RESUMO

Drug nephrotoxicity has high fatality rates and complications. To study this conditional, traditionally, Gentamicin (GM) is used to induce acute injury and establish a nephrotic syndrome model. Baicalin, a flavonoid derived from baicalin with potent anti-inflammatory and antioxidant activity, has been used to treat various inflammatory diseases. This study aims to investigate the process of baicalin-2-ethoxyethyl ester (BAE) synthesis and its therapeutic effect on GM-induced acute kidney injury (AKI). Briefly, baicalin was processed by various reactions to yield BAE. A GM-induced AKI model was established for in vivo evaluation of the protective effect and mechanism of BAE. The results indicated that BAE reduced serum creatinine and urea nitrogen levels and improved pathological alterations, inflammatory responses, and oxidative stress in renal tissues. Furthermore, it was revealed that BAE might exert anti-inflammatory and anti-oxidative responses during AKI via the NF-κB signaling pathway regulation. The findings imply that BAE has a protective impact on the kidneys and might serve as a potent medicine for treating renal damage.

Acute lung injury (ALI) is a serious inflammatory disease with high morbidity and mortality. Rosavin is an anti-inflammatory and antioxidant phenylpropanoid and glucoside, which is isolated from Rhodiola rosea L. However, its potential molecular mechanisms and whether it has protective effects against lipopolysaccharide (LPS)-induced ALI remain to be elucidated. To assess the in vitro anti-inflammatory effects and anti-lung injury activity of rosavin, RAW264.7 and A549 cells were stimulated using 1 µg/mL LPS. Rosavin attenuated LPS-induced activation of the TLR-4/NF-κB signaling pathway in RAW264.7 cells and inhibited LPS-induced release of inflammatory factors in A549 cells. A mouse model of acute lung injury was constructed by intraperitoneal injection of 5 mg/kg LPS to observe the therapeutic effect of rosavin. Transcriptomics analysis and Western blot assays were utilized to verify the molecular mechanism, rosavin (20, 40, and 80 mg/kg) dose-dependently ameliorated histopathological alterations, reduced the levels of inflammatory factors, and inhibited the TLR-4/NF-κB/MAPK signaling pathway and apoptosis activation. Rosavin is a promising therapeutic candidate for acute lung injury by inhibiting the TLR-4/NF-κB/MAPK pathway.

With the increasing incidence of chronic kidney disease (CKD), the development of safe and effective anti-renal fibrosis drugs is particularly urgent. Recently, Baicalin has been considered to have a renal protective effect, but its bioavailability is too low. Therefore, we synthesized baicalin-2-ethoxyethyl ester (BAE) by esterification of baicalin. We hope that this experiment will demonstrate the anti-renal fibrosis effect of BAE and explain its molecular mechanism. In this study, the chronic kidney injury model of SD rats was established by 5/6 nephrectomy, and BAE was given for 28 days. The results showed that after BAE treatment, the serum creatinine and urea nitrogen levels decreased significantly, and the pathological changes in kidneys were improved. In addition, RNA-seq analysis showed that the mechanism of BAE in relieving renal fibrosis was related to the ECM receptor, PI3K/AKT signaling pathway, and inflammatory reaction. The western blotting analysis confirmed that BAE could inhibit the expression of α-SMA, TGF-ß1, p-PI3K, p-AKT, p-IκBα, and NF-κB p65. We found that BAE can inhibit the inflammatory reaction and promote the degradation of the extracellular matrix by inhibiting the activation of the PI3K/AKT/NF-κB pathway, thus alleviating the symptoms of renal fibrosis in 5/6Nx rats, which revealed BAE was a potential compound to relieve renal fibrosis effect.

