The objective of this study was to evaluate the efficacy of two multivalent commercial porcine circovirus (PCV) vaccines against heterologous PCV2d challenges. A total of 24 crossbred male pigs aged 26 days selected from a specific pathogen-free herd were randomly divided into four groups (six pigs per group) and assigned as follows: negative control (unvaccinated/sham-challenge), vaccinated with chimeric PCV1-2a vaccine (PCV1-2a/PCV2d-challenge), vaccinated with chimeric PCV1-2a-2b vaccine (PCV1-2a-2b/PCV2d-challenge) and positive control (unvaccinated/PCV2d-challenge). At 21 days after vaccination, the pigs were intranasally and intramuscularly inoculated with either sham or field isolates of PCV2d (PCV2d/149/TH/2020). After being challenged, blood samples were obtained weekly and analyzed for levels of PCV2d viremia, neutralizing antibodies, and IgG against PCV2. At 30 days post-challenge (DPC), the pigs were euthanized and then subjected to pathological evaluations and molecular analysis. The results indicated that pigs in the PCV1-2a-2b/PCV2d-challenge and the PCV1-2a/PCV2d-challenge groups possessed significantly greater levels of PCV2d-neutralizing antibody titer when compared with the positive control group. Moreover, pigs in the PCV1-2a-2b/PCV2d-challenge group exhibited a lower degree of severity in terms of gross lesion scores and lower levels of PCV2 viremia when compared with the positive control group. This study demonstrated that vaccinating pigs with either the PCV1-2a or PCV1-2a-2b chimeric vaccines elicits a potent immune response against PCV2d infection and reduces viremia after PCV2d inoculation in pigs.

Human norovirus is a major virus that causes acute gastroenteritis in all age groups. Recently, norovirus recombinant strains have been reported as the cause of norovirus outbreaks. This study has investigated the distribution of norovirus genotypes and recombinant strains circulating in children hospitalized with diarrhea in Chiang Mai, Thailand from 2017 to 2018. A total of 882 stool specimens were tested for the presence of norovirus GI and GII by reverse transcription-PCR (RT-PCR) assay. Genotypes of the viruses were assessed by partial VP1 nucleotide sequencing and the representative strains were further characterized for norovirus recombinant strains by amplification of ORF1 (RdRp)/ORF2 (VP1 capsid) junction region. From a total of 882 stool samples, 131 (14.9%) were positive for norovirus, of which the majority of norovirus genogroups were norovirus GII, and only one was identified as norovirus GI. A wide variety of norovirus genotypes were detected in this study, including GI.5, GII.2, GII.3, GII.4, GII.6, GII.7, GII.13, GII.14, and GII.17 with the predominance of GII.2 (62.5%) in 2017 and GII.4 (57.0%) in 2018. Nevertheless, it should be noted that GII.4 remained the most predominant genotype (50.4%) in overall prevalence. Analysis of norovirus recombination revealed that several norovirus recombinant strains (GII.2[P16], GII.3[P16], GII.4[P16], GII.4[P31], GII.6[P7], GII.13[P16], and GII.14[P7]) had been identified with the predominance of GII.2[P16] in 2017 and changed to GII.4[P16] and GII.4[P31] in 2018. In conclusion, this study reported the detection of a wide variety of norovirus genotypes and several norovirus recombinant strains in Chiang Mai, Thailand from 2017 to 2018. IMPORTANCE In the present study, the prevalence of norovirus infection in children with acute gastroenteritis in Chiang Mai, Thailand between 2017 and 2018 was 14.9%. A variety of norovirus genotypes were detected, including GI.5, GII.2, GII.3, GII.4, GII.6, GII.7, GII.13, GII.14, and GII.17 with the predominance of GII.4 genotype. In addition, several norovirus recombinant strains (GII.2[P16], GII.3[P16], GII.4[P16], GII.4[P31], GII.6[P7], GII.13[P16], and GII.14[P7]) had been identified. Our results revealed that GII.2[P16] was a predominant strain till the end of 2017 and then was replaced by GII.4[P16] and GII.4[P31] in 2018. The findings imply that norovirus recombinant strains emerged in Chiang Mai, Thailand and that circulating strains changes over time.

