RESUMO

Phosphorus and nitrogen are important elements in both environmental cycles and biological growth, and their imbalance can lead to serious environmental and biological problems. It is important to be able to monitor the concentration of nitrate and phosphate in the water online. In this paper, a bifunctional boron-doped diamond (BDD) electrode with repeatable electrochemical renewal and modification ability has been developed and used as a shared working electrode for the determination of nitrate and phosphate. First, phosphate can be detected directly with a bare BDD electrode. After a thin copper (Cu) layer was electrodeposited on the BDD electrode, nitrate could be determined. The copper layer is then removed under a positive voltage, and the BDD electrode is renewed and can be used again for phosphate detection. This method enables the detection of both phosphate and nitrate while also improving the stability and repeatability through the renewal of the electrode surface. The segmented linear ranges for phosphate were 0.02-0.4 and 0.4-3 mg/L with a detection limit of 0.004 mg/L. The sensor detected nitrate in a wide concentration range, with segmented linear relationships in the ranges of 0.07-3 and 3-100 mg/L, with a detection limit of 0.065 mg/L. The electrochemical sensor based on the BDD electrode has a good reproducibility for phosphate and nitrate detection. The relative standard deviation (RSD) values of the current responses were 2.98, 2.79, 1.66, 1.81, and 1.23%, respectively, for 35 consecutive tests in 0.05, 0.2, 0.5, 1, and 1.5 mg/L phosphate solution. The RSD values of the current responses were 2.00, 0.97, and 1.03%, respectively, for 25 consecutive tests in 5, 7, and 10 mg/L nitrate solution.

This paper describes an electrochemical sensor based on a Cu-modified boron-doped diamond (BDD) electrode for the detection of nitrate-contaminated water. The sensor utilizes the catalytic effect of copper on nitrate and the stability of the BDD electrode. By optimizing the electrolyte system, the linear detection range was expanded, allowing the sensor to detect highly concentrated nitrate samples up to 100 mg/L with a low detection limit of 0.065 mg/L. Additionally, the stability of the sensor was improved. The relative standard deviation of the current responses during 25 consecutive tests was only 1.03%. The wide detection range and high stability of the sensor makes it suitable for field applications and the on-site monitoring of nitrate-contaminated waters.

An electrochemical sensor based on a thin-layer flow cell and a boron-doped diamond (BDD) working electrode was fabricated for heavy metal ions determination using anodic stripping voltammetry. Furthermore, a fluidic automatic detection system was developed. With the wide potential window of the BDD electrode, Zn2+ with high negative stripping potential was detected by this system. Due to the thin-layer and fluidic structure of the sensor system, the electrodepositon efficiency for heavy metal ions were improved without using conventional stirring devices. With a short deposition time of 60 s, the system consumed only 0.75 mL reagent per test. A linear relationship for Zn2+ determination was displayed ranging from 10 µg/L to 150 µg/L with a sensitivity of 0.1218 µA·L·µg-1 and a detection limit of 2.1 µg/L. A high repeatability was indicated from the relative standard deviation of 1.60% for 30 repeated current responses of zinc solution. The system was applied to determine Zn2+ in real water samples by using the standard addition method with the recoveries ranging from 92% to 118%. The system was also used for the simultaneous detection of Zn2+, Cd2+, and Pb2+. The detection results indicate its potential application in on-site monitoring for mutiple heavy metal ions.

Spider silk is a natural fiber known as "biosteel" with the strongest composite performance, such as high tensile strength and toughness. It is also equipped with excellent biocompatibility and shape memory ability, thus shows great potential in many fields such as biomedicine and tissue engineering. Spider silk is composed of macromolecular spidroin with rich structural diversity. The characteristics of the primary structure of natural spidroin, such as the high repeatability of amino acids in the core repetitive region, the high content of specific amino acids, the large molecular weight, and the high GC content of the spidroin gene, have brought great difficulties in heterologous expression. This review discusses focuses on the relationship between the featured motifs of the microcrystalline region in the repetitive unit of spidroin and its structure, as well as the spinning performance and the heterologous expression. The optimization design for the sequence of spidroin combined with heterologous expression strategy has greatly promoted the development of the biosynthesis of spider silk proteins. This review may facilitate the rational design and efficient synthesis of recombinant spidroin.

