RESUMO

Solid phase extraction combined with ultra-performance liquid chromatography-tandem mass spectrometry was developed for the simultaneous determination of 31 endocrine-disrupting chemicals in fish plasma. The strong anion exchange/primary-secondary amine cartridge and the mixed cation exchange cartridge were used in tandem instead of using a single mixed cation exchange cartridge for sample purification. Suitable eluents were selected for each of the two cartridges: 4.5% ammonia/acetonitrile solution for cartridges in tandem and acetone:n-hexane (V:V = 3:7) for the strong anion exchange/primary-secondary amine cartridge alone. With this optimized Solid phase extraction method, the recoveries of 31 endocrine disrupting chemicals were between 43.0% and 131.3%, the method detection limits were 0.45 to 1.35 ng/ml, and the limits of quantitation were 1.50-4.50 ng/ml. The innovative pretreatment method that connects two cartridges in tandem is well positioned to mitigate the matrix effects of fish plasma, thereby improving the accuracy of multiclass endocrine-disrupting chemicals determination. The significance of this method is to facilitate the application of the fish plasma model for the environmental risk assessment of endocrine-disrupting chemicals.

In order to fully understand the pollution of polychlorinated biphenyls (PCBs) in Dongting Lake, 209 PCB congeners were quantitatively detected and analyzed by isotope-dilution high-resolution mass spectrometry in 21 surface water sampling sites of Dongting Lake and the inflow rivers of the lake. The results showed that a total of 50 PCB congeners were detected in the study area. The ρ(ΣPCBs) ranged from 0.077 to 10 ng·L-1, with an average concentration of 2.7 ng·L-1 and a median concentration of 1.9 ng·L-1. The order of contamination concentrations were as follows:the inflow rivers of the lake > lake region > outlet. Compared with the reports of domestic and foreign studies, PCB pollution in Dongting Lake is at a low level. The main pollutants in the surface water were dichlorobiphenyls and tri- and tetrachlorobiphenyls, with relative abundances of 67%, 14%, and 16%, respectively. PCB11, PCB17, PCB18, PCB20+33, PCB28, PCB47+75, PCB52, and PCB68 were the most widely distributed and detected in more than 80% of samples. Among them, ρ(PCB11) was the highest, with an average concentration of 1.6 ng·L-1, accounting for 59% of ρ(ΣPCBs), from the domestic organic pigment production process of unintentional emissions. Atmospheric transport deposition and historical residue were the main sources of PCBs in water. According to the toxicity equivalent factor method, the ecological toxicity risk of the study area was evaluated, and the results showed that the TEQ in the water was far lower than the relevant standard limit, indicating that the pollution of PCBs in the surface water of Dongting Lake and the inflow rivers would not cause harm to exposed organisms.

In this study, molybdenum disulfide (MoS2) was chosen as a co-catalyst to enhance the removal efficiency of phenacetin (PNT) in water by a ferrous ion-activated peroxymonosulfate (Fe2+/PMS) process. Operating parameters, such as the initial solution pH and chemical dose on PNT degradation efficiency were investigated and optimized. Under an initial pH of 3, an Fe2+ dose of 25 µM, a PMS dose of 125 µM and a MoS2 dose of 0.1 g L-1, the degradation efficiency of PNT reached 94.3%, within 15 min. The presence of common water constituents including Cl-, HCO3 -, SO4 2- and natural organic matter (NOM) will inhibit degradation of PNT in the MoS2/Fe2+/PMS system. Radical quenching tests combined with electron paramagnetic resonance (EPR) results indicated that in addition to free radical species (˙OH, SO4˙- and O2˙-), nonradical reactive species (1O2) were also crucial for PNT degradation. The variations in the composition and crystalline structure of the MoS2 before and after the reaction were characterized by XPS and XRD. Further, the degradation pathways of PNT were proposed according to the combined results of LC/TOF/MS and DFT calculations, and primarily included hydroxylation of the aromatic ring, cleavage of the C-N bond of the acetyl-amino group, and cleavage of the C-O bond of the ethoxy group. Finally, toxicity assessment of PNT and its products was predicted using the ECOSAR program.

