subcritical water extraction (SCWE) and conventional acid extraction (AE), from two different types of peels, fresh peel puree and dried peel powder. SCWE method on fresh peel puree showed an ∼18.88 % increase in pectin yield compared to AE. Extracted pectin is classified as low methoxyl pectin (DE: 8.51-50.64 %), with an average molecular weight ranging from 115.23 kDa to 577.84 kDa and a Gal-A content of 44.09 % - 53.90 %. The potential of pectin from fresh peel puree to be applied as a biodegradable film was further explored. Different pectin concentrations (3-5 % w/v) were used to prepare the films. Regarding the film performance, PF-S5, which was produced from SCWE with 5 % of pectin concentration, exhibits better thermal stability (Tdmax 250 °C, residue of 28.69 %) and higher moisture barrier (WVP 5.59 × 10-11 g.cm-1.s-1.Pa-1). In comparison, PF-A showed lower water solubility (45.14-69.15 %), higher water contact angle (33.01° - 44.35°), and better mechanical properties (TS: 2.12-4.11 MPa, EB: 48.72-61.39 %). Higher molecular weight accompanied by higher DE and Gal-A content contributes to better pectin film properties.

Imidacloprid (IMP) is toxic and a potential carcinogen that is most widely used as an insecticide for pest control and seed treatment. It is important to produce a rapid and sensitive assay for on-site monitoring. We have developed a novel lateral flow assay (LFA) using a sensitive monoclonal antibody (mAb) for monitoring IMP residues on fruits and vegetables. The 50% inhibition concentration result that was found when using the ELISA method was 0.247 ng mL-1, with the cut-off limits using the LFA method the result was 10 ng mL-1 (0.01 M PBS), and in the samples it was 20 ng mL-1 (with a recovery rate of 96-104.7% for Chinese cabbage, cowpea, apple, and pear samples, respectively). All of the results can be determined within seven minutes. The proposed LFA method is a valid, quick, and stable assay for the on-site detection of IMP in large numbers of samples.

Hyoscyamine (HSM), which acts as an antagonist of the acetylcholine muscarinic receptor and can induce a variety of distinct toxic syndromes in mammals (anti-cholinergic poisoning), is hazardous to human health. Therefore, it is urgent to develop a rapid, sensitive, and cost-effective method to determine HSM. A fluorescent microsphere based immunochromatographic assay was developed for this analyte and gold nanoparticles (AuNPs) were used as a comparison. A monoclonal antibody against HSM was prepared with a 50% inhibition concentration (IC50) of 1.17 ng mL-1, with no cross-reactivity with five drugs. Under optimized conditions, the cut off limits using the fluorescence-labeled monoclonal antibody strips were 10 ng mL-1 in 0.01 M PBS and 20 ng mL-1 in pork, pig urine, and honey samples, and the assay could be completed within 10 min. In comparison with a AuNP immunochromatographic assay, the developed method offered a higher coupling rate and lower amounts of antibodies. This approach could be used for simple, sensitive and rapid screening, and is suitable for on-site screening applications.

As the use of non-steroidal anti-inflammatory drugs (NSAIDS) increases, their side effects have also attracted attention. Ibuprofen is one of the most widely-used NSAIDs. In this study, we screened the highly-sensitive and specific antibody 6E10, with an IC50 of 1.92 ng mL-1, and a linear range of 0.53-6.97 ng mL-1. In this study, we developed a rapid lateral flow immunochromatographic assay (ICA) strip method to detect ibuprofen in water or herbal tea. The cut-off limit of the strip is 10 ng mL-1 in water, and concentrations as low as 1 ng mL-1 can be detected in herbal tea samples, with the results obtained by the naked eye within 6 min. All the data were confirmed by high performance liquid chromatography-quadrupole time of flight-mass spectrometry (HPLC-QTOF-MS). This lateral-flow ICA strip is thus a rapid tool for on-site detection and screening of ibuprofen in water and herbal tea.

