RESUMO

Attention deficit hyperactivity disorder (ADHD) is a neurodevelopmental disorder diagnosed with hyperactivity, impulsivity, and a lack of attention inconsistent with the patient's development level. The fact that people with ADHD frequently experience gastrointestinal (GI) dysfunction highlights the possibility that the gut microbiome may play a role in this condition. The proposed research aims to determine a biomarker for ADHD by reconstructing a model of the gut-microbial community. Genome-scale metabolic models (GEM) considering the relationship between gene-protein-reaction associations are used to simulate metabolic activities in organisms of gut. The production rates of dopamine and serotonin precursors and the key short chain fatty acids which affect the health status are determined under three diets (Western, Atkins', Vegan) and compared with those of healthy people. Elasticities are calculated to understand the sensitivity of exchange fluxes to changes in diet and bacterial abundance at the species level. The presence of Bacillota (genus Coprococcus and Subdoligranulum), Actinobacteria (genus Collinsella), Bacteroidetes (genus Bacteroides), and Bacteroidota (genus Alistipes) may be possible gut microbiota indicators of ADHD. This type of modeling approach taking microbial genome-environment interactions into account helps us understand the gastrointestinal mechanisms behind ADHD, and establish a path to improve the quality of life of ADHD patients.

BACKGROUND: Anastomotic leakage (AL) following rectal surgery is associated with increased mortality and morbidity. Neoadjuvant radiotherapy disrupts the wound healing process in rectal surgery. OBJECTIVES: To evaluate the effects of intra-rectal ozone application on rectal anastomoses after radiotherapy. MATERIAL AND METHODS: This study was performed on animals. Thirty-two male Wistar rats were randomly divided into 4 groups: control group, ozone group, radiotherapy group, and radiotherapy/ozone group. Ozone was administered intrarectally in the ozone group and water was administered intrarectally in rthe control group for 5 days. The radiotherapy group received 20 Gy of pelvic radiotherapy. The radiotherapy/ozone group received 20 Gy of pelvic radiotherapy after the administration of ozone. Afterward, colon resection followed by an anastomosis were performed under general anesthesia in all groups. Anastomotic segments were resected to evaluate tissue hydroxyproline (HYP) and myeloperoxidase (MPO) levels, perform a histological evaluation, and measure bursting pressure. RESULTS: There were no statistically significant differences between groups regarding tissue MPO levels (p = 0.55). Tissue HYP levels were significantly decreased in the radiotherapy group (p = 0.04). Bursting pressure was found to be significantly lower in the radiotherapy group (p < 0.05). No significant differences were found between adhesion scores in the control and ozone groups. Exudate formation was significantly lower in the radiotherapy group (p < 0.05). The lowest macrophage scores were found in the radiotherapy group (p < 0.05). Fibroblast scores were the highest in the control group and the lowest in the radiotherapy group (p < 0.05). CONCLUSIONS: Intra-rectal ozone application significantly improved the anastomotic healing process after radiation exposure.

Schizochytrium species is one of the most studied microalgae for production of docosahexaenoic acid (DHA) which is an omega-3 fatty acid with positive effects for human health. However, high cost and low yield in production phase makes optimization of cultivation process inevitable. We focus on the optimization of DHA production using Schizochytrium sp. using different media supplements; glucose, fructose and glycerol as carbon variants, proteose peptone and tryptone as nitrogen variants. The highest biomass (5.61 g/L) and total fatty acid yield (1.74 g/L) were obtained in proteose peptone medium which was used as the alternative nitrogen source instead of yeast extract. The highest DHA yield (0.40 g/L) was achieved with glycerol as the carbon source although it had the second lowest biomass production after ethanol containing medium. Ethanol, as an alternative carbon source and a precursor for acetyl-CoA, increased DHA percentage in total lipid content from 29.94 to 40.04% but decreasing the biomass drastically. Considering different carbon and nitrogen sources during cultivation of Schizochytrium sp. will improve DHA production. Combination of proteose peptone and glycerol as nitrogen and carbon sources, respectively, and addition of ethanol with a proper timing will be useful to have higher DHA yield.
