Negative-pressure-wound-therapy is commonly used in clinical routine for wound management. Aim of the present study was to assess the feasibility and safety of using an additional container to collect wound fluid during ongoing negative-pressure-wound-therapy. In this present prospective observational study, patients with negative-pressure-wound-therapy were included. An additional container was inserted in the connecting tube between the wound and the vacuum generating device. The following 3 days, the container was changed daily and replaced by a new one. Further safety outcome parameters were assessed. A questionnaire was answered by the responsible surgeon. Twenty-two patients with negative-pressure-wound-therapy with a median (IQR) age of 58.5 (53.0-70.0) years were included in the present study. In median, the duration of negative-pressure-wound-therapy was 5.0 (4.6-5.5) days. In mean ± SD the collected volume of the wound fluid in millilitres (mL) was on day one 7 ± 4 on day two 8 ± 7 and 10 ± 11 on day three. In one patient, there was <0.1 mL of clear water in the additional container. No safety concerns due to the additional container were observed. This study demonstrates that collecting wound fluid during ongoing negative-pressure-wound-therapy over a time period of 3 days is feasible and safe. No safety concerns were observed.

A particularly severe equine herpesvirus type 1 (EHV-1) abortion outbreak occurred at a breeding farm in northern Germany. Sixteen of 25 pregnant mares that had received regular vaccination using an inactivated vaccine aborted and two gave birth to weak non-viable foals in a span of three months, with 89% of cases occurring within 40 days after the initial abortion case. Virological examinations revealed the presence of EHV-1 in all cases of abortion and serological follow-up in mares confirmed recent infection. Molecular studies identified a neuropathogenic variant (Pol/ORF30 A2254 to G2254) that belonged to geographical group 4 of EHV-1 isolates. The abortion outbreak was preceded by a case of mild ataxia of unknown cause in a mare that aborted four months after the ataxic episode. Although vaccination of pregnant mares did not prevent abortion, good EHV-1 immune status of the population at the time of outbreak may have had an impact in the failure of manifestation of the neurological form of the disease.

PURPOSE: The application of lithium is well known to have an antimanic-depressive effect, however, the influence it has on the human brain is still insufficiently known. The aim of our work is to develop a method to investigate the lithium concentration in the human brain with a very high sensitivity and a submillimeter resolution. Present methods either do not provide spatial resolution or are not sensitive enough to measure the naturally occurring lithium content in the human brain. Our method provides the opportunity to perform postmortem series measurements and obtain a detailed map of the lithium distribution in the human brain. This way possible correlations of the lithium distribution in the human brain and biological reasons for affective disorder can be clarified. METHODS: To study the lithium distribution in different regions of the human brain the authors developed a method to measure lithium traces postmortem with a submillimeter spatial resolution using the neutron capture reaction (6)Li(n, α)(3)H. The lithium is measured by coincident detection of the alpha particles and tritons, emitted in opposite directions. The general concept, the preparation of the brain samples, the experimental setup at the measurement station of the Forschungs-Neutronenquelle Heinz Maier-Leibnitz, and a first measurement on human brain tissue are presented. RESULTS: A first measurement on a brain tissue sample nicely showed a spatial distribution of lithium down to a few hundreds of pg∕cm(3) with a maximal resolution of about σ(x) = σ(y) ≈ 200 µm. Also a direct correlation of lithium and optical tissue structure is observable. Typical measurement times of a few minutes allow for series measurements of up to 20 × 20 mm(2) large samples with a thickness of w = 10-20 µm in medical studies. CONCLUSIONS: The combination of a very high lithium sensitivity with position resolving measurement makes this method well suited for postmortem studies of the microscopic lithium distribution in the human brain and thus to form a microscopic picture of the impact of lithium in different areas of the human brain.

In order to engineer the choline oxidase from Arthrobacter nicotianae (An_CodA) for the potential application as biological bleach in detergents, the specific activity of the enzyme toward the synthetic substrate tris-(2-hydroxyethyl)-methylammonium methylsulfate (MTEA) was improved by methods of directed evolution and rational design. The best mutants (up to 520% wt-activity with MTEA) revealed mutations in the FAD- (A21V, G62D, I69V) and substrate-binding site (S348L, V349L, F351Y). In a separate screening of a library comprising of randomly mutagenised An_CodA, with the natural substrate choline, four mutations were identified, which were further combined in one clone. The constructed clone showed improved activity towards both substrates, MTEA and choline. Mapping these mutation sites onto the structural model of An_CodA revealed that Phe351 is positioned right in the active site of An_CodA and very likely interacts with the bound substrate. Ala21 is part of an alpha-helix which interacts with the diphosphate moiety of the flavin cofactor and might influence the activity and specificity of the enzyme.

Carrots are one of the highest dietary sources of beta-carotene and are naturally high in the (all-E)-beta-carotene isomer, which has higher bioavailability, provitamin A activity, and antioxidant capacity compared to Z (cis) isomers. The objectives of the present study were to investigate the effects of storage temperature, time, and cooking (boiling for 15 min) on the levels of carotene isomers in 'Stefano' carrots. Storing carrots at either 4 degrees C to simulate long-term storage or 20 degrees C to simulate marketing practices resulted in increases in (all-E)-beta-carotene of 20.3% after 3 days at 4 degrees C and 34.4% after 14 days at 20 degrees C, respectively. The levels of Z isomers in raw carrots were low with (13Z)-beta-carotene and (9Z)-beta-carotene accounting for less that 1.8% of the total beta-carotene present. Levels of (9Z)-beta-carotene decreased during storage at either temperature, whereas storage at 4 degrees C resulted in a 109% increase in (13Z)-beta-carotene after 56 days. Cooking significantly increased the levels of (13Z)-beta-carotene and (9Z)-beta-carotene and resulted in the production of (15Z)-beta-carotene, which was absent in raw carrots. Storage at 4 degrees C for 15 days or more prior to cooking reduced the susceptibility of (all-E)-beta-carotene to thermal isomerization during cooking, resulting in lower levels of all three Z-beta-carotene isomers being generated, while storage at 20 degrees C for up to 21 days resulted in significantly higher levels of (all-E)-beta-carotene before and after cooking but had no effect on Z-isomer production during cooking. Consequently, we conclude that, for the greatest health benefit, fresh carrots can be stored for up to 21 days at 20 degrees C or at 4 degrees C for up to 56 days without significant reduction in (all-E)-beta-carotene and should be consumed raw or boiled for less than 15 min to limit Z-beta-carotene isomer formation.
