Food preparation involves the blending of various food ingredients to make more convenient processed food products. It is a long chain process, where each stage posing a risk of accumulating hazardous contaminants in these food systems. Protecting the public health from contaminated foods has become a demanding task in ensuring food safety. This review focused on the causes, types, and health risks of contaminants or hazardous chemicals during food processing. The impact of cooking such as frying, grilling, roasting, and baking, which may lead to the formation of hazardous by-products, including polycyclic aromatic hydrocarbons (PAHs), heterocyclic amines (HCAs), acrylamide, advanced glycation end products (AGEs), furan, acrolein, nitrosamines, 5-hydroxymethylfurfural (HMF) and trans-fatty acids (TFAs). Potential health risks such as carcinogenicity, genotoxicity, neurotoxicity, and cardiovascular effects are emerging as a major problem in the modern lifestyle era due to the increased uptakes of contaminants. Effects of curing, smoking, and fermentation of the meat products led to affect the sensory and nutritional characteristics of meat products. Selecting appropriate cooking methods include temperature, time and the consumption of the food are major key factors that should be considered to avoid the excess level intake of hazardous contaminants. Overall, this study underscores the importance of understanding the risks associated with food preparation methods, strategies for minimizing the formation of harmful compounds during food processing and highlights the need for healthy dietary choices to mitigate potential health hazards.

Nanopollution (NPOs), a burgeoning consequence of the widespread use of nanoparticles (NPs) across diverse industrial and consumer domains, has emerged as a critical environmental issue. While extensive research has scrutinized the repercussions of NPs pollution on ecosystems and human health, scant attention has been directed towards unraveling its implications for plant life. This comprehensive review aims to bridge this gap by delving into the nuanced interplay between NPOs and plant metabolism, encompassing both primary and secondary processes. Our exploration encompasses an in-depth analysis of the intricate mechanisms governing the interaction between plants and NPs. This involves a thorough examination of how physicochemical properties such as size, shape, and surface characteristics influence the uptake and translocation of NPs within plant tissues. The impact of NPOs on primary metabolic processes, including photosynthesis, respiration, nutrient uptake, and water transport. Additionally, this study explored the multifaceted alterations in secondary metabolism, shedding light on the synthesis and modulation of secondary metabolites in response to NPs exposure. In assessing the consequences of NPOs for plant life, we scrutinize the potential implications for plant growth, development, and environmental interactions. The intricate relationships revealed in this review underscore the need for a holistic understanding of the plant-NPs dynamics. As NPs become increasingly prevalent in ecosystems, this investigation establishes a fundamental guide that underscores the importance of additional research to shape sustainable environmental management strategies and address the extensive effects of NPs on the development of plant life and environmental interactions.

