RESUMO

Ovarian cancer (OV) poses a significant challenge in clinical settings due to its difficulty in early diagnosis and treatment resistance. FOXP4, belonging to the FOXP subfamily, plays a pivotal role in various biological processes including cancer, cell cycle regulation, and embryonic development. However, the specific role and importance of FOXP4 in OV have remained unclear. Our research showed that FOXP4 is highly expressed in OV tissues, with its elevated levels correlating with poor prognosis. We further explored FOXP4's function through RNA sequencing and functional analysis in FOXP4-deficient cells, revealing its critical role in activating the Wnt signaling pathway. This activation exacerbates the malignant phenotype in OV. Mechanistically, FOXP4 directly induces the expression of protein tyrosine kinase 7 (PTK7), a Wnt-binding receptor tyrosine pseudokinase, which causes abnormal activation of the Wnt signaling pathway. Disrupting the FOXP4-Wnt feedback loop by inactivating the Wnt signaling pathway or reducing FOXP4 expression resulted in the reduction of the malignant phenotype of OV cells, while restoring PTK7 expression reversed this effect. In conclusion, our findings underscore the significance of the FOXP4-induced Wnt pathway activation in OV, suggesting the therapeutic potential of targeting this pathway in OV treatment.

The ability of cells to perceive and respond to mechanical cues is essential for numerous biological activities. Emerging evidence indicates important contributions of organelles to cellular mechanosensitivity and mechanotransduction. However, whether and how the endoplasmic reticulum (ER) senses and reacts to mechanical forces remains elusive. To fill the knowledge gap, after developing a light-inducible ER-specific mechanostimulator (LIMER), we identify that mechanostimulation of ER elicits a transient, rapid efflux of Ca2+ from ER in monkey kidney COS-7 cells, which is dependent on the cation channels transient receptor potential cation channel, subfamily V, member 1 (TRPV1) and polycystin-2 (PKD2) in an additive manner. This ER Ca2+ release can be repeatedly stimulated and tuned by varying the intensity and duration of force application. Moreover, ER-specific mechanostimulation inhibits ER-to-Golgi trafficking. Sustained mechanostimuli increase the levels of binding-immunoglobulin protein (BiP) expression and phosphorylated eIF2α, two markers for ER stress. Our results provide direct evidence for ER mechanosensitivity and tight mechanoregulation of ER functions, placing ER as an important player on the intricate map of cellular mechanotransduction.

Mitochondria constantly fuse and divide for mitochondrial inheritance and functions. Here, we identified a distinct type of naturally occurring fission, tail-autotomy fission, wherein a tail-like thin tubule protrudes from the mitochondrial body and disconnects, resembling autotomy. Next, utilizing an optogenetic mitochondria-specific mechanostimulator, we revealed that mechanical tensile force drives tail-autotomy fission. This force-induced fission involves DRP1/MFF and endoplasmic reticulum tubule wrapping. It redistributes mitochondrial DNA, producing mitochondrial fragments with or without mitochondrial DNA for different fates. Moreover, tensile force can decouple outer and inner mitochondrial membranes, pulling out matrix-excluded tubule segments. Subsequent tail-autotomy fission separates the matrix-excluded tubule segments into matrix-excluded mitochondrial-derived vesicles (MDVs) which recruit Parkin and LC3B, indicating the unique role of tail-autotomy fission in segregating only outer membrane components for mitophagy. Sustained force promotes fission and MDV biogenesis more effectively than transient one. Our results uncover a mechanistically and functionally distinct type of fission and unveil the role of tensile forces in modulating fission and MDV biogenesis for quality control, underscoring the heterogeneity of fission and mechanoregulation of mitochondrial dynamics.