Methicillin-resistant Staphylococcus aureus (MRSA) harboring the type-IX staphylococcal cassette chromosome mec (SCCmec) has been found in pigs and humans in Northern Thailand. However, knowledge of the prevalence and acquisition risk factors of this MRSA strain among swine production personnel (SPP) are needed. The nasal swab samples and data were collected from 202 voluntary SPP and 31 swine farms in Chiang Mai and Lamphun Provinces, Thailand in 2017. MRSA were screened and identified using mannitol salt agar, biochemical and antimicrobial susceptibility testing, multiplex PCR, and the SCCmec typing. The prevalence of MRSA was 7.9% (16/202) and 19.3% (6/31) among SPP and swine farms. All isolates were multidrug-resistant, and 55 of 59 isolates (93%) contained the type-IX SCCmec element. Data analysis indicated that education, working time, contact frequency, working solely with swine production, and personal hygiene were significantly related to MRSA acquisition (p < 0.05). The multivariate analysis revealed that pig farming experience, working days, and showering were good predictors for MRSA carriage among SPP (area under the curve (AUC) = 0.84). The biosecurity protocols and tetracycline use were significantly associated with MRSA detection in pig farms (p < 0.05). Hence, the active surveillance of MRSA and further development of local/national intervention for MRSA control are essential.

Porcine reproductive and respiratory syndrome (PRRS), caused by the PRRS virus (PRRSV), remains a major economic threat to swine production throughout the world. The aim of this study was to investigate the humoral and cell-mediated immune responses to PRRSV in 10 PRRSV vaccinated and 10 non-vaccinated young pigs obtained from a PRRSV-seropositive herd under field conditions. On day 35 days of post-vaccination (dpv), two PRRSV seropositive mixed-litter pigs were added to each group to co-mingle the animals. Serum and whole blood samples were collected from all pigs on the first day of vaccination, as well as on the 21, 35, 49, and 63 dpv. The PRRSV-specific humoral and cell-mediated immune response was determined by ELISA and flow cytometry analysis. The PRRSV ELISA sample to positive (S/P) ratio was found to be positive at the threshold level until the age of 84 days in both non-vaccinated and vaccinated groups, whereas the IFN-γ positive staining cytotoxic (CD8+) cells were rapidly expressed in the early periods of vaccination and co-mingling, but were not found to be specific to PRRSV. This result might have been due to an unspecific response to stress antigens. Further studies should be conducted to obtain more immune response data over long-term observation periods and to study the effect of PRRSV endemic strain vaccinations in endemically-infected herds.

Operation of the farrowing house is essential to the productivity of a swine farm, requiring not only good management but also knowledge of the behavior of sows and piglets. Stress can negatively affect production in farm animals and could be a factor in production indexes. The objective of this study was to investigate the effect of artificial sucking sounds on the behavior of piglets and fecal glucocorticoid (FGM) concentrations of sows. A total of 30 sows were divided into two groups: a treatment group (15 sows) was exposed to artificial sucking sounds and a control group (15 sows) was not. Both groups received the same management; the two open-house system locations were separated by a distance of about 270 meters. The study had three key objectives: to compare farrowing indexes and to observe the sucking behavior of piglets using CCTV cameras. Fecal samples were collected daily for 21 days from the period after parturition to weaning to assess adrenal activity. The treatment group had a significantly higher average number of times piglets came to a sow's udder, and sows had a shorter onset time for the first piglet to come to the sow's udder than the control group (both P<0.05). The patterns and levels of FGM between the two groups were not different (both P<0.05), but the treatment group had better farrowing indexes than the control group (P>0.05), particularly for litter weight gain and percent preweaning mortality. In addition, the weaning to first service interval of the treatment group was shorter than the control group (P<0.05). This indicates that the artificial suckling sound probably has no adverse effect on adrenal responses of pig; however, it improves production indexes of postparturition sows.

