RESUMO

Ray tracing in gradient-index (GRIN) media has been thoroughly studied and several ray tracing methods have been proposed. Methods are based on finding the ray path given a known GRIN. In recent decades, the inverse problem, which consists of finding the GRIN distribution for a given light ray path, has been gaining attention. Given that it is not an easy task, the methods proposed in the literature vary in degrees of difficulty. In this work, an alternative method is presented to derive symmetric GRIN distributions whose implementation can be considered the simplest to date. Since it is based on invariants, which result from the symmetries of the system as stated by Fermat's principle, it is an exact numerical method, i.e., the physical system is not approximated. The robustness of the method permits the reconstruction of the GRIN distribution from a ray propagating in three-dimensions. In order to demonstrate its operation, different known symmetric GRIN media are reconstructed using rays that propagate in two and three dimensions.

Ray tracing in gradient-index (GRIN) media has been traditionally performed either by using the analytical or numerical solutions to the Eikonal equation or by creating a layered medium where Snell's law is calculated in each layer. In this paper, an exact general method to perform ray tracing in GRIN media is presented based on the invariants of the system as stated by Fermat's principle when the media presents symmetries. Its advantage, compared with other methods reported in the literature, relies on its easy implementation. Besides the GRIN distribution and the initial conditions of the incident ray, once the invariants of the system are stated the resulting math is simple to solve and interpret. To benchmark the algorithm, ray tracing in typical cases of GRIN media is calculated, finding minimal discrepancies between the analytical solutions and our simulations. The used media are axial refractive index and parabolic index fiber and lenses with spherical gradient-index symmetry, such as: Luneburg's, Gutman's, generalized Maxwell's Fish-eye, Eaton's, and concentrator lenses. Our method can be further applied to distributions with symmetries associated with other common curvilinear orthogonal coordinate systems, in particular to those associated to the separability of the Helmholtz equation that would allow us to investigate wave optics in these GRIN media with the associated geometries.

When a circular aperture is uniformly illuminated, it is possible to observe in the far field an image of a bright circle surrounded by faint rings known as the Airy pattern or Airy disk. This pattern is described by the first-order Bessel function of the first type divided by its argument expressed in circular coordinates. We introduce the higher-order Bessel functions with a vortex azimuthal factor to propose a family of functions to generalize the function defining the Airy pattern. These functions, which we call vortex Jinc functions, happen to form an orthogonal set. We use this property to investigate their usefulness in fitting various surfaces in a circular domain, with applications in precision optical manufacturing, wavefront optics, and visual optics, among others. We compare them with other well-known sets of orthogonal functions, and our findings show that they are suitable for these tasks and can pose an advantage when dealing with surfaces that concentrate a considerable amount of their information near the center of a circular domain, making them suitable applications in visual optics or analysis of aberrations of optical systems, for instance, to analyze the point spread function.

BACKGROUND: Type 2 diabetes mellitus (T2DM) accelerates brain aging and increases the risk for dementia. Insulin is a key neurotrophic factor in the brain, where it modulates energy metabolism, neurovascular coupling, and regeneration. Impaired insulin-mediated brain signaling and central insulin resistance may contribute to cognitive and functional decline in T2DM. Intranasal insulin (INI) has emerged as a potential therapy for treating T2DM-related cognitive impairment. METHODS/DESIGN: Ongoing from 2015, a prospective, two-center, randomized, double-blind, placebo-controlled trial of 210 subjects (120 T2DM and 90 non-diabetic older adults) randomized into four treatment arms (60 T2DM-INI, 60 T2DM-Placebo, 45 Control-INI, and 45 Control-Placebo) evaluating the long-term effects of daily intranasal administration of 40 International Units (IU) of human insulin, as compared to placebo (sterile saline) over 24 weeks and 24 weeks of post-treatment follow-up. Study outcomes are: 1) long-term INI effects on cognition, daily functionality, and gait speed; 2) identifying a clinically relevant phenotype that predicts response to INI therapy; 3) long-term safety. CONCLUSION: This study addresses an important knowledge gap about the long-term effects of intranasal insulin on memory and cognition in older people with T2DM and non-diabetic controls, and may provide a novel therapeutic target for prevention and treatment of cognitive and functional decline and dementia. RESUMO

Intranasal drug administration is a promising method for delivering drugs directly to the brain. Animal studies have described pathways and potential brain targets, but nose-to-brain delivery and treatment efficacy in humans remains debated. We describe the proposed pathways and barriers for nose-to-brain drug delivery in humans, drug properties that influence central nervous system delivery, clinically tested methods to enhance absorption, and the devices used in clinical trials. This review compiles the available evidence for nose-to-brain drug delivery in humans and summarizes the factors involved in nose-to-brain drug delivery.