Low-temperature stress is a key factor limiting the yield and quality of the common bean. 5-aminolevulinic acid (5-ALA), an antioxidant in plants, has been shown to modulate plant cold stress responses. However, the molecular mechanisms of 5-ALA-induced physiological and chemical changes in common bean seedlings under cold stress remains unknown. This study explored the physiological and transcriptome changes of common bean seedlings in response to cold stress after 5-ALA pretreatment. Physiological results showed that exogenous 5-ALA promotes the growth of common bean plants under cold stress, increases the activity of antioxidant enzymes (superoxide dismutase: 23.8%; peroxidase: 10.71%; catalase: 9.09%) and proline content (24.24%), decreases the relative conductivity (23.83%), malondialdehyde (33.65%), and active oxygen content, and alleviates the damage caused by cold to common bean seedlings. Transcriptome analysis revealed that 214 differentially expressed genes (DEGs) participate in response to cold stress. The DEGs are mainly concentrated in indole alkaloid biosynthesis, carotenoid biosynthesis, porphyrin, and chlorophyll metabolism. It is evident that exogenous 5-ALA alters the expression of genes associated with porphyrin and chlorophyll metabolism, as well as the plant hormone signal transduction pathway, which helps to maintain the energy supply and metabolic homeostasis under low-temperature stress. The results reveal the effect that applying exogenous 5-ALA has on the cold tolerance of the common bean and the molecular mechanism of its response to cold tolerance, which provides a theoretical basis for exploring and improving plant tolerance to low temperatures.

Understanding the spatial distribution and path tracing of eutrophication caused by nitrogen (N) enrichment in urban freshwater is crucial for whole-process and precise damage effect control. This study constructed a site-specific life cycle impact assessment (LCIA) model, covering the overall cause-effect chain from source emission to endpoint effect, to assess N-induced eutrophication potential at the species damage level. Applied to Guangzhou city, China, marked spatial disparities in eutrophication potential were derived, with higher values in the downtown areas driven by anthropogenic disturbances, such as wastewater discharge. Spatially differentiated measures were provided through eutrophication hotspot identification and driver tracking. This study offers a necessary complement for eutrophication impact category indicators in LCIA methodology and lays a scientific foundation for potential hotpots diagnosis and targeted mitigation policy-making.

FGFR fusions retaining the FGFR kinase domain are active kinases that are either overexpressed or constitutively activated throughout diverse cancer types. The presence of FGFR translocations enhances tumor cell proliferation and contributes to significant sensitivity to FGFR kinase inhibitors. FGFR2 as an actionable target in intrahepatic cholangiocarcinoma (iCCA) has been tested in many clinical trials. FISH (fluorescence in situ hybridization) and NGS (next-generation sequence) are well-known tools to investigate the translocations of FGFR with multiple or unknown translocation partners. A rapid and robust FISH assay was developed and validated to detect FGFR2 translocations from FFPE specimens in iCCA. The analytical performance of the FISH assay was evaluated for probe localization, probe sensitivity and specificity, and assay precision. Twenty-five archival FFPE specimens from local iCCA patients were tested for FGFR2 translocations. FISH results were correlated with that of NGS on some samples. Biallelic translocations and a novel FGFR2 translocation involving the partner gene, SHROOM3, t(4;10) (q21;q26), were identified in a local iCCA patient.

Substantial evidence indicates that China's afforestation statistically contributed to the ecosystem services (ES) improvement. However, we found the potential challenges behind this improvement, especially in water-limited areas. We propose an attribution analysis method, which can assess the specific contribution of natural, human and cognition degree drivers to ES dynamics. The results found that the ratio of natural and human drivers in the area north of China's 400 mm precipitation isopleth is 2:7. This means local vegetation capacity has already exceeded water limitation, implying a conflict between nature and humans. However, the natural contribution in the area between 400 and 800 mm precipitation isopleth is negative, whereas the human contribution is 91%. This means this area has fragile natural conditions and needs more flexible policies. The ratio of natural and human drivers in the region south of 800 mm precipitation isopleth is 6:3, suggesting the ecological policies here can be maintained.

