RESUMO

ATP-binding cassette (ABC) transporters were crucial for various physiological processes like nutrition, development, and environmental interactions. Selenium (Se) is an essential micronutrient for humans, and its role in plants depends on applied dosage. ABC transporters are considered to participate in Se translocation in plants, but detailed studies in soybean are still lacking. We identified 196 ABC genes in soybean transcriptome under Se exposure using next-generation sequencing and single-molecule real-time sequencing technology. These proteins fell into eight subfamilies: 8 GmABCA, 51 GmABCB, 39 GmABCC, 5 GmABCD, 1 GmABCE, 10 GmABCF, 74 GmABCG, and 8 GmABCI, with amino acid length 121-3022 aa, molecular weight 13.50-341.04 kDa, and isoelectric point 4.06-9.82. We predicted a total of 15 motifs, some of which were specific to certain subfamilies (especially GmABCB, GmABCC, and GmABCG). We also found predicted alternative splicing in GmABCs: 60 events in selenium nanoparticles (SeNPs)-treated, 37 in sodium selenite (Na2SeO3)-treated samples. The GmABC genes showed differential expression in leaves and roots under different application of Se species and Se levels, most of which are belonged to GmABCB, GmABCC, and GmABCG subfamilies with functions in auxin transport, barrier formation, and detoxification. Protein-protein interaction and weighted gene co-expression network analysis suggested functional gene networks with hub ABC genes, contributing to our understanding of their biological functions. Our results illuminate the contributions of GmABC genes to Se accumulation and tolerance in soybean and provide insight for a better understanding of their roles in soybean as well as in other plants.

Immunotherapy has become a very effective treatment for many cancers. It has a unique set of immune system-related adverse effects, collectively known as immune-related adverse events (irAEs). Skin toxicities are the most common irAEs, of which bullous pemphigoid, although rare, is potentially life-threatening and affects patients' survival. In this article, we report the treatment of bullous pemphigoid caused by programmed cell death protein-1 (PD-1) in a case of proficient mismatch repair (pMMR)/microsatellite stable (MSS) colorectal cancer. No significant adverse effects were observed in the patient after methylprednisone was tapered to 4 mg twice a day. No new skin lesions occurred recently in the patient and the original skin lesions healed. In particular, the patient's immunotherapy was not stopped and the best outcome was a partial remission of the disease, lasting for more than 8 months.

Selenium (Se) is an essential element for mammals, and its deficiency in the diet is a global problem. Agronomic biofortification through exogenous Se provides a valuable strategy to enhance human Se intake. Selenium nanoparticles (SeNPs) have been regarded to be higher bioavailability and less toxicity in comparison with selenite and selenate. Still, little has been known about the mechanism of their metabolism in plants. Soybean (Glycine max L.) can enrich Se, providing an ideal carrier for Se biofortification. In this study, soybean sprouts were treated with SeNPs, and a combination of next-generation sequencing (NGS) and single-molecule real-time (SMRT) sequencing was applied to clarify the underlying molecular mechanism of SeNPs metabolism. A total of 74,662 nonredundant transcripts were obtained, and 2109 transcription factors, 9687 alternative splice events, and 3309 long non-coding RNAs (lncRNAs) were predicted, respectively. KEGG enrichment analysis of the DEGs revealed that metabolic pathways, biosynthesis of secondary metabolites, and peroxisome were most enriched both in roots and leaves after exposure to SeNPs. A total of 117 transcripts were identified to be putatively involved in SeNPs transport and biotransformation in soybean. The top six hub genes and their closely coexpressed Se metabolism-related genes, such as adenylylsulfate reductase (APR3), methionine-tRNA ligase (SYM), and chloroplastic Nifs-like cysteine desulfurases (CNIF1), were screened by WGCNA and identified to play crucial roles in SeNPs accumulation and tolerance in soybean. Finally, a putative metabolism pathway of SeNPs in soybean was proposed. These findings have provided a theoretical foundation for future elucidation of the mechanism of SeNPs metabolism in plants.