The high-density particle distribution in capillary was a crucial factor for enhancing SERS properties and a difficult point in the preparation process. The direct high-temperature method was used to fuse the particles and form a uniform and dense particle distribution on the capillary's inner wall, providing a foundation for enhancing Raman signals. The prepared capillary SERS substrate strongly enhances the rhodamine 6G (R6G) signal, and the RSD values of several characteristic peaks of R6G are about 10 %, demonstrating high sensitivity, uniformity, and stability. Using capillary SERS substrate for detecting goat serum. Embedding precious metal particles into capillary SERS substrate can effectively encapsulate the tested liquid and avoid contamination, which improves the disadvantage of traditional substrates exposing the liquid to air. The prepared capillary SERS substrate could be used for field and biomedical sensitivity detection, providing a theoretical and experimental basis for developing the capillary SERS substrate.

Sensitive, accurate, simple and quick monitoring of Salmonella typhimurium (S. typhimurium) in food is significant for preventing food poisoning, but still remains a challenge. Herein, a colorimetry/fluorescence dual-mode sensing strategy was fabricated to detect S. typhimurium by integrating the self-assembly of magnetic covalent organic framework (MCOF) with gold nanoparticles (Au NPs) as the peroxidase-mimicking nanozyme and aggregation-induced emission luminogen (AIEgen). S. typhimurium could competitive bind to aptamer conjugated Au NPs (Au NPs@apt), inhibit the self-assembly of MCOF with Au NPs, and shield the catalytic activity of AuNPs. After adding H2O2 and TPE-4A, the dark green solution changed to light with increasing S. typhimurium concentration, on the contrary, the fluorescent signals were generated. As a result, in colorimetry/fluorescence modes, S. typhimurium could be detected in the linear ranges of 103-108 CFU mL-1 and 101-107 CFU mL-1, with LODs of 1000 and 10 CFU mL-1, respectively. Importantly, different colors consistent with various S. typhimurium concentrations can be accurately classified by smartphone app and linear discriminant analysis (LDA). The smartphone-assisted data interpretation can generate complementary colorimetry and fluorescence signals without any sophisticated equipment and achieve on-site detection. Moreover, the proposed strategy could be explored for S. typhimurium monitoring in milk with satisfactory recoveries (97.6-100.4 %) in colorimetry and fluorescence mode and good classification and prediction performance in smartphone/LDA system, suggesting the feasibility and potential applications of the sensing platform.

Salmonella typhimurium (S. typhimurium) is one of the most common pathogens in the environment, such as in drinking water and soil. Herein, an on-site detection method was developed by combining silver-coated magnetic nanoparticles (Fe3O4@Ag NPs) with the ß-cyclodextrin-capped gold nanoparticles (ß-CD-Au NPs) to achieve sensitive detection of S. typhimurium. After they formed a sandwich structure in the presence of S. typhimurium, the 4-nitrophenol was reduced to 4-aminophenol based on the nitro-reductase activity of ß-CD-Au NPs. The naked eyes were able to observe the color change from yellow to colorless. Under optimal conditions, the detection range of S. typhimurium was 10-107 CFU mL-1, and the limit of detection (LOD) was 10 CFU mL-1. The total detection time was 90 min, showing satisfactory performance in real samples. We combined a smartphone app with the colorimetric method, making it possible to semi-quantitatively detect S. typhimurium by analyzing the grey value. In conclusion, this assay detects S. typhimurium in environmental samples, offering an accurate and sensitive detection method without sophisticated equipment.

