OBJECTIVE: This study aimed to evaluate the effects of trilobatin (TLB) on dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) in mice and further explore the underlying mechanisms from the perspectives of signaling pathway and gut microbiota. METHODS: A mouse model of UC was established using DSS. Trilobatin was administered via oral gavage. Disease severity was assessed based on body weight, disease activity index (DAI), colon length, histological detection, inflammation markers, and colonic mucosal barrier damage. Alternations in the NF-κB and PI3K/Akt pathways were detected by marker proteins. High-throughput 16S rRNA sequencing was performed to investigate the gut microbiota of mice. RESULTS: In the DSS-induced UC mice, TLB (30 µg/g) treatment significantly increased the body weight, reduced the DAI score, alleviated colon length shortening, improved histopathological changes in colon tissue, inhibited the secretion and expression of inflammation factors (TNF-α, IL-1ß, and IL-6), and increased the expression of tight-junction proteins (ZO-1 and occludin). Furthermore, TLB (30 µg/g) treatment significantly suppressed the activation of NF-κB pathway and altered the composition and diversity of the gut microbiota, as observed in the variations of the relative abundances of Proteobacteria, Actinobacteriota, and Bacteroidota, in UC mice. CONCLUSION: TLB effectively alleviates DSS-induced UC in mice. Regulation of the NF-κB pathway and gut microbiota contributes to TLB-mediated therapeutic effects. Our study not only identified a novel drug candidate for the treatment of UC, but also enhanced our understanding of the biological functions of TLB.

As social entities, individuals' perception and behaviors are susceptible to the influence of their social groups. Previous research has consistently shown that the group context in which individuals are situated significantly influences their perceptual processing. We aim to investigate whether the group context in which another individual is situated alters our understanding of their visual perception, which holds profound implications for interpersonal interactions. To address this inquiry, we conducted three experiments. In Experiment 1, participants were presented with a visual scene depicting multiple avatars seated around a table, all facing an arrow positioned at the center of the table. They were instructed to adopt the visual perspective of a specific avatar within the group to perceive the arrow's orientation, and then reproduce its orientation from their own perspectives. We found that participants exhibited a bias towards the group's average perspective when reproducing the arrow's orientation. Experiment 2 further demonstrated that reinforced group processing could elicit an earlier appearance of this bias. In Experiment 3, we investigated an alternative explanation positing that the aforementioned bias originated from visual ensemble perception rather than group influence by instructing participants to reproduce the target avatar's position relative to the arrow's orientation. If the bias indeed originated from ensemble perception, it should also manifest in this task. However, the absence of any reproduction bias refuted this possibility. Through these experiments, we demonstrate that our understanding of an individual's perceptual experiences is influenced by the social context in which they are situated, which manifests as a convergence phenomenon.

Accurate comprehension of others' thoughts and intentions is crucial for smooth social interactions, wherein understanding their perceptual experiences serves as a fundamental basis for this high-level social cognition. However, previous research has predominantly focused on the visual modality when investigating perceptual processing from others' perspectives, leaving the exploration of multisensory inputs during this process largely unexplored. By incorporating auditory stimuli into visual perspective-taking (VPT) tasks, we have designed a novel experimental paradigm in which the spatial correspondence between visual and auditory stimuli was limited to the altercentric rather than the egocentric reference frame. Overall, we found that when individuals engaged in explicit or implicit VPT to process visual stimuli from an avatar's viewpoint, the concomitantly presented auditory stimuli were also processed within this avatar-centered reference frame, revealing altercentric cross-modal interactions.

BACKGROUND: Heilongjiang Province is a high-quality japonica rice cultivation area in China. One in ten bowls of Chinese rice is produced here. Increasing yield is one of the main aims of rice production in this area. However, yield is a complex quantitative trait composed of many factors. The purpose of this study was to determine how many genetic loci are associated with yield-related traits. Genome-wide association studies (GWAS) were performed on 450 accessions collected from northeast Asia, including Russia, Korea, Japan and Heilongjiang Province of China. These accessions consist of elite varieties and landraces introduced into Heilongjiang Province decade ago. RESULTS: After resequencing of the 450 accessions, 189,019 single nucleotide polymorphisms (SNPs) were used for association studies by two different models, a general linear model (GLM) and a mixed linear model (MLM), examining four traits: days to heading (DH), plant height (PH), panicle weight (PW) and tiller number (TI). Over 25 SNPs were found to be associated with each trait. Among them, 22 SNPs were selected to identify candidate genes, and 2, 8, 1 and 11 SNPs were found to be located in 3' UTR region, intron region, coding region and intergenic region, respectively. CONCLUSIONS: All SNPs detected in this research may become candidates for further fine mapping and may be used in the molecular breeding of high-latitude rice.