Polyamine oxidases (PAOs), which are flavin adenine dinucleotide-dependent enzymes, catalyze polyamine (PA) catabolism, producing hydrogen peroxide (H2O2). Several PAO family members have been identified in plants, but their expression in pepper plants remains unclear. Here, six PAO genes were identified in the 'Zunla-1' pepper genome (named CaPAO1-CaPAO6 according to their chromosomal positions). The PAO proteins were divided into four subfamilies according to phylogenetics: CaPAO1 belongs to subfamily I; CaPAO3 and CaPAO5 belong to subfamily III; and CaPAO2, CaPAO4, and CaPAO6 belong to subfamily IV (none belong to subfamily II). CaPAO2, CaPAO4, and CaPAO6 were ubiquitously and highly expressed in all tissues, CaPAO1 was mainly expressed in flowers, whereas CaPAO3 and CaPAO5 were expressed at very low levels in all tissues. RNA-seq analysis revealed that CaPAO2 and CaPAO4 were notably upregulated by cold stress. CaPAO2 and CaPAO4 were localized in the peroxisome, and spermine was the preferred substrate for PA catabolism. CaPAO2 and CaPAO4 overexpression in Arabidopsis thaliana significantly enhanced freezing-stress tolerance by increasing antioxidant enzyme activity and decreasing malondialdehyde, H2O2, and superoxide accumulation, accompanied by the upregulation of cold-responsive genes (AtCOR15A, AtRD29A, AtCOR47, and AtKIN1). Thus, we identified candidate PAO genes for breeding cold-stress-tolerant transgenic pepper cultivars.

Recently, the rapid growth in vehicle activity in rapidly urbanized areas has led to the discharge of large amounts of polycyclic aromatic hydrocarbons (PAHs) into roadside soils and these compounds have gradually accumulated in the soil, which poses a serious threat to national food security and public health. However, previous studies did not clearly investigate the seasonal differences in PAH pollution of roadside soil by different highways. Therefore, based on field investigations, this study collected 84 soil surface samples to compare the pollution characteristics of 16 PAHs in farmland soils located near different roads in different seasons in Guangzhou, China. The results showed that the concentration of Σ16PAHs in farmland soils in spring (with a mean value of 258.604 µg/kg) was much higher than that in autumn (with a mean value of 157.531 µg/kg). There are differences in the PAH compositions in spring (4 ring > 3 ring > 5 ring > 6 ring) and autumn (4 ring > 5 ring > 6 ring > 3 ring). The proportion of 4−6 ring PAHs was much higher than 2−3 ring PAHs in both seasons. The spatial differences were significant. The sampling areas with higher concentrations of 16 PAHs were Tanbu Town, Huadu District (TB), Shitan Town, Zengcheng District (ST), and Huashan Town, Huadu District (HS), while the lowest concentration was in Lanhe Town, Nansha District (LH). The results of the diagnostic ratios showed that the main source of soil PAHs consists of a mixed source from petroleum and biomass combustion. The results from the total pollution assessment method and Nemerow index method indicated that the pollution levels of PAHs in the farmland soils indicated weak contamination. Our study provides a scientific basis for the prevention and control of soil pollution in farmlands near highways.

The COVID-19 pandemic brings a surge in household electricity consumption, thereby enabling extensive research interest on residential carbon emissions as one of the hot topics in carbon reduction. However, research on spatial-temporal driving forces for the increase of residential CO2 emissions between regions still remains unknown in terms of emissions mitigation in post-pandemic era. Therefore, we studied the residential CO2 emissions from the electricity consumption of China during the period 1997-2019. Afterward, the regional specified production emission factors, combining with electricity use pattern, living standard and household size, were modelled to reveal the spatial-temporal driving forces at national and provincial scales. We observed that the national residential electricity-related CO2 increased from 1997 to 2013, before fluctuating to a peak in 2019. Guangdong, Shandong and Jiangsu, from East China were the top emitters with 27% of the national scale. The decomposition results showed that the income improvement was the primary driving force behind the emission increase in most provinces, while the household size and production emission effects were the main negative effects. For the spatial decomposition, differences in the total households between regions further widen the gaps of total emissions. At the provincial scale of temporal decomposition, eastern developed regions exhibited the most significant decrease in production emissions. In contrast, electricity intensity effect showed negative emission influences in the east and central regions, and positive in north-eastern and western China. The research identified the different incremental patterns of residential electricity-related CO2 emissions in various Chinese provinces, thereby providing scientific ways to save energy and reduce emissions.

