RESUMO

In this study, Lactiplantibacillus plantarum X7022 was applied to ameliorate memory impairment of aging mice induced by D-galactose. The strain showed specific choloylglycine hydrolysis ability based on in vitro investigation. Morris water maze test showed L. plantarum X7022 administration improved learning ability and spatial memory of aging mice. The gavage of L. plantarum X7022 displayed a promising ability of relieving cerebral oxidative stress and hippocampal inflammatory condition according to the increased GSH level and SOD activity and decreased MDA level, as well as decreased TNF-α, IL-1ß, and IL-6 levels. The intervention with the strain could protect neuron by regulating cell apoptosis and AChE overexpression and inhibiting amyloid-ß deposition, as well as affect neuron functions by regulating CREB-BDNF signaling pathways and iNOS expression. Besides, the strain could improve fecal SCFA contents and increase the abundance of anti-inflammatory and antioxidant-related genera such as Lactobacillus, Akkermansia, and Adlercreutzia. These results suggest that L. plantarum X7022 could be a prospective therapeutic alternative for the improvement of memory impairment among the elderly.

PURPOSE: Probiotics have been reported to effectively alleviate hyperuricemia and regulate the gut microbiota. The aim of this work was to study the in vivo anti-hyperuricemic properties and the mechanism of a novel strain, Lactiplantibacillus plantarum X7022. METHODS: Purine content and mRNA expression of purine assimilation related enzymes were determined by HPLC and qPCR, respectively. Hyperuricemic mice were induced by potassium oxonate and hypoxanthine. Uric acid (UA), blood urea nitrogen, creatinine and renal inflammation were examined by kits. The expression of renal UA transporters was subjected to western blotting. Kidney tissues were sectioned for histological analysis. The fecal short-chain fatty acids (SCFAs) were determined by HPLC, and gut microbiota was investigated using the 16S rDNA metagenomic sequencing. RESULTS: L. plantarum X7022 possesses a complete purine assimilation pathway and can exhaust xanthine, guanine, and adenine by 82.1%, 33.1%, and 12.6%, respectively. The strain exhibited gastrointestinal viability as 44% at the dose of 109 CFU/mL in mice. After four-week administration of the strain, a significant decrease of 35.5% in the serum UA level in hyperuricemic mice was achieved. The diminished contents of fecal propionate and butyrate were dramatically boosted. The treatment also alleviated renal inflammation and restored renal damage. The above physiological changes may due to the inhibited xanthine oxidase (XO) activity, as well as the expressional regulation of UA transporters (GLUT9, URAT1 and OAT1) to the normal level. Notably, gut microbiota dysbiosis in hyperuricemic mice was improved with the inflammation and hyperuricemia related flora depressed, and SCFAs production related flora promoted. CONCLUSION: The strain is a promising probiotic strain for ameliorating hyperuricemia.

Positive interactions have been hypothesised to influence plant community dynamics and species invasions. However, their prevalence and importance relative to negative interactions remain unclear to understand community change and invasibility. We examined pairwise biotic interactions using over 50 years of successional data to assess the prevalence of positive interactions and their effects on each focal species (either native or exotic). We found that positive interactions were widespread and the relative frequency of positive and negative interactions varied with establishment stage and between native and exotic species. Specifically, positive interactions were more frequent during early establishment and less frequent at later stages. Positive interactions involving native species were more frequent and stronger than those between exotic species, reducing the importance of invasional meltdown on succession. Our study highlights the role of positive native interactions in shielding communities from biological invasion and enhancing the potential for long-term resilience.