BACKGROUND: Acute pancreatitis (AP) is a severe condition with complications that can impact multiple organ systems throughout the body. Specifically, the diffusion of peripancreatic effusion to the pleural cavity is a significant phenomenon in AP. However, its pathways and implications for disease severity are not fully understood. OBJECTIVE: This study aims to investigate the anatomical routes of peripancreatic effusion diffusion into the pleural cavity in patients with AP and to analyze the correlation between the severity of pleural effusion (PE) and the computed tomography severity index (CTSI) and acute physiology and chronic health evaluation II (APACHE II) scoring system. METHODS: 119 patients with AP admitted to our institution were enrolled in this study (mean age 50 years, 74 male and 45 female). Abdominal CT was performed, and the CTSI and APACHE II index were used to evaluate the severity of the AP, Meanwhile, the prevalence and semiquantitative of PE were also mentioned. The anatomical pathways of peripancreatic effusion draining to pleural were analyzed. Finally, the correlation relationship between the severity of AP and the PE was analyzed. RESULTS: In 119 patients with AP, 74.8% of patients had PE on CT. The anatomic pathways of peripancreatic effusion draining to pleural included esophageal hiatus in 33.7% of patients, aortic hiatus in 6.7% of patients and inferior vena cava hiatus in 3.37% of patients. The rating of PE on CT was correlated with CTSI scores (r= 0.449, P= 0.000) and was slightly correlated with the APACHE II scores (r= 0.197, P= 0.016). CONCLUSION: PE is a common complication of AP, which can be caused by anatomic pathways such as diaphragmatic hiatus. Due to its correlation with the CTSI score, the PE may be a supplementary indicator in determining the severity of AP.

In this article, a very effective method is proposed to measure the permittivity of sheet materials with a high dielectric constant and loss. To meet the constraints of the perturbation method, controlling the magnitude of the perturbation by reducing the sample size is necessary. However, this will cause significant changes in the polarization field within the sample and, consequently, affects the accuracy of the calculations. To solve the problem, the effects of the polarized electric field and the non-uniformity of the electric field in the resonant cavity have been considered and an empirical model is developed based on this using curve fitting techniques. Taking the optimized depolarization factor into the resonant perturbation equation, the dielectric properties of arbitrary length of the sample can be accurately calculated when the frequency shift and the quality factor variation are known. Then, the method is validated and analyzed by numerical simulations. Finally, practical tests are carried out on a variety of materials at different sizes, and the results are stable and in excellent agreement with the simulated ones. Therefore, we can conclude that the method can accurately measure the complex permittivity of high dielectric sheet materials.

RESUMO

In the cellular response to stresses, the tumor suppressor p53 is activated to maintain genomic integrity and fidelity. As a transcription factor, p53 exhibits rich dynamics to allow for discrimination of the type and intensity of stresses and to direct the selective activation of target genes involved in different processes including cell cycle arrest and apoptosis. In this review, we focused on how stresses are encoded into p53 dynamics and how the dynamics are decoded into cellular outcomes. Theoretical modeling may provide a global view of signaling in the p53 network by coupling the encoding and decoding processes. We discussed the significance of modeling in revealing the mechanisms of the transition between p53 dynamic modes. Moreover, we shed light on the crosstalk between the p53 network and other signaling networks. This review may advance the understanding of operating principles of the p53 signaling network comprehensively and provide insights into p53 dynamics-based cancer therapy.

Acute lung injury (ALI) is a common respiratory disease in clinics, which is characterized by alveolar-capillary membrane loss, plasma protein leakage, pulmonary edema, massive neutrophil infiltration, and the release of proinflammatory cytokines and mediators. Rhodiola rosea L. an adaptogenic plant rich in phenylethanoloids, phenylpropanoids, monoterpenes, has anti-inflammatory and antioxidant effects. We hope to verify the relieving effect of total glycosides of Rhodiola rosea L. (RTG) on ALI in mice and clarify its mechanism through this study. In this study, we identified the effect and mechanism of RTG on ALI through LPS-induced ALI mice. After RTG treatment, the pathological structure of lung tissue in ALI mice induced by LPS was significantly improved, and the infiltration of inflammatory cells was reduced. In addition, RTG reduced the production of IL-6, IL-1ß, and TNF-α in the serum of ALI mice and reduced the content or activity of MPO, T-SOD, GSH, and MDA in lung tissue. RNAseq analysis showed that RTG ameliorated LPS-induced ALI through anti-inflammatory, reduced immune response, and anti-apoptotic activities. The western blotting analysis confirmed that RTG could down-regulate the expression levels of TLR4, MyD88, NF-κB p65, and p-IκBα/IκBα. These results suggest that RTG can attenuate LPS-induced ALI through antioxidants and inhibition of the TLR4/NF-κB pathway.