In this study, nano zero-valent iron (nZVI) was utilized to activate persulfate (PS) for the degradation of metoprolol (MTP), a commonly used drug for curing cardiovascular diseases, in water. Quenching tests indicated that both the sulfate radical (SO4˙-) and hydroxyl radical (˙OH) contributed to the degradation of MTP, while SO4˙- seemed to play a large role under natural pH conditions. Batch tests were conducted to investigate the effects of several influencing factors, such as PS concentration, initial MTP concentration, pH, temperature and common anions, on the degradation performance of MTP. Generally, lower MTP concentration and pH values, and higher PS concentration and temperature favoured MTP degradation. HCO3 -, NO3 - and SO4 2- were found to inhibit MTP degradation, while Cl- enhanced MTP degradation. Several corrosion products of nZVI, including Fe3O4, Fe2O3 and FeSO4, were formed during the reaction, which was reflected by the combined XRD and XPS analysis. Degradation pathways of MTP were proposed according to the identified transformation products, and the peak areas of the major products along with the time were also monitored. Finally, the toxicity of the reaction solution was assessed by experiments using Aliivibrio fischeri. Overall, it could be concluded that nZVI/PS might be a promising method for the rapid treatment of MTP-caused water pollution.

This study investigated the degradation of propranolol (PRO), a beta (ß)-blockers, by nano zero-valent iron (nZVI) activated persulfate (PS) under ultrasonic irradiation. Effects of several critical factors were evaluated, inclusive of PS concentration, nZVI dosage, ultrasound power, initial pH, common anions, and chelating agent on PRO degradation kinetics. Higher PS concentration, nZVI dosage and ultrasound power as well as acidic pH favored the PRO degradation. Conversely, anions and chelating agent took on the inhibitory effect towards PRO degradation to different extents. Furthermore, the variations of morphology and surface composition of nZVI before and after the reaction were characterized by TEM, XRD and XPS. Finally, on the basis of identified degradation intermediates by LC/MS/MS analysis, this work tentatively proposed the degradation pathways. These encouraging results suggest that US/nZVI/PS process is a promising strategy for the treatment of PRO-induced water pollutant.

In this study, the degradation of propranolol (PRO) by UV/persulfate process was systematically investigated. Direct photolysis of PRO was limited due to its low quantum yield, while the PRO degradation efficiency can be greatly promoted by the combination of persulfate and UV irradiation. Radical scavenging tests showed that both SO4- and OH contributed to the removal of PRO, with SO4- playing a more important role. The degradation rate of PRO was improved by increasing the persulfate dose and initial solution pH consistent with pseudo-first-order reaction kinetics. The effects of common water constituents were species dependent. HCO3- and Cl- promoted PRO degradation. By contrast, NO3- and HA were found to inhibit PRO degradation. A total of nine degradation products were identified by LC/MS/MS, which mainly derived from the ring-opening attack on the naphthalene group or oxidation of the amino moiety by SO4- and OH. Finally, the toxicity of the reaction mixtures was also assessed using luminescent bacteria Vibrio fischeri, and the results indicated that UV/persulfate is capable of controlling the toxicity of PRO degradation.

A confirmatory method for the simultaneous detection of 29 pharmaceuticals in fish muscle and plasma was developed by using solid-phase extraction combined with ultra-high performance liquid chromatography and tandem mass spectrometry. Fish samples were extracted with methanol and enriched using Oasis HLB solid-phase extraction columns in one step. Twenty-nine target pharmaceuticals were quantified by the internal standard method and the calibration curves showed good linearity in a wide range with determination coefficients of greater than 0.913. The detection limits of the pharmaceuticals ranged from 0.01 to 2.00 µg/kg (µg/L). The applicability of the method was checked by precision and recovery experiments. The average recoveries of the 29 pharmaceuticals were between 61 and 111%, and all the relative standard deviations were below 25%. Our reported method has been demonstrated to be sensitive, convenient, rapid, and reliable for the simultaneous determination of 29 pharmaceuticals in fish muscle and plasma. Real sample determination showed that 25 and 9 of the 29 compounds were detected in fish muscle and plasma, respectively.

