RESUMO

Root-knot nematodes (RKNs) pose a serious threat to crop production. Flooding soil with biogas slurry, combined with soil heating before crop planting, has the potential for RKN disease suppression. However, the actual effect of this method has not been verified under field conditions. Here, we present the results of a two-year field experiment in a greenhouse demonstrating the control effect on RKN disease and plant growth using this method, as well as its influence on the soil nematode community. Four treatments were set: untreated control (CK), local control method for RKN (CC), soil flooded with 70 % biogas slurry (BS70), and soil flooded with undiluted biogas slurry (BS100). In the first year, all three RKN control treatments significantly reduced the root-knot index (p < 0.05). In the next year, only BS70 and BS100 still presented significantly suppressed effects (p < 0.05), and it was more obvious under BS70 with a relative control effect of 74.6 %. In the first year, BS70 and BS100 significantly inhibited the plant height of watermelon (p < 0.05). In the next year, however, all three RKN control treatments promoted the growth of watermelon, and their stem diameter was significantly greater than that of CK. The application of biogas slurry (BS70 and BS100) significantly increased nematode richness and the Shannon index in the second year (p < 0.05). However, the structure index showed no significant difference among treatments (p > 0.05), indicating that biogas slurry application did not increase the soil food web complex. Principal component analysis showed that the application of biogas slurry changed the nematode community, especially under BS70, which presented a more lasting influence. The high-level input of biogas slurry also caused soil NH4+-N and heavy-metal and arsenic accumulation in the first year, but these soil-pollution risks disappeared in the second year.

Emerging spatial omics technologies continue to advance the molecular mapping of tissue architecture and the investigation of gene regulation and cellular crosstalk, which in turn provide new mechanistic insights into a wide range of biological processes and diseases. Such technologies provide an increasingly large amount of information content at multiple spatial scales. However, representing and harmonizing diverse spatial datasets efficiently, including combining multiple modalities or spatial scales in a scalable and flexible manner, remains a substantial challenge. Here, we present Giotto Suite, a suite of open-source software packages that underlies a fully modular and integrated spatial data analysis toolbox. At its core, Giotto Suite is centered around an innovative and technology-agnostic data framework embedded in the R software environment, which allows the representation and integration of virtually any type of spatial omics data at any spatial resolution. In addition, Giotto Suite provides both scalable and extensible end-to-end solutions for data analysis, integration, and visualization. Giotto Suite integrates molecular, morphology, spatial, and annotated feature information to create a responsive and flexible workflow for multi-scale, multi-omic data analyses, as demonstrated here by applications to several state-of-the-art spatial technologies. Furthermore, Giotto Suite builds upon interoperable interfaces and data structures that bridge the established fields of genomics and spatial data science, thereby enabling independent developers to create custom-engineered pipelines. As such, Giotto Suite creates an immersive ecosystem for spatial multi-omic data analysis.

The efficient remediation of antibiotic-contaminated soil is critical for agroecosystem and human health. Using the cost-effective and feedstock-independent hydrochar with rich oxygen-containing functional groups as a soil remediation material has become a hot concern nowadays. However, the feasibility and effectiveness of hydrochar amendment in antibiotic-contaminated soil still remain unknown. Therefore, this study investigated the remediation effect and potential mechanisms of different hydrochars from cow manure (H-CM), corn stalk (H-CS) and Myriophyllum aquaticum (H-MA) at two levels (0.5% and 1.0%) in oxytetracycline (OTC)-contaminated soil using a pot experiment. Results showed that compared with CK, OTC content in the soils amended with H-CM and H-MA was decreased by 14.02-15.43% and 9.23-24.98%, respectively, whereas it was increased by 37.03-42.64% in the soils amended with H-CS. Additionally, all hydrochar amendments effectively reduced the OTC uptake in root and shoot of Chinese cabbage by 10.41-57.99% and 31.92-65.99%, respectively. The response of soil microbial community to hydrochar amendment heavily depended on feedstock type rather than hydrochar level. The soil microbial metabolism (e.g., carbohydrate metabolism, amino acid metabolism) was enhanced by hydrochar amendment. The redundancy analysis suggested that TCA cycle was positively related to the abundances of OTC-degrading bacteria (Proteobacteria, Arthrobacter and Sphingomonas) in all hydrochar-amended soils. The hydrochar amendment accelerated the soil OTC removal and reduced plant uptake in soil-Chinese cabbage system by altering soil properties, enhancing OTC-degrading bacteria and promoting microbial metabolism. These findings demonstrated that the cost-effective and sustainable hydrochar was a promising remediation material for antibiotic-contaminated soil.

