RESUMO

Remote photoplethysmography (rPPG) technology is a non-contact physiological signal measurement method, characterized by non-invasiveness and ease of use. It has broad application potential in medical health, human factors engineering, and other fields. However, current rPPG technology is highly susceptible to variations in lighting conditions, head pose changes, and partial occlusions, posing significant challenges for its widespread application. In order to improve the accuracy of remote heart rate estimation and enhance model generalization, we propose PulseFormer, a dual-path network based on transformer. By integrating local and global information and utilizing fast and slow paths, PulseFormer effectively captures the temporal variations of key regions and spatial variations of the global area, facilitating the extraction of rPPG feature information while mitigating the impact of background noise variations. Heart rate estimation results on the popular rPPG dataset show that PulseFormer achieves state-of-the-art performance on public datasets. Additionally, we establish a dataset containing facial expressions and synchronized physiological signals in driving scenarios and test the pre-trained model from the public dataset on this collected dataset. The results indicate that PulseFormer exhibits strong generalization capabilities across different data distributions in cross-scenario settings. Therefore, this model is applicable for heart rate estimation of individuals in various scenarios.

Adenosine 5'-monophosphate-activated protein kinase (AMPK)'s effect in PTEN-induced kinase 1 (PINK1) mutant Parkinson's disease (PD) transgenic flies and the related mechanism is seldom studied. The classic MHC-Gal4/UAS PD transgenic flies was utilized to generate the disease characteristics specifically expressed in flies' muscles, and Western blot (WB) was used to measure the expression of the activated form of AMPK to investigate whether activated AMPK alters in PINK1B9 PD flies. MHC-Gal4 was used to drive AMPK overexpression in PINK1B9 flies to demonstrate the crucial role of AMPK in PD pathogenesis. The abnormal wing posture and climbing ability of PINK1B9 PD transgenic flies were recorded. Mitochondrial morphology via transmission electron microscopy (TEM) and ATP and NADH: ubiquinone oxidoreductase core subunit S3 (NDUFS3) protein levels were tested to evaluate the alteration of the mitochondrial function in PINK1B9 PD flies. Phosphorylated AMPKα dropped significantly in PINK1B9 flies compared to controls, and AMPK overexpression rescued PINKB9 flies' abnormal wing posture rate. The elevated dopaminergic neuron number in PPL1 via immunofluorescent staining was observed. Mitochondrial dysfunction in PINK1B9 flies has been ameliorated with increased ATP level, restored mitochondrial morphology in muscle, and increased NDUFS3 protein expression. Conclusively, AMPK overexpression could partially rescue the PD flies via improving PINK1B9 flies' mitochondrial function.

Angiogenesis promotes neurological recovery after acute ischemic stroke (AIS), and microRNAs play crucial roles in cerebral angiogenesis. This study found that Homo sapiens-microRNA-1303(miR-1303) was reduced in blood specimens of AIS patients and human umbilical vein endothelial cells after suffering from oxygen-glucose deprivation/reperfusion. The experiment detected the effect of miR-1303 on angiogenesis by wound healing assay, tube formation assay, and transwell assay. Down-regulation of miRNA-1303 promotes angiogenesis in vitro experiments, while miR-1303 over-expression reverses this effect. Based on bioinformatics analyses and dual-luciferase reporter assay, the thrombospondin type 1 domain containing 7A (THSD7A) was investigated and further validated as the downstream gene of miR-1303. Furthermore, the knockdown of miR-1303 decreased the protein translation and mRNA transcript levels of THSD7A. Our results reveal a novel miR-1303/THSD7A pathway for angiogenesis and further imply that miR-1303 can be a promising biomarker and therapeutic target for AIS.

Objective: The purpose of this research was to evaluate the influence of immunity on infection in patients with severe hemorrhagic stroke and explore the mechanism underlying this connection. Methods: Clinical data obtained from 126 patients with severe hemorrhagic stroke were retrospectively analyzed, and the factors affecting infection were screened by multivariable logistic regression models. Nomograms, calibration curves, the Hosmer-Lemeshow goodness-of-fit test, and decision curve analysis were used to examine the effectiveness of the models in evaluating infection. The mechanism underlying the reduction in CD4+ T-cell levels in blood was explored by analysis of lymphocyte subsets and cytokines in cerebrospinal fluid (CSF) and blood. Results: The results showed that CD4+ T-cell levels of <300/µL was an independent risk factor for early infection. The models for multivariable logistic regression involving the CD4+ T-cell levels and other influencing factors had good applicability and effectiveness in evaluating early infection. CD4+ T-cell levels decreased in blood but increased in CSF. Similarly, interleukin (IL)-6 and IL-8 levels in CSF had a significant increase, generating a substantial concentration gradient between the CSF and the blood. Conclusion: Reduced blood CD4+ T-cell counts among patients who had severe hemorrhagic stroke increased the risk of early infection. CSF IL-6 and IL-8 may be involved in inducing the migration of CD4+ T cells into the CSF and decreasing blood CD4+ T-cell levels.