Pharmaceuticals in drinking water sources have raised significant concerns due to their persistent input and potential human health risks. The seasonal occurrence of 25 pharmaceuticals including 23 antibiotics, paracetamol (PAR), and carbamazepine (CMZ) in Taihu Lake was investigated; meanwhile, the distribution and removal of these pharmaceuticals in two drinking water treatment plants (DWTPs) and a constructed wetland were evaluated. A high detection frequency (>70%) in the Taihu Lake was observed for nearly all the 25 pharmaceutics. Chlortetracycline (234.7 ng L-1), chloramphenicol (27.1 ng L-1), erythromycin (72.6 ng L-1), PAR (71.7 ng L-1), and CMZP (23.6 ng L-1) are compounds with both a high detection frequency (100%) and the highest concentrations, suggesting their wide use in the Taihu Basin. Higher concentrations of chloramphenicols, macrolides, PAR, and CMZP were observed in dry season than in wet season, probably due to the low flow conditions of the lake in winter and the properties of pharmaceuticals. The overall contamination levels of antibiotic pharmaceutics (0.2-74.9 ng L-1) in the Taihu Lake were lower than or comparable to those reported worldwide. However, for nonantibiotic pharmaceutics, PAR (45.0 ng L-1) and CMZP (14.5 ng L-1), significantly higher concentrations were observed in the Taihu Lake than at a global scale. High detection frequencies of 25 pharmaceuticals were observed in both the two DWTPs (100%) and the wetland (>60%) except for florfenicol and sulfapyridine. The removal efficacies of the studied pharmaceuticals in DWTP B with advanced treatment processes including ozonation and granular activated carbon filtration (16.7-100%) were superior to DWTP A with conventional treatment processes (2.9-100%), except for sulfonamides. Wetland C with the constructed root channel technology was efficient (24.2-100%) for removing most pharmaceuticals. This work suggests that the application of cost-effective technologies such as constructed wetlands should be considered as an efficient alternative for removing pharmaceuticals from water supply sources.

Seventeen polycyclic aromatic hydrocarbon (PAH) compounds were determined in surface sediments collected from the Chaohu Lake (a large shallow lake in eastern China) and its tributaries. Both diagnostic ratios and a receptor model (positive matrix factorization, PMF) were applied to identify and determine the contribution of a local iron-steel manufacturing plant located in the Nanfei River (NFR) to the Chaohu Lake basin. The results show that sites located in the downstream of the steel plant contained concentrations of 17 PAH (Σ17PAH) approximately two orders of magnitudes higher than those from other sites. Five factors were identified by the PMF model, including industrial waste, wood/biomass burning, diagenetic origin, domestic coal combustion, and industrial combustion. Our findings suggest that sediments in the downstream of the plant and in the western part of the Chaohu Lake were predominantly affected by industrial coal combustion. A mixture of pyrolytic origins impacted urban sediments in the upstream of the plant, whereas diagenetic origins along with coal and biomass burning were suggested to influence the eastern part and rural tributaries of the lake. To assess the potential ecological risk and toxicity caused by the iron-steel plant, sediment toxicity was evaluated by the PMF model, sediment quality guideline, and toxic equivalent factors. All of the three approaches suggested PAH accumulation in the NFR sediments could produce significant adverse ecological effects and half of the sediment toxicity in the NFR may be attributed to the emissions from the iron-steel plant. Some rural locations also exhibited PAH concentrations above probable effects, most likely contributed by wood/biomass burning.

