RESUMO

Xelaglifam, developed as a GPR40/FFAR1 agonist, induces glucose-dependent insulin secretion and reduces circulating glucose levels for Type 2 diabetes treatment. This study investigated the effects of Xelaglifam in comparison with Fasiglifam on the in vitro/in vivo anti-diabetic efficacy and selectivity, and the mechanistic basis. In vitro studies on downstream targets of Xelaglifam were performed in GPR40-expressing cells. Xelaglifam treatment exhibited dose-dependent effects, increasing inositol phosphate-1, Ca2+ mobilization, and ß-arrestin recruitment (EC50: 0.76 nM, 20 nM, 68 nM), supporting its role in Gq protein-dependent and G-protein-independent mechanisms. Despite a lack of change in the cAMP pathway, the Xelaglifam-treated group demonstrated increased insulin secretion compared to Fasiglifam in HIT-T15 ß cells under high glucose conditions. High doses of Xelaglifam (<30 mg/kg) did not induce hypoglycemia in Sprague-Dawley rats. In addition, Xelaglifam lowered glucose and increased insulin levels in diabetic rat models (GK, ZDF, OLETF). In GK rats, 1 mg/kg of Xelaglifam improved glucose tolerance (33.4 % and 15.6 % for the 1 and 5 h) after consecutive glucose challenges. Moreover, repeated dosing in ZDF and OLETF rats resulted in superior glucose tolerance (34 % and 35.1 % in ZDF and OLETF), reducing fasting hyperglycemia (18.3 % and 30 % in ZDF and OLETF) at lower doses; Xelaglifam demonstrated a longer-lasting effect with a greater effect on ß-cells including 3.8-fold enhanced insulin secretion. Co-treatment of Xelaglifam with SGLT-2 inhibitors showed additive or synergistic effects. Collectively, these results demonstrate the therapeutic efficacy and selectivity of Xelaglifam on GPR40, supportive of its potential for the treatment of Type 2 diabetes.

The worst forest fires in Korean history broke out on March 4, 2022 and lasted for ten days. In order to monitor the catastrophic forest fires, Geostationary Korea Multi-Purpose Satellite (GK)-2 A Advanced Meteorological Imager (AMI) and GK-2B Geostationary Environment Monitoring Spectrometer (GEMS) data were used in this study. Aerosol optical depth (AOD) irretrievable for the biomass-burning aerosols produced with water vapor classified as could-contaminated, was reconstructed by ultraviolet aerosol index (UVAI). Afterward, aerosol radiative forcing (ARF) at TOA was finally estimated by the correlation of AOD and surface albedo with ARF. Most of the aerosols drifted toward the East Sea by the prevailing westerly winds, and caused a cooling effect on the atmosphere with a maximum daily average radiative forcing of -69.28 Wm-2. Furthermore, the fire-prone conditions for the unprecedented forest fires were discussed in detail as following aspects; 1) the most severe drought caused by a "triple-dip" La Niña; 2) pressure patterns and topographical features that generate strong winds; 3) coniferous forests prone to fires; and 4) increased human activity following the nationwide COVID-19 vaccination. This study demonstrated that the rapid and effective ARF estimation based on the satellite remote sensing can contribute to a better understanding of ARF in the Earth's radiation budget for the global forest fires that will be more frequent, intense, and longer-lasting due to the human-caused climate and environment changes.

Captive propagation and reintroduction are the major steps in the ex-situ conservation of locally extirpated endangered species in a historical region. In a species restoration project conducted in South Korea, we examined temporal changes in demographics and genetic diversity of oriental storks (Ciconia boyciana). Demographic and genetic data from 1996-2018 were analyzed for 80% of all captive and recently reintroduced individuals. Founder establishment and pair formation induced increases in population size and genetic diversity during the early stage of captive propagation. The degree of genetic diversity was found to become saturated and stable with long-term captive propagation. However, this might be a concern for future genetic diversity of both captive and reintroduced populations simultaneously due to the extraction of captive populations at the early stage of reintroduction. Our findings suggest that periodic evaluation of genetic diversity and selection for releasing individuals, using effective genetic markers, would assist in balancing the genetic diversity of the captive and reintroduced oriental storks at the early stage of reintroduction.