A reliable critical-scenario-based safety assessment of autonomous vehicles in China requires a thorough understanding of complex crash scenarios in Chinese background traffic. Based on actual crashes between a vehicle and a powered two-wheeler (PTW) in China, this study generated the autonomous driving testing scenarios from functional, logical and concrete levels. First, 239 video-recorded crash cases were selected from the China In-depth mobility Safety Study - Traffic Accident (CIMSS-TA) database. Using the k-medoids clustering method, six functional scenarios were generalized according to seven crash characteristics (time of day, road type, road surface, obstruction, motion of vehicle, motion of PTW, relative moving direction and position of PTW with respect to vehicle), which contained two straight road scenarios, two T-junction scenarios and two intersection scenarios. Then, using a trajectory analysis program written by Python, the dangerous time instant of each crash was extracted based on the relative trajectory. According to five dynamic parameters of dangerous time instant, namely vehicle velocity (Vehicle_V), PTW X'-coordinate velocity (PTW_VX'), PTW Y'-coordinate velocity (PTW_VY'), PTW X'-coordinate relative position (PTW_LocX') and PTW Y'-coordinate relative position (PTW_LocY'), a crash trigger scheme was built to remain a case challenging when the involved vehicle is replaced by an autonomous vehicle with completely different maneuvers. Using the kernel density estimation (KDE), the logical scenarios were evolved by calculating the distribution of these dynamic parameters in each cluster. The results showed that there were differences in the distribution of dynamic parameters between six functional scenarios. For instance, the Vehicle_V in the scenario where a vehicle turning right impacts with a right/right rear PTW traveling straight ahead was higher than that in the scenario where a vehicle changing to the left lane impacts with a left/left rear PTW traveling straight ahead, with ranges of (10 km/h, 30 km/h) and (5 km/h, 15 km/h), respectively. Finally, considering the correlation of dynamic parameters, a virtual crash generation approach based on the independent component analysis (ICA) representing the original crashes with independent parameters was proposed to obtain sufficient concrete testing scenarios. The results showed that the statistical characteristics of virtual crashes were consistent with those of original crashes. Therefore, the virtual crash generation approach was effective. And a concrete crossing testing scenario with the crash trigger conditions of Vehicle_V = 26.272 km/h, PTW_VX' = 15.567 km/h, PTW_VY' = -1.670 km/h, PTW_LocX' = -27.265 m and PTW_LocY' = 52. 149 m was especially demonstrated. This study provides a theoretical basis for generating autonomous driving testing scenarios and data support for establishing relevant testing schemes tailored to the traffic environment in China.

RESUMO

The driver is one of the most important factors in the safety of the transportation system. The driver's perceptual characteristics are closely related to driving behavior, while electroencephalogram (EEG) as the gold standard for evaluating human perception is non-deceptive. It is essential to study driving characteristics by analyzing the driver's brain activity pattern, effectively acquiring driver perceptual characteristics, creating a direct connection between the driver's brain and external devices, and realizing information interchange. This paper first introduces the theories related to EEG, then reviews the applications of EEG in scenarios such as fatigue driving, distracted driving, and emotional driving. The limitations of existing research have been identified and the prospect of EEG application in future brain-computer interface automotive assisted driving systems have been proposed. This review provides guidance for researchers to use EEG to improve driving safety. It also offers valuable suggestions for future research.

