Larvae of two species of click beetle, Agriotes obscurus and Agriotes lineatus, are important pests of vegetable and field crops in both Europe and North America. Both species have been long and extensively studied, but to date little is known regarding the maturation and egg development of female beetles relative to their swarming periods. This knowledge is important for developing wireworm management tactics that target female beetles, as these would ideally eliminate the beetles before they oviposit. This paper is an attempt to address this knowledge gap. We dissected 2,450 female A. obscurus and 477 female A. lineatus collected in southwestern British Columbia in 2015-2020, and describe how their abdominal lipid content and number of mature eggs change during their swarming period. Based on the presence of mature and immature eggs, active ovarioles, and lipid content, we propose 6 consecutive beetle maturity stages. Beetles collected early in the season have high lipid content and no eggs. Over time, immature, then mature eggs appear and the lipid content decreases dramatically. Ovarioles are generally active throughout the swarming period, even when lipids are no longer present, suggesting that for these species egg laying may continue until the end of the season, and that fecundity depends both on a beetle's original lipid content at emergence, and subsequent diet.

Populations of various economic species of wireworms are increasing in the key cereal crop production areas of Canada and the United States. To address this problem, seed treatments are under development that both provide crop protection and significantly reduce populations equivalent in effectiveness to the formerly used but now deregistered organochlorine lindane. Herein, we evaluated isocycloseram (PLINAZOLIN technology), the first isoxazoline (GABA-gated Chloride Channel Allosteric Modulator) agricultural insecticide, as a seed treatment for the protection of cereal crops from the sugarbeet wireworm, Limonius californicus (Mannerheim). In wheat and barley field trials conducted over 4 years under extreme wireworm pressure, isocycloseram applied as a seed treatment at 5.0-7.5 g AI/100 kg seed was as effective as or more effective than the current industry standard thiamethoxam at 20.0 g AI/100 kg seed in protecting crop stand and yield. Isocycloseram also reduced neonate wireworms (produced from eggs during the growing season) and resident wireworms (in the field at the time of planting) to levels expected from the formerly used seed treatment lindane.

Accurate potential energy models of proteins must describe the many different types of noncovalent interactions that contribute to a protein's stability and structure. Pi-pi contacts are ubiquitous structural motifs in all proteins, occurring between aromatic and nonaromatic residues and play a nontrivial role in protein folding and in the formation of biomolecular condensates. Guided by a geometric criterion for isolating pi-pi contacts from classical molecular dynamics simulations of proteins, we use quantum mechanical energy decomposition analysis to determine the molecular interactions that stabilize different pi-pi contact motifs. We find that neutral pi-pi interactions in proteins are dominated by Pauli repulsion and London dispersion rather than repulsive quadrupole electrostatics, which is central to the textbook Hunter-Sanders model. This results in a notable lack of variability in the interaction profiles of neutral pi-pi contacts even with extreme changes in the dielectric medium, explaining the prevalence of pi-stacked arrangements in and between proteins. We also find interactions involving pi-containing anions and cations to be extremely malleable, interacting like neutral pi-pi contacts in polar media and like typical ion-pi interactions in nonpolar environments. Like-charged pairs such as arginine-arginine contacts are particularly sensitive to the polarity of their immediate surroundings and exhibit canonical pi-pi stacking behavior only if the interaction is mediated by environmental effects, such as aqueous solvation.

Biologic drug discovery pipelines are designed to deliver protein therapeutics that have exquisite functional potency and selectivity while also manifesting biophysical characteristics suitable for manufacturing, storage, and convenient administration to patients. The ability to use computational methods to predict biophysical properties from protein sequence, potentially in combination with high throughput assays, could decrease timelines and increase the success rates for therapeutic developability engineering by eliminating lengthy and expensive cycles of recombinant protein production and testing. To support development of high-quality predictive models for antibody developability, we designed a sequence-diverse panel of 83 effector functionless IgG1 antibodies displaying a range of biophysical properties, produced and formulated each protein under standard platform conditions, and collected a comprehensive package of analytical data, including in vitro assays and in vivo mouse pharmacokinetics. We used this robust training data set to build machine learning classifier models that can predict complex protein behavior from these data and features derived from predicted and/or experimental structures. Our models predict with 87% accuracy whether viscosity at 150 mg/mL is above or below a threshold of 15 centipoise (cP) and with 75% accuracy whether the area under the plasma drug concentration-time curve (AUC0-672 h) in normal mouse is above or below a threshold of 3.9 × 106 h x ng/mL.

