Vertebrate embryogenesis is a remarkable process, during which numerous cell types of different lineages arise within a short time frame. An overwhelming challenge to understand this process is the lack of dynamic chromatin accessibility information to correlate cis-regulatory elements (CREs) and gene expression within the hierarchy of cell fate decisions. Here, we employed single-nucleus ATAC-seq to generate a chromatin accessibility dataset on the first day of zebrafish embryogenesis, including 3.3 hpf, 5.25 hpf, 6 hpf, 10 hpf, 12 hpf, 18 hpf and 24 hpf, obtained 51,620 high-quality nuclei and 23 clusters. Furthermore, by integrating snATAC-seq data with single-cell RNA-seq data, we described the dynamics of chromatin accessibility and gene expression across developmental time points, which validates the accuracy of the chromatin landscape data. Together, our data could serve as a fundamental resource for revealing the epigenetic regulatory mechanisms of zebrafish embryogenesis.

BACKGROUND: Adolescence is a formative period of social development. Adolescents have experienced considerable changes in their lives due to the COVID-19 pandemic. We conducted a longitudinal study to examine the effects of the COVID-19 pandemic on adolescents' prosocial attributes and empathy, as well as their longitudinal bilateral relationships. METHODS: A total of 2,510 students from five junior schools in Sichuan Province were recruited via random cluster sampling. Data were collected in December 2019 (Wave 1, before the outbreak of the pandemic) and July 2020 (Wave 2, during the pandemic) in Chengdu, Sichuan, China. Prosocial attributes and empathy were measured with the Positive Youth Development Scale (GPYDS) subscale and Chinese Empathy Scale, respectively. RESULTS: During the pandemic, both empathy and prosocial attributes decreased significantly from 49.89 (9.12) and 49.89 (8.80) before to 48.29 (8.72) and 49.39 (9.26) (p < 0.001), respectively. A higher level of empathy at Wave 1 significantly predicted higher prosocial attributes at Wave 2 (ß = 0.173, SE = 0.021, t = 8.430, p < 0.001). A lower prosocial attributes score predicted a significantly lower empathy score from Wave 1 to Wave 2 (ß = 0.100, SE = 0.021, t = 4,884, p < 0.001). CONCLUSIONS: The COVID-19 pandemic has had detrimental effects on adolescents' empathy and prosocial attributes. Special attention should be given to these two longitudinally associated factors in any social crisis, such as the COVID-19 pandemic, considering their importance for adolescents' physical, mental, and social development.

A major challenge in understanding vertebrate embryogenesis is the lack of topographical transcriptomic information that can help correlate microenvironmental cues within the hierarchy of cell-fate decisions. Here, we employed Stereo-seq to profile 91 zebrafish embryo sections covering six critical time points during the first 24 h of development, obtaining a total of 152,977 spots at a resolution of 10 × 10 × 15 µm3 (close to cellular size) with spatial coordinates. Meanwhile, we identified spatial modules and co-varying genes for specific tissue organizations. By performing the integrated analysis of the Stereo-seq and scRNA-seq data from each time point, we reconstructed the spatially resolved developmental trajectories of cell-fate transitions and molecular changes during zebrafish embryogenesis. We further investigated the spatial distribution of ligand-receptor pairs and identified potentially important interactions during zebrafish embryo development. Our study constitutes a fundamental reference for further studies aiming to understand vertebrate development.

FeMn bimetallic oxides have been widely used in catalytic adsorption due to their large pore size, large specific surface area and mesoporous structure, which have great potential for high As groundwater remediation. In this study, FeMn composite oxide was synthesized by template-free route and forming mesopores through high temperature calcination, and its efficiency and mechanism for As removal were subsequently investigated. The results showed that the different Fe/Mn molar ratios and calcination temperatures have important effect on FeMn composite oxides performance. For all synthesized materials, the largest specific surface area is 388.6 m2/g of Fe1Mn1-300. The maximum As absorption capacity was also reached by Fe1Mn1-300, which is 59.44 mg/g for As(III) and 31.68 mg/g for As(V), respectively. As removal efficiency was further evaluated through batch adsorption experiments conducted with five variables, initial As concentration, adsorption equilibrium time, pH, solid-to-liquid ratio, and competitive ions. The adsorption capacity of the material reaches to the maximum when the initial As concentration is 40 mg/L, and that for As(III) and As(V) is 74.05 and 38.09 mg/g, respectively. When the pH rises, the adsorption capacity generally shows a decreasing trend, thus acidic conditions are more conducive to the adsorption reaction. The optimum solid-to-liquid ratios for removal 10 mg/L of As(III) and As(V) are 0.3 mg/L and 1 mg/L, respectively. The order of competitive ions effects on As removal is: PO43- > HCO3- > SO42- ≈ NO3- ≈ Cl-. The adsorption mechanisms for As by FeMn composite oxides included adsorption, co-precipitation and oxidative chelation, which was a combination of physical and chemical process.