Objective: This investigation was undertaken to systematically assess the impact of increasingly rapid recovery treatment on the functional status of the knee following anterior cruciate ligament restructuring. Methods: Computer search from the establishment of the database to March 2022 in China Knowledge Network Database (CNKI), PubMed, EMBASE, ScienceDirect, CochraneLibrary, China VIP Database, Wanfang Database, and China Biomedical Literature Database (CBM). The control group was given only traditional rehabilitation training, and the observation group was treated with perioperative accelerated rehabilitation surgery intervention randomized controlled trial (RCT). Data for all included studies were extracted by two independent researchers, and the risk of bias for the quality of each included study was assessed by the Cochrane Handbook 5.1.0 criteria. Meta-analysis of the collected data by using RevMan5.4 statistical software. Results: A total of 593 articles were retrieved from the computer database and 8 RCT articles with a total of 636 samples were finally included for meta-analysis. According to the Jadad scale, the RCT score of 8 articles was ≥4 points. Meta-analysis was performed on the postoperative VAS scores of the ERAS group and the traditional rehabilitation group, and the heterogeneity test showed Chi2 = 288.60, df = 5, P < 0.00001, and I 2 = 99%. There was a statistically significant difference in the postoperative VAS scale between the intervention and the traditional rehabilitation model (P < 0.05). Eight articles reported the effect of accelerated rehabilitation surgery on the recovery of knee joint motion after ACL rehabilitation. After meta-analysis, the heterogeneity test showed Chi2 = 314.98, df = 7, P < 0.00001, and I 2 = 98%, and it can be concluded from the analysis that, compared with the traditional rehabilitation model, the enhanced rehabilitation surgery has an effect on the joint function after anterior cruciate ligament reconstruction. The improvement effect was more significant, and the difference was statistically significant (P < 0.05). Four articles reported the effect of enhanced recovery after surgery intervention on the range of motion of the knee joint of patients, the heterogeneity test showed Chi2 = 117.52, df = 2, P < 0.00001, and I 2 = 98%, through analysis, and compared with the traditional rehabilitation model and the enhanced recovery. The effect of surgery on the range of motion of the knee joint after ACL reconstruction was more significant, and the difference was statistically significant (P < 0.05). The effect of enhanced recovery after surgery and traditional rehabilitation mode on the incidence of postoperative adverse reactions in patients undergoing ACL reoperation was analyzed. The results of heterogeneity test showed that Chi2 = 1.59, df = 2, P=0.66 > 0.05, and I 2 = 98%, and the analysis showed that, compared with the traditional rehabilitation mode, enhanced rehabilitation surgery can significantly reduce the risk of adverse reactions after anterior cruciate ligament reconstruction (P < 0.05). An inverted funnel plot was used to analyze publication bias in studies with quality of life as an outcome measure. The results showed that Egger's test P=0.0005 < 0.001 suggesting that there may be a certain degree of publication bias. Conclusion: The existing research evidence shows that accelerating the reconstruction of anterior cruciate ligament can effectively promote the recovery of knee joint function, reduce the pain of patients, and reduce postoperative complications. However, more research is needed to further verify this.