The combustion characteristics of premixed methane-air flames in a half-open tube with a two-sided 45° branch structure at different ignition positions were investigated by experiments and large eddy simulations. The numerical results were compared with the experimental results to verify the correctness of the model. The results show that the simulation results are highly consistent with the experiment. This study provides a basic understanding of the effects of the branch tube structure and the ignition position on flame dynamics. When the flame propagates to the branch interface, it forms a symmetrical vortex structure at the branch tube with the opposite rotation direction. When the ignition position is at IP0 and IP900, the maximum overpressures obtained in the experiment are 10.1 and 10.7 kPa, respectively, and 9.2 and 10.4 kPa in the simulation, respectively. At IP0, the Karlovitz number indicating the interaction intensity between the flame surface and the turbulence during flame propagation is a maximum of 9.2 and a minimum of 0.04. The premixed flame has a folded small flame, a corrugated small flame, and a thin reaction zone.

BACKGROUND: The association between executive dysfunction, brain dysconnectivity, and inflammation is a prominent feature across major psychiatric disorders (MPDs), schizophrenia, bipolar disorder, and major depressive disorder. A dimensional approach is warranted to delineate their mechanistic interplay across MPDs. METHODS: This single site study included a total of 1543 participants (1058 patients and 485 controls). In total, 1169 participants underwent diffusion tensor and resting-state functional magnetic resonance imaging (745 patients and 379 controls completed the Wisconsin Card Sorting Test). Fractional anisotropy (FA) and regional homogeneity (ReHo) assessed structural and functional connectivity, respectively. Pro-inflammatory cytokine levels [interleukin (IL)-1ß, IL-6, and tumor necrosis factor-α] were obtained in 325 participants using blood samples collected with 24 h of scanning. Group differences were determined for main measures, and correlation and mediation analyses and machine learning prediction modeling were performed. RESULTS: Executive deficits were associated with decreased FA, increased ReHo, and elevated IL-1ß and IL-6 levels across MPDs, compared to controls. FA and ReHo alterations in fronto-limbic-striatal regions contributed to executive deficits. IL-1ß mediated the association between FA and cognition, and IL-6 mediated the relationship between ReHo and cognition. Executive cognition was better predicted by both brain connectivity and cytokine measures than either one alone for FA-IL-1ß and ReHo-IL-6. CONCLUSIONS: Transdiagnostic associations among brain connectivity, inflammation, and executive cognition exist across MPDs, implicating common neurobiological substrates and mechanisms for executive deficits in MPDs. Further, inflammation-related brain dysconnectivity within fronto-limbic-striatal regions may represent a transdiagnostic dimension underlying executive dysfunction that could be leveraged to advance treatment.

BACKGROUND: Sleep deprivation (SD) has emerged as a significant public health concern because of its adverse effects on cognition and behavior. However, the influence of circadian rhythms on SD and brain activities has been less studied. This study employed functional magnetic resonance imaging (fMRI) and functional connectivity density (FCD) metrics to investigate the interaction between sleep pressure and circadian rhythms during SD. METHODS: Thirty-six volunteers with good sleep habits underwent a sleep deprivation trial. Sleepiness was assessed using the Stanford Sleepiness Scale (SSS) at multiple time points, and fMRI scans were conducted to derive global and local FCD (gFCD and iFCD) values. This study focused on specific brain regions and networks, including the thalamus, the frontoparietal network (FPN), and the default mode network (DMN). RESULTS: Analysis indicated significant changes in gFCD and iFCD values in several key brain regions. A strong correlation was found between sleepiness and both gFCD and iFCD values in certain areas, such as the left superior temporal gyrus and left thalamus. The gFCD values in these regions showed a gradual increase across sessions, while iFCD values in the right superior frontal gyrus decreased. CONCLUSIONS: This study revealed that SD leads to enhanced functional activities in the DMN and thalamus and decreased activity in the FPN. These changes in brain activity were significantly correlated with increases in sleepiness, as measured by the SSS. Our findings underscore the importance of understanding the neural underpinnings of SD and could guide future clinical interventions aimed at mitigating its effects.