It is an important way for healthy Selenium (Se) supplement to transform exogenous Se into organic Se through crops. In the present study, Vigna unguiculata was selected as a test material and sprayed with biological nano selenium (SeNPs) and Na2SeO3, and its nutrient composition, antioxidant capacity, total Se and organic Se content were determined, respectively. Further, the response of ABC transporter family members in cowpea to different exogenous Se treatments was analyzed by transcriptome sequencing combined with different Se forms. The results show that the soluble protein content of cowpea increased after twice Se treatment. SeNPs treatment increased the content of cellulose in cowpea pods. Na2SeO3 treatment increased the content of vitamin C (Vc) in cowpea pods. Se treatments could significantly increase the activities of Peroxidase (POD), polyphenol oxidase (PPO) and catalase (CAT) in cowpea pods and effectively maintain the activity of Superoxide dismutase (SOD). SeNPs can reduce the content of malondialdehyde (MDA) in pods. After Se treatment, cowpea pods showed a dose-effect relationship on the absorption and accumulation of total Se, and Na2SeO3 treatment had a better effect on the increase of total Se content in cowpea pods. After treatment with SeNPs and Na2SeO3, the Se species detected in cowpea pods was mainly SeMet, followed by MeSeCys. Inorganic Se can only be detected in the high concentration treatment group. Analysis of transcriptome data of cowpea treated with Se showed that ABC transporters could play an active role in response to Se stress and Se absorption, among which ABCB, ABCC and ABCG subfamilies played a major role in Se absorption and transportation in cowpea. Further analysis by weighted gene co-expression network analysis (WGCNA) showed that the content of organic Se in cowpea treated with high concentration of SeNPs was significantly and positively correlated with the expression level of three transporters ABCC11, ABCC13 and ABCC10, which means that the ABCC subfamily may be more involved in the transmembrane transport of organic Se in cells.

PURPOSE: To determine the association between levator avulsion and urinary stress incontinence (USI) by 3/4D transperineal ultrasound. MATERIALS AND METHODS: 842 patients who were admitted to our hospital from 2016 to 2019 were recruited for our study. 3D/4D transperineal ultrasound was performed. After standard interview and clinical evaluation, general conditions and levator hiatus data were collected and measured to compare with each group. The odds ratio (OR) of USI symptoms or ultrasound features with levator avulsion were calculated. RESULTS: A total of 593 women were studied: 204 suffered from levator avulsion (96 cases of left-side avulsion, 80 cases of right-side avulsion and 28 cases of bilateral avulsion) and 389 women had no avulsion. The gravidity and episiotomy conditions of the avulsion groups were significantly different from the no-avulsion group. Significant differences were found in the transverse diameters and anteroposterior diameters between the levator avulsion group and the no-avulsion group, but there was no difference among the avulsion groups, regardless of whether the patient was at rest or performing the Valsalva maneuver. Interestingly, a significant difference was found in the presence of USI symptoms between the uni-avulsion group and the no-avulsion group. The odds ratio (OR) of USI symptoms in the uni-avulsion group is 2.786 (95 %CI, 1.663-4.669), but 0.939 (95 %CI, 0.276-3.199) for the bilateral avulsion and no-avulsion groups. CONCLUSION: Unilateral levator avulsion may be a risk factor for urinary stress incontinence.

Background: Lung cancer is a malignant tumor with high morbidity and mortality worldwide. At present, the main treatment methods for patients with advanced non-small cell lung cancer (NSCLC) include molecular targeted therapy and immunotherapy. The efficacy rate of immune checkpoint inhibitor (ICI) monotherapy is relatively low. Studies have confirmed that some combination therapies have better anti-tumor efficacy and higher response rates, such as PD-1/PD-L1 inhibitors combined with chemotherapy or targeted therapy. Poly (ADP-ribose) polymerase (PARP) inhibitors have become a new line of cancer therapy in ovarian and breast cancer, but it's not approved in lung cancer. Some reports show that homologous recombination repair (HRR) gene variants may be potential biomarkers for immunotherapy. However, whether lung cancer with HRR gene variants can be benefit from ICIs combined with PARP inhibitors is unknown. Case Description: We present a case of a 30-year-old man who was admitted to hospital with several months of cough and the chest computed tomography (CT) scan showed a mass about 2.6 cm × 2.1 cm in the left lung. Then he was diagnosed with lung adenocarcinoma (LUAD). Next generation sequencing (NGS) revealed that he harbors ROS1 fusion and NBN germline mutation. So, he received platinum-based chemotherapy and ROS1 inhibitors, but the disease continued to progress. Ultimately, the patient was switched to sintilimab combined with niraparib and the efficacy was evaluated as stable disease (SD), with a progression-free survival (PFS) of more than 12 months, and the overall survival (OS) is 23 months up to now. During the treatment, the major adverse events (AEs) observed were lymphopenia, nausea, vomiting, and edema. The AEs were tolerable. Conclusions: This case shows that the combination of small-molecule inhibitors and immunotherapy may improve survival in NSCLC patients with driver genes, and sintilimab combined with niraparib provides a successful clinical case for the treatment of refractory tumors HRR gene mutation, which can be used as a reference for personalized treatment. Of course, more clinical trials are needed to confirm this combination treatment strategy.