In this study, five strains isolated from traditional Inner Mongolian air-dried meat products were used, two Lactobacillus helveticus strains, ZF22 and TR1-1-3, with potent antibacterial activity, acid, salt, and nitrite tolerance, were selected for this study. Lactic acid bacteria (LAB) (Lactobacillus helveticus ZF22 and TR1-1-3) were inoculated into fermented sausages at 107 CFU/g and their volatiles were studied during fermentation and storage. Clustering heat map and principal component analysis (PCA) were used to identify differentiating flavor components in uninoculated and inoculated sausages. The results showed that 72 volatile flavor substances were identified during the fermentation of the fermented sausages and that inoculation with Lactobacillus helveticus ZF22 and TR1-1-3 increased the proportion of acids, ketones and alkanes. Moreover, the clustering heat map demonstrated that esters such as ethyl isobutyrate, ethyl acetate, and ethyl valerate were more abundant in TR1-1-3 and ZF22 than ZR. The PCA analysis showed that the volatile compounds of the three fermented sausages were distributed in separate quadrants, suggesting that the volatile compound compositions of the three fermented sausages differed significantly. Our findings suggest that inoculating fermented sausages with Lactobacillus helveticus TR1-1-3 and ZF22 can improve flavor by enhancing the type and amount of flavor compounds.

Energy saving, emission reduction, and efficiency improvement are important directions for agricultural development in Central Henan Province, the main grain production area in the Huang-huai-hai Plain. Based on the tillage and fertilization positioning experiment in 2010, we investigated the effects of three tillage methods (deep tillage, shallow tillage, and no-tillage) and two fertilization methods (nitrogen fertilizer and nitrogen fertilizer+organic fertilizer) on soil nitrogen balance and greenhouse gas emissions from 2018 to 2019. The results showed that soil nitrogen accumulation increased with organic fertilizer addition. During wheat and maize maturation, soil total nitrogen accumulation in the 0-60 cm layer was the highest under the treatment of shallow tillage+organic fertilizer, being 8058.53 and 8299 kg N·hm-2, respectively, being 3.2%-27.4% and 4.3%-7.2% higher than other treatments. The treatment with organic fertilizer addition resulted in nitrogen surplus. The shallow tillage+organic fertilizer treatment led to the highest nitrogen surplus (13.57 kg N·hm-2), which was 9.52 and 0.18 kg N·hm-2 higher than deep tillage+organic fertilizer and no tillage+organic fertilizer treatments. Nitrate leaching was the main way of nitrogen losses, accounting for 73.4%-76.9% of the total losses. The amount of nitrate leaching was the highest in deep tillage+organic fertilizer treatment (48.37 kg N·hm-2), being 18.9%-35.1% higher than other treatments. Results of greenhouse gases emission during 2018-2019 showed that global warming potential was the highest under the treatment of deep tillage+organic fertilizer, which was 33070 kg N·hm-2, being 6.6%-26.8% higher than other treatments. The treatment of organic fertilizer addition increased the emission of N2O and CO2 and reduced the absorption of CH4. The annual grain yield was highest under the treatment of deep tillage+organic fertilizer, which was 5.0%-17.1% higher than other treatments. The crop harvest index was the highest under the treatment of shallow tillage+organic fertilizer. The recommended cropping mode in Central Henan Pro-vince is shallow tillage+organic fertilizer, which could ensure crop yields, maintain soil nitrogen balance, and reduce greenhouse gas emissions.

Working memory (WM) refers to the holding and manipulation of information during cognitive tasks. Its underlying neural mechanisms have been explored through both functional magnetic resonance imaging (fMRI) and electroencephalography (EEG). Trial-by-trial coupling of simultaneously collected EEG and fMRI signals has become an important and promising approach to study the spatio-temporal dynamics of such cognitive processes. Previous studies have demonstrated a modulation effect of the WM load on both the BOLD response in certain brain areas and the amplitude of P3. However, much remains to be explored regarding the WM load-dependent relationship between the amplitude of ERP components and cortical activities, and the low signal-to-noise ratio (SNR) of the EEG signal still poses a challenge to performing single-trial analyses. In this paper, we investigated the spatio-temporal activities of P3 during an n-back verbal WM task by introducing an adaptive wavelet denoiser into the extraction of single-trial P3 features and using general linear model (GLM) to integrate simultaneously collected EEG and fMRI data. Our results replicated the modulation effect of the WM load on the P3 amplitude. Additionally, the activation of single-trial P3 amplitudes was detected in multiple brain regions, including the insula, the cuneus, the lingual gyrus (LG), and the middle occipital gyrus (MOG). Moreover, we found significant correlations between P3 features and behavioral performance. These findings suggest that the single-trial integration of simultaneous EEG and fMRI signals may provide new insights into classical cognitive functions.
