RESUMO

Complicated chemical reactions occur in the decoction of traditional Chinese medicines(TCMs) which features complex components, influencing the safety, efficacy, and quality controllability of TCMs. Therefore, it is particularly important to clarify the chemical reaction mechanism of TCMs in the decoction. This study summarized eight typical chemical reactions in the decoction of TCMs, such as substitution reaction, redox reaction, isomerization/stereoselective reaction, complexation, and supramolecular reaction. With the "toxicity attenuation and efficiency enhancement" of aconitines and other examples, this study reviewed the reactions in decoction of TCMs, which was expected to clarify the variation mechanisms of key chemical components in this process and to help guide medicine preparation and safe and rational use of medicine in clinical settings. The current main research methods for chemical reaction mechanisms of decoction of TCMs were also summed up and compared. The novel real-time analysis device of decoction system for TCMs was found to be efficient and simple without the pre-treatment of samples. This device provides a promising solution, which has great potential in quantity evaluation and control of TCMs. Moreover, it is expected to become a foundational and exemplary research tool, which can advance the research in this field.

Results show that the astrocytes can not only listen to the talk of large assemble of neurons but also give advice to the conversations and are significant sources of heterogeneous couplings as well. In the present work, we focus on such regulation character of astrocytes and explore the role of heterogeneous couplings among interacted neuron-astrocyte components in a signal response. We consider reduced dynamics in which the listening and advising processes of astrocytes are mapped into the form of group coupling, where the couplings are normally distributed. In both globally coupled overdamped bistable oscillators and an excitable FitzHugh-Nagumo (FHN) neuron model, we numerically and analytically demonstrate that two types of bell-shaped collective response curves can be obtained as the ensemble coupling strength or the heterogeneity of group coupling rise, respectively, which can be seen as a new type of double resonance. Furthermore, through the bifurcation analysis, we verify that these resonant signal responses stem from the competition between dispersion and aggregation induced by heterogeneous group and positive pairwise couplings, respectively. Our results contribute to a better understanding of the signal propagation in coupled systems with quenched disorder.

A moderate degree of diversity, in form of quenched noise or intrinsic heterogeneity, can significantly strengthen the collective response of coupled extended systems. As yet, related discoveries on diversity-induced resonance are mainly concentrated on symmetrically distributed heterogeneity, e.g., the Gaussian or uniform distributions with zero-mean. The necessary conditions that guarantee the arise of resonance phenomenon in heterogeneous oscillators remain largely unknown. In this work, we show that the standard deviation and the ratio of negative entities of a given distribution jointly modulate diversity-induced resonance and the concomitance of negative and positive entities is the prerequisite for this resonant behavior emerging in diverse symmetrical and asymmetrical distributions. Particularly, for a proper degree of diversity of a given distribution, the collective signal response behaves like a bell-shaped curve as the ratio of negative oscillator increases, which can be termed negative-oscillator-ratio induced resonance. Furthermore, we analytically reveal that the ratio of negative oscillators plays a gating role in the resonance phenomenon on the basis of a reduced equation. Finally, we examine the robustness of these results in globally coupled bistable elements with asymmetrical potential functions. Our results suggest that the phenomenon of diversity-induced resonance can arise in arbitrarily distributed heterogeneous bistable oscillators by regulating the ratio of negative entities appropriately.

OBJECTIVE: To clarify the influence of obstructive sleep apnea (OSA) on postoperative cognitive dysfunction (POCD) in elderly patients undergoing joint replacement. METHODS: This study retrospectively enrolled130 patients who underwent joint replacement in the Department of Orthopaedics of Taizhou Municipal Hospital between January 2019 and March 2021 for analysis. According to polysomnography (PSG) results, 80 patients without OSA were included in group A and 50 with OSA were assigned to group B. The two groups were compared with respect to the following items: surgical indications (length of stay (LOS), intraoperative blood loss (IBL) and operation time (OT), incidence of postoperative delirium (POD), postoperative cognitive function (Mini-mental State Examination, MMSE), neurological function recovery (National Institutes of Health Stroke Scale, NIHSS) and (Scandinavian Stroke Scale, SSS)), mental health (Self-Rating Anxiety Scale (SAS) and Self-Rating Depression Scale (SDS)), compliance, overall response rate (ORR), complications and patient satisfaction. RESULTS: The LOS and OT were shorter, and the IBL was less in group A compared with those in group B. Group A also showed reduced NIHSS and SSS scores as well as SAS and SDS scores when compared with group B. In addition, lower incidence of POD, and higher compliance, ORR and satisfaction were observed in group A than in group B. In terms of cognitive function, although the MMSE score in both groups decreased after surgery, patients in group B had a lower MMSE score and a milder form of POCD. CONCLUSIONS: OSA may affect the postoperative cognitive function and adversely influence the treatment outcome of elderly patients undergoing joint replacement.