The nutrient content in the muscle and edible skin parts of rainbow trout (Oncorhynchus mykiss) fillets, sampled at two growth stages, was evaluated. The average concentrations of protein and essential amino acids were higher in the muscle than in the skin. The chemical scores reached a value of 1.0 for the amino acids in the muscle and ranged from 0.40 (tryptophan) to 0.94 (threonine) in the skin. The average lipid content and the saturated fatty acids/polyunsaturated fatty acids and n-6/n-3 ratios were higher in the skin than in the muscle, whereas the proportion of docosahexaenoic acid (C22:6 n-3) was higher in the muscle. Significant differences were found for the essential minerals analysed, except for Cu. The concentrations of Na, K and Mg were higher and those of Ca, P, Fe, Mn and Zn were lower in the muscle than in the skin. Significant effects of the fish growth on the composition were detected.

We report on the design and experimental demonstration of array-enhanced nanoantennas for polarization-controlled multispectral nanofocusing in the near-IR spectral range. We design plasmonic double bow-tie nanoantennas-coupled to multiple-periodic nanoparticle arrays to harvest radiation of designed wavelengths from a large spatial area and to focus it into a targeted nanoscale region. Near-field calculations were performed on a gold nanoantenna array using three-dimensional finite difference time domain simulations. Cross-shaped optical nanoantennas were fabricated on glass substrates using electron beam lithography. The optical characterization of the fabricated nanoantennas was performed using second harmonic excitation spectroscopy that demonstrates multiwavelength photonic coupling in good agreement with the antenna modeling. The nanoantenna structures introduced in this Letter provide the ability to focus optical energy into deep subwavelength areas and to address multiple spectral regions with polarization control. Such attributes are highly desirable in optical biosensing, enhanced Raman scattering, and for nonlinear plasmonic applications.

BACKGROUND AND STUDY AIMS: The fanning technique for endoscopic ultrasound-guided fine-needle aspiration (EUS - FNA) involves sampling multiple areas within a lesion with each pass. The aim of this study was to compare the fanning and standard techniques for EUS - FNA of solid pancreatic masses. PATIENTS AND METHODS: Consecutive patients with solid pancreatic mass lesions were randomized to undergo EUS - FNA using either the standard or the fanning technique. The main outcome measure was the median number of passes required to establish diagnosis. The secondary outcome measures were the diagnostic accuracy, technical failure, and complication rate of the two techniques. RESULTS: Of 54 patients, 26 were randomized to the standard technique and 28 to the fanning technique. There was no difference in diagnostic accuracy (76.9 % vs. 96.4 %; P = 0.05), technical failure or complication rates (none in either cohort). There was a significant difference in both the number of passes required to establish diagnosis (median 1 [interquartile range 1 - 3] vs. 1 [1 - 1]; P = 0.02) and the percentage of patients in whom a diagnosis was achieved on pass one (57.7 % vs. 85.7 %; P = 0.02) between the standard and fanning groups, respectively. CONCLUSIONS: The fanning technique of FNA was superior to the standard approach because fewer passes were required to establish the diagnosis. If these promising data are confirmed by other investigators, consideration should be given to incorporating the fanning technique into routine practice of EUS - FNA. Tungsten disulfide (WS(2)) nanometer sheets, spheres, fibers and tubes were generated by a synthetic pathway that avoids the use of H(2)S as the source of sulfur and employs instead CS(2) vapor, carried by an Ar or N(2)/H(2) stream in a heated tubular furnace, for the reaction with WO(3) precursor powders. The experiments were conducted at temperatures between 700 and 1000 °C, while the reaction times expanded between 30 min and 24 h. Characterization methods used to analyze the products of the synthesis include TEM, SEM, XRD and EDX. We found a strong correlation between precursor and product microstructure, although the temperature and reaction times play a critical role in the products' microstructural features as well. WS(2) inorganic fullerene (IF) nanospheres are generated in a wide window of conditions, while nanotubes and nanofibers are only produced at high temperatures or long reaction times. A proposed growth mechanism based on the CS(2) synthetic approach is presented. Nanoindentation and nano-impulse techniques were used to characterize the mechanical properties of polymer matrix-WS(2) nanotube composites, finding them superior to equivalent SWCNT composites. The improvements in toughness of nanocomposites based on WS(2) can be attributed to geometrical and morphological effects that assisted several toughening mechanisms such as crack pinning and the formation of an immobilized polymeric interphase around the nanotubes.