Wireworms are primary pests of potatoes in Canada. Presently, the highly toxic organophosphate phorate (i.e., Thimet 20G) is the only effective insecticide in use in Canada. As such, there is an urgent need for novel alternative treatments that provide competitive tuber blemish protection and wireworm reduction with a safer human and environmental portfolio. Herein we evaluated broflanilide, a novel meta-diamide insecticide for both tuber protection and wireworm mortality. When evaluated in field trials in Agassiz, British Columbia over 6 yr, broflanilide applied as a seed piece treatment (SPT) to mother tubers at 1.5-2.0 g AI/100 kg seed (approx. 50 g AI/ha), or as an in-furrow spray (IFS) at 0.23-0.25 g AI/100 m row (approx. 25 g AI/ha) was as effective at reducing blemishes to daughter tubers by wireworms (Agriotes obscurus) as phorate (Thimet 20G at 3230 g AI/ha), bifenthrin (Capture 2EC IFS at 300 g AI/ha) and clothianidin (Titan ST at 312.5 g AI/ha). In addition, broflanilide SPT and IFS applied at the above rates reduced resident wireworms (in the field at the time of planting) by 95.4-99.0% and neonate wireworms (produced from eggs laid during the growing season) by 98.1-100%. Similar results were obtained when broflanilide IFS (nonsystemic) was paired with clothianidin SPT (systemic) for broad-spectrum potato insect pest control. Strategies for the use of broflanilide on wheat (e.g., Teraxxa F4) in rotation with potatoes (Cimegra), both registered in Canada in 2020 are discussed.

Protein phase separation is increasingly understood to be an important mechanism of biological organization and biomaterial formation. Intrinsically disordered protein regions (IDRs) are often significant drivers of protein phase separation. A number of protein phase-separation-prediction algorithms are available, with many being specific for particular classes of proteins and others providing results that are not amenable to the interpretation of the contributing biophysical interactions. Here, we describe LLPhyScore, a new predictor of IDR-driven phase separation, based on a broad set of physical interactions or features. LLPhyScore uses sequence-based statistics from the RCSB PDB database of folded structures for these interactions, and is trained on a manually curated set of phase-separation-driving proteins with different negative training sets including the PDB and human proteome. Competitive training for a variety of physical chemical interactions shows the greatest contribution of solvent contacts, disorder, hydrogen bonds, pi-pi contacts, and kinked beta-structures to the score, with electrostatics, cation-pi contacts, and the absence of a helical secondary structure also contributing. LLPhyScore has strong phase-separation-prediction recall statistics and enables a breakdown of the contribution from each physical feature to a sequence's phase-separation propensity, while recognizing the interdependence of many of these features. The tool should be a valuable resource for guiding experiments and providing hypotheses for protein function in normal and pathological states, as well as for understanding how specificity emerges in defining individual biomolecular condensates.

The power of structural information for informing biological mechanisms is clear for stable folded macromolecules, but similar structure-function insight is more difficult to obtain for highly dynamic systems such as intrinsically disordered proteins (IDPs) which must be described as structural ensembles. Here, we present IDPConformerGenerator, a flexible, modular open-source software platform for generating large and diverse ensembles of disordered protein states that builds conformers that obey geometric, steric, and other physical restraints on the input sequence. IDPConformerGenerator samples backbone phi (φ), psi (ψ), and omega (ω) torsion angles of relevant sequence fragments from loops and secondary structure elements extracted from folded protein structures in the RCSB Protein Data Bank and builds side chains from robust Monte Carlo algorithms using expanded rotamer libraries. IDPConformerGenerator has many user-defined options enabling variable fractional sampling of secondary structures, supports Bayesian models for assessing the agreement of IDP ensembles for consistency with experimental data, and introduces a machine learning approach to transform between internal and Cartesian coordinates with reduced error. IDPConformerGenerator will facilitate the characterization of disordered proteins to ultimately provide structural insights into these states that have key biological functions.