AIM: To evaluate the accuracy, efficiency, and reliability of a handheld infrared eccentric autorefractor (hICA) with artificial intelligence (AI) by comparing its refraction measurements to those recorded using hICA and a clinical table-mounted automatic refractor (TAR). METHODS: A cross-sectional study using three optometers, including hICA with or without AI and TAR, for refractometry of adults (aged 19-49 years old) with no signs of ocular disease or trauma in the absence of cycloplegia. Right and left eye refraction data were recorded, including the spherical equivalent (SE), diopter of spherical power (DS), diopter of cylindrical power (DC) decomposed into vectors J0 and J45, and measurement times. To avoid analytical difficulties associated with the interdependence of observations between eyes from the same individual, the Generalized Estimation Equation was used to compare the SE, DS, J0 and J45 measurements, and the times thereof, among the different groups. The intraclass correlation coefficient (ICC) and Spearman's rank correlation coefficient were used to evaluate correlations among the measurements recorded by the three different instruments. Bland-Altman were used to analyze the precision of the equipment by the agreement. RESULTS: A total of 70 patients (140 eyes; mean age: 31.37y; range: 19-49y) were assessed using refractometry. In a brightly lit environment, there was no significant difference between the mean SE recorded using TAR and that recorded using hICA with AI or without AI (both P>0.05). In an intense-light environment, hICA equipped with AI showed a better detection rate than without AI. Light intensity had a greater effect on dioptric measurements recorded using hICA without AI (P<0.001) than on those recorded using the one equipped with AI (P<0.05). Measurement times varied significantly between the different light intensities and instruments (P<0.05). CONCLUSION: For the normal human eyes, AI may improve the accuracy, efficiency, and reliability of measurements recorded using hICA in various light environments.

Macrophages substantially influence the development, progression, and complications of inflammation-driven diseases. Although numerous studies support the critical role of Notch signaling in most inflammatory diseases, there is limited data on the role of Notch signaling in TLR4-induced macrophage activation and interaction of Notch signaling with other signaling pathways in macrophages during inflammation, such as the NF-κB pathway. This study confirmed that stimulation with lipopolysaccharide (LPS), a TLR4 ligand, upregulated Notch1 expression in monocyte/macrophage-like RAW264.7 cells and bone marrow-derived macrophages (BMDMs). LPS also induced increased mRNA expression of Notch target genes Notch1 and Hes1 in macrophages, suggesting that TLR4 signaling enhances activation of the Notch pathway. The upregulation of Notch1, Notch1 intracellular domain (NICD), and Hes1 proteins by LPS treatment was inhibited by DAPT, a Notch1 inhibitor. Additionally, the increased TNF-α, IL-6, and IL-1ß expression induced by LPS was inhibited by DAPT and rescued by jagged1, a Notch1 ligand. Furthermore, suppression of Notch signaling by DAPT upregulated Cylindromatosis (CYLD) expression but downregulated TRAF6 expression, IκB kinase (IKK) α/ß phosphorylation, and subsequently, phosphorylation and degradation of IκB-α, indicating that DAPT inhibited NF-κB activation triggered by TLR-4. Interestingly, DAPT did not inhibit the increased MyD88 expression induced by LPS. Our study findings demonstrate that macrophage stimulation via the TLR4 signaling cascade triggers activation of Notch1 signaling, which regulates the expression patterns of genes involved in pro-inflammatory responses by activating NF-κB. This effect may be dependent on the CYLD-TRAF6-IKK pathway. Thus, Notch1 signaling may provide a therapeutic target against infectious and inflammation-driven diseases.