Interspecific hybridization occurs among birds, and closely related sister taxa tend to hybridize at a high rate. Genomic hybridization markers are useful for understanding the patterns and processes of hybridization and for conserving endangered species in captivity and the wild. In this study, we developed genomic hybridization markers for the F1 progeny of the sister taxa feral pigeons (Columba livia var. domestica) and endangered hill pigeons (Columba rupestris) (family Columbidae). Using whole-genome re-sequencing data, we performed genome-wide analysis for insertion/deletion (InDel) polymorphisms and validated using primers. We conducted polymerase chain reaction (PCR) and agarose gel electrophoresis to identify species-specific InDels. We produced eight F1 hybrids of hill and feral pigeons, and their samples were tested by re-performing analyses and sequencing using 11 species-specific InDel polymorphisms. Eight InDel markers simultaneously amplified two DNA fragments from all F1 hybrids, and there was no abnormality in the sequencing results. The application of genomic tools to detect hybrids can play a crucial role in the assessment of hybridization frequency in the wild. Moreover, systematic captive propagation efforts with hybrids can help control the population decline of hill pigeons.

Understanding possible follow-up actions on in vitro findings helps determine the necessity of labeling for drug interactions. We analyzed information for in vitro findings on transporter-mediated interactions of drugs approved by the U.S. Food and Drug Administration's Center for Drug Evaluation and Research for the last five years (i.e., 2017-2021) and their follow-up actions for labeling. Higher R values than the pre-defined cut-off were observed with 3.7-39.1% inhibitor drugs in a simple prediction. Among these drugs, 16-41.7% were labeled with their potential drug interactions, while results of supporting studies or scientific rationales were submitted for the other drugs leading to no interaction labeling. In vitro transporter substrates were reported with 1.7-67.6% of drugs. The interaction labels for these substrate drugs were observed in up to 40% of drugs, while the other drugs were not labeled on the drug interactions with claims for their low interaction potential, evidenced by clinical studies or scientific rationales. The systematic and comprehensive analysis in this study will provide insight into the management of in vitro findings for transporter substrate or inhibitor drugs.

The latitudinal and elevational patterns of species richness of resident and migrant birds have been of interest to researchers over the past few decades, and various hypotheses have been proposed to explain the factors that may affect these patterns. This study aimed to shed light on the elevational distribution patterns of resident and migrant bird species richness by examining biotic and abiotic factors such as climate, and habitat heterogeneity using a piecewise structural equation model (pSEM). The overall pattern of resident species richness showed a decreasing trend with increasing elevation, whereas that of migrant species richness showed an increasing trend. The mid-peak pattern of species richness was affected by a combination of resident and migrant species and not by either resident or migrant species. Our results showed that resident species were distributed in lower elevation regions with higher mean spring temperatures, whereas migrant species were found in higher elevation regions with lower mean spring temperatures and higher overstory vegetation coverage. Although high elevation conditions might adversely affect the reproduction of migrant birds, higher overstory vegetation coverage at high elevations seemed to compensate for this by providing a better nesting and roosting environment. Despite the significance of habitat diversity and understory vegetation coverage in univariate linear regression models, multiple regression models of the interconnection of ecological processes demonstrated that mean spring temperature and overstory vegetation coverage were more explanatory than other variables.

