RESUMO

OBJECTIVE: To establish a recombinase polymerase amplification(RPA) method for extensively resistant pathogen screening and rapid detection in the field for rapid amplification of the metallo-beta-lactamase gene bla_(NDM). METHODS: Specific conservative sequence had been selected as target genes by sequence comparative analysis. The primers and probes for RPA assays were designed according to the principle of RPA amplification requirements. A descending gradient diluted template genes were used for RPA detection to determine the sensitivity. Reference templates of other resistant types of bacteria were used to analysis the specificity of amplification reaction systems. The amplification reaction systems were also conducted repeatedly for verifying the repeatability. RESULTS: Three of the RPA reaction systems could effectively amplify the target genes, the sensitivities reach 2×10~2 copies. No one cross reaction existed with the other drug-resistant bacteria DNA. All the reactions can be completed between two to seven minutes. CONCLUSION: The RPA assays of the metallo-beta-lactamase gene bla_(NDM) are established, which may amply target genes fast and have a lower detection limit, and be potentially useful for in field pathogens detection.

RESUMO

AIMS: The aim of this study was to investigate the impact of the signal transducer and activator of transcription 3 (STAT3) gene and additional STAT3 gene-smoking interaction on Crohn's disease (CD) risk based on a Chinese population. METHODS: A total of 1012 participants (491 men, 521 women), were selected, including 502 CD patients and 510 normal controls. The mean age of all participants was 42.3 ± 11.2 years. Logistic regression model was used to examine the association between single nucleotide polymorphism (SNP) of STAT3 and CD risk; the odds ratio (OR) and 95 % confident interval (95 % CI) were calculated. Generalized multifactor dimensionality reduction was employed to analyze the interaction among several SNPs. RESULTS: Logistic analysis showed the significant association between genotypes of variants in two SNP and decreased CD risk, after covariates adjustment. The carriers of homozygous mutant of two SNP polymorphism revealed decreased CD risk than those with wild-type homozygotes; OR (95 % CI) was 0.75 (0.59-0.93) and 0.68 (0.57-0.91), respectively. There was a significant two-locus model (p = 0.0107) involving rs744166 and smoking, indicating a potential gene-environment interaction between rs744166 and smoking. Overall, the cross-validation consistency was 10/10, and the testing accuracy was 62.17 %, and never smokers with TC or CC genotype have the lowest CD risk, compared to smokers with TT genotype; OR (95 % CI) was 0.52 (0.31-0.82), after covariate adjustment. CONCLUSIONS: Our results support an important association of rs744166 and rs4796793 with decreased CD risk, and additional interaction between rs744166 and smoking.

RESUMO

OBJECTIVE: To explore the expression of costimulatory molecule CD40 in thyroid tissue of Graves' disease patients and understand its immune pathogenetic significance. METHODS: From January 2008 to December 2011, 8 patients undergoing partial thyroidectomy for Graves' disease (n = 3) or non-toxic goiter (n = 5) at Affiliated Suzhou Hospital of Nanjing Medical University and Third Affiliated Hospital of Soochow University were enrolled. Using the method of immunohistochemistry, the expression of CD40 was detected in their thyroid tissues. Variation in CD40 expression on thyroid follicular (TFC) in primary cultures was analyzed in the absence (no stimulation group) or presence of inflammatory cytokines including interleukin interferon-γ (IFN-γ) (IFN-γ stimulation group), interferon-6 (IL-6) (IL-6 stimulation group)and tumor necrosis factor (TNF)-α (TNF-α stimulation group) with flow cytometry. IFN-γ-stimulated TFC were cultured with agonist CD40 monoclonal antibody (5C11) (IFN-γ + CD40 group) or isotypic mouse IgG (mIgG) antibody (IFN-γ + mIgG group). And the proliferation of TFC was assessed by 3-(4,5)-dimethyl-thiazolyl-3,5-di-phenytetrazoliumromide (MTT) assays for each donor. The production of free triiodothyronine (FT3) and free thyroxine (FT4) and the release of thyroglobulin (Tg) were measured with radioimmunoassays. RESULTS: The expression of CD40 on infiltrated lymphocytes and TFC were detected in Graves' disease but not in non-toxic goiter patient tissues. Compared with no stimulation group (23.7% ± 7.3%), the expression of CD40 on TFC increased in IFN-γ stimulation group (86.4% ± 4.6%), IL-6 stimulation group (90.0% ± 4.2%) and TNF-α stimulation group (87.3% ± 4.2%). Compared with the IFN-γ + mIgG group (0.75 ± 0.06), the TFC proliferation of IFN-γ + CD40 group (1.14 ± 0.14) significantly increased (P < 0.01). The levels of FT3, FT4 and Tg secretion of IFN-γ + CD40 group were (1.10 ± 0.15) pmol/L, (0.80 ± 0.14) pmol/L and (30.23 ± 1.60) µg/L respectively. They were all significantly increased compared with the IFN-γ + mIgG group, of which the FT3, FT4 and Tg production were (0.76 ± 0.07) pmol/L, (0.63 ± 0.09) pmol/L and (21.37 ± 3.22) µg/L respectively (all P < 0.05). CONCLUSIONS: CD40 is abnormally expressed in thyroid tissue of Graves' disease patients. And its costimulatory signal may take part in the immunopathologic mechanism of Graves' disease.