Plant growth is affected by various abiotic stresses, including water, temperature, light, salt, and heavy metals. Selenium (Se) is not an essential nutrient for plants but plays important roles in alleviating the abiotic stresses suffered by plants. This article summarizes the Se uptake and metabolic processes in plants and the functions of Se in response to water, temperature, light, salt, and heavy metal stresses in plants. Se promotes the uptake of beneficial substances, maintains the stability of plasma membranes, and enhances the activity of various antioxidant enzymes, thus alleviating adverse effects in plants under abiotic stresses. Future research directions on the relationship between Se and abiotic stresses in plants are proposed. This article will further deepen our understanding of the relationship between Se and plants.

BACKGROUND: Ovarian cancer (OC) is a common malignancy in the gynecological tumor. Standard treatment for ovarian cancer is surgery and chemotherapy based on paclitaxel and platinum. However, traditional chemotherapy for ovarian cancer is limited by drug resistance and systemic side effects. It is imperative to explore effective treatment options for refractory ovarian cancer. CASE PRESENTATION: A 52-year-old female initially presented with lower abdominal distension and migratory pain. After the laparoscopic exploration and biopsy, immunohistochemistry showed poorly differentiated adenocarcinoma originated from ovarian (cT3NxM1, stage IV, peritoneal and abdominal wall metastasis). The next generation sequence detected ERRFI1 (T187A, exon4) mutation. RESULTS: The patient received first-line chemotherapy (paclitaxel, nedaplatin plus avastin), followed by maintenance therapy with gefitinib, achieving a 15-month progression-free survival (PFS). After disease progression and second-line treatment failure, endostar plus apatinib was administered for 14 cycles and she obtained a PFS of 14 months without long-term adverse events. CONCLUSION: We believe that the ERRFI1 gene may be a potential target of gefitinib. Importantly, endostar combined with apatinib is worth recommending for maintenance treatment in refractory ovarian cancer.

The contamination of water systems by heavy metals greatly threatens human health and ecological safety. An efficient adsorbent is critical for the removal of these contaminants. In this work, magnetic Ni3Si2O5(OH)4 nanotubes (NTs) have been synthesized via in situ hydrothermal reduction and further functionalized by grafting poly(4-vinyl pyridine) (P4VP) brushes on its surface via atom transfer radical polymerization. Characterizations by Fourier transform infrared, X-ray diffraction, thermogravimetric analysis, transmission electron microscopy, and X-ray photoelectron spectroscopy proved that P4VP was successfully grafted on the surface of magnetic Ni3Si2O5(OH)4 NTs. The resultant Ni3Si2O5(OH)4-g-P4VP NTs are efficient nanosorbents for removing Cr(VI) anions from water. The Cr(VI) adsorption capacity of Ni3Si2O5(OH)4-g-P4VP NTs reaches 1.49 mmol/g at a pH of 3. The pseudo-second-order kinetic model and the Freundlich isothermal model are suitable to describe the adsorption process. The analysis using Weber-Morris and Boyd models indicates that both intraparticle diffusion and external film diffusion affect the Cr(VI) adsorption process. The adsorption enthalpy is estimated to be 18.37 kJ/mol. More than 90% of the Cr(VI) adsorption capacity of the Ni3Si2O5(OH)4-g-P4VP NTs remains after eight adsorption and desorption cycles.