Most studies show that self-processing in schizophrenia is impaired at the supraliminal level. Schizophrenic patients generally lack the ability to prioritize the processing of self-related information, such as their own face. However, some evidence suggests that schizophrenic patients may retain intact subliminal processing abilities even though their conscious experiences are compromised. We conducted the first study exploring schizophrenic patients' subliminal self-face processing. Using a breaking continuous flash suppression (bCFS) paradigm, we interocularly suppressed face images (self, famous, and unknown faces). Participants' reaction times to detect the faces when they broke the suppression were recorded as an index for the subliminal processing of faces. Unlike the healthy controls, schizophrenic patients did not demonstrate a processing advantage for their own face when it broke interocular suppression; only a face familiarity effect was found. These findings contribute to the understanding of self-processing deficits in schizophrenia.

How people process gaze cues from multiple others is an important topic but rarely studied. Our study investigated this question using an adapted gaze cueing paradigm to examine the cueing effect of multiple gazes and its neural correlates. We manipulated gaze directions from two human avatars to be either convergent, created by the two avatars simultaneously averting their gazes to the same direction, or non-convergent, when only one of the two avatars shifted its gaze. Our results showed faster reaction times and larger target-congruency effects following convergent gazes shared by the avatars, compared with the non-convergent gaze condition. These findings complement previous research to demonstrate that observing shared gazes from as few as two persons is sufficient to enhance gaze cueing. Additionally, ERP analyses revealed that (1) convergent gazes evoked both left and right hemisphere N170, while non-convergent gazes evoked N170 mainly in the hemisphere contralateral to the cueing face; (2) effects of target congruency on target-locked N1 and P3 were modulated by gaze convergence. These findings shed light on temporal features of the processing of multi-gaze cues.

Past research on level 2 visual perspective-taking (VPT) has mostly focused on understanding the mental rotation involved when one adopts others' perspective; the mechanisms underlying how the visual world of others is mentally represented remain unclear. In three studies, we addressed this question by adopting a novel VPT task with motion stimuli and exploring the aftereffect on motion discrimination from the self-perspective. Overall the results showed a facilitation aftereffect when participants were instructed to take the avatar's perspective. Meanwhile, participants' self-reported perspective-taking tendencies correlated with the aftereffect for both instructed and spontaneous VPT tasks, when the "to-be-adopted" perspective required the participants to mentally transform their self-body clockwise. Specifically, while facilitation was induced for participants with low self-reported perspective-taking tendencies (e.g., viewing a leftward motion stimulus under another's perspective enhanced subsequent perception of leftward motion from the self-perspective), those with high self-reported perspective-taking tendencies showed an adaptation aftereffect (e.g., viewing a leftward motion stimulus under another's perspective weakened subsequent perception of leftward motion from the self-perspective). For these individuals, the adaptation effect indicated the engagement of direction-selective neurons in processing of the subsequent congruent-direction motion from self's perspective. These findings suggest that motion perception from different perspectives (self vs. another) may share the same direction-selective neural circuitry, and this possibility depends on observers' general perspective-taking tendencies.

BACKGROUND: Drought is one of major abiotic stresses constraining crop productivity worldwide. To adapt to drought stress, plants have evolved sophisticated defence mechanisms. Wild barley germplasm is a treasure trove of useful genes and offers rich sources of genetic variation for crop improvement. In this study, a proteome analysis was performed to identify the genetic resources and to understand the mechanisms of drought tolerance in plants that could result in high levels of tolerance to drought stress. RESULTS: A greenhouse pot experiment was performed to compare proteomic characteristics of two contrasting Tibetan wild barley genotypes (drought-tolerant XZ5 and drought-sensitive XZ54) and cv. ZAU3, in response to drought stress at soil moisture content 10% (SMC10) and 4% (SMC4) and subsequently 2 days (R1) and 5 days (R2) of recovery. More than 1700 protein spots were identified that are involved in each gel, wherein 132, 92, 86, 242 spots in XZ5 and 261, 137, 156, 187 in XZ54 from SMC10, SMC4, R1 and R2 samples were differentially expressed by drought over the control, respectively. Thirty-eight drought-tolerance-associated proteins were identified using mass spectrometry and data bank analysis. These proteins were categorized mainly into photosynthesis, stress response, metabolic process, energy and amino-acid biosynthesis. Among them, 6 protein spots were exclusively expressed or up-regulated under drought stress in XZ5 but not in XZ54, including melanoma-associated antigen p97, type I chlorophyll a/b-binding protein b, glutathione S-transferase 1, ribulosebisphosphate carboxylase large chain. Moreover, type I chlorophyll a/b-binding protein b was specifically expressed in XZ5 (Spots A4, B1 and C3) but not in both of XZ54 and ZAU3. These proteins may play crucial roles in drought-tolerance in XZ5. Coding Sequences (CDS) of rbcL and Trx-M genes from XZ5, XZ54 and ZAU3 were cloned and sequenced. CDS length of rbcL and Trx-M was 1401 bp (the partial-length CDS region) and 528 bp (full-length CDS region), respectively, encoding 467 and 176 amino acids. Comparison of gene sequences among XZ5, XZ54 and ZAU3 revealed 5 and 2 SNPs for rbcL and Trx-M, respectively, with two 2 SNPs of missense mutation in the both genes. CONCLUSIONS: Our findings highlight the significance of specific-proteins associated with drought tolerance, and verified the potential value of Tibetan wild barley in improving drought tolerance of barley as well as other cereal crops.