Rapid qualitative and quantitative detection of Salmonella typhimurium (S. typhimurium) takes an important role in ensuring food safety. Herein, a colorimetric assay aptasensor for S. typhimurium utilizing intrinsic peroxidase-like activity of gold nanoparticles embedded spherical covalent organic framework and the affinity and specificity of S. typhimurium-aptamer has been explored. This aptasensor can capture the S. typhimurium via the selective binding effect of aptamer, and the catalytically active sites were shielded. As a result, the colorimetric signals of the 3,3',5,5'-tetramethylbenzidine-H2O2 system were turned off. Under optimum conditions, the aptasensor gave a linear response over the range of 10 to 107 CFU/mL for S. typhimurium. The detection limit of 7 CFU/mL was obtained within 45 min and was effectively applied to detect S. typhimurium in milk and lake water samples with recoveries in the range from 96.4 to 101.0%. More importantly, combined with a self-developed smartphone-based image analysis system, the proposed aptasensor can be used for point-of-care testing applications.

Background: Leptin is a multifunctional hormone secreted from adipose tissue, which plays a core role in regulating energy intake and expenditure. Evidence has demonstrated that leptin receptors are located in brain areas involved in emotional processing, and major depressive disorder (MDD) is characterized by dysfunction of emotional processing. Taken together, these features suggest that leptin may play a potential role in the pathophysiology of MDD. However, the precise roles of leptin in modulating depressive symptoms in MDD remain unclear. Methods: Participants [18 drug-naïve MDD patients, 15 unaffected first-degree relatives of MDD patients (FDR-MDD), and 40 healthy controls] completed clinical assessments and provided blood samples for measurement of leptin levels. We evaluated the effect of leptin on clinical status (MDD or FDR-MDD) and symptomatic dimensionalities of MDD using mediation analysis. Results: We found that leptin was increased in MDD patients and this only predicted "somatic anxiety" symptoms. Furthermore, leptin was a significant and indirect mediator of the association between clinical status (MDD or FDR-MDD) and "somatic anxiety" symptoms. Conclusion: Our finding that leptin was a significant and indirect mediator of clinical status (MDD or FDR-MDD) and "somatic anxiety" symptoms suggests that leptin may indirectly affect somatic depressive symptoms in MDD. Our findings may provide a theoretical basis for novel clinical interventions in MDD.

Introduction: Medication-related osteonecrosis of the jaw (MRONJ) is an oral complication in cancer patients being treated with either antiresorptives, mainly denosumab and bisphosphonates, or antiangiogenic drugs. Osimertinib is a third-generation epidermal growth factor receptor (EGFR)-tyrosine kinase inhibitor (TKI) for the treatment of patients with EGFR T790M advanced non-small-cell lung cancer (NSCLC). TKI-induced osteonecrosis of the jaw has been reported in recent years, but these cases almost occur in combination with bisphosphonates, and the data on MRONJ associated to osimertinib is scarce. Case report: We reported a case of MRONJ associated only with osimertinib. A 69-year-old female patient with NSCLC developed MRONJ after 4 years of treatment with osimertinib. Six months ago, she felt persistent pain and swelling in the right maxilla. After 3 months of pain, her dentist extracted one tooth in the right maxilla under local anesthesia. We examined her gingiva and found fistula and pus spillage. A digital volume tomography scan revealed sequestrum. The patient underwent surgical debridement of the necrotic bone under general anesthesia and administered intravenous antibiotics at the hospital. Histopathological analysis of the bone biopsy revealed a diagnosis of MRONJ. Conclusion: This report provides evidence that osimertinib monotherapy can cause MRNOJ, and has a contribution to explore the formation mechanism of MRONJ. For those patients who take osimertinib, routine oral examinations and monitoring should be performed before and during treatment, as well as prompt closure of wounds and antibiotic treatment to avoid infection after invasive oral surgery such as tooth extraction.