BACKGROUND AND AIMS: Allyl isothiocyanate(AITC) has been shown to play an important role in the improved symptoms of chronic obstructive pulmonary disease(COPD) and the inhibition of inflammation, but the role in COPD lipid metabolism disorder and the molecular mechanism remains unclear. We aimed to explore whether and how AITC affects COPD by regulating lipid metabolism and inflammatory response. METHODS: The COPD rat model was established by cigarette smoke exposure. Cigarette smoke extract stimulated 16HBE cells to induce a cell model. The effect of AITC treatment was detected by lung function test, H&E staining, Oil red O staining, immunohistochemistry, ELISA, CCK-8, HPLC, fluorescence efflux test, siRNA, RT-PCR, and Western blotting. Biological analysis was performed to analyze the results. Graphpad Prism 8.0 software was used for statistical analysis. RESULTS: AITC can improve lung function and pathological injury in COPD rats. The levels of IL-1 ß and TNF- α in the AITC treatment group were significantly lower than those in the model group(P < 0.05), and the lipid metabolism was also improved (P < 0.05). AITC reverses CSE-induced down-regulation of LXR α, ABCA1, and ABCG1 expression and function in a time-and concentration-dependent manner (P < 0.05). AITC regulates the cholesterol metabolism disorder induced by CSE in NR8383 cells and attenuates macrophage inflammation (P < 0.05). In addition, after silencing LXR α with siRNA, the effect of AITC was also inhibited. CONCLUSION: These results suggest that AITC improves COPD by promoting RCT process and reducing inflammatory response via activating LXR pathways.

The phase transition of epidemic spreading model on networks is one of the most important concerns of physicists to theoretical epidemiology. In this paper, we present an analytical expression of epidemic threshold for interplay between epidemic spreading and human behavior on multiplex networks. The threshold formula proposed in this paper reveals the relation between the threshold on single-layer networks and that on multiplex networks, which means that the theoretical conclusions of single-layer networks can be used to improve the threshold accuracy of multiplex networks. To verify how well our formula works in different networks, we build a network model with constant total number of edges but gradually changing the heterogeneity of the network, from scale-free network to Erdos-Rényi random network. By use of theoretical analysis and computer simulations, we find that the heterogeneity of information layer behaves as a "double-edged sword" on the epidemic threshold: The strong heterogeneity can effectively improve the epidemic threshold (which means the disease outbreak requires a higher infection probability) when the awareness probability α is low, while the opposite effect takes place for high α. Meanwhile, the weak heterogeneity of the information layer is effective in suppressing the epidemic prevalence when the awareness probability is neither too high nor too low.

We investigate the occurrence of synchronous population activities in a neuronal network composed of both excitatory and inhibitory neurons and equipped with short-term synaptic plasticity. The collective firing patterns with different macroscopic properties emerge visually with the change of system parameters, and most long-time collective evolution also shows periodic-like characteristics. We systematically discuss the pattern-formation dynamics on a microscopic level and find a lot of hidden features of the population activities. The bursty phase with power-law distributed avalanches is observed in which the population activity can be either entire or local periodic-like. In the purely spike-to-spike synchronous regime, the periodic-like phase emerges from the synchronous chaos after the backward period-doubling transition. The local periodic-like population activity and the synchronous chaotic activity show substantial trial-to-trial variability, which is unfavorable for neural code, while they are contrary to the stable periodic-like phases. We also show that the inhibitory neurons can promote the generation of cluster firing behavior and strong bursty collective firing activity by depressing the activities of postsynaptic neurons partially or wholly.

