RESUMO

Brucellosis is a zoonotic caused by the Brucella which is a well-known infectious disease agent in domestic animals and if transmitted, it can cause infection in humans. Because brucellosis is contagious, its control depends on the eradication of the animal disease in farms. There are two vaccines based on the killed and/or weakened bacteria against B. melitensis and B. abortus, but no recombinant vaccine is available for preventing the disease. The present study was designed to develop a multi-epitope vaccine against of B. melitensis and B. abortus using virB10, Omp31 and Omp16 antigens by the prediction of T lymphocytes, T cell cytotoxicity and IFN-γ epitopes. 50S L7/L12 Ribosomal protein from Mycobacterium tuberculosis was used as a bovine TLR4 and TLR9 agonist. GPGPG, AAY and KK linkers were used as a linker. Brucella construct was well-integrated in the pET-32a Shuttle vector with BamHI and HindIII restriction enzymes. The final construct contained 769 amino acids, that it was soluble protein of about ∼82 kDa after expression in the Escherichia coli SHuffle host. Modeled protein analysis based on the tertiary structure validation, molecular docking studies, molecular dynamics simulations results like RMSD, Gyration and RMSF as well as MM/PBSA analysis showed that this protein has a stable construct and is capable being in interaction with bovine TLR4 and TLR9. Analysis of the data obtained suggests that the proposed vaccine can induce the immune response by stimulating T- and B-cells, and may be used for prevention and remedial purposes, against B. melitensis and B. abortus.Communicated by Ramaswamy H. RESUMO

Organophosphate compounds (OPCs) are a diverse class of chemicals utilized in both industrial and agricultural settings. The exact molecular pathways that OPCs-induced toxicity is caused by are still being investigated, despite the fact that studies on this topic have been ongoing for a long time. As a result, it's important to identify innovative strategies to uncover these processes and further the understanding of the pathways involved in OPCs-induced toxicity. In this context, determining the role of microRNAs (miRs) in the toxicity caused by OPCs should be taken into consideration. Recent research on the regulation function of miRs presents key discoveries to identify any gaps in the toxicity mechanisms of OPCs. As diagnostic indicators for toxicity in people exposed to OPCs, various expression miRs can also be used. The results of experimental and human studies into the expression profiles of miRs in OPCs-induced toxicity have been compiled in this article.

INTRODUCTION: This meta-analysis study assessed the effects of Berberis vulgaris L. and Berberis aristata L. in patients with metabolic syndrome. METHODS: Data were analysed through "random-effects meta-regression" performance. RESULTS: The findings indicated that LDL was 0.68 and 2.92 lower in the B. vulgaris L. and B. aristata L.-treated groups versus the controls. The HDL was 0.71-fold higher in the B. aristata L.-treated group versus the controls. The total-cholesterol levels were 1.02 and 2.25 folds lower in the B. vulgaris L. and B. aristata L.-treated groups versus the matched control groups. The triglyceride levels were 1.35 and 1.16-fold lower in the B. vulgaris L. and B. aristata L.-treated groups versus the controls. Glucose was 0.96 and 0.54 folds lower in the B. vulgaris L. and B. aristata L.-treated groups versus the control groups, respectively. CONCLUSION: B. vulgaris L. and B. aristata L. have beneficial effects in patients with metabolic syndrome.

This study aimed to assess the effects of Buprenorphine (BUP) on oxidative parameters in pups born to mothers exposed to the drug during gestation and lactation. Pregnant and lactating rats received BUP, 0.5 or 0.1 mg/kg subcutaneously for 21 and 28 days, respectively. At the end of the study, the pups were anesthetized, and the hearts were dissected out to measure oxidative stress indices, including the levels of Malondialdehyde (MDA), Nitric oxide (NO), Glutathione (GSH), and the activity of Superoxide dismutase (SOD). Our findings indicated that BUP did not alter MDA, NO, GSH levels, nor SOD activity in the cardiac tissue of pups exposed to this drug during the fetal period and through breast milk. We suggest performing additional studies to determine the association between BUP and oxidative modifications in cardiac tissues of pups born to mothers under BUP therapy during gestation and lactation.

BACKGROUND & OBJECTIVE: Age-dependent Organophosphates (OPs) toxicity is a controversial topic. The present study was designed to investigate the effect of the sub-acute exposure to diazinon (DZN), one of the main OPs insecticides, on the hematological alterations in adult and aged male rats. METHODS: For the aim of this approach, the adult and aged rats were administered with DZN (15 mg/kg, orally) for 4 weeks. Then, the blood samples were collected from the retro-orbital sinus for measuring red blood cell (RBC), hemoglobin (Hb), hematocrit (Hct), platelets (PLT), MCV (mean corpuscular volume), mean corpuscular hemoglobin (MCH), and mean corpuscular hemoglobin (MCHC). RESULTS: The obtained results indicated that DZN significantly decreased RBCs (4.93 ± 0.41), Htc (28.12 ± 1.21), Hb (10.31 ± 0.36), MCHC (30.51 ± 2.04), MCV (62.86 ± 2.58), and PLT (265.6 ± 34.81) values in the adult and aged rats versus the age-matched control rats. Moreover, RBC, Hb, and Htc levels decreased significantly in the aged rats versus adult rats. However, no significant differences were observed between MCHC, MCV, and PLT levels in adult and aged rats. Moreover, the MCH concentration did not change in any group. Additionally, DZN did not deteriorate the hematological alterations in the aged rats versus adult rats. CONCLUSION: the present study showed that the toxicity of DZN is not associated with age. However, more studies should be conducted to confirm this finding.

BACKGROUND: Diabetes and its complications are age-related diseases. Low-grade inflammation plays the main role in the aging processes. Diazinon (DZN), an organophosphate pesticide, has been found to induce metabolic disturbances. OBJECTIVE: The present study was designed to investigate the impact of DZN on age-related changes on inflammatory cells, blood glucose concentration, lipid profile, and liver and kidney function indices in adult and aged rats. METHODS: Male rats (2 and 16 month old) were orally administrated with DZN (15 mg/kg) for 4 weeks. Then the blood was obtained for measuring inflammatory cells, lipid profile, glucose and serum biochemical indices such as liver enzymes, albumin, total protein, creatinine (Cr), urea, and uric acid in the serum of adult and aged male rats. RESULTS: DZN increased the blood levels of glucose and the percentage of lymphocytes and also serum levels of TChol, TG, LDL-c, AST, ALT, ALP, LDH, Cr, urea, and uric acid in the adult and aged rats versus the aged matched control rats (p< 0.001). A marked reduction in HDL-c levels, total protein, albumin, and in the percentage of neutrophils were seen in the adult and aged animals exposed to DZN versus the aged matched control rats. DZN also increased the levels of LDL-c and ALT in the aged rats versus adult animals. CONCLUSION: The present study indicated that DZN can cause metabolic disturbance. However, the age-dependent effects of DZN on metabolic indices were not be confirmed by the present data.
