RESUMO

Based on the career theory of Cognitive Information Processing (CIP), we selected scale items from literature reviews and expert guidance. The scale consisted of 28 items with 4 factors (interests, abilities, values, personality). To test the scale's factor structure, we used confirmatory factor analysis (CFA), and the model was modified according to CFA results. The second-order confirmatory factor analysis was applied to the model of the scale to prove the rationality of the total score. The internal consistency were evaluated using Cronbach's alpha coefficients. In addition, the composite reliability (CR) and average variance extraction (AVE) of the scale were also calculated to test the convergent validity. After related analyses, the scale was proved to have good psychometric properties, which can be used to measure junior high school students' career planning level in information technology course from the aspects of interest, ability, values, and personality. The effect of the first-order confirmatory factor analysis model constructed in this study is not ideal. Therefore, on this basis, a second-order confirmatory factor analysis model is constructed in combination with existing literatures, and the rationality of the model is verified through data, which highlights the novelty of this study.

As an emerging technology in precision medicine, the patient-derived organoid (PDO) technology has been indicated to provide novel modalities to judge the sensitivity of individual tumors to cancer drugs. In this work, an in vitro model of colorectal cancer (CRC) was established using the PDO culture, and it is demonstrated that the PDO samples preserved, to a great extent, the histologic features and marker expression of the original tumor tissues. Subsequently, cancer drugs 5-FU, oxaliplatin, and irinotecan were selected and screened on five CRC PDO samples, while the patient-derived organoid xenograft (PDOX) model was applied for comparison. The receiver operating characteristic (ROC) curve was drawn according to the IC50 data from the PDO model and the relative tumor proliferation rate (T/C%) from PDOX. Interestingly, the area under the ROC curve was 0.84 (95% CI, 0.64-1.04, P value = 0.028), which suggested that the IC50 of cancer drugs from the PDO model was strongly correlated with PDOX responses. In addition, the optimal sensitivity cutoff value for drug screening in CRC PDOs was identified at 10.35 µM, which could act as a reference value for efficacy evaluation of 5-FU, oxaliplatin, and irinotecan in the colorectal cancer drug screening. Since there are no unified criteria to judge the sensitivity of drugs in vitro, our work provides a method for establishing in vitro evaluation criteria via PDO and PDOX model using the patient tissues received from local hospitals, exhibiting potential in clinical cancer therapy and precision medicine.

Long non-coding RNA associated with poor prognosis of hepatocellular carcinoma (lncRNA-AWPPH) is a newly discovered lncRNA that has important functions in the pathogenesis of several malignancies. However, its role in the development of colorectal adenocarcinoma is unknown. The current study therefore investigated the function of AWPPH in colorectal adenocarcinoma. A total of 86 patients with colorectal adenocarcinoma and 56 healthy controls were included. Tumor tissues and adjacent healthy tissues were collected from patients with colorectal adenocarcinoma, and blood was collected from both patients and healthy controls. Expression of AWPPH in tissues and blood was detected by the reverse transcription-quantitative polymerase chain reaction. Receiver operating characteristic curve analysis was used to evaluate the diagnostic value of serum AWPPH for colorectal adenocarcinoma. All patients were followed up for 5 years, and survival curve analysis was performed to investigate the association between serum level of AWPPH and patients' survival. The effects of AWPPH overexpression and silencing in colorectal adenocarcinoma cell lines were investigated. Effects on cell proliferation and viability were detected by the cell counting kit-8 and MTT assays, respectively. Effects on transforming growth factor ß1 (TGF-ß1) expression were determined by western blotting. AWPPH was significantly upregulated in tumor tissues compared with adjacent healthy tissues. AWPPH expression levels in blood increased in patients with colorectal adenocarcinoma compared with healthy controls, suggesting that AWPPH may be a sensitive and accurate diagnostic and prognostic biomarker for colorectal adenocarcinoma. AWPPH overexpression in colorectal adenocarcinoma cell lines promoted cell proliferation and increased cell viability, while AWPPH silencing resulted in opposite effects. AWPPH overexpression promoted and silencing inhibited TGF-ß1 expression. Therefore, lncRNA-AWPPH promoted colorectal adenocarcinoma by promoting tumor growth, increasing tumor cell viability and activating the TGF-ß1 signaling.

OBJECTIVE: To evaluate the endourethral surgery for the complicated urethra stenosis and urethratresia. METHODS: The endourethral surgery, such as internal urethrotomy transurethral scar electrosectomy or transurethral scar plasmakinetic bipolar electrocautery (PKR) or transurethral laser cicatrectomy, were carried out in 46 cases suffering from the complicated urethra stenosis and urethratresia. RESULTS: The curative rate in this series being achieved by once and twice or three times'operation were 80.43% (39/46) and 13.04% (6/46) respectively. Three cases of treatment failure were caused by long-segment stricture and urethratresia or severe malposition of the urethral proximal and distal to a narrow-caliber area or post-operation infection. Thirty-nine cases have been followed up for 6 to 84 months. Satisfactory voiding has been achieved in all patients. CONCLUSION: Endoscopic surgery was believed to be a safe and efficient therapeutic choice for the complicated urethra stenosis and urethratresia. The success of the treatment depends on understanding the length of the stricture before operation, resecting completely the scar tissue with electric or PKR or laser technique during the process, preventing infection and managing appropriately the urethral catheterization after operation.
