Objectives: In this study we aimed to evaluate the in vitro antibacterial activity and wound healing properties of Achillea millefolium essential oil (AMEO) in full-thickness wound model in rat. The antibacterial activity of AMEO was evaluated against Staphylococcus aureus and Pseudomonas aeruginosa using the broth dilution method. Methods: The 2 cm × 2 cm full-thickness excisional wounds were created on the back of animals. Topical therapy was applied twice a day using 1%, 2%, and 3% w/w AMEO ointments, and the measurement of the wounds area was carried out every 3 days, after that the wound closure percentage was calculated in these days. Hydroxyproline content and histopathological evaluation of wound tissue samples were carried out on day 7 and 14 post wounding. Eucerin was used for the treatment of vehicle control group and negative control group received no treatment. Results: Our results revealed the bacteriostatic activity of AMEO against S. aureus and P. aeruginosa. Wound healing activity evaluation of AMEO showed the significant increase (p < 0.05) in the wound closure percentages in rats treated with AMEO 1% and 2% comparing to those of non-treatment group. In addition, hydroxyproline contents of tissue significantly (p < 0.01) increased in AMEO 1% and 2% comparing to non-treatment group. Histopathological evaluations of wound tissue samples on day 7 and 14 demonstrated higher accumulation of collagen fibers, reduction of edema and inflammation and also formation of tissue appendages in 1% and 2% AMEO treated groups in comparison with non-treatment group. Conclusion: The results of this study indicated that AMEO has the potential to be used as a safe and effective wound healing agent.

Chlorogenic acid (CGA) is a phenolic compound widely found in plants. Several studies have shown that CGA possesses antioxidant, antibacterial, anti-inflammatory and wound healing properties. Because of their three-dimensional structure, good permeability, excellent biocompatibility and moisturizing properties, hydrogels are ideal candidates for wound dressing. The aim of the present study was to preparation and characterization of Polyvinyl alcohol (PVA) hydrogel containing CGA microspheres and evaluation its wound healing activity. The double-emulsion solvent evaporation technique was applied for preparing the CGA containing microspheres. The microspheres were characterized using scanning electron microscopy (SEM) and Fourier transformation infrared spectroscopy (FTIR) and subsequently incorporated in the structure of a PVA hydrogel. The effects of prepared hydrogel on NIH3T3 cell line viability were evaluated using MTT method and wound healing activity was investigated in full thickness wound model in rabbit. SEM images showed formation of homogenous CGA microspheres with diameters in the range of 1-2 µm, embedded in the porous structure of the hydrogel. Infra-red results indicated successful incorporation of CGA microspheres into PVA hydrogel. The NIH3T3 cell viability percentage in CGA 2.5% hydrogel treated group significantly (p < .05) increased after 24 h and 48 h comparing to control group. In vivo studies showed that CGA hydrogel significantly (p < .001) stimulated the rate of wounds closures. Histological studies revealed that administration of CGA hydrogel significantly increased epithelialization and production of collagen fibers compared to the control group. It can be concluded that the CGA microsphere loaded PVA hydrogel has the potential for wound healing.

Objectives: Hypertrophic scars (HSs) are caused by abnormal wound healing. To date, no standard treatment has been made available for HSs. Scrophularia striata has been reported to accelerate wound healing and has the potential to prevent HS formation. In this study, we investigated the anti-scarring effects of S. striata extract (SSE) in a rabbit ear model of scarring. Methods: In this study, New Zealand white rabbit (weight 2.3-2.5 kg) were used. In the prevention phase of the study, three test groups received 5%, 10%, and 15% ointments of SSE in the Eucerin base, the fourth group received Eucerin, and the fifth group received no treatment. The samples were obtained on day 35 after wounding. In the treatment phase of the study, the test groups received an intralesional injection of SSE (5%, 10%, and 15%), the fourth group received an intralesional injection of triamcinolone, the fifth group received a solvent (injection vehicle), and the sixth group received no treatment. To evaluate the anti-scarring effects of SSE, the scar elevation index (SEI), epidermis thickness index (ETI), collagen deposition, and MMP2 and MMP9 gene expression were evaluated. Results: A significant reduction in SEI, ETI, and collagen deposition was noted in animals treated with SSE compared with the control groups. In addition, topical SSE stimulated MMP2 and MMP9 gene expression. Conclusion: The findings of this study demonstrate the potential for SSE in the prevention and treatment of HS. SSE could be prepared as an appropriate formulation to treat wounds and prevent abnormal scarring.

BACKGROUND: Quince seed mucilage (QSM) has been used in Iranian folk medicine in the treatment of wounds and burns. Experimental and clinical studies showed its wound healing activity. However, the mechanism by which this agent affects cells involved in the wound healing process is unknown. OBJECTIVES: In this study, we investigated the effects of QSM at concentrations of 50, 100, 200, and 400 µg/mL on human skin fibroblast proliferation as an aspect of promotion of wound healing. MATERIALS AND METHODS: Human skin fibroblast cell line (HNFF-P18) was used in the experiment. Cell proliferation assay was measured by a MTT assay. RESULTS: Cells treated with QSM at concentrations less than 400 µg/mL increased their proliferative activity. The concentration of 50 µg/mL was the most effective dose after 72 hours treatment. CONCLUSIONS: QSM has the ability to stimulate proliferation of human skin fibroblast. This effect suggests that this compound can act as a wound healing agent.

OBJECTIVE: Quince seed mucilage (QSM) has been used in Iranian traditional medicine for the treatment of skin wounds and burns. Recent studies indicated that QSM accelerated wound healing. The present study was undertaken to investigate the healing efficiency of QSM formulated as 5%, 10%, and 20% creams in eucerin base with especial attention on growth factors involving in wound healing. METHODS: Full thickness wounds were created in Iranian male rabbits divided into five experimental groups (n = 6), as negative control, eucerin and treatments. Negative control group did not receive any treatment. Eucerin group received topical eucerin, twice a day. Treatment groups were treated topically by creams of QSM 5%, 10% and 20% (w/w) in eucerin base, twice daily. The efficacy of treatment was evaluated based on wound contraction, haydroxyproline content, tensile strength of wound tissue. The levels of epidermal growth factor (EGF), transforming growth factor-ß1 (TGF-ß1), vascular endothelial growth factor (VEGF) and platelet derived growth factor (PDGF) were also determined in serum and wound fluid of tested animals. RESULTS: Results showed that there were statistically significant differences in wound contraction between QSM 10 and 20% creams treatments groups and control groups (P < 0.05) in most of the days. Rabbits treated with QSM 20% cream had the best results (completed healing in 13 days, higher hydroxyproline content, higher tissue resistance and higher wound fluid levels of evaluated growth factors). CONCLUSION: We concluded tha QSM in 10-20% concentrations have a good potential for promote wound healing thus supports its traditional use.