BACKGROUND: Hill pigeons (Columba rupestris) are close to local extinction (ca. less than 100 individuals) in South Korea where a variety of conservation management procedures are urgently required. OBJECTIVE: This study was aimed at determining the conservation direction of captive propagation and reintroduction of hill pigeons using genetic information based on mitochondrial DNA. We also evaluated the extent of hybridization between hill pigeons and cohabiting domestic pigeons. METHODS: We used 51 blood samples of hill pigeons from Goheung (GH), Gurye (GR), and Uiryeong (UR), and domestic pigeons cohabiting with hill pigeon populations. Genetic diversity, pairwise Fst, analysis of molecular variance, and haplotype network analysis were used to examine the genetic structure of hill pigeons. RESULTS: Hill pigeons that inhabited South Korea were not genetically distinct from Mongolian and Russian populations and showed relatively low genetic diversity compared with other endangered species in Columbidae. The GR population that exhibited the largest population size showed lower genetic diversity, compared to the other populations, although the pairwise Fst values of the three populations indicated low genetic differentiation. The GH and GR populations were confirmed to lack hybridization, relatively, whereas the UR population was found to exhibit some degrees of hybridization. CONCLUSION: To conserve hill pigeons with low genetic diversity and differentiation in South Korea, the conservation process of captive propagation and reintroduction may require artificial gene flows among genetically verified populations in captivity and wildness. The introduction of foreign individuals from surrounding countries is also considered an alternative strategy for maintaining genetic diversity.

The long-tailed goral, Naemorhedus caudatus (Mammalia: Bovidae), is one of the endangered animals in the Republic of Korea (Korea). Sarcoptic mange mites infested in diverse species of mammals, including humans, but no case has been reported in long-tailed gorals. We report 2 cases of mange mite, Sarcoptes scabiei, infestation in long-tailed gorals. Mange mites were sampled in the skin legions of the 2 long-tailed gorals, which were rescued in 2 different regions, Uljin-gun, Gyeongsangbuk-do and Cheorwon-gun, Gangwon-do, Korea. Our results showed that the ectoparasite was the itch mite that burrowed into skin and caused scabies on the morphological inspection and placed within the phylogenetic relations of the species. The present study confirmed for the first time in Korea that mange mites are pathogenic scabies of long-tailed goral. Closer surveillance of this pathogenic ectoparasite in zoonotic and infectious ecosystems is warranted.

Satellite-derived aerosol optical depth (AOD) products are one of main predictors to estimate ground-level particulate matter (PM10 and PM2.5) concentrations. Since AOD products, however, are only provided under high-quality conditions, missing values usually exist in areas such as clouds, cloud shadows, and bright surfaces. In this study, spatially continuous AOD and subsequent PM10 and PM2.5 concentrations were estimated over East Asia using satellite- and model-based data and auxiliary data in a Random Forest (RF) approach. Data collected from the Geostationary Ocean Color Imager (GOCI; 8 times per day) in 2016 were used to develop AOD and PM models. Three schemes (i.e. G1, A1, and A2) were proposed for AOD modeling according to target AOD data (GOCI AOD and AERONET AOD) and the existence of satellite-derived AOD. The A2 scheme showed the best performance (validation R2 of 0.74 and prediction R2 of 0.73 when GOCI AOD did not exist) and the resultant AOD was used to estimate spatially continuous PM concentrations. The PM models with location information produced successful estimation results with R2 of 0.88 and 0.90, and rRMSE of 26.9 and 27.2% for PM10 and PM2.5, respectively. The spatial distribution maps of PM well captured the seasonal and spatial characteristics of PM reported in the literature, which implies the proposed approaches can be adopted for an operational estimation of spatially continuous AOD and PMs under all sky conditions.

Some seabirds commonly use artificially reclaimed lands, which are frequently located next to mainland environments, for breeding. Nest predation risk caused by birds or mammals from the mainland has negative influence on fitness-related costs and distribution of seabirds. Here, we sought to link potential factors, specifically those related to nest predation and nest environment, with breeding performance and colony movements of the Saunders's gull (Saundersilarus saundersi), a vulnerable species, on a large reclaimed area (1350 ha) in Incheon in Republic of Korea. This reclaimed area has experienced rapid changes in communities of nest predators from the mainland and vegetation ranging from halophytes to terrestrial plants after reclamation. Additionally, changes in the surrounding of used nest sites were retrospectively examined to determine whether colony movement was reversible in this reclaimed area. Our results indicated that high nest predation in a previous year induced colony movements in a consecutive year while the breeding colony exhibited a gradual reduction in clutch size. However, such movement after high nest predation seemed to be irreversible due to ongoing habitat degradation caused by construction and vegetation alteration. This study highlights that high nest predation may exert strong pressure on breeding colonies of Saunders's gulls. It also has anthropogenic impacts, leading to continuous dispersal of colonies to new areas for this vulnerable seabird in a reclaimed land.

