RESUMO

BACKGROUND: Previous study implied that local M2 polarization of macrophage promoted mucosal edema and exacerbates Th2 type inflammation in chronic rhinosinusitis with nasal polyps (CRSwNP). However, the specific pathogenic role of M2 macrophages and the intrinsic regulators in the development of CRS remains elusive. OBJECTIVE: We thought to investigate the regulatory role of SIRT5 in the polarization of M2 macrophages and its potential contribution to the development of CRSwNP. METHODS: RT-qPCR and Western blot analyses were performed to examine the expression levels of SIRT5 and markers of M2 macrophages in sinonasal mucosa samples obtained from both CRS and control groups. Wild-type and Sirt5 knockout mice were used to establish nasal polyp model with Th2 inflammation and investigate the effects of SIRT5 in macrophages on disease development. Furthermore, in vitro experiments were conducted to elucidate the regulatory role of SIRT5 in polarization of M2 macrophages. RESULTS: Clinical investigations showed that SIRT5 was highly expressed and positively correlated with M2 macrophages markers in eosinophilic polyps. The expression of SIRT5 in M2 macrophages was found to contribute to the development of the disease, which was impaired in Sirt5 deficiency mice. Mechanistically, SIRT5 was shown to enhance the alternative polarization of macrophages through promoting glutaminolysis. CONCLUSIONS: SIRT5 plays a crucial role in promoting the development of CRSwNP by supporting the alternative polarization of macrophage and thus provides a potential target for CRSwNP interventions.

Urban trees possess different capacities to mitigate ozone (O3) pollution through stomatal uptake. Stomatal closure protects trees from oxidative damage but limits their growth. To date, it is unclear how plant hydraulic function affect stomatal behaviour and determine O3 resistance. We assessed gas exchange and hydraulic traits in three subtropical urban tree species, Celtis sinensis, Quercus acutissima, and Q. nuttallii, under nonfiltered ambient air (NF) and elevated O3 (NF60). NF60 decreased photosynthetic rate (An) and stomatal conductance (gs) only in Q. acutissima and Q. nuttallii. Maintained An in C. sinensis suggested high O3 resistance and was attributed to higher leaf capacitance at the full turgor. However, this species exhibited a reduced stomatal sensitivity to vapour pressure deficit and an increased minimal gs under NF60. Such stomatal dysfunction did not decrease intrinsic water use efficiency (WUE) due to a tight coupling of An and gs. Conversely, Q. acutissima and Q. nuttallii showed maintained stomatal sensitivity and increased WUE, primarily correlated with gs and leaf water relations, including relative water content and osmotic potential at turgor loss point. Our findings highlight a trade-off between O3 resistance and stomatal functionality, with efficient stomatal control reducing the risk of hydraulic failure under combined stresses.

Objective: Differentiating 2 types of chronic rhinosinusitis with nasal polyps (CRSwNP) is important for the treatment. The current diagnostic methods using single indicators, including peripheral blood eosinophils and traditional sinus computed tomography (CT) scores, are not accurate. In this study, we aimed to investigate the diagnostic value of combining peripheral blood eosinophils and improved sinus CT scores for eosinophic chronic rhinosinusitis (ECRS). Study Design: Retrospective cohort. Setting: Tertiary medical center. Methods: We conducted a study involving 81 patients with CRSwNP. Peripheral blood samples were collected from the non-ECRS and ECRS groups. Improved three-dimensional volume image analysis and Lund-Mackay scoring system were performed to quantify the thickening of sinus mucosa. Multivariate binary logistic regression analysis was carried out to detect the predictive value of the scoring indicators. For significant indexes, receiver operating characteristic (ROC) curve analysis was applied. Results: The ECRS group had higher levels of blood eosinophil percentage and count, ethmoid sinus score, total sinus score, the ratio of ethmoid sinus score and maxillary sinus score, and the difference between ethmoid and maxillary score, compared to the non-ECRS group (P < 0.05). Binary logistic regression analysis demonstrated that both blood eosinophil percentage and the improved E - M score (subtraction of ethmoid and maxillary sinus scores) were significant predictors of ECRS diagnosis (P < .01). ROC curve analysis indicated that the combination of improved E - M score and blood eosinophil percentage had a higher diagnostic value compared to either factor alone (area under the curve = 0.874). Conclusion: Our study suggested the combination of improved total ethmoid sinus-maxillary score and blood eosinophil percentage is more accurate in predicting the diagnosis of ECRS.

