RESUMO

The utilization of atomically confined plasmonic fields has revolutionized the imaging technique. According to the fundamental position-momentum uncertainty principle, such a narrow spatial distribution certainly leads to a broad momentum distribution in the fields, which has however been overlooked. Here we propose a novel exploitation for the momentum distribution by adaptively satisfying the conservation law of momentum in inelastic Raman scatterings in periodic systems, providing a unique optical means of directly measuring the whole phonon dispersions. The proposed technique is particularly useful for measuring phonon dispersions of low-dimensional hydrogen-rich materials, which are completely inaccessible via other techniques. The numerical results for a single all-trans polyacetylene chain demonstrate that all phonon dispersion branches can be conclusively measured from their Raman images for the first time. Our findings highlight a unique advantage of the emerging momentum-based nanophotonics and open the door for exploiting highly confined plasmonic fields in another dimension.

INTRODUCTION: Acute primary angle closure (APAC) is often characterized by acute elevation of intraocular pressure (IOP) accompanied by severe ocular and systemic symptoms. Excessive collagen accumulation, which can be caused by upregulated heat shock protein 47 (HSP47) expression, can produce scarring in rat conjunctival blebs. Meanwhile, the presence of HSP47 in human aqueous humor and its levels are yet to be determined. METHODS: We examined 32 consecutive patients with APAC and 16 age-matched participants without APAC scheduled for cataract surgery who were enrolled as a control group. Aqueous humor samples were collected from all subjects at the time of surgery and compared between the subjects with and without APAC. RESULTS: The levels of HSP47 in the aqueous humor of patients with APAC (1,210.4 ± 450.2 pg/mL) were found to be significantly increased (P = 0.001) compared with those in the control group (863.4 ± 240.0 pg/mL). Notably, the levels of HSP47 negatively correlated with the age of patients with APAC (P = 0.023). CONCLUSION: HSP47 was upregulated in the aqueous humor of patients with APAC and may play a role in scarring after trabeculectomy for APAC.

Nuclear receptor subfamily 4 group A1 (NR4A1) is an orphan nuclear receptor, which is expressed in the majority of cells. NR4A1 expression in peripheral blood mononuclear cells is low during the preclinical stage of multiple sclerosis. Knockout of the Nr4a1 gene in mice can aggravate the symptoms of experimental autoimmune encephalomyelitis (EAE), which is an animal model of multiple sclerosis. In this study, we intragastrically administered the NR4A1 agonist cytosporone B (Csn-B) to mice after inducing EAE. After treatment with Csn-B, the clinical symptoms in the EAE mice were substantially attenuated compared with that in PBS-treated control mice. The percentages of CD4+ T cells and F4/80+ cells in the central nervous system were decreased. In addition, interferon-γ and interleukin-17 production by proinflammatory Th1/Th17 cells in the central nervous system and interferon-γ levels in splenocytes were decreased after Csn-B treatment. These findings suggest that the NR4A1 agonist Csn-B can alleviate nerve injury after EAE induction, and, therefore, may be useful as a potential treatment for multiple sclerosis.

OBJECTIVE: To identify potential tumor antigen by immunoscreening the urinary bladder cancer cDNA library with monoclonal antibodies. METHODS: Monoclonal antibodies were prepared. A cDNA expression library was constructed from bladder cancer cell line BLZ211. Immunogenic proteins were identified by immunoscreening the cDNA library with ten monoclonal antibodies. RESULTS: The cDNA library of BLZ211 cells was established using lambdaZAP as vector. The titer of unamplified cDNA library was 1.39x10(6) pfu/ml with a recombinant rate of 97.72%, and titer of amplified one was 8.4x10(9) pfu/ml. After immunoscreening, ten positive clones representing ten different antigens were identified, which include two proteins with unknown function; coactosin-like 1, eukaryotic translation elongation factor, HNRPA1, histidine triad nucleotide binding protein, KRT7, LCN2, TSTA3, zinc finger protein, C11orf48 and HSPC148. CONCLUSION: The cDNA library was of high quality and can be used in further study. By immunoscreening the bladder cancer cDNA library with ten monoclonal antibodies, we identified ten immunogenic proteins that otherwise would not have been identified as potential diagnostic marker and vaccinogens of bladder cancer using the gene discovery effort.