Salmonella spp. is acknowledged as a significant zoonotic foodborne pathogen throughout the world. Contaminated pork consumption is considered as a main cause of human salmonellosis. In the later stage of the pig production chain, poor hygiene and unsuitable storage conditions in retail outlets are considered to be key factors linked to the risk of Salmonella infection. The purpose of current study, which was conducted throughout April 2014 to September 2014, was to determine the prevalence and characteristics of Salmonella spp. in pork sold at the retail stage in wet markets and supermarkets in the Chiang Mai urban area of Thailand. Additionally, clonal relations between Salmonella strains described in this study and those identified in earlier study from the same geographical area were considered. It is provided as a means of contributing to current knowledge regarding Salmonella epidemiology with an ultimate aim of improved food security and consumer protection in this region. From a total of 82 pork samples analyzed in this study, 41% were positive for Salmonella, with prevalence of 73.2% from wet markets (n=30/41) and 9.8% from supermarkets (n=4/41). Twelve Salmonella serovars were identified, S. Rissen being the most commonly encountered. Antibiotic resistance of the isolates was highest for ampicillin and tetracycline (53%), followed by streptomycin (44%). Pulsed-field gel electrophoresis (PFGE) and subsequent geographical distribution analysis indicated that the clonal Salmonella strains originated from multiple sources had been spread over a wide area. The existence of a common pig supply chain "farm-slaughterhouse-retail" transmission route is inferred. Continuous monitoring of Salmonella along the entire production chain is needed to reduce contamination loads and to ensure the safety of pork products for end consumers.

Pigs and pork products are well known as an important source of Salmonella, one of the major zoonotic foodborne pathogens. The emergence and spread of antimicrobial resistance is becoming a major public health concern worldwide. Integrons are genetic elements known to have a role in the acquisition and expression of genes conferring antibiotic resistance. This study focuses on the prevalence of class 1 integrons-carrying Salmonella, the genetic diversity of strains of those organisms obtained from swine production chains in Chiang Mai and Lamphun provinces, Thailand, using multilocus sequence typing (MLST) and comparison of genetic diversity of sequence types of Salmonella from this study with pulsotypes identified in previous study. In 175 Salmonella strains, the overall prevalence of class 1 integrons-carrying-Salmonella was 14%. The gene cassettes array pattern "dfrA12-orfF-aadA2" was the most frequently observed. Most of the antimicrobial resistance identified was not associated with related gene cassettes harbored by Salmonella. Six sequence types were generated from 30 randomly selected strains detected by MLST. Salmonella at the human-animal-environment interface was confirmed. Linkages both in the farm to slaughterhouse contamination route and the horizontal transmission of resistance genes were demonstrated. To reduce this problem, the use of antimicrobials in livestock should be controlled by veterinarians. Education and training of food handlers as well as promotion of safe methods of food consumption are important avenues for helping prevent foodborne illness.

This study was conducted to analyze the prevalence and quantitative loads of Salmonella spp. on pig farms in Chiang Mai, Lamphun, Thailand to assess loading levels before slaughtering. The serotype diversity, antimicrobial-resistance pattern and pulse-field type of Salmonella spp. were also characterized to assess the dynamic propagation of the pathogen. The Salmonella-positive prevalence was 246/805 (30.56%), and the quantitative loads varied from 1.48 ˜ 4.04 Log10MPN/g, with a mean ± standard deviation of 2.11 ± 0.57. AMP/S/TE (ampicillin/streptomycin/tetracycline) was the highest frequency antimicrobial resistance pattern found in this study. In addition, Salmonella Rissen was the primary serotype in this region. PFGE results indicated the occurrence of infection by cross contamination among pig farms. Our study showed that pork is easily contaminated with this pathogen. Farm control programs must be based on strict biosecurity and hygienic measures, which could further reduce the contamination pressure at slaughterhouses or retail shops.