Rationale: Gustation is important to several biological functions in mammals. However, chemotherapy drugs often harm taste perception in cancer patients, while the underlying mechanism is still unclear for most drugs and there is no effective way to restore taste function. This study investigated the effects of cisplatin on the taste cell homeostasis and gustatory function. Methods: We used both mice and taste organoid models to study the effect of cisplatin on taste buds. Gustometer assay, gustatory nerve recording, RNA-Sequencing, quantitative PCR, and immunohistochemistry was performed to analyze the cisplatin-induced alteration in taste behavior and function, transcriptome, apoptosis, cell proliferation and taste cell generation. Results: Cisplatin inhibited proliferation and promoted apoptosis in the circumvallate papilla, leading to significant impairment in taste function and receptor cell generation. The transcriptional profile of genes associated with cell cycle, metabolic process and inflammatory response was significantly altered after cisplatin treatment. Cisplatin inhibited growth, promoted apoptosis, and deferred taste receptor cell differentiation in taste organoids. LY411575, a γ-secretase inhibitor, reduced the number of apoptotic cells and increased the number of proliferative cells and taste receptor cells, potentially suggesting as a taste tissue protective agent against chemotherapy. LY411575 treatment could offset the increased number of Pax1+ or Pycr1+ cells induced by cisplatin in the circumvallate papilla and taste organoids. Conclusion: This study highlights the inhibitory effects of cisplatin on taste cell homeostasis and function, identifies critical genes and biological processes regulated by chemotherapy, and proposes potential therapeutic targets and strategy for taste dysfunction in cancer patients.

Lake Taihu, located in a densely populated and highly industrialized area in eastern China, has experienced dramatic changes in water quality since the reform and opening-up in the 1980s. Landsat data can be used to trace water quality changes over approximately 40 years. However, chlorophyll-a (Chla) estimation, which characterizes the trophic status, has not been thoroughly explored (especially in turbid water using wide bandwidth Landsat) due to the interference of suspended particulate matter (SPM) to Chla. In this study, we used Landsat TM/OLI for turbid water Chla inversion and to analyze the spatiotemporal variation of Chla in Lake Taihu for 38 years and its influencing factors. An optical classification algorithm based on Rrs(green)/Rrs(red) was used to exclude highly turbid waters dominated by SPM; Chla was estimated only in waters with low SPM. We constructed an exponential estimation model based on Rrs(NIR)/Rrs(red), and verified the accuracy of the model using the measured Chla synchronized with satellite data. The model was applied to Landsat images to calculate the Chla concentration in Lake Taihu during 1984-2021, and its spatiotemporal distribution patterns were further analyzed. Spatially, the Chla concentrations in the western and northern regions of Lake Taihu were higher than those in other regions, probably because these areas are estuaries with large exogenous pollutant discharge and more nutrients are imported from exogenous sources. Chla showed an overall significant upward trend from 1984 to 2021 probably because of temperature rise, wind speed reduction, and nutrient increase. The results of the spatial and temporal variation of Chla and the influencing factors in this study provide supporting data for eutrophication monitoring and management in Lake Taihu. The proposed Chla estimation method can be extended to assess the spatial and temporal distribution of eutrophication in other inland waters with similar optical properties.

OBJECTIVE: We aimed to evaluate the effectiveness of topical tranexamic acid (TXA) versus topical vasoconstrictors in the management of epistaxis via a systematic review and meta-analysis. METHODS: The Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) standards were followed for the meta-analysis. We systematically searched Embase, Web of Science, Cochrane Library, CNKI, and PubMed for randomized controlled trials (from inception to August 2022; no language restrictions), comparing the effect of topical TXA and topical vasoconstrictors on the treatment of epistaxis. The Q test was used to evaluate heterogeneity, and funnel plots were utilized to identify bias. For the meta-analysis, the fixedeffects model was employed, and the t-test was utilized to determine significance. RESULTS: Of 1012 identified studies, 5 were found to be eligible for our analysis. In total, 598 patients were included; 297 of them received TXA and 301 received vasoconstrictors. Hemostasis was more likely to be achieved at the first re-assessment in patients treated with TXA. Subgroup analysis indicated patients treated with TXA to have less likelihood of bleeding recurrence, compared to patients treated with vasoconstrictors. The detected time interval of rebleeding was 10 min, between 24 h to 72 h, and after 7 days, respectively, and the differences were significant between the two groups of patients treated with TXA and vasoconstrictors. CONCLUSION: Topical TXA was associated with better post-treatment hemorrhagic arrest rates compared to topical vasoconstrictors in the management of epistaxis.