Despite the increasing prevalence of inflammatory bowel disease (IBD), classified as immune-mediated disorders, the exact biological mechanisms leading to its development are undetermined, and treatment strategies remain elusive. Probiotics have been proposed as potential alternatives for treating IBD. The purpose of this research was to find therapeutic candidates of probiotics for colitis. We adopted dextran sulfate sodium (DSS)-induced colitis model to demonstrate the therapeutic effects of ID-JPL934, a mixture of three live bacterial strains at a 1:1:1 ratio: Lactobacillus johnsonii IDCC9203, Lactobacillus plantarum IDCC3501, and Bifidobacterium animalis subspecies lactis IDCC4301, on IBD. The severity was scored according to the disease activity index (DAI) for colitis by observing body weight (BW) and stool status of each mouse once a day. BALB/c mice given 3.5% DSS in drinking water suffered from symptoms of colitis such as weight loss, diarrhea, and bloody excrement. In our study, administration of ID-JPL934 reduced the DAI scores in a dose-dependent manner, and treatments with ID-JPL934 108 and 109 colony-forming unit per mouse per day showed similar inhibition compared with those of sulfasalazine 500 mg per kg BW per day. Moreover, the contraction of colon length improved. ID-JPL934 also suppressed inflammatory lesions such as infiltration of immune cells in mucosa and submucosa, severe crypt damage, and loss of goblet and epithelial cells on the histological analysis. These results might be due to downregulation of the expression of proinflammatory cytokines, including tumor necrosis factor-α, interleukin (IL)-1ß, and IL-6. From these results, ID-JPL934 might be an effective therapeutic candidate for IBD.

Osteoarthritis is a disease that affects the articular cartilage and osseous tissue, and can be worsened by aging, overweight status, and post-traumatic arthritis. The present study aimed to evaluate the effect of ID-CBT5101 (tyndallized Clostridium butyricum) on bone metabolism and the inflammatory response in a monosodium iodoacetate-induced rat model of osteoarthritis. ID-CBT5101 was administered orally at doses of 108 or 1010 CFU/day for 2 weeks before direct injection of monosodium iodoacetate (3 mg/50 µl of 0.9% saline) into the intra-articular space of the rats' right knees. The rats subsequently received the same doses of oral ID-CBT5101 for another 4 weeks. We evaluated the treatment effects based on serum biomarkers, mRNA expression, morphological and histopathological analyses of the knee joints, and weight-bearing distribution analysis. Compared with those in control rats, the ID-CBT5101 treatments significantly reduced the serum concentration of inflammation and bone metabolism markers (i.e., COX-2, IL-6, LTB4, and COMP), and significantly increased the concentration of IFN-γ and glycosaminoglycans. In addition, the ID-CBT5101 treatments inhibited the mRNA expression of matrix metalloproteinases and tissue inhibitors of metalloproteinases (i.e., MMP-2, MMP-3, MMP-9, MMP-13, TIMP-1, and TIMP-2). Furthermore, the ID-CBT5101 treatments effectively preserved the knee cartilage and synovial membrane, and significantly decreased the amount of fibrous tissue. Moreover, compared with that of the negative control group, the ID-CBT5101 treatments increased the weight-bearing distribution by ≥20%. The results indicate that ID-CBT5101 prevented and alleviated osteoarthritis symptoms. Thus, ID-CBT5101 may be a novel therapeutic option for the management of osteoarthritis.