Thick, viscous respiratory secretions are a major pathogenic feature of COVID-19, but the composition and physical properties of these secretions are poorly understood. We characterized the composition and rheological properties (i.e., resistance to flow) of respiratory secretions collected from intubated COVID-19 patients. We found the percentages of solids and protein content were greatly elevated in COVID-19 compared with heathy control samples and closely resembled levels seen in cystic fibrosis, a genetic disease known for thick, tenacious respiratory secretions. DNA and hyaluronan (HA) were major components of respiratory secretions in COVID-19 and were likewise abundant in cadaveric lung tissues from these patients. COVID-19 secretions exhibited heterogeneous rheological behaviors, with thicker samples showing increased sensitivity to DNase and hyaluronidase treatment. In histologic sections from these same patients, we observed increased accumulation of HA and the hyaladherin versican but reduced tumor necrosis factor-stimulated gene-6 staining, consistent with the inflammatory nature of these secretions. Finally, we observed diminished type I interferon and enhanced inflammatory cytokines in these secretions. Overall, our studies indicated that increases in HA and DNA in COVID-19 respiratory secretion samples correlated with enhanced inflammatory burden and suggested that DNA and HA may be viable therapeutic targets in COVID-19 infection.

We evaluated the relative efficacy of six pheromone-baited traps used in trapping Agriotes obscurus (L.) click beetles (Coleoptera: Elateridae): original Yatlor Traps, Yatlor Funnel Traps, Vernon Beetle Traps, Unitraps, Baited Pitfall Traps, and Vernon Pitfall Traps. Traps were rated according to quantitative and qualitative criteria of importance for each of four trap uses: general surveys, scientific studies, IPM monitoring, and mass trapping. Measurable quantitative categories included: total catch of A. obscurus; time for assembly, installation, and inspection; exclusion of nontarget invertebrates; and cost. Qualitative criteria were small mammal exclusion, flooding, design and handling variability, and convenience for various field uses. The most desirable characteristics were determined for the above four uses, and the cumulative ranking based on quantitative criteria and all four uses was Vernon Pitfall Trap, Baited Pitfall Trap, Original Yatlor Trap, Vernon Beetle Trap, Yatlor Funnel Trap, and Unitrap.

Although many studies have focused on a role for hyaluronan (HA) of interstitial extracellular matrix (presumably produced by non-vascular "stromal" cells) in regulating vascular growth, we herein examine the influence of "autocrine HA" produced by vascular endothelial cells themselves on tubulogenesis, using human umbilical vein endothelial cells (HUVECs) in angiogenic and vasculogenic three-dimensional collagen gel cultures. Relative to unstimulated controls, tubulogenic HUVECs upregulated HAS2 mRNA and increased the synthesis of cell-associated HA (but not HA secreted into media). Confocal microscopy/immunofluorescence on cultures fixed with neutral-buffered 10% formalin (NBF) revealed cytoplasmic HAS2 in HUVEC cords and tubes. Cultures fixed with NBF (with cetylpyridinium chloride added to retain HA), stained for HA using "affinity fluorescence" (biotinylated HA-binding protein with streptavidin-fluor), and viewed by confocal microscopy showed HA throughout tube lumens, but little/no HA on the abluminal sides of the tubes or in the surrounding collagen gel. Lumen formation in angiogenic and vasculogenic cultures was strongly suppressed by metabolic inhibitors of HA synthesis (mannose and 4-methylumbelliferone). Hyaluronidase strongly inhibited lumen formation in angiogenic cultures, but not in vasculogenic cultures (where developing lumens are not open to culture medium). Collectively, our results point to a role for autocrine, luminal HA in microvascular sprouting and lumen development. (J Histochem Cytochem 69: 415-428, 2021).

