AIMS: It has been reported that allopregnanolone (APα) promotes the neurogenesis of the neural progenitor cells (NPCs) in the subventricular zone (SVZ) and prevents the decrease of dopaminergic neurons in 6-hydroxydopamine (6-OHDA)-treated mice by binding to γ-aminobutyric acid A receptor (GABAAR) and then opening voltage-gated L-type Ca2+ channel, but the underlying mechanisms remain elusive. The aim of this study was to explore the possible involvement of GABAAR and calcium/calmodulin-dependent protein kinase II delta 3 (CaMKIIδ3) in this process. METHODS: 6-OHDA-treated mice and primary cultured midbrain cells were administrated with APα and GABAAR antagonist bicuculline (Bic), and the proliferation and differentiation of NPCs, the tyrosine hydroxylase (TH)-positive neurons and their fibers, the expression levels of CaMKIIδ3 and brain-derived neurotrophic factor (BDNF), and motor functions were measured using ELISA, immunohistochemical staining, real-time RT-PCR, Western blot, and behavioral test. RESULTS: Allopregnanolone significantly promoted the phosphorylation of cytoplasmic CaMKIIδ3 and its nuclear translocation by binding to GABAAR, which, in turn, increased the expression levels of BDNF. This may account for the findings that the exogenous APα enhanced the proliferation and differentiation of NPCs, and ameliorated the nigrostriatal system and behavioral performance in 6-OHDA-treated mice. CONCLUSIONS: Allopregnanolone may directly activate GABAAR, which, in turn, enhance the proliferation and differentiation of NPCs via upregulating the expression levels of CaMKIIδ3, and finally contribute to the restoration of dopaminergic neurons in 6-OHDA-treated mice.

Pesticides are among the most widespread organic contaminants in aquatic environments. In this work, a new green fluorescence application was proposed for the simultaneous determination of four widely employed pesticides in environmental water samples. To overcome the highly overlapped spectra within the analytes, and with the tissue matrix interferences in complex solutions, we have used the multivariate calibration methods such as parallel factor analysis (PARAFAC) and unfolded partial least squares coupled to residual bilinearization (U-PLS/RBL). These four pesticides can be identified simultaneously, and the correlation coefficients between resolved and actual spectra are all above 0.95. The second-order advantage allowed the determination of four pesticides at the ng mL-1 level, even in the presence of humic acid (HA). The best results were obtained with the limits of detection of 1.72-18.69 for Carbendazim (CBZ), 0.30-5.19 for carbaryl (CAR), 0.35-6.32 for chlorothalonil (CHL), and 4.92-29.96 for tsumacide (TSU) (ng mL-1), which can fully meet the quantitative detection and analysis requirements of trace pesticides in water samples. The real water sample of Bohai Seawater was used to check the performance of this approach in practical applications, which have achieved good prediction results of U-PLS/RBL. This study demonstrated the proposed method is rapid, accurate, sensitive, low detection limit, and environmentally friendly to determinate multi-pesticide residues in environmental water samples.

A novel fluorescence application for simultaneous determination of two common fungicide pesticides (carbendazim and chlorothalonil) in peanut oil is presented. Using the strategy of combining excitation-emission matrix (EEM) fluorescence with three-way calibration methods, the proposed approach successfully achieved quantitative analysis of carbendazim and chlorothalonil pesticide residues in peanut oil, even with highly overlapped spectra. It needs little preparation, using "mathematical separation" instead of "analytical separation" to achieve concentration prediction of target analytes in complex systems. Each analyte was performed using fluorescence spectroscopy after instrument spectral correction and scatter removal. Then the data were modeled with two three-way calibration algorithms, including alternating trilinear decomposition (ATLD) and alternating penalty trilinear decomposition (APTLD). The results indicated that APTLD performed slightly better than ATLD for this system. The carbendazim and chlorothalonil can be recognized simultaneously with the correlation coefficients all above 0.96 between resolved spectra and actual spectra. Satisfactory results have been achieved with the average recoveries (mean ±â€¯standard deviation) of carbendazim and chlorothalonil being 100.2 ±â€¯6.7% and 99.7 ±â€¯6.7%, respectively. Moreover, as for carbendazim and chlorothalonil, the sensitivity (SENs) are 1.50 × 102 and 3.80 × 102 mL ng-1, the limits of detection (LODs) are 11 ng mL-1 and 4.3 ng mL-1, the limit of quantitation (LOQ) are 33.33 ng mL-1 and 13.03 ng mL-1, respectively. The above results demonstrated that the proposed method is sensitive, fast and accurate for direct quantitative analysis of multiple pesticide residues in complex matrix such as that of peanut oil.

As an essential part of programmed cell death, pyroptosis is an inflammatory response that is elicited upon infection by intracellular pathogens. Metabolic diseases, atherosclerosis and vital organ damage occur if pyroptosis is over-activated. Macrophages are the main cells that induce pyroptosis with the help of intracellular pattern-recognition receptors stimulated by danger signals and pathogenic microorganisms in the cytosol of host cells. Activated inflammatory caspases induce pyroptosis and produce pro-inflammatory cytokines, such as interleukin-1ß and interleukin-18. Inflammatory programmed cell death is classified as canonical or non-canonical based on inflammatory caspases, which includes caspase-1 (in human and mouse) and caspase-11 (in mouse) or caspase-4 and -5 (in humans). Activated inflammatory caspases cleave the pore-forming effector protein, gasdermin-D, inducing osmotic pressure deregulation of internal fluids and subsequently rupturing the cell membranes. Inflammatory caspases could be attractive therapeutic targets for inflammatory bowel disease (IBD) in which pyroptosis may play an important role. This article reviews the current understanding of the mechanism of pyroptosis, focusing on the regulation of inflammatory caspases and therapeutic strategies for IBD.