The Tibetan Plateau (TP) is a region sensitive to global climate change and has been experiencing substantial environmental changes in the past decades. Lake ice phenology (LIP) is a perceptible indicator reflecting changes of lake thermodynamics in response to global warming. Lake ice phenology over the Tibetan Plateau is however rarely observed and recorded. This research presents a dataset containing 39-year (1978-2016) lake ice phenology data of 132 lakes (each with area >40 km2) over the Tibetan Plateau by combining the strengths of both remote sensing (MOD11A2, MOD10A1) and numerical modelling (air2water). Data validation shows that the ice phenology data derived by our method is highly consistent with that based on existing approaches (with R2 > 0.75 for all phenology index and RMSE < 5d). The dataset is valuable to investigate the lake-atmosphere interactions and long-term hydrothermal change of lakes across the Tibetan Plateau.

Suspended particulate matter (SPM) affects the optical properties of water, which can be used as a marker of water quality. The water quality of Lake Taihu has changed immensely since the 1980's. However, despite the link between water quality and SPM, long-term systematic studies on SPM have not been conducted in this lake. Here, we used Landsat-5 TM and Landsat-8 OLI data to model changes in the SPM concentration of Lake Taihu from 1984 to 2020. Various models were generated, calibrated, and finally validated using in situ SPM, remote sensing reflectance (Rrs) data, and synchronous satellite data. After comparing various commonly used models, it was found that the exponential model based on band combination [Rrs(red) + Rrs(NIR)/Rrs(green)] had the highest accuracy, with an average unbiased relative error greater than 35%. Subsequently, the SPM products of Lake Taihu during 1984-2020 were generated. Overall, the SPM concentration showed a downward trend over the study period, which might be primarily attributable to a decline in wind speed. These findings may assist in the conservation of Lake Taihu and its associated water resources.

Objective: To investigate the characteristics of sleep disorders and anxiety in patients with tinnitus, their influencing factors, and the role of sleep disorders as mediators. Methods: The general conditions and disease characteristics of 393 patients with tinnitus presented to the Changzheng Hospital of the Naval Medical University from 2018 to 2021 were collected. All patients accepted questionnaires such as Tinnitus Handicap Inventory (THI), Pittsburgh Sleep Quality Index (PSQI) and Self-rating Anxiety Scale (SAS), and then the characteristics and the influencing factors of sleep disorders and anxiety were analyzed. Results: Among the 393 tinnitus patients, 213 cases (54.19%) were diagnosed with sleep disorders, and 78 cases (19.85%) were diagnosed with anxiety, including 25 men (32.1%) and 53 women (67.9%). Binary regression showed that gender, hearing loss, tinnitus severity, and sleep disorders severity were positively associated with anxiety. Multiple logistic regression analysis showed that female gender (OR = 2.526, P = 0.008), hearing loss (OR = 2.901, P = 0.003, tinnitus severity (OR = 1.863, P = 0.003) and sleep disorders (OR = 2.510, P = 0.001) were the independent risk factors of anxiety. The mediating effect of sleep disorders between tinnitus severity and anxiety accounted for 27.88% of the total effect size. Conclusion: Females patients with hearing loss, moderate to severe tinnitus, and sleep disorders were at greater risk for anxiety, with sleep disorders partially mediating the anxiety associated with tinnitus.

Eutrophication is a severe environmental pollution problem for inland waters and poses significant threats to the water safety. Monitoring trophic state of inland waters using optical remote sensing generally requires the inversion of water quality parameters, such as chlorophyll-a, secchi depth, etc. However, the accurate inversion of these individual indicators remains challenging, while the associated retrieval errors can propagate and degrade the evaluation of trophic state. Hence, we proposed a novel monitoring method by developing a Trophic State Index (TSI) based on optical remote-sensing parameters, i.e., Forel-Ule index (FUI) and non-water absorption coefficient at 674 nm (referred to as at-w(674)) retrieved from Sentinel-3 Ocean and Land Color Instrument (OLCI) imagery. The estimated TSI showed favorable correspondence with observed water quality data, including coefficient of determination (r2 = 0.91), root mean squared error (RMSE = 5.54), and mean absolute percentage error (MAPE = 10.69%). Using the Sentinel-3 OLCI data, the proposed method also had very good performance in the field spectrum (MAPE = 5.25 % , RMSE = 3.36). The monthly trophic state evaluation also showed congruence (MAPE = 12.51 % , RMSE = 6.41) with surface water quality monthly report (SWQMR) from the Ministry of Environment and Ecology of the People's Republic of China. The monthly TSI showed favorable agreement for 23 ungauged lakes (RMSE = 7.26, MAPE = 12.78%), indicating potential utility for regional lake water quality monitoring. The proposed method was then applied to 47 other large (>50 km2) water bodies in the Middle-and-Lower watershed of Yangtze River and the Huaihe watershed to evaluate the spatial and temporal variation of trophic state from 2016 to 2020. The TSI results revealed several lakes, such as Lake Honghu and Lake Luoma, with rapidly deteriorating water quality during the study period, while other lakes show relative improvement (e.g., Xiashan Reservoir), indicating unbalanced environmental pressure over the region. Overall, this study showed promising performance and potential for satellite-based monitoring of regional aquatic environments.