Many pairwise additive force fields are in active use for intrinsically disordered proteins (IDPs) and regions (IDRs), some of which modify energetic terms to improve the description of IDPs/IDRs but are largely in disagreement with solution experiments for the disordered states. This work considers a new direction-the connection to configurational entropy-and how it might change the nature of our understanding of protein force field development to equally well encompass globular proteins, IDRs/IDPs, and disorder-to-order transitions. We have evaluated representative pairwise and many-body protein and water force fields against experimental data on representative IDPs and IDRs, a peptide that undergoes a disorder-to-order transition, for seven globular proteins ranging in size from 130 to 266 amino acids. We find that force fields with the largest statistical fluctuations consistent with the radius of gyration and universal Lindemann values for folded states simultaneously better describe IDPs and IDRs and disorder-to-order transitions. Hence, the crux of what a force field should exhibit to well describe IDRs/IDPs is not just the balance between protein and water energetics but the balance between energetic effects and configurational entropy of folded states of globular proteins.

The extracellular matrix glycosaminoglycan hyaluronan (HA) accumulates in human and mouse islets during the onset of autoimmune type 1 diabetes (T1D). HA plays a critical role in T1D pathogenesis, as spontaneous disease is blocked in mice fed the HA synthesis inhibitor 4-methylumbelliferone (4MU). The present study demonstrates the involvement of HA in T cell-mediated autoimmune responses to transplanted islets and in in vivo and in vitro T cell activation. Scaffolded islet implants (SIs) loaded with RIP-mOVA mouse islets expressing chicken ovalbumin (OVA) on their ß cells were grafted into T and B cell-deficient RIP-mOVA mice, which subsequently received CD4+ T cells from DO11.10 transgenic mice bearing OVA peptide-specific T cell receptors (TcRs), followed by injection of OVA peptide to induce an immune response to the OVA-expressing islets. By affinity histochemistry (AHC), HA was greatly increased in grafted islets with T cell infiltrates (compared to islets grafted into mice lacking T cells) and a portion of this HA co-localized with the infiltrating T cells. Transferred T cells underwent HA synthase (HAS) isoform switching - T cells isolated from the SI grafts strongly upregulated HAS1 and HAS2 mRNAs and downregulated HAS3 mRNA, in contrast to T cells from graft-draining mesenteric lymph nodes, which expressed HAS3 mRNA only. Expression of HAS1 and HAS2 proteins by T cells in SI infiltrates was confirmed by immunohistochemistry (IHC). DO11.10 mice fed 4MU had suppressed in vivo T cell immune priming (measured as a reduced recall response to OVA peptide) compared to T cells from control mice fed a normal diet. In co-cultures of naïve DO11.10 T cells and OVA peptide-loaded antigen-presenting cells (APCs), pre-exposure of the T cells (but not pre-exposure of APCs) to 4MU inhibited early T cell activation (CD69 expression). In addition, T cells exposed to 4MU during activation in vitro with anti-CD3/CD28 antibodies had inhibited phosphorylation of the CD3ζ subunit of the TcR, a very early event in TcR signaling. Collectively, our results demonstrate that T cell-derived HA plays a significant role in T cell immune responses, and that expression of T cell HAS isoforms changes in a locale-specific manner during in vivo priming and functional phases of the T cell response.

We present findings of a general survey of pest wireworms in Alberta, Saskatchewan, and Manitoba conducted from 2004 to 2019; the first such survey of the Canadian Prairie provinces since that published by Glen et al. (1943). Samples were collected from 571 farmland locations where crop damage from wireworms was observed or suspected, and a total of 5,704 specimens (3,548 larvae, 2,156 beetles) were identified. Most specimens (96.9%) were identified as Hypnoidus bicolor (3,278), Selatosomus aeripennis destructor (1,280), Limonius californicus (842), and Aeolus mellillus (125). This suggests that H. bicolor has replaced S. a. destructor as the predominant species and that the relative importance of L. californicus as a pest species has increased since earlier reports. Despite the relatively small number of specimens collected per location (approx. 10), H. bicolor and S. a. destructor, and S. a. destructor and L. californicus were frequently collected at or near the same location (within 1 km). We provide species records and incidence of co-occurrence at different spatial scales, discuss potential reasons for and implications of shifts in species composition, implications of species co-occurrence for managing wireworm pests in crop production, and outline pertinent research needs. A path analysis approach used to correlate incidence of the main species with various soil characteristics indicated that organic matter, cation exchange capacity, and water retention capacity all had a significant species-specific influence on wireworm presence.