The role of ocean carbon sinks in global climate change mitigation and carbon neutrality is still affected by lack of research. Aiming at overcoming the present limitations, a comprehensive and holistic framework and accounting method of ocean carbon sink evaluation are proposed in this study, which consider both carbon sink types and their characteristic carbon storage cycle timescales. The results show that (1) China's total ocean carbon sink is 69.83-106.46 Tg C/year, among which the mariculture, coastal wetlands, and offshore carbon sinks are 2.27-4.06, 2.86-5.85, and 64.70-96.55 Tg C/year, respectively; (2) ocean-based solutions such as coastal protection and restoration, mariculture development, ocean alkalization, ocean fertilization, and marine bioenergy with carbon capture and storage have substantial mitigation potential, but further investigation is required before large-scale deployment; (3) although China's ocean carbon sinks only counterbalanced 3.27-4.99% of its fossil fuel emissions, their tremendous enhancing potential and specific advantages cannot be ignored, and enhancing measures must be taken according to regional characteristics; (4) some uncertainties and limitations still exist, and problems such as double counting, carbon sink offset, and so forth need to be further considered. In a word, this study provides a basis for the development of ocean-based solutions on closing climate mitigation gaps.

Ultraviolet (UV), particularly UVB, is widely used in the treatment of skin diseases including psoriasis, atopic dermatitis, vitiligo, mycosis fungoides and pruritus. Recently, there has been a trend of replacing broad-band UVB (BB-UVB) units with narrow-band UVB (NB-UVB), as studies have demonstrated that NB-UVB is more efficacious in the treatment of psoriasis. The purpose of this study is to evaluate the biological effects and transcriptome changes induced by light-emitting diode-based NB-UVB (NB-UVB LED) phototherapy. Cell viability and the cell migration ability were significantly decreased posttreatment, as well as apoptosis and ROS levels were remarkably increased. NB-UVB-induced S phase arrest was observed 12 h postirradiation. Bioinformatics analysis of transcriptome sequencing data revealed that NB-UVB LED irradiation induced dose-depended changes in multiple key signaling pathways, such as PI3K and cytoskeletal-related pathways. The depolymerization of cytoskeleton induced by NB-UVB was observed 24 h posttreatment. In addition, the expression levels of cytoskeleton-related proteins FN1, ITGB4, ITGA1, RAC2 and DOCK1 decreased significantly 12 h after irradiation. Our results indicated that NB-UVB LED may serve as a novel option for the development of NB-UVB phototherapy devices.

AIMS: Mechanical stress overload in the temporomandibular joint (TMJ) is an important cause of TMJ osteoarthritis (TMJOA). Whether secreted frizzled-related proteins (SFRPs) play important roles in the development of mechanical stress-induced TMJOA remains controversial. In this study, we investigated the roles of the Wnt/ß-catenin signaling and SFRPs in the progression of mechanical stress-induced TMJOA. METHODS: We investigated the progression of mechanical stress-induced TMJOA using an in vivo model via modified increased occlusal vertical dimension (iOVD) malocclusion and an in vitro model in which isolated chondrocytes were subjected to mechanical stress. The effects of inhibition of Wnt/ß-catenin signal on TMJOA induced by mechanical stress were studied by in vitro drug added and in vivo intra-articular injection of XAV-939. TMJOA progression, Wnt/ß-catenin signaling and SFRPs was assessed by Cone beam computed tomography (CBCT) analysis, histochemical and immunohistochemical (IHC) staining, quantitative real-time PCR (qRT-PCR), Western blotting (WB), and immunofluorescence (IF) staining. RESULTS: Our in vivo results showed that iOVD-induced mechanical stress in the TMJ disrupted mandible growth, induced OA-like changes in TMJ cartilage, and increased OA-related cytokine expression. In addition, iOVD activated Wnt/ß-catenin signaling and suppressed Sfrp1, Sfrp3, and Sfrp4 expression in condylar cartilage. Moreover, our in vitro study showed that stress disrupted homeostasis, activated Wnt/ß-catenin signaling and inhibited SFRP3 and SFRP4 expression in chondrocytes. Suppression of Wnt/ß-catenin signaling with XAV-939 promoted SFRP3 and SFRP4 expression and rescued mechanical stress-induced cartilage degeneration in vivo and in vitro. CONCLUSIONS: Our work suggests that mechanical stress reduces SFRPs expression both in vivo and in vitro and promotes TMJOA via Wnt/ß-catenin signaling. Suppression of Wnt/ß-catenin signaling promotes SFRPs expression, especially SFRP3 and SFRP4 expression, and rescues mechanical stress-induced cartilage degeneration. Wnt/ß-catenin signaling and SFRPs may represent potential therapeutic targets for TMJOA.