AIM: Exercise therapy is a key intervention in the management of knee osteoarthritis (OA). This study aimed to test the 6-month effectiveness of Wu Qin Xi Qigong (WQXQ) exercise versus a conventional physical therapy (control group [CG]) on physical functioning in patients with early knee OA. METHODS: This study was a 6-month follow-up from a randomized controlled trial. Participants with knee OA were randomly allocated to the WQXQ or CG. Data from the Berg Balance Scale, Timed Up and Go Test, 6-Minute Walk Test, 30-Second Chair Stand Test, the Western Ontario and McMaster Universities Osteoarthritis Index, knee extension strength and knee flexion strength were collected before and after the 6-month intervention. RESULTS: Both treatment groups demonstrated large (20%-50%) and clinically relevant reductions in activity limitations, pain and knee instability, which were sustained at 6 months post-treatment. No differences in effectiveness between experimental and control treatment were found on Timed Up and Go Test, 6-Minute Walk Test, knee extension strength and knee flexion strength except for a higher Berg Balance Scale score (P = 0.029) and lower Western Ontario and McMaster Universities Osteoarthritis Index pain score (P = 0.031) in the WQXQ group. CONCLUSIONS: Both WQXQ and conventional physical therapy exercise programs were highly effective in reducing activity limitations and pain, and promoting balance and muscle power. WQXQ was found to be more effective in promoting balance and reducing pain than conventional physical therapy exercise in patients with knee OA. Geriatr Gerontol Int 2020; 20: 899-903.

Fopius arisanus is a solitary endoparasitoid that parasitizes a variety of tephritid species. Native to the Indo-Australian region, it is currently exploited worldwide as a biological control agent due to its exceptional efficiency in reducing pest populations. The efficiency of any biological control program is affected by the host location ability of the parasitoids. The present study used a Y-tube olfactometer to test the behavioural responses of female F. arisanus to four fruit species which had undergone different types of damages: undamaged, damaged through Bactrocera dorsalis ovipositioning (i.e., infested), or different levels of mechanical damage. Our results suggest that F. arisanus females were significantly attracted to mangoes and pears (vs. purified air), regardless of their condition; however, whilst infested mangoes did not attract more female parasitoids compared to healthy or mechanically damaged fruits, infested pears attracted significantly more. For citrus fruits and peaches, oviposition damage caused them to be more attractive to parasitoid females. In terms of the longevity of the effects, infested mango fruits remained attractive for up to 5 days after infestation, whereas for infested peaches, pears, and citrus fruits, the attractiveness tended to decrease as time passed. Regarding mechanical damage, mango fruits that had undergone any intensity of damage were equally attractive to parasitoid females; however, peach and citrus fruits with high levels of mechanical damage were more attractive, and pears were found to be most attractive with slight mechanical damage. Additional to the above, we also tested the effect of insecticides on behavioural responses using mangoes. We found that the treatment of infested fruits with lambda-cyhalothrin and cypermethrin remained attractive to F. arisanus females, albeit to different extents, which is in contrast to spinosad, cyantraniliprole, and acetamiprid. Finally, we suggest that the host-searching behaviour of F. arisanus females is mainly mediated by oviposition-induced volatiles, either emitted from the fruit or left by the fruit fly.

The existence of Pb(ii) ions in water systems poses significant potential hazards to public health and the environment. In the present study, poly(2-acrylamido-2-methylpropanesulfonic acid) (PAMPS) brush-modified Ni3Si2O5(OH)4 nanotubes were prepared, and their adsorption efficiency against the Pb(ii) ions was investigated. The characterization results of FTIR spectroscopy, TGA, TEM, and XPS indicated the successful grafting of PAMPS on the surface of free Ni3Si2O5(OH)4 NTs, and the prepared PAMPS-g-Ni3Si2O5(OH)4 NTs exhibited a 6-8 nm grafting layer, which could provide abundant binding sites for metal adsorption. During the Pb(ii) removal process, a pH-dependent adsorption behavior was observed, and the adsorption processes fitted well with the pseudo-second-order kinetic model and the Langmuir isotherm model. Compared with unmodified Ni3Si2O5(OH)4, the PAMPS-g-Ni3Si2O5(OH)4 NTs exhibited obviously faster adsorption of Pb(ii) and higher equilibrium adsorption capacity for the removal of Pb(ii). The maximum adsorption capacity calculated via the Langmuir isotherm model was 0.653 mmol g-1 (135.3 mg g-1) at 298 K. In a metal coexisting system, the total adsorption capacity of the NTs was increased; this indicated the potential of the proposed NTs in the removal of Pb(ii) from metal coexisting wastewater. This study showed the significant potential of PAMPS-g-Ni3Si2O5(OH)4 NTs in the effective removal of Pb(ii).