A coat of pericellular hyaluronan surrounds mature dendritic cells (DC) and contributes to cell-cell interactions. We asked whether 4-methylumbelliferone (4MU), an oral inhibitor of HA synthesis, could inhibit antigen presentation. We find that 4MU treatment reduces pericellular hyaluronan, destabilizes interactions between DC and T-cells, and prevents T-cell proliferation in vitro and in vivo. These effects were observed only when 4MU was added prior to initial antigen presentation but not later, consistent with 4MU-mediated inhibition of de novo antigenic responses. Building on these findings, we find that 4MU delays rejection of allogeneic pancreatic islet transplant and allogeneic cardiac transplants in mice and suppresses allogeneic T-cell activation in human mixed lymphocyte reactions. We conclude that 4MU, an approved drug, may have benefit as an adjunctive agent to delay transplantation rejection.

Following the deregistration of lindane, several neonicotinoid insecticides have been registered as seed treatments for controlling wireworms in cereal crops. Unlike lindane, which did both, neonicotinoids provide crop protection but do not reduce wireworm populations. Hence populations of various economic species are growing in key wheat production areas of the United States and Canada, and there is a need for novel seed treatments that match the efficacy of lindane. Herein we evaluated broflanilide, a novel GABA-gated Cl- channel allosteric modulator that differs from the cyclodienes (e.g., lindane) in that it binds to a unique site in the GABA receptor. As such, broflanilide confers a new mode of insecticidal action (Group 30 MOA), and is the first meta-diamide insecticide developed. When evaluated in field trials over 7 yr at various rates, broflanilide at 5.0 g [AI]/100 kg wheat seed was as effective at protecting wheat stand from wireworm (Agriotes obscurus) injury as the current industry standard thiamethoxam at 20-30 g [AI]/100 kg seed. In addition, broflanilide at 5.0 g reduced neonate wireworms (produced from eggs laid in established wheat during the growing season) and resident wireworms (in the field at time of planting) by 73.1 and 81.1%, respectively, which is comparable to that reported for the previous industry standard lindane (75.3 and 57.6%, respectively). These studies show that broflanilide at 5.0 g [AI] will provide consistent wheat stand protection (equal to thiamethoxam at 20-30 g [AI]), and A. obscurus wireworm population reduction (equal to lindane at 59 g [AI]), and will do so at far lower dosages per hectare.

Endeavoring toward a transferable, predictive coarse-grained explicit-chain model for biomolecular condensates underlain by liquid-liquid phase separation (LLPS) of proteins, we conducted multiple-chain simulations of the N-terminal intrinsically disordered region (IDR) of DEAD-box helicase Ddx4, as a test case, to assess roles of electrostatic, hydrophobic, cation-π, and aromatic interactions in amino acid sequence-dependent LLPS. We evaluated three different residue-residue interaction schemes with a shared electrostatic potential. Neither a common hydrophobicity scheme nor one augmented with arginine/lysine-aromatic cation-π interactions consistently accounted for available experimental LLPS data on the wild-type, a charge-scrambled, a phenylalanine-to-alanine (FtoA), and an arginine-to-lysine (RtoK) mutant of Ddx4 IDR. In contrast, interactions based on contact statistics among folded globular protein structures reproduce the overall experimental trend, including that the RtoK mutant has a much diminished LLPS propensity. Consistency between simulation and experiment was also found for RtoK mutants of P-granule protein LAF-1, underscoring that, to a degree, important LLPS-driving π-related interactions are embodied in classical statistical potentials. Further elucidation is necessary, however, especially of phenylalanine's role in condensate assembly because experiments on FtoA and tyrosine-to-phenylalanine mutants suggest that LLPS-driving phenylalanine interactions are significantly weaker than posited by common statistical potentials. Protein-protein electrostatic interactions are modulated by relative permittivity, which in general depends on aqueous protein concentration. Analytical theory suggests that this dependence entails enhanced interprotein interactions in the condensed phase but more favorable protein-solvent interactions in the dilute phase. The opposing trends lead to only a modest overall impact on LLPS.