Some nest predators visually assess parental activities to locate a prey nest, whereas parents modify fitness-related traits to reduce the probability of nest predation, and/or nestlings fledge early to escape the risky nest environment. Here, we experimentally tested if the parental and fledging behaviours of oriental tits (Parus minor) that bred in the nest-box varied with cavity conditions associated with nest predation risk during the nestling period. The entrance of experimental nest-boxes was enlarged to create a long-term risk soon after clutch competition. A short-term risk, using simulated playbacks with a coexisting control bird and avian nest predator sound, was simultaneously applied to the nest-boxes whether or not the long-term risk existed. We found that the parents reduced their hourly feeding trips, and the nestlings fledged early with the long-term risk, although the nest mortality of the two nest-box types was low and did not differ. While this study presents a portion of prey-predator interactions with the associated uncertainties, our results highlight that the entrance size of cavities for small hole-nesting birds may play an important role in determining their fitness-related traits depending upon the degree of perceived risk of nest predation.

The therapeutic effect of oral administration of Lactobacillus rhamnosus IDCC 3201 tyndallizate (RHT3201) on atopic dermatitis (AD)-like skin lesions in NC/Nga mice were investigated. After induction of dermatitis in NC/Nga mice with house-dust mite extract, each group was fed RHT3201 with 1 × 10(8) , 1 × 10(9) , or 1 × 10(10) cells orally once a day for 8 weeks. Dermatitis scores and frequency of scratching were improved by oral feeding with RHT3201. In contrast to the control group, RHT3201-fed mice showed significantly down-regulated mast cell numbers and serum immunoglobulin E (IgE) concentrations had significantly less IL4 in their axillary lymph node cells. The therapeutic effect of RHT3201 was found to be dose-dependent. These findings indicate that RHT3201 has potential for treating AD.

We have previously reported amidopiperidine derivatives as a novel peptide deformylase (PDF) inhibitor and evaluated its antibacterial activity against Gram-positive bacteria, but poor pharmacokinetic profiles have resulted in low efficacy in in vivo mouse models. In order to overcome these weaknesses, we newly synthesized aminopiperidine derivatives with remarkable antimicrobial properties and oral bioavailability, and also identified their in vivo efficacy against methicillin-resistant Staphylococcus aureus (MRSA), vancomycin-resistant Enterococcus (VRE) and penicillin-resistant Streptococcus pneumoniae (PRSP).

Parental behaviors that vary by sex and breeding stage facilitate the survival of offspring in birds. Females invest in reproducing according to the level of direct and/or indirect benefits provided by males. Males face trade-offs from defending territories or nests, providing for the young, and seeking additional mating opportunities. We examined whether post-mating sexual behaviors such as courtship display and nest-building were associated with reproductive investments made by males and females in the current study. The oriental stork (Ciconia boyciana) is known as a solitary breeder that nests in tall trees and provides biparental care with long post-hatching development. We filmed parental behaviors in seven stork pairs in captivity for 655 hours (245 hours during nest-building, 218 hours during incubation, 328 hours during nestling, and 192 hours during the fledgling period). We found that paired mating behaviors such as synchronous allopreening and bill-clattering were highly skewed to the pre-incubation period. Males participated in nest-building more than females although both sexes shared similar parental care during the incubation and nestling period. Male nest-building was negatively correlated with male nest attentiveness only during the nestling period. Our results suggest that male oriental storks with higher nest-building effort might spend more time taking precautions against conspecific intruders or nest predators near the nests with chicks while the females were attending the nests in the form of biparental care.