To reveal grain physio-chemical and proteomic differences between two barley genotypes, Zhenong8 and W6nk2 of high- and low-grain-Cd-accumulation, grain profiles of ultrastructure, amino acid and proteins were compared. Results showed that W6nk2 possesses significantly lower protein content, with hordein depicting the greatest genotypic difference, compared with Zhenong8, and lower amino acid contents with especially lower proportion of Glu, Tyr, Phe and Pro. Both scanning and transmission electron microscopy observation declared that the size of A-type starch molecule in W6nk2 was considerably larger than that of Zhenong8. Grains of Zhenong8 exhibited more protein-rich deposits around starch granules, with some A-type granules having surface pits. Seventeen proteins were identified in grains, using 2-DE coupled with mass spectrometry, with higher expression in Zhenong8 than that in W6nk2; including z-type serpin, serpin-Z7 and alpha-amylase/trypsin inhibitor CM, carbohydrate metabolism, protein synthesis and signal transduction related proteins. Twelve proteins were less expressed in Zhenong8 than that in W6nk2; including barley trypsin inhibitor chloroform/methanol-soluble protein (BTI-CMe2.1, BTI-CMe2.2), trypsin inhibitor, dehydroascorbate reductase (DHAR), pericentrin, dynein heavy chain and some antiviral related proteins. The data extend our understanding of mechanisms underlying Cd accumulation/tolerance and provides possible utilization of elite genetic resources in developing low-grain-Cd barley cultivars.

A hydroponic experiment was conducted to study the ameliorative effects of separate or combined application of exogenous glutathione (GSH), selenium (Se) and zinc (Zn) upon 20 µM cadmium (Cd) plus 20 µM chromium (Cr) heavy metal stress (HM) in rice seedlings. The results showed that HM caused a marked reduction in seedling height, chlorophyll content (SPAD) and biomass, and activities of catalase (CAT) and ascorbate peroxidase (APX) in leaves and H(+)-ATPase in roots/leaves, but elevated superoxide dismutase (SOD) and guaiacol peroxidase (POD) activities in leaves with elevated malondialdehyde (MDA) accumulation both in leaves and roots over the control. The best mitigation effect was recorded in HM+GSH+Zn and HM+GSH (addition of GSH+Zn and GSH to HM solution), which greatly alleviated HM-induced growth inhibition and oxidative stress. Compared with HM alone, HM+GSH and HM+GSH+Zn markedly reduced Cr uptake and translocation but not affected Cd concentration; improved H(+)-ATPase activity and Fe, Zn, Mn uptake and translocation, and repressed MDA accumulation. Meanwhile exogenous GSH and GSH+Zn counteracted HM-induced response of antioxidant enzymes, via suppressing HM-induced dramatic increase of root/leaf SOD and leaf POD activities, and elevating stress-depressed leaf APX and leaf/root CAT activities.

In order to establish a high efficient regeneration system of sunflower, we optimized the process of callus induction, differentiation and rooting by screening the optimum genotype, explant materials, hormone and cytokine concentration and additives. The results indicated that hybrid sunflowers were easier to regenerate than selfing ones; The best explant was four days cotyledon. The optimum induction medium was Murashige and Skoog (MS) + 2.0 mg/L 6-benzyladenine (6-BA) + 0.5 mg/L naphthaleneacetic acid (NAA) + 1.0 mg/L kinetin (KT). The maximum rate of callus induction was 100%. The optimum differentiation medium was MS +0.2 mg/L 6-BA + 0.5 mg/L NAA + 0.3 mg/L KT + 0.3 mg/L silver nitrate (AgNO3) + 0.2 g/L active carbon (AC), and the buds differentiation rate was up to 71%. The best rooting culture medium was 1/2 MS + 0.6 mg/L indolebutyric acid (IBA). The highest rooting rate was 77%. The analysis of variance showed that genotype, explants growth time, different kinds and concentration of hormone, AC concentration had a significant effect on sunflower regeneration.