Because of increasing levels of obesity and the development of social network platforms, the online sale of slimming beverages has become very common. However, such beverages are often reported to contain hazardous drugs, such as phenolphthalein. In the present study, we established a rapid and sensitive immunochromatography gold-labeled monoclonal antibody (mAb) (IGM) method and a fluorescence-labeled mAb (IFM) method for determining phenolphthalein in ten types of slimming tea. The monoclonal antibodies and coating antigens were produced in our laboratory. Under optimal conditions, the cut-off limits were 250 ng mL-1 (IFM strip) and 500 ng mL-1 (IGM strip) in both 0.01 M phosphate-buffered saline (PBS) and samples of slimming tea. The mean recoveries were 96.2 to 104.7% for the IGM strip and 90.7 to 104.7% for the IFM strip. The data showed that the IFM strip was more sensitive than the IGM strip and that results could be generated within 10 min. Consequently, this novel technique represents a rapid and convenient method with which to detect phenolphthalein.

Tadalafil (TDL) is an illegal additive drug found in drinks and functional foods that could threaten public health. There was a great concern whether the adulteration occurred in coffee added with similar type of herbs. Here we have developed a rapid, simple, sensitive, and semi-quantitative lateral flow immunoassay (LFIA) based on gold and fluorescence labelled monoclonal antibody (mAb) for detection of TDL in coffee sample. Under optimal conditions, the cut off limits using gold nanoparticles labelled mAb (GLM) was found to be 250 ng/mL and 100 ng mL using fluorescent labelled mAb (FLM) in coffee samples. The coffee samples were spiked with TDL, and the LFIA with GLM gave average recoveries of 92-105.3% (intra-assay) and 96.6-105.9% (inter-assay), meanwhile with FLM gave recoveries 97.9-107.3% (intra-assay) and 98.3-108.9% (inter-assay). Results gave LFIA with FLM more sensitive than with GLM and all the test can be completed within 10 min, which would be an option for convenient and rapid assay of TDL detection.

A semi-quantitative and quantitative multi-immunochromatographic (ICA) strip detection assay was developed for the simultaneous detection of twenty types of mycotoxins from five classes, including zearalenones (ZEAs), deoxynivalenols (DONs), T-2 toxins (T-2s), aflatoxins (AFs), and fumonisins (FBs), in cereal food samples. Sensitive and specific monoclonal antibodies were selected for this assay. The semi-quantitative results were obtained within 20 min by the naked eye, with visual limits of detection for ZEAs, DONs, T-2s, AFs and FBs of 0.1-0.5, 2.5-250, 0.5-1, 0.25-1 and 2.5-10 µg kg(-1), and cut-off values of 0.25-1, 5-500, 1-10, 0.5-2.5 and 5-25 µg kg(-1), respectively. The quantitative results were obtained using a hand-held strip scan reader, with the calculated limits of detection for ZEAs, DONs, T-2s, AFs and FBs of 0.04-0.17, 0.06-49, 0.15-0.22, 0.056-0.49 and 0.53-1.05 µg kg(-1), respectively. The analytical results of spiked samples were in accordance with the accurate content in the simultaneous detection analysis. This newly developed ICA strip assay is suitable for the on-site detection and rapid initial screening of mycotoxins in cereal samples, facilitating both semi-quantitative and quantitative determination.

A rapid, simple, and sensitive immunochromatographic test strip has been developed for testing residues of ciprofloxacin (CIP). A specific and sensitive monoclonal antibody (mAb) for CIP was generated by immunizing BALB/c mice with well-characterized CIP-Keyhole limpet haemocyanin. Under the optimized conditions, the cut-off limits of test strips for CIP were found to be 5 ng/mL in phosphate-buffered saline and 2.5 ng/mL in milk samples. Each test can be evaluated within 3 min. The cross-reactivities of the CIP test strip to enrofloxacin (ENR), norfloxacin (NOR), nadifloxacin (NDF), danofloxacin (DANO), pefloxacin (PEX), lomefloxacin (LOME), enoxacin (ENO), and sarafloxacin (SAR) were 71.4%, 71.4%, 66%, 50%, 33%, 20%, 12.5%, and 6.25%, respectively. The data indicate that the method is sensitive, specific, and has the advantages of simplicity and speed, therefore, this test strip is a useful screening method for the detection of CIP residues in milk samples.