Thermostable phytases, which are active over broad pH ranges, may be useful as feed additives, since they can resist the temperatures used in the feed-pelleting process. We designed new beta-propeller phytases, using a structure-guided consensus approach, from a set of amino acid sequences from Bacillus phytases and engineered Pichia pastoris strains to overproduce the enzymes. The recombinant phytases were N-glycosylated, had the correct amino-terminal sequence, showed activity over a pH range of 2.5 to 9, showed a high residual activity after 10 min of heat treatment at 80°C and pH 5.5 or 7.5, and were more thermostable at pH 7.5 than a recombinant form of phytase C from Bacillus subtilis (GenBank accession no. AAC31775). A structural analysis suggested that the higher thermostability may be due to a larger number of hydrogen bonds and to the presence of P257 in a surface loop. In addition, D336 likely plays an important role in the thermostability of the phytases at pH 7.5. The recombinant phytases showed higher thermostability at pH 5.5 than at pH 7.5. This difference was likely due to a different protein total charge at pH 5.5 from that at pH 7.5. The recombinant beta-propeller phytases described here may have potential as feed additives and in the pretreatment of vegetable flours used as ingredients in animal diets.

1. A 35 d feeding trial with 480 d-old male broiler chickens (Cobb) was conducted to evaluate the effects of graded levels of inulin (0, 5, 10, 15 and 20 g/kg diet) in a maize-soybean meal based diet on growth performance, AME(N) and apparent ileal digestibility (AID) of crude protein, amino acids, starch, crude fat and fatty acids. A commercial product (Inulina 95) containing 9140 g/kg of inulin and a degree of polymerisation (DP) ranging from DP 1-10 to DP 51-60 was used. 2. The performance of birds (feed intake, BW gain and feed to gain ratio) was not affected by the different dietary inclusions of inulin. 3. Dietary inulin significantly improved the AID coefficient of crude protein and crude fat. Also, there was a significant effect of inulin on the digestibility of most amino acids (10 out of 15) and major fatty acids (oleic and linoleic acids). The AID coefficient of starch and the AME(N) content of diets were not affected by inulin inclusion. 4. In conclusion, the current study showed that inulin supplementation improved the digestibility of protein and fat in a maize-soybean meal based diet, but had no effect on the performance of broiler chickens.

A study was conducted to evaluate the effect of adding inulin to diets containing 2 different types of fat as energy sources on performance, blood serum metabolites, liver lipids, and fatty acids of abdominal adipose tissue and breast and thigh meat. A total of 240 one-day-old female broiler chicks were randomly allocated into 1 of 6 treatments with 8 replicates per treatment and 5 chicks per pen. The experiment consisted of a 3 x 2 factorial arrangement of treatments including 3 concentrations of inulin (0, 5, and 10 g/kg of diet) and 2 types of fat [palm oil (PO) and sunflower oil (SO)] at an inclusion rate of 90 g/kg of diet. The experimental period lasted from 1 to 34 d. Dietary fat type did not affect BW gain but impaired feed conversion (P < 0.001) in birds fed the PO diets compared with birds fed the SO diets. The diets containing PO increased abdominal fat deposition and serum lipid and glucose concentrations. Triacylglycerol contents in liver were higher in the birds fed PO diets. Dietary fat type also modified fatty acids of abdominal and i.m. fat, resulting in a higher concentration of C16:0 and C18:1n-9 and a lower concentration of C18:2n-6 in the birds fed PO diets. The addition of inulin to diets modified (P = 0.017) BW gain quadratically without affecting feed conversion. Dietary inulin decreased the total lipid concentration in liver (P = 0.003) and that of triacylglycerols and very low density lipoprotein cholesterol (up to 31%) in blood serum compared with the control groups. The polyunsaturated fatty acid:saturated fatty acid ratio increased in abdominal and i.m. fat when inulin was included in the SO-containing diets. The results from the current study suggest that the addition of inulin to broiler diets has a beneficial effect on blood serum lipids by decreasing triacylglyceride concentrations The results also support the use of inulin to increase the capacity of SO for enhancing polyunsaturated fatty acid:saturated fatty acid ratio of i.m. fat in broilers.

The cloning and expression of a native gene encoding a Bacillus subtilis phytase using Pichia pastoris as the host is described. In addition, the influence of N-glycosylation on the biochemical properties of the B. subtilis phytase, the influence of pH on the thermostability of the recombinant and native B. subtilis phytases, and the resistance of both phytases to shrimp digestive enzymes and porcine trypsin are also described. After 48 h of methanol induction in shake flasks, a selected recombinant strain produced and secreted 0.82 U/ml (71 mg/liter) recombinant phytase. This phytase was N-glycosylated, had a molecular mass of 39 kDa after N-deglycosylation, exhibited activity within a pH range of 2.5 to 9 and at temperatures of 25 to 70 degrees C, had high residual activity (85% +/- 2%) after 10 min of heat treatment at 80 degrees C and pH 5.5 in the presence of 5 mM CaCl(2), and was resistant to shrimp digestive enzymes and porcine trypsin. Although the recombinant Bacillus phytase had pH and temperature activity profiles that were similar to those of the corresponding nonglycosylated native phytase, the thermal stabilities of the recombinant and native phytases were different, although both were calcium concentration and pH dependent.