Boucerosia diffusa Wight. is an important endangered medicinal plant belonging to the family Asclepiadaceae. In this study, an efficient protocol has been developed for B. diffusa using nodal explants for callus induction and direct organogenesis. The optimal callus induction (83.7%) was observed on 0.6 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D) in Murashige and Skoog medium. The shoot regeneration was observed on different concentrations and combinations of 6-benzylaminopurine (BAP) and 2,4-D using shoot induction (88.5%) was observed on 0.5 mg/L BAP and 0.6 mg/L 2,4-D. Maximum root induction frequency (85.6%) was obtained on 0.6 mg/L α-naphthalene-acetic acid (NAA) and 0.5 mg/L BAP. The fully developed plants were acclimatized (98.86% survival rate) and transferred to natural photoperiod conditions. The phytochemical and pharmacological activity was determined in in vitro-regenerated plants (IRP) and was compared to in vivo wild plants (IWP). The primary and the secondary metabolite contents of bioactive compounds were significantly higher in the methanolic extract of IRP. A comparative antioxidant activity study shows IRP exhibited better scavenging activity. The antidiabetic activity of α- amylase (IC50 - 71.56 ± 15.4 µg/mL) and α-glucosidase (IC50 - 82.94 ± 12.84 µg/mL) inhibitor activity also exhibited maximum in methanolic extract of IRP. Furthermore, chemical composition was analyzed using gas chromatography-mass spectroscopy (GC-MS). Antibacterial activity against human pathogenic bacteria, IRP methanolic extracts showed a maximum zone of inhibition (75 µg/mL) observed against Salmonella typhi (23.5 ± 0.5 mm) compared to the IWP. Molecular docking analysis of B. diffusa inhibition of antidiabetic activity showed better affinity in ß-Sitosterol. The Striga angustifolia (D. Don) C.J. Saldanha was used as an Ayurvedic and homeopathic medicine for cancer by the tribal peoples of the Maruthamalai Hills, Coimbatore, India. Hence, the traditional use that has been proven to be effective lacks convincing scientific references. This present study was conducted to investigate the presence of potentially bioactive compounds from S. angustifolia and provides a scientific basis for the ethnobotanical utility. The organosulfur compound 5,5'-dithiobis(1-phenyl-1H-tetrazole) (COMP1) was isolated from S. angustifolia extracts, and the structures of COMP1 were elucidated and characterized by using 13C and 1H nuclear magnetic resonance (NMR) and single crystal X-ray powder diffraction (XRD). Our findings showed that COMP1 significantly reduced cell proliferation of breast and lung cancer cells, but not that of non-malignant epithelial cells. Further analysis revealed that COMP1 promoted cell cycle arrest and apoptosis of lung cancer cells. Mechanistically, COMP1 facilitates p53 activity and inhibits mammalian target of rapamycin (mTOR) signaling, thereby inducing cell cycle arrest and apoptosis of lung cancer cells by inhibiting cell growth. Our findings suggest that COMP1 may serve as a potential drug for lung cancer through the regulation of p53/mTOR pathways.

Several tribal medicinal systems assert anti-migraine and common headache-remedying properties in all parts of Priva cordifolia (L.f.) Druce. Therefore, there are no clear scientific references to the validated traditional use of this plant. The present study provides a scientific basis for the ethnobotanical utility of P. cordifolia whose whole-plant extracts were evaluated against target proteins (PACAP 38 and PAC1-R) that cause migraine. Understanding the polarity-based distribution and oxidative stress scavenging ability was reported higher in ethyl acetate extracts due to the moderate distribution of secondary metabolites. Based on the preliminary analysis anti-migraine activity in the wet and dry lab experiments was compared with a commercial drug Sumatriptan. The GC-MS analysis revealed that two lead volatile compounds Bicyclo(3.2.1)oct-3-en-2-one,3,8-Dihydroxy-1-methoxy-7-(7-methoxy-1,3- and -Hexyl-2-nitrocyclohexane, present in the ethyl acetate extract showed favourable in silico anti-migraine efficiency. Notably, the ex-vivo results also showed considerable downregulation of the extract-induced mRNA expression of PACAP38. The conclusion of our study justifies that P. cordifolia has valuable plant metabolites that portray it as an efficient anti-oxidant and anti-migraine source. BACKGROUND & AIMS: Human beings are often driven to exhibit dietary preference according to their hedonic characteristics. Though previous studies proposed that the fat taste preference of an obese individual was associated with BMI, the perception of fat taste differs for every individual. The genetic variation among populations in taste receptor genes such as CD36 may be a contributing factor for this difference. Satiety peptides can also play a role in the regulation of fat taste perception. Generally, this hormone helps us to feel the sense of satiety. METHODS: We have analysed the relationship among oro-gustatory perception of dietary lipids, salivary peptide-YY and genetic polymorphism in CD36. Oral fatty acid sensitivity analysis was performed by alternative forced choice method. Salivary peptide-YY concentration was analysed by ELISA and single nucleotide polymorphism (SNP) in CD36 gene was determined by Real-Time PCR experiments. RESULTS: We observed that the SNP at rs1761667 of CD36 and oral detection threshold for linoleic acid (LA) are associated with choice of food, lipid profiles, peptide-YY as well as adiposity parameters in obese population. Obese peoples had significantly low levels of peptide YY than people with BMI less than 25. These factors possibly play a role in preference for energy rich diets, development of obesity and associated complications. CONCLUSION: This study provides a solid foundation for understanding the alterations in the dietary fat intake and levels of peptide-YY, which are associated with polymorphism in fat taste receptor. This is the first report that shows a significant relationship between the satiety hormone level, SNP in CD36 gene and oral fat detection threshold in human subjects.

