RESUMO

Aim: To investigate the antibacterial effects of Corydalis Saxicola bunting total alkaloid (CSBTA) on Porphyromonas gingivalis. Methods: SEM, chemical staining, RT-qPCR and ELISA were used to detect effects of CSBTA on P. gingivalis. Results: CSBTA treatment caused shrinkage and rupture of P. gingivalis morphology, decreased biofilm density and live bacteria in biofilm, as well as reduced mRNA expression of virulence genes hagA, hagB, kgp, rgpA and rgpB of P. gingivalis. Furthermore, NOK cells induced by CSBTA-treated P. gingivalis exhibited lower IL-6 and TNF-α expression levels. Conclusion: CSBTA is able to kill free P. gingivalis, disrupt the biofilm and weaken the pathogenicity of P. gingivalis. It has the potential to be developed as a drug against P. gingivalis infection.

RESUMO

To increase the boiling heat transfer limit, we disrupted the previously nonevaporating region and increased the vaporization activity of "inert" liquid molecules by introducing nano ridges on the boiling surface. This solved the paradox of no heat transfer occurring through the thinnest liquid film in boiling bubbles; thus, the internal heat transfer limit of the bubbles was exceeded. We found that vigorous boiling occurred immediately once the nonevaporating region was activated, and the bubble frequency increased by an order of magnitude, reaching 1186 Hz, which has not been previously reported. With an increase in heat flux, the boiling curve exhibited a "return". We achieved an extremely high bubble frequency by experimentally quantifying the major influence of nonevaporating region disruption on boiling heat transfer. The mechanism behind the generation of the ultrahigh-frequency bubbles was discovered. This study also reveals a new mechanism for the reversed boiling curve.

RESUMO

People in Eastern countries hold a tradition of soaking herbal medicine in wine; however, the efficacy and safety of herbal wine have not been rigorously assessed. By assessing the efficacy of resveratrol (RSV) in ethanol against alcoholic liver disease (ALD) in mice, we aimed to offer a perspective on the use of herbal wine. To simulate the behaviour of herbal wine users, RSV (15 mg/kg) soaked in ethanol (RSV-alcohol) was administrated via gavage to the mice, here with alcohol consumption-induced ALD. RSV soaked in water (RSV-water) was the treatment control. The efficacy and safety of RSV on ALD were evaluated. Compared with the RSV-water group, a higher rate of mortality was found in the RSV-alcohol group (50.0% vs. 20.0%), which also exhibited more severe liver injury. RSV significantly increased the exposure of alcohol by 126.0%, which was accompanied by a significant inhibition of the ethanol metabolic pathway. In contrast, alcohol consumption significantly reduced exposure to RSV by 95.0%. Alcohol consumption had little effect on the expression of drug-metabolizing enzymes in RSV; however, alcohol seemed to reduce the absorption of RSV. RSV in liquor exacerbates alcoholic liver injury and has a reduced therapeutic effect, suggesting that the habit of herbal wine use without supervision is risky.

RESUMO

Polyphenylene sulfide (PPS) is a high-performance, low-cost special engineering polymer. The mechanical properties of three-dimensionally (3D) printed PPS samples are affected by the mass of the melt extruded filament and the printing pattern in the inner layer. The present study demonstrates the effects of the melt extrusion and filament alignment parameters on tensile, bending, and impact strengths of 3D printed PPS samples. The results indicate that by increasing the melt extrusion parameters, the tensile and bending strengths of 3D printed PPS samples can be improved and internal voids between adjacent filaments can be eliminated. However, the impact strength was restricted by excess melt extrusion. By measuring the strength of 3D printed PPS samples with different filament alignment, we observed that the tensile and bending strengths increase with decreasing angle between the filament and load direction. Moreover, the local tensile strain between adjacent filaments and slippage of the microstructure cell formed by aligning the filament are thought to absorb the impact energy. This study provides a useful guide for selection of appropriate printing parameters to realize a diverse range of mechanical properties for 3D printed PPS samples.

RESUMO

RATIONALE: Understanding the association between Henoch-Schönlein purpura (HSP) and malignancy is essential for early diagnosis and treatment of the potential lethal disease. To the best of our knowledge, there has been only one published case of HSP coexisting with oesophageal cancer. Here, we report another patient diagnosed with HSP and oesophageal squamous carcinoma simultaneously. PATIENT CONCERNS: A 60-year-old Chinese male was referred to our hospital because of intermittent abdominal pain, abdominal distension, melena, lower extremities purpura. Positive laboratory values included pancytopenia, microscopic hematuria, nephrotic proteinuria, hematochezia, hypoalbuminemia, hyperlipidaemia, hypocomplementemia, and increased levels of hepatobiliary enzymes and immunoglobulin (Ig) A. Gastrocolonoscopy showed multiple erosion lesion on descending duodenum, terminal ileum, and ileal flap. Biopsy of these lesions suggested non-specific inflammation. DIAGNOSES: HSP (IIIb type) was diagnosed based on renal pathology examination in accordance with the International Study of Kidney Disease in Children (ISKDC) classification. Liver biopsy confirmed the diagnosis of nodular cirrhosis (Ishak 5). Gastroscopy unintentionally revealed three oesophagus lesions. Pathology study suggested intermediate differentiated squamous cell carcinoma (cTNM IB). INTERVENTIONS: Before admission, he was administered intravenous Ig 10 g once daily(qd) for 10 days, methylprednisolone 40 mg qd for a week, followed by prednisolone 50 mg qd for almost 8 weeks. Endoscopic submucosal dissection (ESD) was performed to remove all lesions with negative margin after prednisolone was tapered (5 mg per week until 10 mg qd). OUTCOMES: Despite prednisone being tapered to 2.5 mg qd within 2 months, complete remission of HSP and esophageal malignancy was achieved after the resection of the esophagus lesions during 12 months follow-up. LESSONS: We report a rare case of oesophageal squamous cell carcinoma initially presented as HSP. This case suggests the importance of evaluating adult patients with HSP for an underlying malignancy.