BACKGROUND: Red cell distribution width (RDW) is associated with a poor prognosis and adverse events in cardiovascular diseases. The aims of this study were to investigate the relationship between serum RDW levels and outcomes after percutaneous coronary intervention and to identify potential novel laboratory markers for evaluating the risk of in-stent restenosis (ISR) with stable angina pectoris. METHODS: A total of 261 patients with coronary heart disease from Dongfeng General Hospital implanted with a coronary drug-eluting stent (DES) were enrolled in the study. We retrospectively analysed the role and prognosis values of serum parameters that were measured before angiography at the first admission. According to the results of the second angiogram, the patients were divided into two groups as follows: the non-ISR group (n=143) and the ISR group (n=118). The clinical characteristics and all laboratory data were considered for univariate and multivariate logistic regression analyses. RESULTS: The white cell count, RDW, neutrophil count, C-reactive protein (CRP), total cholesterol, low-density lipoprotein cholesterol (LDL-C), blood urea nitrogen and uric acid levels were higher in the ISR group than in the non-ISR group. There were no differences in the rates of hypertension, fasting plasma glucose, red cell count, neutrophil to lymphocyte ratio, platelet count, triglyceride, high-density lipoprotein cholesterol and creatinine levels. In the univariate regression analysis, age, diabetes, white cell count, neutrophil count, RDW, CRP, total cholesterol, LDL-C, blood urea nitrogen, Gensini score and number of stents were predictors of ISR. According to the multiple logistic regression analysis, age, RDW and number of stents were independent predictors of ISR. CONCLUSIONS: Preprocedural blood parameters can independently predict ISR. Our study results demonstrated that a high preprocedural RDW is an independent predictor of DES restenosis.

AIM: The present study investigated the effectiveness of Baduanjin Qigong on symptoms related to gait, functional mobility and sleep in Parkinson disease (PD) patients. METHODS: A total of 100 patients (age 67.53 ± 8.56 years, range 55-80 years) with mild to moderate PD were randomly assigned to two groups. Participants in the Baduanjin Qigong group (BQG) received a Baduanjin Qigong program, consisting of four 45-min sessions each week and daily walking 30 min for 6 months. Participants in the control group were carried out daily walking for 30 min. Pre- and post-intervention testing was carried out to assess sleep quality, fatigue, functional mobility and gait performance in these participants. RESULTS: After the 6-month Baduanjin Qigong intervention, the BQG showed sleep quality improvements in the Unified Parkinson's Disease Rating Scale score (P = 0.049), Parkinson's Disease Sleep Scale-2 (PDSS-2) total score (P = 0.039), Motor Symptoms at Night (PDSS-2) score (P = 0.039), PD Symptoms at Night (PDSS-2) score (P = 0.029), Disturbed Sleep (PDSS-2) score (P = 0.037). The BQG showing functional mobility capacity greater improvements in the Berg Balance Scale (P = 0.041) and 6-minute walk test (P = 0.042), and greater decrease in the Timed Up & Go (s; P = 0.046). The BQG showing gait function increased in the gait speed (m/s; P = 0.011). However, this was not the case for the control group, which remained at the same level as pretest performance. CONCLUSIONS: BQG improved the gait performance, functional mobility and sleep quality in older adults with PD at the 6-month follow up. It is as an alternative home exercise program for older adults in rehabilitation for PD. Geriatr Gerontol Int 2016; 16: 911-919.