ADP-ribosylation is a reversible post-translational modification where an ADP-ribose moiety is covalently attached to target proteins by ADP-ribosyltransferases (ARTs). Although best known for its nuclear roles, ADP-ribosylation is increasingly recognized as a key regulatory strategy across cellular compartments. ADP-ribosylation of mitochondrial proteins has been widely reported, but the exact nature of mitochondrial ART enzymes is debated. We have identified neuralized-like protein 4 (NEURL4) as a mitochondrial ART enzyme and show that most ART activity associated with mitochondria is lost in the absence of NEURL4. The NEURL4-dependent ADP-ribosylome in mitochondrial extracts from HeLa cells includes numerous mitochondrial proteins previously shown to be ADP-ribosylated. In particular, we show that NEURL4 is required for the regulation of mtDNA integrity via poly-ADP-ribosylation of mtLIG3, the rate-limiting enzyme for base excision repair (BER). Collectively, our studies reveal that NEURL4 acts as the main mitochondrial ART enzyme under physiological conditions and provide novel insights in the regulation of mitochondria homeostasis through ADP-ribosylation.

BACKGROUND: Knee osteoarthritis (KOA) is a common cause of disability among the elderly. We aimed to explore the effects of aldehyde dehydrogenase (ALDH) 2 on the progression of KOA and identifying the potential mechanisms. METHODS: First, ALDH2 expression in knee joint effusion of patients with KOA and the levels of oxidative stress-related markers were determined. After ALDH2 overexpression in monosodium iodoacetate (MIA)-treated SW1353 cells, cell viability was tested with CCK-8 assay. Subsequently, oxidative stress and inflammation-associated factors were measured. Meanwhile, cell apoptosis was assessed with TUNEL staining and expression of apoptosis-related proteins was detected by western blotting. To analyze the mechanism of ALDH2 in KOA, aquaporin 4 (AQP4) expression was determined using western blotting following ALDH2-upregulation. Subsequently, AQP4 was overexpressed to evaluate the changing of oxidative stress, inflammation and apoptosis in SW1353 cells exposed to MIA with ALDH2 overexpression. RESULTS: Results indicated that knee joint effusion with higher ALDH2 expression displayed lower oxidative stress. In addition, significantly upregulated ALDH2 expression was observed in MIA-treated SW1353 cells. ALDH2 overexpression oxidative stress, inflammation and apoptosis in SW1353 cells exposed to MIA. Moreover, MIA-triggered elevated expression of AQP4, which was reduced by ALDH2 overexpression. By contrast, AQP4-upregulation abrogated the inhibitory effects of ALDH2 on oxidative stress, inflammation and apoptosis in MIA-induced SW1353 cells. CONCLUSIONS: ALDH2 inactivates the expression of AQP4, by which mechanism the MIA-induced oxidative stress, inflammation and apoptosis injuries were alleviated, which provides a novel insight for understanding the mechanism of KOA and a promising target for the treatment of this disease.

OBJECTIVE: To compare accuracy of anterior cervical pedicle screws between assist of rapid prototyping 3D guide plate and free-hand insertion, and evaluate the safety of two methods. METHODS: Eight adult cervical cadaver specimens after formaldehyde immersion, including 4 males and 4 females, aged 32 to 65(40.3±5.6) years old. After X-ray examination to exclude bone damage and deformity, 4 of them (3D guide plate group) randomly selected were for CT scan to obtain DICOM format data, and the data was imported into Mimics software for model, designed the ideal entry point and nail path for anterior cervicaltranspedicular screw (ATPS). After obtaining the personalized guide plate of the nail channel, it was exported as STL data, and the individual guide plate was printed by rapid prototyping and 3D printing technology. In turn, with the assistance of 3D guide plates, one-to-one personalized ATPS screws were placed on the four lower cervical cadaver specimens. Another 4 (free-hand group) lower cervical cadaver specimens were implanted with ATPS screws using free-hand technique. All specimens were performed CT thin-layer scanning and three-dimensional reconstruction after operation. The Tomasino method was used to evaluate the safety of the screws on the CT cross-sectional and sagittal images, to determine whether there was a cortical puncture of the lower and inner edges of the pedicle. According to the CT rating results, gradeⅠandⅡwere safe, and grade Ⅲ- Ⅴ were dangerous.And the accuracy of screws was recorded and analyzed between two groups. RESULTS: Two screws were inserted in each segment from C3 to C7 in 8 adult cadavers. A total of 80 screws were inserted, 40 in the 3D guide plate group, and 40 in the free-hand group. The Tomasino screw rating method was used to evaluate the safety of screw, 21 screws were gradeⅠ, 14 screws were gradeⅡ, 3 screws were grade Ⅲ, 1 screw was grade Ⅳ, 2 screws were grade Ⅴ in 3D guide plate group, while 14 screws were gradeⅠ, 8 screws were gradeⅡ, 8 screws were grade Ⅲ, 6 screws were grade Ⅳ, 2 screws were grade Ⅴ in free-hand group. The safety rate of 3D guide plate group was 87.5%, and 55.0% of the free hand group (χ2=8.7, P=0.003). CONCLUSION: The 3D printing rapid prototyping guide plate assisted insertion of the anterior cervical pedicle screw can significantly improve the accuracy and safety, and provide a theoretical basis for further clinical application.

