OBJECTIVE: In our previous study, we identified a notable increase in miR-548ag content after obesity, which contributes to the progression of Type 2 diabetes Mellitus(T2DM) through the up-regulation of Dipeptidyl Peptidase-4(DPP4) expression within the liver. However, the precise molecular mechanisms underlying the upregulation of DPP4 by miR-548ag remain elusive. Mature miRNAs rich in GU sequences can activate the TLR(7/8)/NF-κB signalling pathway, which transcriptionally activates DPP4 expression. Notably, the proportion of GU sequences in hsa-miR-548ag was found to be 47.6%. The study proposes a hypothesis suggesting that miR-548ag could potentially increase DPP4 expression in hepatocytes by activating the TLR(7/8)/NF-κB signalling pathway. METHODS: Male C57BL/6J mice were fed normal chow diet (NCD, n = 16) or high-fat diet (HFD, n = 16) for 12 weeks. For a duration of 6 weeks, NCD mice received intraperitoneal injections of a miR-548ag mimic, while HFD mice and db/db mice (n = 16) were administered intraperitoneal injections of a miR-548ag inhibitor. qRT-PCR and Western Blot were used to detect the expression level of miR-548ag, DPP4 and the activation of TLR(7/8)/NF-κB signalling pathway. HepG2 and L02 cells were transfected with miR-548ag mimic, miR-548ag inhibitor, TLR7/8 interfering fragment, and overexpression of miR-548ag while inhibiting TLR7/8, respectively. RESULTS: (1) We observed elevated levels of miR-548ag in the serum, adipose tissue, and liver of obese mice, accompanied by an upregulation of TLR7/8, pivotal protein in the NF-κB pathway, and DPP4 expression in the liver. (2) miR-548ag promotes DPP4 expression in hepatocytes via the TLR(7/8)/NF-κB signalling pathway, resulting in a reduction in the glucose consumption capacity of hepatocytes. (3) The administration of a miR-548ag inhibitor enhanced glucose tolerance and insulin sensitivity in db/db mice. CONCLUSIONS: MiR-548ag promotes the expression of DPP4 in hepatocytes by activating the TLR(7/8)/NF-κB signalling pathway. MiR-548ag may be a potential target for the treatment of T2DM.

Photovoltaic solid waste, particularly diamond-wire sawing silicon powder (DWSSP), has emerged as a significant concern in the industry. Consequently, the recycling and reuse of such waste have become a prominent research focus. In this study, focusing on achieving direct recycling and reuse of DWSSP, the key driving factors for effective purification were identified. It was found that simply increasing the melting temperature and time was insufficient to completely remove volatile impurities, and the migration process in the melt had to be taken into consideration. Additionally, this study focused on analyzing the instability model of inter-granular grain growth and its impact on the stable migration of impurities, with particular attention to the microstructure of inter-granular micro-regions. The thermal control directional solidification technique focused on controlling the temperature gradient during the melt solidification process. This approach helped stabilize the microstructure, enhance impurity migration, and ultimately led to a more effective purification of DWSSP.

Previous studies have explored the effects of time poverty and money worship on intertemporal decision making based on a resource scarcity perspective. However, how the pace of life affects intertemporal decision making has not been examined. Furthermore, manipulating time perceptions can influence intertemporal decision-making preferences. Based on the perspective of time perception differences, it remains unknown how views of time or temporal focus affect the intertemporal decision making of individuals with different pace of life. To address these issues, study 1 adopted a correlational study to initially explore the relationship between the pace of life and intertemporal decision making. Studies 2 and 3 used manipulation experiments to examine the effects of the pace of life and view of time and temporal focus and pace of life on intertemporal decision making. The results suggest that the faster the life pace, the more recent rewards are preferred. Views of time and temporal focus manipulations can influence the intertemporal decision making of faster-paced individuals, making them prefer smaller-sooner (SS) payoffs under a linear view of time or future temporal focus and larger-later (LL) payoffs under a circular view of time or past temporal focus. However, the manipulation does not affect the intertemporal decision of slower-paced individuals. Our study examined the effect of the pace of life on intertemporal decision making based on a resource scarcity perspective, and found boundary conditions for the influence of the view of time and temporal focus on intertemporal decision making based on the perspective of differences in people's perception of time.