Recently, electromagnetic radiation is a serious threat to equipment accuracy, military safety and human health. The combination with different materials to fabricate absorber composites with well-designed morphology is expected to ameliorate this issue. In here, CuS/Fe3O4@polypyrrole (CuS/Fe3O4@PPy) flower-like composites are constructed by the combination of hydrothermal method, solvothermal method and in-situ polymerization. CuS with flower-like structure consisting of nanosheets can provide a conductive backbone and large specific surface area. Hollow Fe3O4 microspheres play a key role in deciding magnetic loss, and electromagnetic waves can penetrate their hollow structure, result in multiple reflection and refraction. PPy coating can enhance the combined strength of composite, and effectively consume microwaves by scattering and multiple refraction in the intercalated structure. As expected, the minimum reflection loss (RLmin) of CuS/Fe3O4@PPy composites is -74.12 dB at 8.16 GHz with a thickness of 2.96 mm, and the effective absorption bandwidth (EAB) is 4.6 GHz (13.4-18.0 GHz) at 1.68 mm. The excellent electromagnetic wave absorption performances are attributed to the synergy effect of different components. This work provides a unique strategy for the structural design of flower-like microspheres in the field of electromagnetic wave absorption.

The clinical application of gentamicin may lead to acute kidney injury (AKI), and the nephrotoxicity of gentamicin is related to the pathological mechanism of several oxidative and inflammatory cytokines. Plant-derived essential oils have good anti-inflammatory and antioxidant properties. This study aimed to clarify the protective effect of Amomum tsao-ko essential oils (AOs) on gentamicin-induced AKI in rats and its possible mechanism. The rat AKI model was induced by intraperitoneal injection of gentamicin. After 14 days of oral AO treatment, the renal function and pathological changes of the kidney tissues were evaluated, and the level of kidney tissue oxidative stress was detected. The content of inflammatory cytokines was measured by ELISA. The expression of ERK1/2, JNK1/2, p38, NF-κB, caspase-3, and Bax/Bcl-2 proteins were estimated by Western blot analysis. The results showed that taking AO reduced the contents of serum urea and creatinine in AKI rats and improve the pathological changes and oxidative stress of the kidney tissue in rats. At the same time, AO reduced inflammation and apoptosis during AKI by regulating the MAPK pathway. The data show that AO has a protective effect on the kidneys and may be a potential drug for treating kidney injury.

Epithelial-to-mesenchymal transition (EMT) plays a significant role in cancer metastasis. A series of models have focused on EMT regulation by TGF-ß network. However, how EMT is regulated under hypoxia is less understood. We developed a model of HIF-1α network to explore the potential link between EMT and the network topology. Our results revealed that three positive feedback loops, composed of HIF-1α and its three targets SNAIL, TWIST, and miR-210, should be sequentially activated to induce EMT under aggravating hypoxia. We suggested that the number of the positive feedback loops is critical for determining the number of stable states in EMT. Our work may advance the understanding of the significance of network topology in the regulation of EMT.

The root cause behind the development of chronic obstructive pulmonary disease (COPD) is cigarette smoke that induces the inflammation of the lung tissue and alveolar destruction. Long-term cigarette smoking can lead to deterioration in lung parenchymal function and cause structural changes in the lung, further resulting in pulmonary fibrosis. Rhodiola rosea L., a traditional medicinal perennial herb, is well known for its numerous pharmacological benefits, including anti-inflammation, antioxidant, antifatigue, antidepressive, and antifibrotic properties. Here, we evaluated the pharmacological effects and mechanisms of the Rhodiola rosea L. (RRL) macroporous resin extract on COPD caused by lipopolysaccharide (LPS) and cigarette smoke (CS) in rats. The RRL significantly improved the pathological structure of the lung tissue. Additionally, RRL decreased the infiltration of inflammatory cells and, subsequently, oxidative stress. Furthermore, the RNAseq assay indicated that RRL attenuated the CS and LPS-induced COPD via anti-inflammatory, antifibrotic, and antiapoptotic activities. Western blot analysis substantiated that the RRL resulted in upregulated levels of Nrf2 and HO-1 as well as downregulated levels of IκBα, NF-κB p65, α-SMA, and TGF-ß1. Interestingly, the RRL could protect rats from CS and LPS-induced COPD by inhibiting the ERK1/2 and Smad3 signaling pathways and apoptosis. Thus, the RRL could attenuate CS and LPS-induced COPD through inflammation inhibition and antioxidant and antifibrosis pathways.