Immunomagnetic separation based on Fe3O4 magnetic nanoparticles (MNPs) has been widely performed in sample pretreatment. The oriented conjugation strategy can achieve a better capture effect than the N-(3-dimethylamlnopropyl)-N'-ethylcarbodiimide hydrochloride (EDC) /N-hydroxysuccinimide (NHS) method. However, immunoglobulin yolk (IgY) cannot be oriented through an SPA strategy like immunoglobulin G (IgG). In this article, an oriented conjugation nanoprobe was prepared for the enrichment of bacteria based on pH adjusting. The main factors affecting the enrichment efficiency were studied, such as the pH of the buffer system, the concentration of IgY, the concentration of nanoprobe, and the enrichment time. Under the optimal conditions, the enrichment efficiency toward target bacteria could reach 92.8%. Combined with PCR, the limit of detection (LOD) was found to be 103 CFU/ml, which was lower than the PCR only. In conclusion, we provided a new protocol for the oriented conjugation of IgY and high sensitivity detection with simple pretreatment.

We developed an effective and specific colorimetric strategy to detect Salmonella typhimurium (S. typhimurium) based on hexadecyl trimethyl ammonium bromide (CTAB)-induced supramolecular assembly of ß-cyclodextrin-capped gold nanoparticles (ß-CD-AuNPs). In this study, ssDNA aptamer of S. typhimurium could combine with CTAB to form the supramolecular ssDNA-CTAB composite, so the ssDNA aptamer was applied to control the concentration of CTAB. In the presence of S. typhimurium, ssDNA aptamers selectively bound to S. typhimurium but not to CTAB, leading to the host-guest chemistry reaction of CTAB and ß-CD resulting in ß-CD-AuNP supramolecular assembly aggregation with an obvious color change. The ratio of absorption at 650 and 520 nm (A650nm/A520nm) has a linear correlation to the log scale of the concentration of the bacteria (1 × 102-1 × 107 CFU/mL) with a low limit of detection (LOD) of 13 CFU/mL. In addition, this optical sensor has good selectivity and practicability. In milk samples, the recovery was 93.55-111.32%, which suggested its potential application in real samples.

As shown in studies conducted in recent decades, polyoxometalates (POMs), as inorganic metal oxides, have promising biological activities, including antitumor, anti-infectious and anti-Alzheimer's activities, due to their special structures and properties. However, some side effects impede their clinical applications to a certain extent. Compared with unmodified POMs, POM-based inorganic-organic hybrids and POM-based nanocomposite structures show significantly enhanced bioactivity and reduced side effects. In this review, we introduce the biological activities of POMs and their derivatives and highlight the side effects of POMs on normal cells and organisms and their possible mechanisms of action. We then propose a development direction for overcoming their side effects. POMs are expected to constitute a new generation of inorganic metal drugs for the treatment of cancer, infectious diseases, and Alzheimer's disease.Graphical abstract.

OBJECTIVE: Clinical heterogeneity in major depressive disorder likely reflects the range of etiology and contributing factors in the disorder, such as genetic risk. Identification of more refined subgroups based on biomarkers such as white matter integrity and lipid-related metabolites could facilitate precision medicine in major depressive disorder. METHODS: A total of 148 participants (15 genetic high-risk participants, 57 patients with first-episode major depressive disorder and 76 healthy controls) underwent diffusion tensor imaging and plasma lipid profiling. Alterations in white matter integrity and lipid metabolites were identified in genetic high-risk participants and patients with first-episode major depressive disorder. Then, shared alterations between genetic high-risk and first-episode major depressive disorder were used to develop an imaging x metabolite diagnostic panel for genetically based major depressive disorder via factor analysis and logistic regression. A fivefold cross-validation test was performed to evaluate the diagnostic panel. RESULTS: Alterations of white matter integrity in corona radiata, superior longitudinal fasciculus and the body of corpus callosum and dysregulated unsaturated fatty acid metabolism were identified in both genetic high-risk participants and patients with first-episode major depressive disorder. An imaging x metabolite diagnostic panel, consisting of measures for white matter integrity and unsaturated fatty acid metabolism, was identified that achieved an area under the receiver operating characteristic curve of 0.86 and had a significantly higher diagnostic performance than that using either measure alone. And cross-validation confirmed the adequate reliability and accuracy of the diagnostic panel. CONCLUSION: Combining white matter integrity in corpus callosum, superior longitudinal fasciculus and corona radiata, and unsaturated fatty acid profile may improve the identification of genetically based endophenotypes in major depressive disorder to advance precision medicine strategies.