BACKGROUND: Horse gram (Macrotyloma uniflorum (Lam.) Verdc.) is an underutilized pulse crop with good drought resistance traits. It is a rich source of protein. Conventional breeding methods for high yielding and abiotic stress tolerant germplasm are hampered by the scarcity of morphological data sets. Thus, horse gram cultivars considered for this study is classified based on prevailing growth factors showing homogenous genotype in various agro ecological zones. Nowadays, several machine learning (ML) methods are used in the field of plant phenotyping. RESULTS: We adopted unsupervised learning techniques from the K-means clustering algorithm to analyze important morphological traits: plant shoot length, total plant height, flowering percentage, number of pods per plant, pod length, number of seeds per plant, and seed length variants between germplasm. Unsupervised clustering revealed that 20 germplasm accessions were grouped in four clusters in which high-yielding traits were predominantly observed in cluster 2. CONCLUSION: These findings could guide ML-based classification to characterize suitable germplasms on the basis of high-yielding varieties for different agro-ecological zones. © 2020 Society of Chemical Industry.

BACKGROUND: Recalcitrant nature is a major constraint for the in vitro regeneration and genetic transformation of leguminous species members. Therefore, an improved genetic transformation in horse gram has been developed via in planta method, in which Agrobacterium strain harboring binary vector pCAMBIA2301 was used for the transformation. Several factors affecting in planta transformations were put forth viz. Agrobacterium cell density, co-cultivation, and sonication combined with vacuum infiltration duration which were optimized. RESULTS: Germinated seeds were sonicated and vacuum infiltrated with different densities of Agrobacterium culture and co-cultivated in half-strength MS medium with 100 µM of acetosyringone for 48 h. Seedlings were washed with cefotaxime and sowed in vermiculite soil for maturation. T1 plants were subjected to histochemical and molecular analysis to ensure transformation efficiency. Among various combinations analyzed, maximum transformation efficiency (20.8%) was attained with seeds of 5 min sonication combined with vacuum infiltration with 0.6 optical density of Agrobacterium culture. CONCLUSIONS: It concludes that a different Agrobacterium cell density with sonication combined with vacuum infiltration has improved transgenic efficiency in horse gram plants. This simple and efficient method is feasible for the stable expression of foreign genes that could be beneficial for future food security.

A major challenge in taste research is to overcome the flavour imperfections in food products and to build nutritious strategies to combat against obesity as well as other related metabolic syndromes. The field of molecular taste research and chemical senses has contributed to an enormous development in understanding the taste receptors and mechanisms of taste perception. Accordingly, the development of taste-modifying compounds or taste modulators that alter the perception of basic taste modalities has gained significant prominence in the recent past. The beneficial aspects of these substances are overwhelming while considering their potential taste-modifying properties. The objective of the present review is to provide an impression about the taste-modulating compounds and their distinctive taste-modifying properties with reference to their targets and proposed mechanisms of action. The present review also makes an effort to discuss the basic mechanism involved in oro-gustatory taste perception as well as on the effector molecules involved in signal transduction downstream to the activation of taste receptors.