While endoscopic ultrasound (EUS)-guided fiducial placement is technically feasible, its role in the delivery of image-guided radiation therapy (IGRT) is unclear. In this study, we evaluated the feasibility and safety of using EUS-guided fiducial markers for delivering IGRT in patients with pancreatic cancer. Following computed tomography (CT) simulation, a radiation therapy treatment plan was prepared for each patient. A kilovoltage (kv) image obtained before each treatment session was aligned with images generated during treatment planning, based on the position of fiducials. Radiation therapy was then delivered. Image analysis was also performed by alignment of bony landmarks (current standard of care). Daily shifts of fiducials between the two sets of images represented movement of the pancreas relative to the bony landmarks. EUS-guided fiducial placement was technically successful in all nine patients and no complications were encountered. The fiducials were easily identified for image fusion and IGRT was delivered successfully to all patients. The movement of the pancreas relative to bones was 0.2 cm (0.1-0.5 cm), 0.5 cm (0.2-1.5 cm), and 0.4 cm (0.2-0.9 cm) in the anterior-posterior, superior-inferior, and left-right axes, respectively. This technique may have significant clinical implications in terms of radiation dosimetry.

A study was undertaken to examine the effects of inulin, alone or in combination with enzyme complex (primarily xylanase and beta-glucanase), on growth performance, ileal and cecal microflora, cecal short-chain fatty acids, and d-lactic acid and jejunal histomorphology of broiler chickens fed a wheat- and barley-based diet from 7 to 35 d of age. A total of 240 seven-day-old male Cobb broilers were allocated to 1 of 6 treatments, with 8 replicate pens per treatment and 5 birds per pen. The experiment consisted of a 3x2 factorial arrangement of the treatments with 3 concentrations of inulin (0, 10, or 20 g/kg of diet) and 2 concentrations of enzyme complex (0 or 100 mg/kg of diet). At the end of the experiment, 8 birds per treatment (one from each pen) were randomly chosen and slaughtered. Birds fed inulin-containing diets exhibited significantly (P=0.043) improved final BW gain. Dietary inulin had a positive and significant (P<0.002 to 0.009) effect on bifidobacteria and lactobacilli counts in both ileal and cecal contents and, to an extent, also altered the fermentation patterns in the ceca, increasing the concentration of n-butyric and d-lactic acids and the n-butyric acid:acetic acid ratio. Inulin inclusion had no effect on villus height and crypt depth or microvillus length, width, and density in the jejunum. Enzyme supplementation of the control diet and inulin-containing diets had no effect on many of the variables studied and only resulted in a decrease in crypt depth and an increase in villus height:crypt depth ratio in the jejunum.

Single- and multi-analyte detection of two gonadotropic hormones (follicle stimulating hormone (hFSH) and luteinizing hormone (hLH)) was achieved by a Surface Plasmon Resonance (SPR) immunoassay on untreated human urine samples. Multi-analyte detection was accomplished using two alternative formats which are based in the individual or simultaneous immobilization of the hormones on the sensor surface. The lowest detection limit for both hormones in urine was found to be 1 ng mL(-1), which in international units (IU) in terms of the World Health Organization (WHO) standards represents 8 mIU mL(-1) of hLH and 14 mIU mL(-1) of hFSH, respectively. The reliability of the assay was demonstrated by intra- and inter-assay variabilities < 6%, chip-to-chip variabilities < 5%, recoveries in the range of 80-120% and stability of the sensor response through more than 100 measurements. The sensitivity of this biosensing methodology renders it in a useful technique for the diagnosis of reproductive disorders, as well as for fertility monitoring.

RESUMO

1. A 5-week feeding trial with 240 one-day-old male broiler chickens was conducted to investigate the responses of performance, intestinal tract measurements, relative apparent retention of minerals (Ca, Mg, Fe, Zn and Cu), and tibial bone measurements and mineral content (ash and Ca) to dietary graded levels of inulin. Treatments consisted of a maize-soybean meal control diet without or with supplemental antibiotic (flavomycin, 40 mg/kg diet) and 4 diets containing inulin at concentrations of 5, 10, 15 and 20 g/kg. 2. Growth performance of birds and morphological measurements of intestinal tract (duodenum, jejunum, ileum and caeca) showed no response to dietary levels of inulin as compared with the control diet and the control diet with flavomycin. 3. Dietary inulin improved the relative apparent retention of Ca, Zn and Cu (up to 18.4, 35.5 and 466%, respectively), did no affect that of Mg and impaired the retention of Fe. Differences among inulin treatments and control with flavomycin were significant only for Mg content. 4. Dietary inulin increased the concentration of ash and Ca in the tibiae. This effect was not reflected in the tibial bone morphology (weight, length and width). In conclusion, the results from the current study suggest that feeding inulin to broiler chickens may have a beneficial effect on bone quality.