Utilizing the latest SCAN-rVV10 density functional, we thoroughly assess the electrochemical properties of 35 Mo-based ordered double transition metal MXenes, including clean Mo2MC2 (M = Sc, Ti, V, Zr, Nb, Hf, Ta) and surface functionalized structures Mo2MC2T2 (T = H, O, F and OH), for the potential use as anode materials in lithium ion batteries (LIBs). The first principles molecular dynamics simulations in combination with the calculations of the site adsorption preferences for Li atoms on all investigated MXenes reveal that both Li-saturated adsorption structures and theoretical capacities of Mo-based MXenes are fundamentally influenced by the surface terminations. We find that the adsorption of Li atoms on either -OH or -F functionalized MXenes is chemically unstable. In particular, the F-groups prefer to form a separate fluoride layer with Li atoms, detaching from the Mo2MC2 substrates. The Li atoms could form a stable single adsorption layer on the -H, -O and intrinsic MXenes surface, exhibiting theoretical capacities in the range from 121 mA h g-1 to 195 mA h g-1. Besides -F and -OH terminations, the remaining Mo-based MXenes also possess superior flat open circuit voltage (OCV) profiles with the most reversible storage capacity below 1.0 V during the charging/discharging cycles. We further predict the low barrier heights of Li-ion diffusion, at a range of 0.03-0.06 eV for most Mo-based MXenes except -O and -H terminations, exceeding that of graphene or Ti3C2. Furthermore, combining the Vineyard transition state theory (TST) with the phonon spectra obtained from density functional perturbation theory (DFPT), the mean planar diffusion coefficient is calculated to be 2 × 10-8 m2 s-1 at 300 K for intrinsic Mo2MC2 monolayers. Although the overall specific capacity is fundamentally restricted with the relatively heavy molecular mass of MXenes, we conclude that Mo-based structures, especially the intrinsic Mo2MC2 (M = Sc, Ti, V) monolayers, might be promising anode materials from the aspect of fast charging/discharging application for LIBs.

BACKGROUND: Aberrant expression of up-regulated long non-coding RNA [LncRNA highly upregulated in liver cancer (HULC)] has been observed to play an important regulatory role in the development of multiple human diseases. However, the molecular mechanism underlying the role of HULC and miR-377-5p in HCC needs to be urgently explored. METHODS: The mRNA and protein expression levels of HULC were detected by quantitative real-time polymerase chain reaction (qRT-PCR) and western blot in hepatocellular carcinoma (HCC) cell line HB611, HepG2 and H22, respectively. HULC-shRNA was transfected into HepG-2 cells, which were randomly divided into the control, shRNA-NC, and sh-HULC groups. The correlation between HULC and miR-377-5p was analyzed by performing a luciferase reporter assay. The targeting relationship between miR-377-5p and hypoxia-inhibitory factor-1α (HIF-1α) was also investigated using a luciferase reporter assay. Sh-HULC and miR-377-5p inhibitors were transfected either alone or together into HepG2 cells, and which were divided into the control group, the sh-HULC group, the miR-377-5p inhibitor, and the sh-HULC + inhibitor group for subsequent experiments. HepG2 cell proliferation and invasion were measured by 5-Ethynyl-2-Deoxyuridine (EdU) staining and Transwell invasion assay, respectively. Western plot was carried out to detect the protein expression levels of Ki67, PCNA, E-cadherin, and N-cadherin. Tumor xenograft mouse models were established to confirm the effect of HULC down-regulation on the development of HCC in vivo. RESULTS: The mRNA and protein expression levels of HULC were markedly increased, whereas the mRNA expression levels of miR-377-5p were decreased in HCC cell lines. HepG2 cell proliferation and invasion were suppressed in the Sh-HULC group, while miR-377-5p showed the opposite. Further experiments exhibited that miR-377-5p was targeted by HULC, and an negative correlation between HULC and miR-377-5p was observed. Importantly, the in vivo experiments indicated that down-regulation of HULC could inhibit tumor growth. Taken together, our research demonstrated that down-regulation of HULC plays an anti-cancer role through restrainingHepG2 cell proliferation and invasion. CONCLUSIONS: In summary, our in vitro and in vivo findings confirmed HULC to play a role in the progression of HCC, with the underlying mechanism possibly involving the miR-377-5p/HIF-1α pathway.