RESUMO

For bone tissue engineering, the porous scaffold should provide a biocompatible environment for cell adhesion, proliferation, and differentiation and match the mechanical properties of native bone tissue. In this work, we fabricated porous polyetherimide (PEI) scaffolds using a three-dimensional (3D) printing system, and the pore size was set as 800 µm. The morphology of 3D PEI scaffolds was characterized by the scanning electron microscope. To investigate the mechanical properties of the 3D PEI scaffold, the compressive mechanical test was performed via an electronic universal testing system. For the in vitro cell experiment, bone marrow stromal cells (BMSCs) were cultured on the surface of the 3D PEI scaffold and PEI slice, and cytotoxicity, cell adhesion, and cell proliferation were detected to verify their biocompatibility. Besides, the alkaline phosphatase staining and Alizarin Red staining were performed on the BMSCs of different samples to evaluate the osteogenic differentiation. Through these studies, we found that the 3D PEI scaffold showed an interconnected porous structure, which was consistent with the design. The elastic modulus of the 3D PEI scaffold (941.33 ± 65.26 MPa) falls in the range of modulus for the native cancellous bone. Moreover, the cell proliferation and morphology on the 3D PEI scaffold were better than those on the PEI slice, which revealed that the porous scaffold has good biocompatibility and that no toxic substances were produced during the progress of high-temperature 3D printing. The osteogenic differentiation level of the 3D PEI scaffold and PEI slice was equal and ordinary. All of these results suggest the 3D printed PEI scaffold would be a potential strategy for bone tissue engineering.

RESUMO

The purpose of this work was to investigate the porous polyetherimide scaffold (P-PEIs) as an alternative biopolymer for bone tissue engineering. The P-PEIs was fabricated via solvent casting and particulate leaching technique. The morphology, phase composition, roughness, hydrophilicity, and biocompatibility of P-PEIs were evaluated and compared with polyetherimide (PEI) and Ti6Al4V disks. P-PEIs showed a biomimetic porous structure with a modulus of 78.95 ± 2.30 MPa. The water contact angle of P-PEIs was 75.4 ± 3.39°, which suggested that P-PEIs had a wettability surface. Moreover, P-PEIs provides a feasible environment for cell adhesion and proliferation. The relative cell adhesion capability and the cell morphology on P-PEIs were better than PEI and Ti6Al4V samples. Furthermore, the MC3T3-E1 cells on P-PEIs showed faster proliferation rate than other groups. It was revealed that the P-PEIs could be a potential material for the application of bone regeneration.

RESUMO

The success of the 3D-printing process depends upon the proper selection of process parameters. However, the majority of current related studies focus on the influence of process parameters on the mechanical properties of the parts. The influence of process parameters on the shape-memory effect has been little studied. This study used the orthogonal experimental design method to evaluate the influence of the layer thickness H, raster angle θ, deformation temperature Td and recovery temperature Tr on the shape-recovery ratio Rr and maximum shape-recovery rate Vm of 3D-printed polylactic acid (PLA). The order and contribution of every experimental factor on the target index were determined by range analysis and ANOVA, respectively. The experimental results indicated that the recovery temperature exerted the greatest effect with a variance ratio of 416.10, whereas the layer thickness exerted the smallest effect on the shape-recovery ratio with a variance ratio of 4.902. The recovery temperature exerted the most significant effect on the maximum shape-recovery rate with the highest variance ratio of 1049.50, whereas the raster angle exerted the minimum effect with a variance ratio of 27.163. The results showed that the shape-memory effect of 3D-printed PLA parts depended strongly on recovery temperature, and depended more weakly on the deformation temperature and 3D-printing parameters.

RESUMO

The rapid development of transgenic biotechnology has greatly promoted the breeding of genetically engineered (GE) rice in China, and many GE rice lines are in the pipeline for commercialization. To understand field performances of GE rice, key agronomic traits of two insect-resistant Bt rice lines that have been granted biosafety certificates for commercial production in China were evaluated together with their nontransgenic counterparts under environmental conditions with significant differences in insect pressure. Results from the experiments showed enhanced field performances of the Bt GE rice lines compared with the non-GE counterparts for yield-related traits such as number of panicles and filled seeds per plant, under environmental conditions with no insecticide application. No detectable underlying cost of the Bt transgene was observed in the two insect-resistant GE rice lines, particularly in the GE hybrid rice line. Results further indicated significantly greater yield performances of the two insect-resistant GE rice lines under environmental conditions with non-target insect control compared with no insect control. It is concluded from this study that insect-resistant Bt GE rice, particularly the hybrid line, has great potential to maintain its high yield when ambient insect pressure is high. In addition, proper application of insecticides to control non-target insects will guarantee optimal performance of insect-resistant Bt GE rice.