Water colour is the result of its constituents and their interactions with solar irradiance; this forms the basis for water quality monitoring using optical remote sensing data. The Forel-Ule Index (FUI) is a useful comprehensive indicator to show the water colour variability and water quality change in both inland waters and oceans. In recent decades, lakes around the world have experienced dramatic changes in water quality under pressure from both climate change and anthropogenic activities. However, acquiring consistent water colour products for global lakes has been a challenge. In this paper we present the first time series FUI dataset for large global lakes from 2000-2018 based on MODIS observations. This dataset provides significant information on spatial and temporal changes of water colour for global large lakes during the past 19 years. It will be valuable to studies in search of the drivers of global and regional lake colour change, and the interaction mechanisms between water colour, hydrological factors, climate change, and anthropogenic activities.

With the development of citizen science, digital cameras and smartphones are increasingly utilized in water quality monitoring. The smartphone application HydroColor quantitatively retrieves water quality parameters from digital images. HydroColor assumes a linear relationship between the digital pixel number (DN) and incident radiance and applies a grey reference card to derive water leaving reflectance. However, image DNs change with incident light brightness non-linearly, according to a power function. We developed an improved method for observing and calculating water leaving reflectance from digital images based on multiple reflectance reference cards. The method was applied to acquire water, sky, and reflectance reference card images using a Cannon 50D digital camera at 31 sampling stations; the results were validated using synchronously measured water leaving reflectance using a field spectrometer. The R2 for the red, green, and blue color bands were 0.94, 0.95, 0.94, and the mean relative errors were 27.6%, 29.8%, 31.8%, respectively. The validation results confirm that this method can derive accurate water leaving reflectance, especially when compared with the results derived by HydroColor, which systematically overestimates water leaving reflectance. Our results provide a more accurate theoretical foundation for quantitative water quality monitoring using digital and smartphone cameras.

Secchi disk depth (ZSD) and Forel-Ule index (FUI) are the two oldest and easiest measurements of water optical properties based on visual determination. With an overarching objective to obtain water inherent optical properties (IOPs) using these historical measurements, this study presents a model for associating remote-sensing reflectance (Rrs) with FUI and ZSD. Based upon this, a scheme (FZ2ab) for converting FUI and ZSD to absorption (a) and backscattering coefficients (bb) is developed and evaluated. For a data set from HydroLight simulations, the difference is <11% between FZ2ab-derived a and known a, and <28% between FZ2ab-derived bb and known bb. Further, for a data set from field measurements, the difference is < 30% between FZ2ab-derived a and measured a. These results indicate that FZ2ab can bridge the gap between historical measurements and the focus of IOP measurements in modern marine optics, and potentially extend our knowledge on the bio-optical properties of global seas to the past century through the historical measurements of FUI and ZSD.

During the atmospheric correction of remote sensing data in inland waters, the original Second Simulation of the Satellite Signal in the Solar Spectrum-Vector version (6SV) model does not eliminate the specular reflection of downward skylight radiance at the air-water interface. Thus, we propose a modified version of the 6SV model (M6SV) that does remove reflected skylight at the air-water interface. We apply the new model to the atmospheric correction of a Landsat 8 Operational Land Imager (OLI) image over Taihu Lake, China, where the aerosol optical depth is known. In situ reflectance measurements acquired concurrently with the L8/OLI image are used to validate the performance of the new M6SV algorithm. To further analyze the merits and demerits of M6SV, the model is compared with two short-wave infrared (SWIR)-based atmospheric correction models: the Sea-Viewing Wide Field-of-View Sensor Data Analysis System short-wave infrared (SD-SWIR) model and the Vanhellemont & Ruddick short-wave infrared with a per scene fixed aerosol type (VR-SWIR-F) model. Comparisons of results from all three L8/OLI image atmospheric corrections with the in situ remote sensing reflectance data show that M6SV produces reliable atmospheric corrections in the green and red spectral bands and is an effective alternative for Landsat 8 OLI atmospheric correction in inland waters.