OBJECTIVE: To evaluate the efficacy and safety of electrical stimulation at the rabbit sciatic nerve trunk with the body-insulated acupuncture needle whose body is painted with insulating material. METHODS: Eighteen male New Zealand rabbits were randomized into the body-insulated acupuncture needle (BIAN), the general acupuncture needle (GAN), and the blank control groups,with 6 rabbits in each group. The rats'sciatic nerve trunks in BIAN and GAN groups were stimulated by electroacupuncture with the body-insulated acupuncture needle (only allowing the uncoated needle handle and tip to conduct electricity) and the general acupuncture needle, separately. The current intensity was recorded when regular plantarflexion reflexes (sciatic nerve effector reflexes) were observed in the rabbit's foot. The pathological changes of the sciatic nerve at the acupuncture site were observed by H.E. staining, and the ultrastructural changes of the sciatic nerve trunk were observed by transmission electron microscope. RESULTS: The intensity of the current causing the regular plantar flexion reflection in BIAN group (ï¼»0.29±0.07ï¼½ mA) was significantly lower than that in the GAN group(ï¼»0.86±0.08ï¼½ mA, P<0.01). H.E. staining revealed nerve axon degeneration, forming eosinophilic bodies, nerve fiber edema, and focal loss of myelin sheath in the GAN group. While the nerve fiber damage was not obvious, and axons were only degenerated in a few areas in the BIAN group. Transmission electron microscopy observations showed that the nerve myelin sheath structure was separated, the layers were arranged disorderly and bubbled in the GAN group. while the nerve myelin sheath structure of the BIAN group was normal, and it presents a concentric circle-like light and dark lamellar structure, with fewer myelin vacuoles and fissures, only a small part of the mitochondria, microfilaments, and microtubules of the nerve axons were abnormal, and the overall vacuole-like degeneration was significantly reduced, with few of the myelinated fibers were slightly degenerated, and axonal disease was not obvious. CONCLUSION: Insulated acupuncture needle is more accurate and safer than ordinary acupuncture needle for electrical stimulation of rabbit sciatic nerve trunk, and the required electric current intensity is smaller.

PTEN-induced putative kinase 1 (PINK1) mutation induces autosomal recessive Parkinson's Disease (PD), mitochondrial dysfunction is the central pathogenic process. However, more and more studies presented the bulk of the damage to neurons with mitochondrial dysfunction stems from the endoplasmic reticulum (ER) stress. In mitochondria damaged PINK1B9 fly model how protein kinase RNA-like ER kinase (PERK) arm of ER stress functions remains a mystery. Thus, we generated both PERK overexpressed (PEK OE) and down expressed (PEK RNAi) PINK1B9 flies and monitored their motor activity. We found PEK OE decreased the abnormal wing posture rate and rescued PINK1B9 flies' motor activity. Furthermore, we observed the increased number of dopaminergic neurons of protocerebral posterior lateral 1 (PPL1) and the tyrosine hydroxylase (TH) protein levels in PINK1B9 flies. When testing the mitochondrial morphology in flight muscle with TEM, we found that the shape of the mitochondria became normal. The ATP levels of flight muscle tissues were significantly elevated in PEK OE PINK1B9 flies with the increased function of mitochondrial Electron Transport Chain (ETC) Complex I (CI) but not Complex Ⅱ (CⅡ) which is further confirmed by oxygen consumption experiments, Western Blot, and RT-PCR to examine the corresponding subunits. We suggest that overexpression of PERK can rescue PINK1B9 PD flies' pathogenic phenotypes and it is linked with the improved mitochondrial function especially CI of ETC but not CⅡ. Our findings may pave a way for the target of the drug for alleviating the suffering of PINK1 mutant autosomal recessive PD patients.

Seeking an efficient treatment method for bisphenol A ( BPA), a representative endocrine disrupting compound, is important for environmental remediation and human health. Herein, the degradation of BPA by means of photoelectrocatalysis was investigated. Fe doped-TiO2 nanotube arrays ( Fe/TNA ) served as the photoanode, and a xenon lamp simulated the solar light source. First, undoped TiO2 nanotube arrays (TNA) and a series of Fe/TNA were characterized by field emission scanning electron microscopy, X-ray diffraction and UV-Vis diffuse reflectance spectroscopy. The UV-Vis absorption spectra of Fe/TNA showed a red-shift and an enhancement of the absorption in the visible-light region compared to TNA. Then, experimental conditions including Fe doping content, current intensity and aeration rate were varied to demonstrate their effects on the elimination of BPA. It was observed that the degradation of BPA could be fitted to the quasi-first-order equation. Under the following conditions: Fe/TNA prepared by 0.9 mol x L(-1) Fe(NO3)3 solution dip-coating as photoanode, titanium foil as cathode, current intensity of 1.15 mA x cm(-2) and initial BPA concentration of 10 mg x L(-1), 72.3% BPA was decomposed during 4 h reaction, with a rate constant of 5.32 x 10(-3) min(-1). Aeration enhanced the removal rate of BPA to 82.7% and 94.1% with an aerating rate of 1.0 L x min(-1) using titanium foil as cathode and an aerating rate of 0.2 L x min(-1) using carbon cloth as cathode, respectively, and the corresponding rate constants were 7.20 x 10(-3) min(-1) and 11.6 x 10(-3) min(-1), respectively.