Epizootic Ulcerative Syndrome (EUS) caused by Aphanomyces invadans which is a primary fungal parasitic pathogen, inflicts serious economic loss in tropical freshwater fish including snakehead murrel, Channa striatus. In the present study with an aim to circumvent the adverse effects of the traditional measures in graded levels (2%, 4%, and 6%) of Zeolite enriched diet on growth performance, hematology, immunological response, and disease resistance in C. striatus against A. invadans is reported. The final weight (FW), specific growth rate (SGR), feed conversion ratio (FCR), protein efficiency ratio (PER), and average daily gain (ADG) were significantly high in infected fish fed with 4% or 6% Zeolite incorporated diets on 4th week. The maximum survival rates (SR) of 96% and 98% were observed when fed with 2% or 4% diets on 4th week. Similarly, the white blood cell (WBC), red blood cell (RBC), hematocrit (Hct), mean corpuscular volume (MCV), and mean corpuscular haemoglobin concentration (MCHC) were significantly high when fed with any Zeolite enriched diet. However, the haemoglobin (Hb) and mean corpuscular haemoglobin (MCH) were significantly high with 4% and 6% Zeolite diets. The total protein and globulin were significantly high with 4% and 6% diets; the albumin, glucose, cholesterol, and triglyceride were significantly elevated with any enriched diet. The 4% and 6% Zeolite diets significantly enhanced the phagocytic activity on 2nd week but the 2% diet could increase it on 4th week. The respiratory burst (RB) activity, complement activity, and lymphocyte proliferation level were significantly enhanced with 4% and 6% Zeolite diets on weeks 1 and 2 while with 2% diet on 4th week. All enriched diets significantly increased the lysozyme activity during the experimental period. Superoxide anion (SOA) production significantly enhanced with 6% diet on weeks 1 and 2 whereas with 2% diet on week 4. Lower cumulative mortality of 10% and 15% was found with 4% and 6% Zeolite diets whereas a higher mortality (20%) was observed with 2% diet. The present study suggests that dietary supplementation with 4% or 6% Zeolite significantly increase growth performance, hemato-biochemical changes, immunological response, and disease resistance in C. striatus against A. invadans.

BACKGROUND: In plants, carotenoids serve as the precursors to C13-norisoprenoids, a group of apocarotenoid compounds with diverse biological functions. Enzymatic cleavage of carotenoids catalysed by members of the carotenoid cleavage dioxygenase (CCD) family has been shown to produce a number of industrially important volatile flavour and aroma apocarotenoids including ß-ionone, geranylacetone, pseudoionone, α-ionone and 3-hydroxy-ß-ionone in a range of plant species. Apocarotenoids contribute to the floral and fruity attributes of many wine cultivars and are thereby, at least partly, responsible for the "varietal character". Despite their importance in grapes and wine; carotenoid cleavage activity has only been described for VvCCD1 and the mechanism(s) and regulation of carotenoid catabolism remains largely unknown. RESULTS: Three grapevine-derived CCD-encoding genes have been isolated and shown to be functional with unique substrate cleavage capacities. Our results demonstrate that the VvCCD4a and VvCCD4b catalyse the cleavage of both linear and cyclic carotenoid substrates. The expression of VvCCD1, VvCCD4a and VvCCD4b was detected in leaf, flower and throughout berry development. VvCCD1 expression was constitutive, whereas VvCCD4a expression was predominant in leaves and VvCCD4b in berries. A transgenic population with a 12-fold range of VvCCD1 expression exhibited a lack of correlation between VvCCD1 expression and carotenoid substrates and/or apocarotenoid products in leaves, providing proof that the in planta function(s) of VvCCD1 in photosynthetically active tissue is distinct from the in vitro activities demonstrated. The isolation and functional characterisation of VvCCD4a and VvCCD4b identify two additional CCDs that are functional in grapevine. CONCLUSIONS: Taken together, our results indicate that the three CCDs are under various levels of control that include gene expression (spatial and temporal), substrate specificity and compartmentalisation that act individually and/or co-ordinately to maintain carotenoid and volatile apocarotenoid levels in plants. Altering the expression of VvCCD1 in a transgenic grapevine population illustrated the divergence between the in vitro enzyme activity and the in planta activity of this enzyme, thereby contributing to the efforts to understand how enzymatic degradation of carotenoids involved in photosynthesis occurs. The identification and functional characterisation of VvCCD4a and VvCCD4b suggest that these enzymes are primarily responsible for catalysing the cleavage of plastidial carotenoids.