Background: Vaccination is effective to prevent hepatitis B virus (HBV) infection. However, there is still a risk of infection after vaccination. In clinical work, we found that newborns were positive for HBV surface antigen (HBsAg) after vaccination. Objectives: To determine the effect of hepatitis B vaccination on the detection of HBsAg trend in newborns. Methods: We collected data at birth, history of vaccination for hepatitis B, quantitative HBsAg results, and other information about newborns born in our hospital from July 2017 to July 2020. Serum samples from healthy neonates were randomly selected to be supplemented with recombinant hepatitis B vaccine on a concentration gradient, and HBsAg was measured quantitatively. Results: Data from 1417 neonates were included in the study; 306 (21.6%) were HBsAg positive within 8 d after vaccination, with levels ranging from 0.104 IU/mL to 0.339 IU/mL. The proportion of neonates with HBsAg-positive serum was significantly correlated with the level of hepatitis B surface antibodies (anti-HBs) in the serum of their mothers (P < 0.01). Experiments in vitro showed that the proportion of neonates with HBsAg-positive serum was correlated with the dose of the hepatitis B vaccine, and when the concentration of the hepatitis B vaccine reached 5 ng/mL and 10 ng/mL, the serum HBsAg levels showed a significant negative correlation with the original concentration of serum anti-HBs. Conclusions: Hepatitis B vaccination can affect the level of HBsAg detected in neonatal serum, and the effect could be mitigated by delaying the measurement. Moreover, maternal anti-HBs offset the effects of neonatal vaccination on HBsAg serum levels.

Efficient and noble metal-free co-catalyst loading is an effective solution for separating and transferring photo-generated carriers and lowering the overpotential in photocatalytic H2 evolution activity. In this work, we designed and prepared a series of novel NiCoP/Mn0.3Cd0.7S (NCP/MCS) composites by modifying MCS nanorods with the co-catalyst NCP using a simple calcination method. Notably, the 10-NCP/MCS composite displays the optimum photocatalytic H2 evolution rate of 118.5 mmol g-1 h-1 under visible-light irradiation. This is approximately 3.39 times higher than that of pure MCS. The corresponding apparent quantum efficiency is 10.2% at 420 nm. The superior photocatalytic activity of the NCP/MCS composites can be attributed to the efficient separation of photogenerated carriers caused by the intimate heterojunction interface between NCP and MCS, smaller transfer resistance, and lower overpotential of NCP. Moreover, the NCP/MCS composites exhibit remarkable photostability. A plausible mechanism is proposed.

OBJECTIVE: To investigate the influence of storage time and temperature on plasma insulin levels and to establish a correction formula. METHODS: Venous blood samples were taken from 20 volunteers and processed as follows: whole blood samples, centrifuged samples, and separated plasma samples were stored at 4°C or 25°C. Insulin levels were determined by direct chemiluminescence at 0, 0.5, 1, 2, 4, and 8 hours. According to the correlation between the insulin concentration ratio and storage time, correction formulas for the insulin concentration were established. To verify the test, the venous blood samples of another 33 volunteers were processed in the same way. The insulin levels of the samples were corrected after 3, 6, 12, and 24 hours and compared with the value at 0 hours to verify the feasibility of the corrected formula. RESULTS: With the prolongation of storage time, the insulin levels of the whole blood samples at 4°C or 25°C and of the centrifuged samples at 25°C decreased gradually (P < .001), and the insulin level correction formulas were Ccorrection  = Cdetermination /0.991e-0.069 x , Ccorrection  = Cdetermination /1.048e-0.126 x , and Ccorrection  = Cdetermination /[-0.068ln(x) + 0.9242]. There was no significant difference between the corrected insulin results and the original results at any time within 12 hours (P > .05). CONCLUSIONS: The insulin levels of the whole blood samples at 4°C or 25°C and of the plasma samples at 25°C gradually decreased with storage time. The effect of storage time on the insulin level can be reduced with the correction formulas.