Drug addiction is a widespread social problem, which not only brings adverse consequences to the human body, but also causes great burden to the society. However, it's still unclear how the long-term and sustained cocaine exposure will affect clock genes' expression in the reward related brain areas. We hypothesize that chronic cocaine exposure causes changes in the circadian rhythmic expression of clock genes in brain regions associated with reward, since previous studies have shown that cocaine use causes circadian disorders. Sprague-Dawley male rats were administrated with cocaine 20 mg/kg at ZT4 through intraperitoneal injection for 21 consecutive days. Twenty-four hours after the last cocaine administration brain samples were collected at 4-h intervals for 24 h (every 4h: ZT 0; ZT 4; ZT 8; ZT 12; ZT 16; ZT 20) to examine expression of rPer1, rPer2, rPer3, rCry, rBmal1 and rClock by quantitative real-time reverse-transcription polymerase chain reaction (RT-PCR). We found that chronic cocaine exposure to rats resulted in significantly disturbances in expression of clock genes in reward related brain areas compared to saline - treated rats. In cocaine-treated rats, rPer1 expression in the suprachiasmatic nucleus (SCN), prefrontal cortex (PFC) and ventral tegmental area (VTA); rPer2 expression in the nucleus accumbens (NAc) shell and hippocampus; rPer3 expression in the NAc core; rCry expression in the SCN and PFC; rBmal1 expression in the SCN and NAc core showed robust circadian rhythms that were essentially identical to those in control rats. However, robust circadian rhythm in rPer1 expression in the SCN and rCry expression in the PFC was nearly completely phase - reversed in cocaine-treated rats. A blunting of circadian oscillations of rPer1 expression occurred in the NAc core and shell and hippocampus; of rPer2 expression occurred in the SCN, PFC, NAc core and hippocampus; of rPer3 expression occurred in the SCN, PFC, NAc shell, hippocampus and VTA; of rCry expression occurred in the NAc core and shell, hippocampus and VTA; of rBmal1 expression occurred in the PFC, NAc shell, hippocampus and VTA in cocaine - treated rats. These rhythm changes accompanied by significant increase in rPer1, rPer2, rPer3 and rBmal1 in the PFC, rPer1, rPer2 and rBmal1 in the hippocampus; significant decrease in rPer2, rPer3 and rCry in the SCN, rPer3, rCry, rBmal1 and rClock in the NAc core compared to control rats. rClock expression in cocaine - treated rats showed no rhythmic change, identical to control rats.These results suggest that chronic cocaine exposure results in disturbances in clock genes' expression in reward related areas.

In this work, we use the approximate-master-equation approach to study the dynamics of the Kinouchi-Copelli neural model on various networks. By categorizing each neuron in terms of its state and also the states of its neighbors, we are able to uncover how the coupled system evolves with respective to time by directly solving a set of ordinary differential equations. In particular, we can easily calculate the statistical properties of the time evolution of the network instantaneous response, the network response curve, the dynamic range, and the critical point in the framework of the approximate-master-equation approach. The possible usage of the proposed theoretical approach to other spreading phenomena is briefly discussed.

Pigeon paramyxovirus type-1(PPMV-1) is an enzootic in pigeon flocks and causes severe economic losses in the poultry industry in many countries. A PPMV-1 isolate, abbreviated as PPMV-1/QL-01/CH/15, was isolated from a great spotted woodpecker in the northwest region of China in 2015. The complete genome was sequenced, and the results showed that the virus genome was 15,192 nt in length, in the gene arrangement 3'-NP-P-M-F-HN-L-5'. Several amino acid mutations were identified in the functional domains of the F and HN proteins. The pathogenicity index of the isolate was evaluated, and the mean death time (MDT) was 72 h and the intracerebral pathogenicity index (ICPI) was 0.925, indicating that this isolate was mesogenic. Sequencing results showed that it had a virulent Newcastle disease virus cleavage motif 112R-R-Q-K-R-F117 at the fusion protein cleavage site. Morbidity and mortality were 70% and 50%, after inoculation of pigeons, whereas this virus was nonpathogenic in chickens. Different immune responses of pigeons and chickens were induced in vivo, which led to different HI serum antibody titers. The results of phylogenetic and evolutionary distance analysis showed that this PPMV-1 strain belonged to sub-genotype VIa in class II. To our knowledge, this is the first identification and analysis of PPMV-1 co-circulation among wild birds and domestic pigeon flocks in China. The data from this study highlight the important role of wild birds in the dissemination of PPMV-1 and provide useful references for improving our understanding of the distribution and evolution of PPMV-1 in China.