BACKGROUND: Increasing evidence suggests that major psychiatric disorders, including major depressive disorder (MDD), bipolar disorder (BD) and schizophrenia (SZ) share biological, neuropsychological and clinical features, despite the criteria for their respective diagnoses being different. Neuroimaging studies have shown disrupted 'static' neural connectivity in these disorders. However, the changes in brain dynamics across the three psychiatric disorders remain unknown. METHODS: We aim to examine the connections and divergencies of the dynamic amplitude of low-frequency fluctuation (dALFF) in MDD, BD and SZ. In total, 901 participants [MDD, 229; BD, 146; SZ, 142; and healthy controls (HCs), 384] received resting-state functional magnetic resonance imaging. The dALFF was calculated using sliding-window analysis and compared across three psychiatric disorders. RESULTS: We found significant increases of dALFF in the right fusiform, right hippocampus, right parahippocampal in participants with MDD, BD and SZ compared to HC. We also found specific increased dALFF changes in caudate and left thalamus for SZ and BD and decreased dALFF changes in calcarine and lingual for SZ and MDD. CONCLUSION: Our study found significant intrinsic brain activity changes in the limbic system and primary visual area in MDD, BD, and SZ, which indicates these areas disruptions are core neurobiological features shared among three psychiatric disorders. Meanwhile, our findings also indicate that specific alterations in MDD, BD, and SZ provide neuroimaging evidence for the differential diagnosis of the three mental disorders.

The development of powerful, simple and cost-effective signal amplifiers has significant implications for biological research and analysis. Hybridization chain reaction (HCR) has attracted increasing attention because of its enzyme-free, simple, and efficient amplification. In the HCR process, an initiator probe triggered a pair of metastable hairpins through a cross-opening process to propagate a chain reaction of hybridization events, yielding a long-nicked double-stranded nucleic acid structure. To achieve more noticeable signal amplification, nanomaterials, including graphene oxide, quantum dots, gold, silver, magnetic, and other nanoparticles, were integrated with HCR. Various types of colorimetric, fluorescence, plasmonic analyses or chemiluminescence optical sensing strategies incorporating nanomaterials have been developed to analyze various targets, such as nucleic acids, small biomolecules, proteins, and metal ions. This review summarized the recent advances of HCR technology pairing diverse nanomaterials in optical detection and discussed their challenges.

Due to its wide applications in tire and rubber products, carbon black (CB) implicates concerns on its safety during production, collection, and handling. Here we report that exposure CB, increases coagulation-thrombosis potential in a splenic extramedullary hemopoiesis (EMH)-dependent manner. Adult C57BL/6 mice are kept in whole-body inhalation chambers, and exposed to filtered room air (FRA) or CB for 28 consecutive days. CB exposure resulted in splenic EMH characterized with platelet precursor cells, megakaryocytes (MKs), hyperplasia and enhanced in vivo blood coagulation ability. Metabolomics analysis suggests significant enhance in PGE2 production but reduction in folic acid (FA) levels in murine serum following CB exposure. Mechanistically, activation of COX-dependent PGE2 production promotes IL-6 expression in splenic macrophages, which subsequently results in splenic EMH and increased platelet counts in circulation. Administration of FA protects the mice against CB-induced splenic EMH through inhibiting prostaglandin-endoperoxide synthase 2 (Ptgs2 or Cox2) and prostaglandin E synthase (Ptges) expression in splenic macrophages, eventually recover the coagulation capacity to normal level. The results strongly suggest the involvement of splenic EMH in response to CB exposure and subsequently increased coagulation-thrombosis potential. Supplementation with FA may be a candidate to prevent thrombosis potential attributable to CB exposure.