CONTEXT: The rhizome of Polygonatum sibiricum Redoute (Liliaceae) has long been used to treat diabetes-associated complications. However, the pharmacological mechanism of P. sibiricum on metabolic disorders is not clear. OBJECTIVE: This study investigates the effect of an ethanol extract of P. sibiricum rhizomes (designated ID1216) on obesity conditions including weight loss in high-fat (HF) diet-fed mice and explores the potential underlying mechanisms. METHODS: To identify the metabolic impact of the P. sibiricum rhizome extract, HF diet-fed mice were administered ID1216 orally at doses of 250 and 1000 mg/kg/d for 10 weeks, and various factors related to metabolic syndrome were analyzed. We also examined the effects of ID1216 on the expression of genes involved in adipogenesis and lipolysis in 3T3-L1 cells, as well as genes associated with energy homeostasis in C2C12 myocytes. RESULTS: ID1216 administration led to significant decreases in body weight gain (37.5%), lipid accumulation in adipose tissues (52.8%), and the levels of plasma triglycerides (26.4%) and free fatty acids (28.1%) at a dose of 250 mg/kg/d, compared with the vehicle-treated group, as well as improved insulin resistance. In addition, ID1216 was found to regulate the expression of genes related to adipogenesis and fatty acid oxidation in 3T3-L1 cells and enhance the expression of genes that modulate energy homeostasis in C2C12 myocytes. CONCLUSIONS: ID1216 may be a promising therapeutic agent for improving obesity conditions through the sirtuin-1 and peroxisome proliferator-activated receptor γ coactivator-1α pathway.

Various selection pressures induce the degree and direction of sexual size dimorphism in animals. Selection favors either larger males for contests over mates or resources, or smaller males are favored for maneuverability; whereas larger females are favored for higher fecundity, or smaller females for earlier maturation for reproduction. In the genus of Larus (seagulls), adult males are generally known to be larger in size than adult females. However, the ontogeny of sexual size dimorphism is not well understood, compared to that in adults. The present study investigates the ontogeny of sexual size dimorphism in Saunders's gulls (Larus saundersi) in captivity. We artificially incubated fresh eggs collected in Incheon, South Korea, and measured body size, locomotor activity, and foraging skill in post-hatching chicks in captivity. Our results indicated that the sexual differences in size and locomotor activity occurred with the post-hatching development. Also, larger males exhibited greater foraging skills for food acquisition than smaller females at 200 days of age. Future studies should assess how the adaptive significance of the sexual size dimorphism in juveniles is linked with sexual divergence in survival rates, intrasexual contests, or parental effort in sexes.

Comparative hormone studies can reveal how physiology underlies life history variation. Here, we examined seasonal variation in plasma testosterone concentration between populations of male orange-crowned warblers (Vermivora celata) breeding in Fairbanks, Alaska (V. c. celata) and on Santa Catalina Island, California (V. c. sordida). These populations face different ecological constraints and exhibit different life histories. Alaska birds have a short breeding season, low annual adult survival, and high reproductive rates. In contrast, Catalina Island birds exhibit high adult survival and low reproductive rates despite having a long breeding season. We examined seasonal variation in male testosterone concentrations as a potential mechanism underlying differences in male reproductive strategies between populations. From 2006 to 2008, we sampled males during the pre-incubation, incubation, and nestling stages. Alaska males exhibited a seasonal testosterone pattern typical of northern passerines: testosterone levels were high during pre-incubation and declined during incubation to low levels during nestling provisioning. Testosterone concentrations in Catalina Island males, however, did not vary consistently with breeding stage, remained elevated throughout the breeding season, and were higher than in Alaska males during the nestling stage. We hypothesize that in Alaska, where short seasons and high adult mortality limit breeding opportunities, the seasonal testosterone pattern facilitates high mating effort prior to incubation, but high parental investment during the nestling stage. On Catalina Island, elevated testosterone levels may reflect the extended mating opportunities and high population density facing males in this population. Our results suggest that population variation in seasonal testosterone patterns in orange-crowned warblers may be a function of differences in life history strategy and the social environment.

Frequencies of spontaneous mutation from inducible resistance to constitutive resistance were determined for the four clinical isolates of erythromycin-resistant enterococci, including one isolate with ermB gene and three clinical isolates with ermA gene. The rate of ermB mutation was higher than that of ermA mutation by more than 10 fold. Sequence analysis of the regulatory regions of erm genes revealed that mutation type of ermB was just point mutation, by contraries the mutation type of ermA was either deletion or tandem duplication. These results showed distinct characteristics in mutation patterns of ermB and ermA.