Hyaluronan and proteoglycan link protein 1 (HAPLN1) stabilizes interactions between two important extracellular matrix (ECM) macromolecules, versican and hyaluronan, which facilitate proliferation of fibroblasts and their conversion to myofibroblasts. However, the role of HAPLN1 in these events has not been studied. Using immunocytochemistry, cellular and ECM locations of HAPLN1 were evaluated in cultured human lung fibroblasts during proliferation and conversion to myofibroblasts. HAPLN1 localized to pericellular matrices, associating with both versican and hyaluronan in the ECM and on the cell surface. Nuclear and total HAPLN1 immunostaining increased after myofibroblast induction. Confocal microscopy showed HAPLN1 predominant in the ECM under cells while versican predominated above cells. Versican and HAPLN1 were also juxtaposed in columnar inclusions in the cytoplasm and nucleus. Nuclear HAPLN1 staining in interphase cells redistributed to the cytosol during mitosis. In the absence of TGF-ß1, addition of exogenous bovine HAPLN1 (together with aggrecan G1) facilitated myofibroblast formation, as seen by significant upregulation of α-smooth muscle actin (SMA) staining, while adding full-length bovine versican had no effect. Increased compaction of hyaluronan-rich ECM suggests that HAPLN1 plus G1 addition affects hyaluronan networks and myofibroblast formation. These observations demonstrate changes in both extracellular and intracellular localization of HAPLN1 during fibroblast proliferation and myofibroblast conversion suggesting a possible role in fibrotic remodeling.

BACKGROUND: The microvasculature (MV) of brains with Alzheimer's disease neuropathologic change (ADNC) and cerebral amyloid angiopathy (CAA), in the absence of concurrent pathologies (e.g., infarctions, Lewy bodies), is incompletely understood. OBJECTIVE: To analyze microvascular density, diameter and extracellular matrix (ECM) content in association with ADNC and CAA. METHODS: We examined samples of cerebral cortex and isolated brain microvasculature (MV) from subjects with the National Institute on Aging-Alzheimer's Association (NIA-AA) designations of not-, intermediate-, or high ADNC and from subjects with no CAA and moderate-severe CAA. Cases for all groups were selected with no major (territorial) strokes, ≤ 1 microinfarct in screening sections, and no Lewy body pathology. MV density and diameter were measured from cortical brain sections. Levels of basement membrane (BM) ECM components, the protein product of TNF-stimulated gene-6 (TSG-6), and the ubiquitous glycosaminoglycan hyaluronan (HA) were assayed by western blots or HA ELISA of MV lysates. RESULTS: We found no significant changes in MV density or diameter among any of the groups. Levels of BM laminin and collagen IV (col IV) were lower in MV isolated from the high ADNC vs. not-ADNC groups. In contrast, BM laminin was significantly higher in MV from the moderate-severe CAA vs. the no CAA groups. TSG-6 and HA content were higher in the presence of both high ADNC and CAA, whereas levels of BM fibronectin and perlecan were similar among all groups. CONCLUSIONS: Cortical MV density and diameter are not appreciably altered by ADNC or CAA. TSG-6 and HA are increased in both ADNC and CAA, with laminin and col IV decreased in the BM of high ADNC, but laminin increased in moderate-severe CAA. These results show that changes in the ECM occur in AD and CAA, but independently of one another, and likely reflect on the regional functioning of the brain microvasculature.