The dissimilarity and hierarchy of trait values that characterize niche and fitness differences, respectively, have been increasingly applied to infer mechanisms driving community assembly and to explain species co-occurrence patterns. Here, we predict that limiting similarity should result in the spatial segregation of functionally similar species, while functionally similar species will be more likely to co-occur either due to environmental filtering or due to competitive exclusion of inferior competitors (hereafter hierarchical competition).We used a fully mapped 50-ha subtropical forest plot in southern China to explore how pairwise spatial associations between saplings and between adult trees were influenced by trait dissimilarity and hierarchy in order to gain insight into assembly mechanisms. We assessed pairwise spatial associations using two summary statistics of spatial point patterns at different spatial scales and compared the effects of trait dissimilarity and trait hierarchy of different functional traits on the interspecific spatial associations. These comparisons allow us to disentangle the effects of limiting similarity, environmental filtering, and hierarchical competition on species co-occurrence.We found that trait dissimilarity was generally negatively related to interspecific spatial associations for both saplings and adult trees across spatial scales, meaning that species with similar trait values were more likely to co-occur and thus supporting environmental filtering or hierarchical competition. We further found that trait hierarchy outweighed trait dissimilarity in structuring pairwise spatial associations, suggesting that hierarchical competition played a more important role in structuring our forest community than environmental filtering across life stages.This study employed a novel method, by offering the integration of pairwise spatial association and trait dissimilarity as well as trait hierarchy, to disentangle the relative importance of multiple assembly mechanisms in structuring co-occurrence patterns, especially the mechanisms of environmental filtering and hierarchical competition, which lead to indistinguishable co-occurrence patterns. This study also reinforced the importance of trait hierarchy rather than trait dissimilarity in driving neighborhood competition.

A classic theory proposes that plant xylem cannot be both highly efficient in water transport and resistant to embolism, and therefore a hydraulic efficiency-safety trade-off should exist. However, the trade-off is weak, and many species exhibit both low efficiency and low safety, falling outside of the expected trade-off space. It remains unclear under what climatic conditions these species could maintain competitive fitness. We compiled hydraulic efficiency and safety traits for 682 observations of 499 woody species from 178 sites world-wide and measured the position of each observation within the proposed trade-off space. For both angiosperms and gymnosperms, observations from sites with high climatic seasonality, especially precipitation seasonality, tended to have higher hydraulic safety and efficiency than observations from sites with low seasonality. Specifically, high vapour pressure deficit, high solar radiation, and low precipitation during the wet season were driving factors. Strong climatic seasonality and drought in both dry and wet seasons appear to be ecological filters that select for species with co-optimized safety and efficiency, whereas the opposite environmental conditions may allow the existence of plants with low efficiency and safety.

Studies that test community assembly hypotheses in observational communities frequently evaluate patterns for plots or entire communities, yet studies that examine assembly patterns across spatial scales show that they are greatly influenced by scale. Here, we test the spatial dependency of patterns of relatedness and plant height for all individual herbaceous plants along five 40-m old-field transects (Southern Ontario, Canada). We identified each individual plant and measured its distance along the transect and its height, and we constructed a molecular phylogeny for all observed species. To uncover the scale at which community phylogenetic and trait similarities shift, we used partial Mantel correlograms and distance-based Moran Eigenvector Maps (dbMEMs). We found that communities shift from significantly overdispersed at relatively smaller scales (i.e., < 15 m) to spatially clustered at larger scales, showing that assembly mechanism influence depends on scale of observation. This pattern was observed for both phylogeny and height, but was the strongest when considering phylogeny only. These results reveal the importance of spatial scale when examining community phylogenetic or trait patterns, where finding support for one assembly mechanism at a single scale does not necessarily mean that other mechanisms are also not important for structuring community composition and diversity.

Phylogenetic analyses are essential for disentangling how environmental filtering and competition determine species coexistence across spatial scales. Inner Mongolia steppe has strong environmental gradients, but how the phylogenetic relatedness of co-occurring species and phylogenetic signals of functional traits change across spatial scales remains unclear. We investigated the phylogenetic structure of grass assemblages along environmental gradients from regional to local scales, and measured functional traits within assemblages. We compared phylogenetic signals of plant traits between the same numbers of species randomly selected from the regional pool and species observed at the local scale, did phylogenetic principal component analysis to infer the main factors driving species coexistence, and examined the key plant trait-environment relationships across the phylogeny to reveal ecological adaptation mechanisms. Regionally, grass species were phylogenetically clustered with contrasting climate preferences. With decreasing spatial scales, species richness declined, changing from phylogenetically clustered to overdispersed, and phylogenetic signals of plant traits became weaker. At the local scale, grass assemblages were structured by soil water content and neighbor density, and the trait-environment relationships were less clear than those at the regional scale. This study demonstrated that at smaller scales, co-occurring grass species in the steppe tended to be more phylogenetically overdispersed, and that phylogenetic signals of plant functional traits became weaker with increasing abiotic and biotic interactions. Our findings contributed evidence for understanding species coexistence and maintenance at scales spanning regional to local communities in the East Asia steppe biome.