The quality control of Saussurea involucrata has been greatly improved by macroscopic and microscopic identification and chemical profiling described in Chinese Pharmacopoeia since 2005. However, these methods have their own limitations, e.g., their dependence on personal experience and expertise, and it is a huge challenge to identify closely related species that share similar or identical morphological characteristics and chemical profiles. A novel and generally accepted identification strategy is urgently needed as a complement to regulations for protecting the public health interests. In this work, a comprehensive chromatographic fingerprint method was developed and tested on 43 samples from four haplotypes of S. involucrata according to DNA barcoding. Three common patterns consisting of 20, 14, and 7 common peaks were generated by frequency filters of median, upper quartile, and 100%, respectively. Based on two formerly screened patterns, S. involucrata can be effectively identified from its five easily confused snow lotus species, including the most closely related plant (S. orgaadayi) in the orthogonal partial least-squares discriminant analysis (OPLS-DA) models. The model is supported by good R and Q coefficients. In addition, different haplotypes of S. involucrata can be discriminated in the OPLS-DA model using the 20 common peaks. Among them, peaks 9, 11, 16 (zaluzanin C), and 18 (dehydrocostus lactone) have been identified as fingerprint markers of S. involucrata via S-plots and VIP values (>1). Additionally, peaks 19 and 20 were identified as linolenic acid and linoleic acid with anti-inflammatory activity, and they were isolated from the herb for the first time. Collectively, the chromatographic fingerprint of S. involucrata can be an effective and integrated method for the identification of authentic herbs from adulterant species or related plants, and discrimination of its different haplotypes provides an objective and reliable tool for quality control.

OBJECTIVE: To identify, rapidly and accurately, the chemical composition of the traditional Uighur formulation Baixuan Xiatare (BXXTR-FU). METHODS: We investigated if application of three-stage infrared (IR) spectroscopy enabled identification of the main chemical constituents (and their origins) in BXXTR-FU. RESULTS: The characteristic peaks of herbal material(s) and BXXTR-FU were assigned. In Fourier transform-IR (FT-IR) spectroscopy of BXXTR-FU, peaks at 1616 and 1605 cm-1 of BXXTR-FU were considered to denote anthraquinones and their derivatives; 1066 cm-1 was regarded as the characteristic absorption peak of resin glycosides. In second-derivative IR (SD-IR) spectroscopy, the main carbonyl types of BXXTR-FU in the range 1743-1636 cm-1 were assigned: 1651 cm-1 belonged to the carbonyl stretching vibrations of flavonoids and chromones; 1717 cm-1 belonged to tannins; 1699 cm-1 belonged to carboxylic acids; 1636 cm-1 belonged to anthraquinones and their derivatives. SD-IR spectroscopy further confirmed that the characteristic absorption peaks at 1636, 1618 and 1603 cm-1 could be used as markers that BXXTR-FU contained anthraquinones and their derivatives. Synchronous 2D-IR correlation spectra of chemical groups further confirmed the results of FT-IR and SD-IR spectroscopy. CONCLUSION: Our study strongly supported the necessity and importance of three-stage IR spectroscopy owing to its rapid and accurate identification of herbal formulations.

Wavefield drift or wobbling occurs quite often in coherent scanning systems such as satellite laser communication, laser pointing of high-power lasers, or microscopy. The uncertainty of wavefront positions might result in blurred images or large measurement errors. Here we propose an iterative approach that can retrieve both the drift positions and complex-valued distribution of the wavefield from a ptychographic diffraction intensity dataset. We demonstrate the feasibility and effectiveness of the method in numerical simulation and an optical experiment. The method requires little a priori knowledge and thus would open up new opportunities in many fields.