We investigated the effects of allelochemical ferulic acid (FA) on a series of physiological and biochemical processes of blue-green algae Microcystis aeruginosa, in order to find sensitive diagnostic variables for allelopathic effects. Algal cell density was significantly suppressed by FA (0.31-5.17 mM) only after 48 h exposure. Inhibitions of photosynthetic parameters (F(v)/F(m) and F(v)'/F(m)') occurred more rapidly than cell growth, and the stimulation of non-photochemical quenching was observed as a feed-back mechanisms induced by photosystem II blockage, determining by PAM fluorometry. Inhibitions on esterase activity, membrane potential and integrity, as well as disturbance on cell size, were all detected by flow cytometry with specific fluorescent markers, although exhibiting varied sensitivities. Membrane potential and esterase activity were identified as the most sensitive parameters (with relatively lower EC50 values), and responded more rapidly (significantly inhibited only after 8 h exposure) than photosynthetic parameters and cell growth, thus may be the primary responses of cyanobacteria to FA exposure. The use of PAM fluorometry and flow cytometry for rapid assessment of those sensitive variables may contribute to future mechanistic studies of allolepathic effects on phytoplankton.

UV irradiation-activated sodium persulfate (UV/PS) was studied to degrade florfenicol (FLO), a phenicol antibiotic commonly used in aquaculture, in water. Compared with UV/H2O2 process, UV/PS process achieves a higher FLO degradation efficiency, greater mineralization, and less cost. The quantum yield for direct photolysis of FLO and the second-order rate constant of FLO with sulfate radicals were determined. The effects of various factors, namely PS concentration, anions (NO3 (-), Cl(-), and HCO3 (-)), ferrous ion, and humic acid (HA), on FLO degradation were investigated. The results showed that the pseudo-first-order rate constant increased linearly with increased PS concentration. The tested anions all adversely affected FLO degradation performance with the order of HCO3 (-) > Cl(-) > NO3 (-). Coexisting ferrous ions enhanced FLO degradation at a Fe(2+)/PS molar ratio under 1:1. HA significantly inhibited FLO degradation due to radical scavenging and light-screening effect. Toxicity assessment showed that it is capable of controlling the toxicity for FLO degradation. These findings indicated that UV/PS is a promising technology for water polluted by antibiotics, and the treatment is optimized only after the impacts of water characteristics are carefully considered.

Environmental problems as well as their related ecosystem stress and human health risk in China have raised wide concerns along with the rapid economic development in recent years. Numerous studies with a sharp increase in publication number have addressed the ubiquitous of anthropogenic chemicals in various environmental compartments and human tissues. However, very few data were available to clarify the temporal trend and to give the retrospective analysis of chemical pollution in China. Environmental Specimen Bank (ESB) is a system for the systematic collection and long-term storage of specimens, which has been established since the 1970s in developed counties and recognized as a fundamental complement for environmental monitoring and scientific research. Currently, the value of ESB is becoming more broadly recognized globally, and China is still at the early stage. This article described the history and status and put forwarded the future key points of Chinese ESB development for illustrating the intensive environmental changes in China and the world.

This study aimed to develop a sensitive and reliable multi-residue method for the determination of trace amounts of endocrine disrupting chemicals including five phthalate esters (PAEs), five monoalky phthalate esters (MPEs), four alkylphenols (APs) and bisphenol A (BPA) in seafood. Ultrasonic liquid extraction was selected for extraction based on acetonitrile, instead of frequently-used n-hexane, due to its lower background of PAEs. Application of solid phase extraction (SPE) with primary secondary amine (PSA, 1g/6 mL) cartridge achieved the relatively low matrix effects for MPEs and BPA in seafood. To our knowledge, it is the first study reporting about simultaneous extraction and purification of PAEs, MPEs, APs and BPA in biota samples. To obtain the maximum sensitivity, both liquid chromatography tandem mass spectrometry (LC-MS/MS) and gas chromatography tandem mass spectrometry (GC-MS/MS) were applied for analysis. This method was validated and tested on fish, mollusk and prawn. Sufficient linearity was verified by Mandel's fitting test for the matrix-matched calibrations used in this study for MPEs, APs and BPA, between 0.5 ng/g and 200 ng/g or 400 ng/g. And correlation coefficients of all calibrations suppressed 0.99 for all analytes. Good recoveries were obtained, ranging from 60% to 127% for most compounds. The sensitivity was good with method detection limits (MDLs) of 0.015-2.2 ng/g wet weight (ww) for all compounds. Most MDLs are much lower than those in previous reports. The sensitive method was then applied on real fish, mollusk and prawn samples from the Yangtze River Delta sea area (China), and all the target compounds were detected with the maximum concentrations of PAEs, MPEs, APs and BPA up to 219.3 ng/g ww, 51.4 ng/g ww, 62.0 ng/g ww and 8.6 ng/g ww, respectively.