BACKGROUND: Mailuoning is widely used in the treatment of acute ischaemic stroke in China. Animal experimental studies and clinical pharmacological research indicate that mailuoning might improve blood circulation, prevent ischaemic injury, and protect heart and brain tissue. This review was last published in 2009. As new data have become available, it is necessary to reassess the evidence from randomised controlled trials. OBJECTIVES: To determine the effects and safety of mailuoning agents (injection or oral liquid) in the treatment of people with acute ischaemic stroke. SEARCH METHODS: We searched the Cochrane Stroke Group Trials Register (May 2014), the Cochrane Central Register of Controlled Trials (CENTRAL;2014, Issue 4), MEDLINE (1966 to May 2014), Embase (1980 to May 2014), AMED (1985 to May 2014), the Chinese Stroke Trials Register (June 2014), the China Biological Medicine Database (CBM-disc; 1979 to June 2014), China Science and Technology Journal database (CSTJ; 1979 to June 2014), Wanfang Data Chinese databases (1979 to June 2014), and the China National Knowledge Infrastructure (1979 to June 2014). We searched clinical trials and research registers, handsearched 10 Chinese journals including relevant conference proceedings, scanned reference lists, and contacted the pharmaceutical company that manufactures mailuoning. We also attempted to contact trial authors to obtain further data. SELECTION CRITERIA: Randomised controlled trials comparing mailuoning with placebo or mailuoning plus other treatment compared with that other treatment in people with acute ischaemic stroke. DATA COLLECTION AND ANALYSIS: Two review authors independently selected trials for inclusion, assessed trial quality, and extracted data. MAIN RESULTS: We included 21 trials, involving 1746 participants, in this update; six trials were new. The included trials did not report the numbers of dead and dependent participants at the end of at least three months' follow-up. Of the 12 trials that reported adverse events, five events occurred in two trials. There was no significant difference between the treatment group and the control group. We assessed 20 trials to be of a poor quality: When analysing these trials together, mailuoning was associated with a significant increase in the number of participants with an improved neurological deficit (risk ratio (RR) 0.31, 95% confidence interval (CI) 0.23 to 0.42) and showed a significant improvement of neurological deficit with the European Stroke Scale (ESS) (mean difference (MD) (fixed) 8.29, 95% CI 3.44 to 13.15). One placebo-controlled trial, assessed to be of a better methodological quality, failed to show a significant improvement of neurological deficit at the end of three months' follow-up (MD (fixed) 2.49, 95% CI -1.45 to 6.43) or in quality of life. One trial, which reported cognitive function using the Montreal Cognitive Assessment as a continuous scale, showed a significant improvement of cognitive function (MD (fixed) 2.68, 95% CI 1.82 to 3.54). Two trials assessed activities of daily life: One trial showed a significant improvement, but the other did not. AUTHORS' CONCLUSIONS: This review did not provide sufficient evidence to support the routine use of mailuoning for the treatment of people with acute ischaemic stroke. High-quality large-scale randomised controlled trials are needed to confirm the efficacy of mailuoning.

BACKGROUND: A proliferation-inducing ligand (APRIL) is a tumor-necrosis factor (TNF) family member and is a novel cytokine crucial in sustaining lymphocytic leukemia B cell survival and proliferation. However, its role in gastric cancer (GC) remains unclear. In this study, we investigated the expression pattern and prognostic role of APRIL in GC. METHODS: Expression of APRIL was assessed by immunohistochemistry and real-time PCR. Prognostic role of APRIL expression was evaluated. We also discovered the effect of APRIL on chemo-resistance in GC cells and the underlying mechanisms. RESULTS: APRIL mRNA levels were significantly increased in GC tissues compared with adjacent tissues and high expression levels of APRIL in tumor cells significantly correlated with poor overall survival in patients receiving cisplatin adjuvant treatment. Overexpression of APRIL in AGS cells significantly attenuated the therapeutic efficacy of cisplatin in vitro and in vivo. In contrast, silence of APRIL in SGC7901 cells enhanced cisplatin-induced tumor suppression. Our data further revealed that the canonical NF-κB pathway was involved in APRIL-mediated chemo-resistance. In addition, expression of APRIL was regulated by miR-145 in GC cells. CONCLUSION: APRIL is a novel clinical chemo-resistance biomarker for gastric cancer and might be a promising therapeutic target for GC patients.