RESUMO

A novel method was developed for the rapid determination of 22 representative pesticide residues in vegetables and fruits based on accelerated solvent extraction (ASE) coupled with online gel permeation chromatography-gas chromatography-mass spectrometry (GPC-GC-MS). The sample was extracted by accelerated solvent extraction with dichloromethane-acetone (1:1, v/v) and purified with a carbon/NH2 column, evaporated to dryness by nitrogen, then dissolved in cyclohexane-acetone (7:3, v/v), and finally identified and quantified by GPC-GC-MS system in selected ion monitoring (SIM) mode. The results showed that the linearities of the 22 pesticides were good in their linear ranges. The limits of detection (S/N = 3) were 0.3-1.8 microg/kg. The limits of quantification (S/N = 10) ranged from 1-6 microg/kg. The recoveries for all at three spiked levels in Chinese cabbages and apples ranged from 70.5% to 107.5% with the relative standard deviations (RSDs) of 2.1%-8.7%. The proposed method is accurate, sensitive and highly efficient in the extraction, and can be used for the quick determination of the pesticide multiresidues in vegetables and fruits.

RESUMO

We analyzed the impact of a GAGA element on a transgenic promoter in Drosophila melanogaster that was activated by proteins composed of the Tet(on) DNA binding domain and either the heat shock factor (HSF) activation domain or a potent subdomain of VP16. Permanganate footprinting was used to monitor polymerase II (Pol II) on the transgenic promoters in vivo. Activation by Tet(on)-HSF but not by Tet(on)-VP16(A2) required the GAGA element; this correlated with the ability of the GAGA element to establish a paused Pol II. Although the GAGA element was not required for activation by Tet(on)-VP16(A2), the GAGA element greatly accelerated the rate of activation. The permanganate data also provided evidence that Pol II encountered different rate-limiting steps, following initiation in the presence of Tet(on)-HSF and Tet(on)-VP16(A2). The rate-limiting step in the presence of Tet(on)-HSF was release of Pol II paused about 20 to 40 nucleotides downstream from the start site. The rate-limiting step in the presence of Tet(on)-VP16(A2) occurred much closer to the transcription start site. Several biochemical studies have provided evidence for a structural transition shortly after Pol II initiates transcription. The behavior of Pol II in the presence of Tet(on)-VP16(A2) provides the first evidence that this transition occurs in vivo.

RESUMO

An inactivated vaccine for severe acute respiratory syndrome (SARS)-associated coronavirus (SARS-CoV) was evaluated in rhesus monkeys. The monkeys were inoculated intramuscularly (i.m.) with 0.5, 5, 50, or 5000 microg of vaccine, or PBS as control, and boosted on day 7. After 3 weeks, they were challenged with the NS-1 strain of SARS-CoV. The humoral and mucosal immune responses, clinical signs, chemical indices and viremia were monitored following the immunization and challenge. The control animals who received PBS developed atypical SAR-CoV infection after viral challenge, according to clinical, virological and pathological findings. No systematic side effects were observed in vaccinated animals post-immunization, even in at the high dose of 5000 microg. The 50 microg dosage of vaccine elicited SARS-CoV specific immune responses against viral infection as compared to the partial immunity elicited by 0.5 and 5 microg doses. The results show that this inactivated vaccine can induce effective concomitant humoral and mucosal immunity against SARS-CoV infection, is safe in monkeys, and the vaccine maybe a good candidate for clinical trials.
