RESUMO

Pyraclostrobin, a widely used fungicide, poses significant risks to both the environment and human health. However, research on the microbial degradation process of pyraclostrobin was scarce. Here, a pyraclostrobin-degrading strain, identified as Burkholderia sp. Pyr-1, was isolated from activated sludge. Pyraclostrobin was efficiently degraded by strain Pyr-1, and completely eliminated within 6 d in the presence of glucose. Additionally, pyraclostrobin degradation was significantly enhanced by the addition of divalent metal cations (Mn2+ and Cu2+). The degradation pathway involving ether bond and N-O bond cleavage was proposed by metabolite identification. The sodium alginate-immobilized strain Pyr-1 had a higher pyraclostrobin removal rate from contaminated lake water than the free cells. Moreover, the toxicity evaluation demonstrated that the metabolite 1-(4-chlorophenyl)-1H-pyrazol-3-ol significantly more effectively inhibited Chlorella ellipsoidea than pyraclostrobin, while its degradation products by strain Pyr-1 alleviated the growth inhibition of C. ellipsoidea, which confirmed that the low-toxic metabolites were generated from pyraclostrobin by strain Pyr-1. The study provides a potential strain Pyr-1 for the bioremediation in pyraclostrobin-contaminated aquatic environments.

BACKGROUND: Circular RNAs (circRNAs) are often dysregulated in cancers and closely related to cancer progression, including cutaneous squamous cell carcinoma (CSCC). However, the role and mechanism of circ_0068631 in CSCC progression have not been reported. METHODS: The expression of circ_0068631, microRNA-139-5p (miR-139-5p), and homeobox B7 (HOXB7) was measured by real-time quantitative polymerase chain reaction (RT-qPCR). Cell counting kit-8 (CCK-8) assay, 5-ethynyl-2'-deoxyuridine (EdU) assay, and colony formation assay were used to measure cell proliferation. Cell apoptosis was assessed by flow cytometry. Cell migration was detected by transwell assay. The interaction between miR-139-5p and circ_0068631 or HOXB7 was confirmed by dual-luciferase reporter assay. A xenograft tumor model was established to confirm the function of circ_0068631 in vivo. RESULTS: Circ_0068631 was upregulated in CSCC tissues and cells, and its silencing could inhibit CSCC cell proliferation and metastasis while promoting apoptosis in vitro, as well as restrain CSCC tumor growth in vivo. Circ_0068631 acted as a sponge of miR-139-5p, and miR-139-5p inhibition reversed the repressive effect of circ_0068631 knockdown on CSCC cell progression. Furthermore, HOXB7 was a target of miR-139-5p, and miR-139-5p inhibited the malignant behaviors by downregulating HOXB7 expression in CSCC cells. Further, circ_0068631 sponged miR-139-5p to regulate HOXB7 expression. CONCLUSION: Circ_0068631 functioned as a novel oncogene in CSCC progression by regulating miR-139-5p/HOXB7 axis, suggesting that circ_0068631 may be a potential target for CSCC treatment. HIGHLIGHTS: Circ_0068631 was overexpressed in CSCC tissues and cells. Circ_0068631 downregulation suppressed CSCC progression via miR-139-5p. Circ_0068631 regulated HOXB7 via sponging miR-139-5p.

Objective: To evaluate the different actions of crude and prepared Typhonii Rhizoma through pharmacological and statistical methods. Methods: Spontaneous activity test, sleep test, and seizure induced by dimefline in mice were used to examine the sedative and anticonvulsant effects of different prepared products of Typhonii Rhizoma. Twisting induced by acetic acid, law pain induced by formaldehyde, and ear swelling caused by xylene were applied to examine the anti-inflammatory and analgesic effects of those different prepared products. Then the pharmacological effects of different prepared Typhonii Rhizoma were comprehensively analyzed by principal components analysis method. Results: Typhonii Rhizoma significantly reduced the spontaneous activities of mice, prolonged the convulsions latency,decreased twisting times, licking time and ear swelling. The principal components analysis results showed that the sample order from strong to weak was Typhonii Rhizoma prepared by ginger and alum, prepared by alum,crude Typhonii Rhizoma, and those prepared by ginger. Conclusion: Crude and prepared Typhonii Rhizoma exert sedative, anticonvulsant, analgesic and anti-inflammatory effects in different degrees.

Hazardous Air Pollutants (HAPs) such as exhaust particulate matter (PM), carbon monoxide (CO), sulfur dioxide (SO2), nitrogen oxide (NOx), mercury (Hg) and polychlorinated dibenzo-p-dioxins and polychlorinated dibenzo-furans ( PCDD/Fs) are emitted by the process of cremation and the burning of oblation. Risks to health posed by emissions of hazardous air pollutants from crematories are emerging concerns. Through field investigation and data collection, we obtained the related activity levels and monitored the concentrations of air pollutants from typical cremators, so as to better understand the current pollutants emission levels for crematory. Using the emission factor method, we calculated the emission inventory of HAPs for crematory of Beijing in 2012 and quantified the range of uncertainty. Using atmospheric diffusion model ADMS, we evaluated the influence of crematories on the surrounding environment, and identified the characteristics of air pollution. The results showed that: for the cremators installed with flue gas purification system, the emission concentration of exhaust PM was rather low, and the CO emission concentration fluctuated greatly. However, relative high emission concentrations of PCDD/Fs were detected mainly due to insufficient combustion. Exhaust PM, CO, SO2, NOx, Hg and PCDD/Fs emitted by crematory of Beijing in 2012 were estimated at about 11. 5 tons, 41.25 tons, 2.34 tons, 7.65 tons, 13.76 kg and 0.88 g, respectively; According to the results of dispersion model simulation, the concentration contributions of exhaust PM, CO, SO2, NOx, Hg, PCDD/Fs from crematories were 0.05947 microg x m(-3), 0.2009 microg x m(-3) and 0.0126 microg x m(-3), 0.03667 microg x m(-3) and 0.06247 microg x m(-3), 0.004213 microg x m(-3), respectively.