Advanced modulation and multiple access schemes with high spectral efficiencies are desirable to overcome the bandwidth limitation in low-cost optical and electrical devices to fulfill the high-data rate requirements in passive optical networks (PONs). We propose a non-orthogonal multiple access (NOMA) scheme, known as power-code division NOMA (PCD-NOMA), for the next-generation PON, where the optical network units (ONUs) are divided into several groups with similar path losses. The ONUs per groups are allocated in the same power domain multiplexing layer with different codebooks. We show by experimental demonstration that the proposed PCD-NOMA with a high spectral efficiency offers improved overall system performance and reduced required transmission power in the next-generation PON, particularly in flexible PON where ONUs have different path losses. For PON with a power difference loss of 14 dB, the reduced required transmission powers are 5 and 11 dB for downstream and upstream, respectively, compared with orthogonal frequency division multiple access.

In order to increase understanding of the hydrogeochemical effects that influence changes in the quality of salt water, we investigated the distribution of saline and fresh water in an inland plain area and, in particular, the scarcity of fresh water resources. Taking the inland plain in Jiyang County as a specific case study, samples of undisturbed clay and underground saline water from different depths were collected to examine hydrogeological changes. A wide variety of methods was used to analyze the blocking effect of clay on the chemical characteristics of the groundwater. These include real-time monitoring for field water quality, tests for isothermal adsorption, a factor analysis model, physiochemical analysis, and correlation analysis. Our results show that the optimal adsorption isotherm of clay for Na⁺, Ca2+ and Mg2+ in groundwater conform to the established Henry and Langmuir equations for adsorption isotherms. The influence of clay mineral types and content on the blockage of Na⁺, Ca2+ and Mg2+ in groundwater samples were evident at different depths, with the clay adsorption capacity increasing in line with increases in the clay mineral content. Clay at different depths was found to have the strongest blocking effect on Na⁺ in groundwater, being systematically greater than its effect on Ca2+ and Mg2+. It is believed that the blocking effect of clay has an important influence on the hydrochemical zoning of groundwater in inland plains and the formation of saline water in groundwater systems. This study therefore provides concrete evidence in support of this supposed effect.

Composting is regarded as an effective treatment to suppress pathogenic organisms and stabilize the organic material in sewage sludge. This study investigated the use of biochar as an amendment to improve the composting effectiveness and reduce the bioavailability of heavy metals and loss of nitrogen during composting. Biochar of 0%, 1%, 3%, 5% and 7% were added into a mixture of sludge and straw, respectively. The use of biochar, even in small amounts, altered the composting process and the properties of the end products. Biochar addition resulted in a higher pile temperature (66°C) and could reduce nitrogen loss by transforming ammonium into nitrite. In the 5% biochar group, the final product from sludge composting, ammonia nitrogen, decreased by 22.4% compared to the control, and nitrate nitrogen increased by 310.6%. Considering temperature and N transformation, the treatment with 5% biochar is suggested for sludge composting.