RESUMO

An efficient oxydifluoroalkylation of hydroxyl-containing alkenes using a copper catalytic system has been developed. This reaction proceeded through a one-pot process of difluoroalkylation followed by nucleophilic attack of the appended hydroxyl group. This strategy has the advantages of a low-cost catalyst and broad substrate scope, which provides a facile access to various fluoroalkylated tetrahydrofurans and tetrahydropyrans.

RESUMO

An efficient and facile method for the construction of tri- or tetrasubstituted exocyclic alkenes is achieved via a Cu(I)-catalytic system. This protocol exhibits mild conditions, low-cost catalyst, good functional group tolerance, and good yields. The selectivity toward tri- or tetrasubstituted alkenes can be delicately controlled by adjustment of base and solvent. A preliminary mechanism study manifested that the reaction undergoes a radical process, where B2pin2 plays an indispensable role.

A nitridation treatment technology with a urea/ammonia complex nitrogen source improved resistive switching property in HfO2-based resistive random access memory (RRAM). The nitridation treatment produced a high performance and reliable device which results in superior endurance (more than 109 cycles) and a self-compliance effect. Thus, the current conduction mechanism changed due to defect passivation by nitrogen atoms in the HfO2 thin film. At a high resistance state (HRS), it transferred to Schottky emission from Poole-Frenkel in HfO2-based RRAM. At low resistance state (LRS), the current conduction mechanism was space charge limited current (SCLC) after the nitridation treatment, which suggests that the nitrogen atoms form Hf-N-Ox vacancy clusters (Vo+) which limit electron movement through the switching layer.

The objective of this study was to explore a novel method to alter the sex-ratio balance of mouse offspring by silencing the paralogous genes Zfx/Zfy (Zinc finger X/Y-chromosomal transcription factor gene) during spermatogenesis. Four recombined vectors PRZ1, PRZ2, PRZ3, and PRZ4 (RNAi-Ready-pSIREN-RetroQ-ZsGreen) were constructed for interrupting the Zfx gene. Additionally, a recombined vector Psilencer/Zfy-shRNA was constructed for interrupting the Zfy gene. Male mice were randomly divided into 8 groups, with 20 animals per group. Five groups of mice were injected with PRZ1, PRZ2, PRZ3, PRZ4, and Psilencer/Zfy-shRNA vectors, respectively. The three control groups were injected with an equal volume of physiological saline, empty RNAi-Ready-pSIREN-RetroQ-ZsGreen vector, and empty Psilencer/Zfy-shRNA vector, respectively. All groups were injected every 7 days for a total of four injections. Fourteen days after the fourth injection, 10 male mice from each group were mated individually with 10 females. Testicular tissue of 10 male mice in each group was collected, and the expression level of Zfx/Zfy mRNA was determined by qRT-PCR. Results showed that, compared with the empty RNAi-Ready-pSIREN-RetroQ-ZsGreen vector and the physiological saline group, expression of Zfx mRNA decreased significantly after injection of PRZ1 (p < 0.01), PRZ3 (p < 0.01), and PRZ4 (p < 0.01), and 78.75 ± 7.50% of the offspring were male in PRZ4 group, significantly higher than the offspring derived from the empty RNAi-Ready-pSIREN-RetroQ-ZsGreen vector and physiological saline group (p < 0.01). In the PRZ1 group, the expression of Zfx mRNA was also significantly lower (p < 0.01), but the male rate of offspring was not different (p > 0.05). Conversely, the expression of Zfy mRNA decreased significantly after injection of Psilencer/Zfy-shRNA (p < 0.01) and 31.00 ± 11.00% of the offspring were male, significantly lower than in the physiological saline group (p < 0.01). In conclusion, our findings show that RNAi-mediated disruption of Zfx/Zfy in mouse testis affected X/Y spermatogenesis. Additionally, results suggest that the paralogous genes Zfx/Zfy play an important role in the process of X and Y sperm development. The individual interference of Zfx/Zfy may predict the outcome of X and Y haploid sperms. Presented herein is an advanced method developed to control mouse X/Y spermatogenesis and sex ratio of offspring.