The photocatalytic-degradation behavior of tetracycline hydrochloride (TTC) was studied. The catalyst used was photosensitive semiconductor titanium dioxide (TiO2). The results showed that the photocatalytic degradation of TTC was well fitted to first order reaction kinetics model, and the adsorption was the control step of the whole photocatalytic-degradation process, indicating that the main degradation path was the photocatalytic reaction of TTC adsorbed on the surface of TiO2. Besides, through photocatalytic-degradation of the mixed solution of TTC and sulfamethoxazole or amoxicillin, the degradation of the two antibiotics showed obvious selectivity when the pH, TiO2 dosage and other conditions were changed.

Polybrominated diphenyl ethers (PBDEs) are widely used flame retardants and are banned around the world as potent environmental contaminants. PBDE-47 is the most concerned PBDE with its environmental prevalence and various toxicity characteristics including neurotoxicity. In this paper, we studied larval zebrafish behavioral alterations caused by PBDE-47 neurotoxicity. The light-dark cycle stimulation was used to investigate the locomotor changes of zebrafish larvae at different ages (4-6 day post-fertilization, dpf) after PBDE-47 exposure (5, 50, 500 µg L(-1)). Three exposure modes, namely continuous exposure, early pulse exposure and interval exposure, were adopted to assess and compare the impacts of exposure modes on larval zebrafish locomotion. Our results showed that locomotor effects upon PBDE exposure depended on the specific exposure mode studied. In the early pulse exposure mode, the locomotion of zebrafish larvae did not change significantly at all PBDE-47 concentrations tested. In contrast, for both the continuous exposure and interval exposure modes, the highest dose of PBDE-47 (500 µg L(-1)) elicited pronounced hypoactivity at 5 dpf during dark periods except for the initial one. However, at 6 dpf, hypoactivity was only observed in the continuously exposed zebrafish larvae (to an even higher degree compared to 5 dpf), but not in the interval exposure treatment group. Our results suggested that the conventional, continuous exposure mode might not be enough to evaluate the toxicity of chemicals in the real environments.

The contamination of fresh water with pharmaceutical and personal care products (PPCPs) has risen during the last few years. The adsorption of some PPCPs namely, Diclofenac-Na, Naproxen, Gemfibrozil and Ibuprofen from aqueous solution has been studied, magnetic nanoparticles coated zeolite (MNCZ) has been used as the adsorbent. Batch adsorption experiment was conducted to study the influences of different adsorption parameters such as contact time, solution pH and PPCPs concentrations in order to optimize the reaction conditions. The removal was favored at low pH values. Thus, as pH turns from acidic to basic conditions these compounds were less efficiently removed. The initial concentration does not appear to exert a noticeable effect on the removal efficiency of the studied PPCPs at low concentrations, but it showed less removal efficiency during high concentration of PPCPs especially for Ibuprofen. The removal of Diclofenac-Na was independent on time, while the contact time was of significant effect on the adsorption of Naproxen, Gemfibrozil and Ibuprofen even though these compounds were removed up to 95% during 10 min using MNCZ. From the isotherm adsorption study, the adsorption of PPCPs studied on MNCZ was best fitted with Freundlich isotherm equation. Pseudo-second order model providing the best fit model with the experimental data. Column adsorption study was conducted to compare the removal efficiency of MNCZ with other processes used at drinking water treatment plants (DWTPs), MNCZ showed high removal efficiency (>99%) than other used processes at DWTPs.