The content of elements in fifteen different regions of Nitraria roborowskii samples were determined by inductively coupled plasma-atomic emission spectrometry(ICP-OES), and its elemental characteristics were analyzed by principal component analysis. The results indicated that 18 mineral elements were detected in N. roborowskii of which V cannot be detected. In addition, contents of Na, K and Ca showed high concentration. Ti showed maximum content variance, while K is minimum. Four principal components were gained from the original data. The cumulative variance contribution rate is 81.542% and the variance contribution of the first principal component was 44.997%, indicating that Cr, Fe, P and Ca were the characteristic elements of N. roborowskii.Thus, the established method was simple, precise and can be used for determination of mineral elements in N.roborowskii Kom. fruits. The elemental distribution characteristics among N.roborowskii fruits are related to geographical origins which were clearly revealed by PCA. All the results will provide good basis for comprehensive utilization of N.roborowskii.

Polygoni Multiflori Radix is the dried root of Polygonum multiflorum Thunb officially recorded in the Chinese Pharmacopoeia as HeShouWu (HSW) in Chinese pinyin. The processed HSW are commonly used in TCM to treat liver disease and Chinese Pharmacopoeia has described the actions of it to tonify liver-kidney, replenish essence and blood, blacken beard and hair, strengthen sinew and bone, and resolve turbidity and lower lipid hence making it use not only as a herbal medicine in TCM but also as supplementary food in health care. AIM OF THE STUDY: Concerns about the hepatotoxicity in association with Polygoni Multiflori Radix and its processed products have been reported in some countries. In the present study, we aim to investigate the potential hepatotoxicity of HSW in rats with oral administration of 95% ethanol-extracts of Polygoni Multiflori Radix by using metabolomics method. MATERIALS AND METHODS: Here, male rats with 150-180g body weight were received vehicle control or Polygoni Multiflori Radix extracts (HSW-Ex) orally at 19.2 (low dose), 192 (medium dose), or 1920mg/kg/day (high dose), respectively, for 28 consecutive days. Signs of HSW-induced toxicity were monitored by traditional toxicity assessments (e.g., clinical pathology and histopathology). Metabolomics investigation of serum was performed to identify potential endogenous metabolites which may be relevant to liver injury. RESULTS: Rats received High and Medium dose of HSW-Ex showed greater sign of liver injury with increased levels of ASP, ALT, and AST, as well as reduced SOD activity when compared to vehicle control. In contrast, there are no significant changes relevant to liver injury observed in rats by receiving the low dose of HSW-Ex. Metabolomics analyses have identified ten potential endogenous metabolites varied significantly among the treatment groups with varying doses of HSW-Ex, of which might be related to liver injury. CONCLUSION: Our data has further suggested that liver damage resulting from HSW-Ex consumption is dosage dependent in rats. It is possible that disruption in amino acid and energy metabolism might lead to subsequent oxidative damage in the liver of rats. Because the clinic practice often use low dose in a short time, therefore HSW usage in TCM still keep safe currently, but we present a warning to the clinical doctors and make them has some concern about high dose of HSW usage in a long term that has potential danger to damage liver.

Esophageal squamous cell carcinoma (ESCC) is one of the most common types of malignancies in China. Most ESCC patients are diagnosed at middle to late stages with poor prognosis due to the lack of an effective method for early diagnosis. MicroRNAs (miRNAs) are a family of endogenous small non-coding RNAs that can regulate ESCC development and progression by repressing their specific target genes' expression. Compared to traditional biomarkers (e.g., mRNAs and proteins), miRNAs are more stable and can be readily screened and accurately quantitated and analyzed, making them ideal new-generation of biomarkers for early cancer detection and prognostic evaluation. Recent studies have shown that the changes of the expression levels of some serum miRNAs from ESCC patients significantly correlate with their diagnostic and prognostic outcome. In this review, we summarize the trend of the expression changes of miRNAs in ESCC patients' serum and discuss the possibility of detecting these miRNAs' expression changes as a novel method for ESCC early diagnosis and prognostic evaluation. Notably, the results of serum miRNAs from different detection methods are not completely consistent. Thus, we also discuss several possible reasons for such inconsistency.

SDAU18, an amphiploid of Ae.ventricosa with Ae.cylindrica, was identified by cytological analysis, seed storage protein electrophoresis, genomic in situ hybridization (GISH) and inoculation assessment. The results are as follows: The chromosome number of root tip cells (RTCs) of SDAU18 plants varied from 52 to 56. 28 bivalents were observed in most PMCs MI of SDAU18 with 56 chromosomes, meanwhile, a few univalents, multivalents also existed in some PMCs MI, and the average chromosome configuration was 2n = 56 = 3.21 I +19.78 II, (Ring)+6.50 II (Rod)+0.01 III +0.04 IV (Ring)R+0.01 IV (Rod). There were both Ae. ventricosa-specific bands and Ae. cylindrica-specific bands in the seed storage protein electrophoretogram of SDAU18, furthermore, SDAU18 had one novel HMW-GS not found in the parents and two novel ones not found in common wheats. By labeling the total genomic DNA of Ae. ventricosa and Ae. cylindrica as probes respectively, and using that of another parent as block, GISH of RTCs spread of SDAU18 was carried out. The green hybridization signal was observed in 14 chromosomes respectively, within 56 ones in RTCs of SDAU18. SDAU18 was immune to powdery mildew and stripe rusts. SDAU18 was an amphiploid of Ae. ventricosa with Ae. cylindrica, and had very important significance in wheat breeding and genetic improvement.