BACKGROUND: Unilateral posterior crossbite, one of the most frequent malocclusions, is often associated with functional lateral shift of the mandible. Although the effects of functional lateral shift on the mandible and temporomandibular joint have been examined in various animal experiments, cranial and maxillary changes have received less attention. OBJECTIVE: The aim of this study was to investigate the effects of functional lateral shift on the craniofacial complex in growing rats. METHODS: Eighty 5-week-old male Sprague-Dawley rats were randomly divided into an experimental group (n = 40), which received an oblique guide appliance that shifted the mandible to the left during closure, and a control group (n = 40). The rats were scanned by cone-beam computed tomography at 3 days and 1, 2, 4 and 8 weeks. The dimensions of the mandibular bone, condyle, maxilla and cranium were measured. RESULTS: The mandibles of rats in the experimental group were smaller than those of the rats in the control group and were asymmetrical. The condyles of the rats in the experimental group were thinner than those of the control rats. The condylar length on the ipsilateral side was shorter and wider than that on the contralateral side from 4 to 8 weeks. No significant differences in cranial length or height were observed between the experimental and control groups. The height of the upper first molar and alveolar bone on the contralateral side was significantly smaller than that on the ipsilateral side and in the controls from 4 to 8 weeks. CONCLUSION: Functional shift in the mandible produces morphological asymmetries in the mandible and maxillary region and may cause bilateral condylar degenerative changes.

Elevated air pollution, along with rapid urbanization, have imposed higher health risks and a higher disease burden on urban residents. To accurately assess the increasing exposure risk and the spatial association between PM2.5 and lung cancer incidence, this study integrated PM2.5 data from the National Air Quality Monitoring Platform and location-based service (LBS) data to introduce an improved PM2.5 exposure model for high-precision spatial assessment of Guangzhou, China. In this context, the spatial autocorrelation method was used to evaluate the spatial correlation between lung cancer incidence and PM2.5. The results showed that people in densely populated areas suffered from higher exposure risk, and the spatial distribution of population exposure risk was highly consistent with the dynamic distribution of the population. In addition, areas with PM2.5 roughly overlapped with areas with high lung cancer incidence, and the lung cancer incidence in different locations was not randomly distributed, confirming that lung cancer incidence was significantly associated with PM2.5 exposure. Therefore, dynamic population distribution has a great impact on the accurate assessment of environmental exposure and health burden, and it is necessary to use LBS data to improve the exposure assessment model. More mitigation controls are needed in highly populated and highly polluted areas.

Mandibular deviation affects the biomechanical environment of the temporomandibular joint (TMJ) and causes thinning of cartilage on the deviated side. We aimed to evaluate, using a rat model, the effect of mandibular functional deviation on the TMJ in relation to the functional roles of integrin ß family members. The effects of experimental functional deviation on the TMJ of 6-week-old Sprague-Dawley female rats, randomly assigned to control (n = 42) and experimental groups (n = 42), were evaluated at 3 days and 1, 2, 4, and 8 weeks by histological staining, immunofluorescence, real-time quantitative polymerase chain reaction, and micro-computed tomography. The results showed that the experimental functional shift changed the shape of condyles, thinned the cartilage, and increased the proportion of the hypertrophic layer on the deviated sides of condyles. In addition, the extracellular matrix of the condyle cartilage exhibited degradation at 1 week and subchondral trabecular bone was lost at 4 and 8 weeks. Osteoarthritis (OA)-like changes occurred in the left and right condyles of rats in the experimental group and were aggravated over time. Integrin ß family expression, especially integrin ß2 , was altered from week 1, possibly related to the OA-like changes. These data may provide insight into the onset of TMJ OA.