OBJECTIVE: To study relationships between serum ferritin and bone metabolism in patients with hip fragility fractures. METHODS: This cross-sectional study included 76 postmenopausal women with hip fracture from Feburary 2011 to June 2012. The mean age of the women was (73 ± 10) years (range, 55-93 years) and the mean duration of menstruation was (22 ± 10)years (range, 5-50 years). Serum concentrations of ferritin, transferrin, alkaline phosphatase (ALP), amino-terminal extension peptide of type I collagen (P1NP), C-terminal telopeptides of type I collagen (ß-CTX)and femoral and lumbar bone mineral density by dual-energy X-ray absorptiometry were measured. Bone metabolism was compared between normal and elevated ferritin groups with t-test, Pearson linear, partial correlation and multiple regression analysis examined associations between iron- and bone-related markers. RESULTS: Serum ferritin concentration raised to (230 ± 146)µg/L, transferrin concentration reduced to (1.89 ± 0.33)g/L. P1NP concentration raised to (61 ± 32) ng/L when the concentration of serum ALP and ß-CTX were in the normal range. T-scores for bone mineral density in the femoral neck (-2.0 ± 1.1) and lumbar (-2.1 ± 1.2) were below the normal ranges(-1.0-1.0). The subjects were divided into two groups according to serum ferritin concentration, normal group(serum ferritin concentration ≤ 150 µg/L, n = 25) and elevated group(serum ferritin concentration > 150 µg/L, n = 51). Patients of elevated group had lower bone mineral density in femoral neck and lumbar than normal group(t = 3.13,2.89, P < 0.01), and higher P1NP, ß-CTX concentration (t = -2.38, -3.59, P < 0.05) . In partial correlation analysis adjusted for confounders, serum ferritin concentration was correlated negatively with bone mineral density in both femoral neck and lumbar (r = -0.335,-0.295, P < 0.05), and positively with P1NP and ß-CTX (r = 0.467,0.414, P < 0.05), but not correlated with ALP (r = 0.188, P > 0.05). Transferrin concentration tended to be correlated positively with bone mineral density in both femoral neck and lumbar (r = 0.444, 0.262, P < 0.05) and negatively with ALP, P1NP and ß-CTX(r = -0.326,-0.285,-0.278, P < 0.05). CONCLUSIONS: Iron overload has a high prevalence in postmenopausal women with fragility fracture. Increased iron stores, which might lead to bone loss and lower bone mineral density by enhancing the activity of bone turnover, could be an independent factor to take effects on bone metabolism on postmenopausal women.

An open field experiment was conducted to study the effects of applying controlled-release fertilizer blended with rapidly available chemical N fertilizer on Chinese cabbage yield and quality as well as nitrogen losses, including ammonia volatilization and NO3- -N accumulation and leaching in Beijing suburb. The results showed that a combined application of 2:1 controlled-release fertilizer and urea fertilizer (total N rate 150 kg x hm(-2)) did not induce the reduction of Chinese cabbage yield, and decreased the leaf nitrate and organic acid contents significantly, compared with conventional urea N application (300 kg x hm(-2)), and had no significant difference in the cabbage yield and leaf nitrate content, compared with applying 150 kg x hm(-2) of urea N. The combined application of 2:1 controlled-release fertilizer and urea fertilizer improved the N use efficiency of Chinese cabbage, and reduced the ammonia volatilization and NO3- -N leaching. At harvest, the NO3- -N concentrations in 20-40, 60-80 and 80-100 cm soil layers were significantly lower in the combined application treatment than in urea N treatment.

The study of nitrogen (N) balance in stock farming system is the basis of nutrient management in animal husbandry production, and the determination of N cycling parameters is the foundation of this study. Through documents checking and data collecting, this paper analyzed the items and values of input/output parameters of N cycling in stock farming system, and ascertained the estimation parameters of N balance in the system, which could offer the theoretical foundation for nutrient management in animal husbandry production.