Compared to far-field ptychography, near-field ptychography can reduce the requirement on the detector dynamic range, while it is able to cover a larger field of view with a fewer number of sample scans. However, its spatial resolution is limited by the detector pixel size. Here, we utilize a pixel-super-resolved approach to overcome this limitation. The method has been applied to four types of experiment configurations using planar and divergent illuminations together with two different cameras with highly contrast specifications. The proposed method works effectively for up-sampling up to 6 times. Meanwhile, it can achieve ∼5.9-fold and ∼3.1-fold resolution improvement over the 6.5-µm and 2.4-µm detector pixel size. We also demonstrate the precisely quantitative phase imaging capability of the method by using a phase resolution target. The presented method is believed to have great potential in X-ray tomography and on-chip flow cytometry.

Using SCAN-rVV10+U, we show Cr2TiC2 and Cr2TiC2T2 (T = -F and -OH) MXenes are moderate band gap semiconductors mostly in the antiferromagnetic state. All investigated MXene structures show large Seebeck coefficients (>400 µV/K), especially Cr2TiC2 (>800 µV/K) and Cr2TiC2F2 (>700 µV/K). The hole relaxation time of p-type Cr2TiC2(OH)2 is found to be ∼8 ps, ensuring its superior electron transport properties in comparison to other investigated MXenes. It is also discovered that the surface functionalization could decrease the phonon thermal conduction and that Cr2TiC2(OH)2 has the smallest lattice thermal conductivity (∼6.5 W/m·K) and the largest electron thermal conduction (>50 W/m·K with n = 1019 cm-3). We predict the ZT value of p-type Cr2TiC2(OH)2 can reach 3.0 at 600 K with the maximum thermoelectric conversion efficiency of 20%. Overall, the thermoelectric property of Cr-based ordered double transition metal MXenes is far superior to that of any known two-dimensional structures in the MXene family.

Cervical cancer is among the most frequent cancer types in women worldwide. Radiotherapy, including external beam radiation and brachytherapy, is one of the commonly used treatment options for cervical cancer. However, the adverse effects of radiation therapy on cervical cancer survival have been poorly investigated with inconclusive results. Therefore, the aim of this study was to determine the suitable radiotherapy modality according to patients' characteristics. A retrospective survival analysis of 44,602 patients was performed using the Surveillance, Epidemiology, and End Results (SEER) database. Multivariate proportional hazard Cox model was used to evaluate the prognostic impact of different radiotherapy modalities, primary surgery, age, TNM stage, and tumor size. Our results indicated that patients without primary surgery, diagnosed at older age (≥45 years' old), at advanced TNM stages (III/IV) or with larger tumor size (≥3 cm) could benefit from radiotherapy. However, radiotherapy was detrimental in patients with primary surgery, diagnosed at younger age (<45 years' old), at earlier TNM stages (I/II) or with smaller tumor size (<3 cm). In addition, external beam radiation was in most cases less effective compared with combined external beam and brachytherapy. These results highlighted the necessity of realizing personalized radiotherapy treatments for patients with cervical cancer.

Idiopathic Pulmonary fibrosis (IPF) is diagnosed as a life-threatening, progressive and incurable lung disease characterized by accumulation of extracellular matrix and myofibroblasts, resulting in the function degradation and structural alterations in normal lung parenchyma. Notably, Pulmonary Fibrosis has been considering as a difficult problem in clinical with high mortality and effective treatment strategies. Rosavin, a benzylPropylene glycoside, is isolated from Rhodiola rosea L., exhibiting nootropic, anti-depressant, anti-cancer, anti-inflammatory and anti-oxidative activities. In this study, we attended to elucidate the pharmacological activity of Rosavin for treatment of pulmonary fibrosis induced by bleomycin in mice. The results indicated that Rosavin could significantly ameliorate the lung index and Pathological structure of mice with Pulmonary fibrosis by bleomycin-induced. Additionally, Rosavin could evidently decreased inflammatory cells infiltration in bronchoalveolar lavage fluid and pro-inflammatory cytokines expression in lung tissue specimens induced by bleomycin. Rosavin could down-regulate the expression of hydroxyproline and malondialdehyde and increased the activities of superoxide dismutase, glutathione peroxidase in lung tissue. The expression of Nrf2 were increased, and the expression of NF-κB p65, TGF-ß1 and α-SMA were inhibited. The findings revealed the protective effects and the primary mechanism of rosavin on bleomycin-induced pulmonary fibrosis, which provided a scientific foundation for Rosavin as a promising candidate for Pulmonary fibrosis treatment.