BACKGROUND: We report a case and its 4-year follow-up of Osteoglophonic dysplasia (OD), a rare disease that disturbs both skeletal and dental development, which is usually caused by heterozygous FGFR1 mutations. CASE PRESENTATION: This article presents a case where a 6-year-old male patient suffered dysregulation of tooth eruption and was diagnosed with osteogenic dysplasia from a fibroblast growth factor receptor 1 (FGFR1) heterozygote mutation. However, the number of teeth is within the normal range, and their roots are well developed. Several interventions were implemented with varying degrees of results. The details of the 4-year follow-up showed that the signs of OD were more pronounced, including dwarfism, frontal bossing, delayed skeletal maturation, anteverted nares, micrognathia, and prominent ears, but the patient's impacted teeth and edentulous jaws remained unchanged. CONCLUSIONS: FGFR1 heterozygote mutation and OD present significant difficulty for teeth eruption and subsequent intervention. Further measures ought to be taken in recognizing various symptoms presented by the patient. This case supports the significance of careful inquiry, comprehensive physical examination and correct diagnosis as indispensable steps for clinical practice in patients with unerupted teeth. Additionally, the detailed case and its 4-year follow-up length may provide new insights into osteogenic dysplasia and patients with impacted teeth while encouraging further exploration in treatment methods.

Many nations are taking measures to address the negative impacts of development projects in order to achieve the goal of No Net Loss (NNL) of ecosystem services. It still faces the most fundamental and critical problem- the unified accounting of ecosystem service. To address this problem, we conducted an emergy-based ecosystem service assessment to tackle the challenges and obstacles of current accounting methods and established a new accounting framework to provide a unified measurement for ecosystem service marketing. We adopted the Credit-Debit method of wetland mitigation banking and the emergy-based method to evaluate the ecosystem services of 82 ecological projects. The results have shown that: (1) The emergy-based credit covers more types of ecosystem services, and shows advantages in terms of objectivity, accuracy, dynamic monitoring, and generalization; (2) The new accounting method can provide cross-scale and cross-type ecosystem service; (3) The Em-credit helps to promote the trans-regional eco-bank system in a large scale and at multiple levels, and it can improve the effective supervision and predict the eco-bank in a long period.

Asparagus bean (Vigna unguiculata ssp. sesquipedialis) is an important cowpea subspecies. We assembled the genomes of Ningjiang 3 (NJ, 550.31 Mb) and Dubai bean (DB, 564.12 Mb) for comparative genomics analysis. The whole-genome duplication events of DB and NJ occurred at 64.55 and 64.81 Mya, respectively, while the divergence between soybean and Vigna occurred in the Paleogene period. NJ genes underwent positive selection and amplification in response to temperature and abiotic stress. In species-specific gene families, NJ is mainly enriched in response to abiotic stress, while DB is primarily enriched in respiration and photosynthesis. We established the pan-genomes of four accessions (NJ, DB, IT97K-499-35 and Xiabao II) and identified 20,336 (70.5%) core genes present in all the accessions, 6,507 (55.56%) variable genes in two individuals, and 2,004 (6.95%) unique genes. The final pan genome is 616.35 Mb, and the core genome is 399.78 Mb. The variable genes are manifested mainly in stress response functions, ABC transporters, seed storage, and dormancy control. In the pan-genome sequence variation analysis, genes affected by presence/absence variants were enriched in biological processes associated with defense responses, immune system processes, signal transduction, and agronomic traits. The results of the present study provide genetic data that could facilitate efficient asparagus bean genetic improvement, especially in producing cold-adapted asparagus bean.

Objectives. Safety assessment helps the development of continuous improvement strategies in construction safety, especially coping with dynamic changes to the on-site environment with uncertainties. This article proposes a composite safety assessment based on on-site conditions to facilitate improved and proactive construction safety management. Methods. First, based on evident rectification records, we utilized set pair analysis, a grey rough approach and a coevolution approach to quantify overall safety performance. Second, we incorporated two safety performance indicators into a composite assessment framework, using rough set theory and fluid dynamics. Finally, the assessment results of the seven completed projects were compared. Results. The coevolution approach had novel advantages in assessing rectification performance and the fluid dynamics approach could enhance the proactive warning ability of the safety assessment. Conclusion. Theoretically, the research contributes to new insights into the quantification of construction safety assessment under dynamic on-site conditions. Practically, it also contributes to the active and objective measurement of management performance and promotes the dynamic and stable safety performance evaluation for on-site construction.