The distributions and effects of 31 selected endocrine disrupting chemicals (EDCs) in two drinking water factories were analyzed in this study. The distributions of EDCs were analyzed by solid phase extraction (SPE) combined with liquid chromatography tandem mass spectrometry (LC-MS/MS). The concentrations of these EDCs were from lower than the LOD (limit of detection) to 23.13 ng L (- 1) in the samples; most of them were lower than 1 ng L (- 1). The highest concentration (23.13 ± 1.45 ng L (- 1)) was detected in the raw water. Twenty-six chemicals were found in the raw water and only five in the finished water of drinking water factory A, while 25 chemicals were detected in the raw water and two in the finished water of drinking water factory B. The results indicate that most of the EDCs can be removed by the water treatment process. In the advanced treatment process, the ozonation processes have the highest removal efficiency. Separate analyses in May and September show similar results. Apart from the chemical analysis, yeast strain transformed when the estrogen receptor α (ERα) gene was employed to test the estrogenic effects of the water samples. Due to the low concentrations of these EDCs, no significant estrogenic effects were found from the samples.

Copper pollutions are typical heavy metal contaminations, and their ability to move up food chains urges comprehensive studies on their effects through various pathways. Currently, four exposure pathways were prescribed as food-borne (FB), water-borne plus clean food (WCB), water-food-borne (WFB) and water-borne (WB). Caenorhabditiselegans was chosen as the model organism, and growth statuses, feeding abilities, the amounts of four antioxidant enzymes, and corresponding recovery effects under non-toxic conditions with food and without food were investigated. Based on analysis results, copper concentrations in exposure were significantly influenced by the presence of food and its uptake by C.elegans. Both exposure and recovery effects depended on exposure concentrations and food conditions. For exposure pathways with food, feeding abilities and growth statuses were generally WFBFB>WCB>WFB (p<0.05), while the antioxidant activities were all inhibited in a concentration-dependent fashion. In conclusion, contaminated food was the primary exposure pathway, and various pathways caused different responses of C.elegans.

A novel analytical method employing MCX (mixed-mode cationic exchange) based solid phase extraction (SPE) coupled with liquid chromatography tandem mass spectrometry (LC-MS/MS) was developed to detect 31 endocrine-disrupting compounds (EDCs) in surface water samples simultaneously. The target EDCs belong to five classes, including seven estrogens, eight androgens, six progesterones, five adrenocortical hormones and five industrial compounds. In order to simultaneously concentrate the target EDCs and eliminate matrix interferences in the water samples, MCX SPE cartridges were employed for SPE, and then followed by a simple and highly efficient three-step sequential elution procedure. Two electrospray ionization (ESI) detection modes, positive (ESI+) and (ESI-), were optimized for HPLC-MS/MS analysis to obtain the highest sensitivity for all the EDCs. The limits of detection (LODs) were 0.02-1.9 ng L(-1), which are lower than or comparable to these reported in references. Wide linear ranges (LOD-100 ng L(-1) for ESI+ mode, and LOD-200 ng L(-1) for ESI- mode) were obtained with determination coefficients (R(2)) higher than 0.99 for all the compounds. With five internal standards, good recoveries (84.4-103.0%) of all the target compounds were obtained in selected surface water samples. The developed method was successfully applied to investigate the EDCs occurrence in the surface water of Shanghai by analyzing surface water samples from 11 sites. The results showed that nearly all the target compounds (30 in 31) were present in the surface water samples of Shanghai, of which three industrial compounds (4-t-OP, BPA, and BPF) showed the highest concentrations (median concentrations were 11.88-23.50 ng L(-1)), suggesting that industrial compounds were the dominating EDCs in the surface water of Shanghai, and much more attention should be paid on these compounds. Our present research demonstrated that SPE with MCX cartridges combined with HPLC-MS/MS was convenient, efficient and reliable for multiclass analysis of EDCs in surface water.
