RESUMO

The plant Carpobrotus edulis has traditionally been known for its wide applications in diseases, especially vitiligo, which is characterized by patches and white macules caused by the loss of melanocytes. One of the chemical treatments for vitiligo consists mainly of skin repigmentation and usually leads to a non-durable effect by inhibiting the Janus kinase (JAK) signal transduction (STAT pathway). JAK inhibitors generally block multiple JAK tyrosine kinases, which leads to secondary effects. In this study, natural molecules from Carpobrotus edulis were extracted and tested using a structure-based drug-design approach and pharmacophore modeling. The best-fit candidate from the extracted molecules was compared to the chemical molecules used. The results indicated a similarity between the chemical and natural ligands which suggested the potential use of the natural product against vitiligo. The main finding of this research work was the discovery of a new molecule extracted from a natural plant and the detection of its anti-vitiligo activity using an in-silico approach. This method can significantly reduce the cost of searching for potential medicinal molecules.

Microplastics (MPs) are emerging pollutants of global concern due to their pervasiveness, high sorption ability for persistent organic pollutants (POPs) and direct and indirect toxicity to marine organisms, ecosystems, as well as humans. As one of the major coastal interfaces, beaches are considered among the most affected ecosystems by MPs pollution. The morphological characteristics of MPs (pellets and fragments) collected from four beaches along the Tunisian coast and sorbed POPs, including polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs), were investigated in this study. The results showed that the MPs varied greatly in color, polymer composition and degradation degree. The color varied from colored to transparent and the most prevalent polymer identified using Raman spectroscopy was polyethylene. Scanning electron microscope (SEM) images exhibited various surface degradation features including cavities, cracks, attached diatom remains, etc. The concentrations of Σ12PCBs over all beaches ranged from 14 to 632 ng g-1 and 26 to 112 ng g-1 in the pellets and fragments, respectively, with a notable presence and dominance of highly-chlorinated PCBs such as CB-153 and -138. Among the OCPs, γ-HCH is the only compound detected with concentrations ranging from 0.4 to 9.7 ng g-1 and 0.7 to 4.2 ng g-1 in the pellets and fragments, respectively. Our findings indicate that MPs found on the Tunisian coast may pose a chemical risk to marine organisms as the concentrations of PCBs and γ-HCH in most of the analysed samples exceeded the sediment-quality guidelines (SQG), especially the effects range medium (ERM) and the probable effects level (PEL). As the first report of its kind, the information gathered in this study can serve as the baseline and starting point for future monitoring work for Tunisia and neighbouring countries, as well as for stakeholders and coastal managers in decision-making processes.

We investigated the occurrence, origin, and potential ecological risk of dissolved polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyl (PCBs) and organochlorine pesticides (OCPs) in 27 surface water samples collected from a highly anthropized and industrialized area in the Gulf of Gabès (Tunisia, Southern Mediterranean Sea) in October-November 2017. The results demonstrated a wide range of concentrations (ng L-1) with the following decreasing order: Æ©16 PAHs (17.6-71.2) > Æ©20 PCBs (2.9-33.7) > Æ©6 DDTs (1.1-12.1) > Æ©4 HCHs (1.1-14.8). Selected diagnostic ratios indicated a mixture of both pyrolytic and petrogenic sources of PAHs, with a predominance of petrogenic sources. PCB compositions showed distinct contamination signatures for tetra- to hepta-chlorinated PCBs, characteristic of contamination by commercial (Aroclor) PCB mixtures. The dominant OCP congeners were γ-HCH, 2,4'-DDD and 2,4'-DDE, reflecting past use of Lindane and DDTs in the study area. Agricultural, industrial and domestic activities, as well as atmospheric transport are identified as potential sources of PAHs, PCBs and OCPs in surface waters of the Gulf of Gabès. Toxic equivalents (TEQs) suggested a low carcinogenic potential for PAHs in seawater samples (mean of 0.14 ng TEQ L-1). Evaluation of risk coefficients revealed low risk for PAHs and PCBs, and moderate to severe risk for OCPs.

Microbial production of bioplastics, derived from poly(3-hydroxybutyrate) (PHB), have provided a promising alternative towards plastic pollution. Compared to other extremophiles, halophilic archaea are considered as cell factories for PHB production by using renewable, inexpensive carbon sources, thus decreasing the fermentation cost. This study is aimed at screening 33 halophilic archaea isolated from three enrichment cultures from Tunisian hypersaline lake, Chott El Jerid, using starch as the sole carbon source by Nile Red/Sudan Black staining and further confirmed by PCR amplification of phaC and phaE polymerase genes. 14 isolates have been recognized as positive candidates for PHA production and detected during both seasons. The identification of these strains through 16S rRNA gene analyses showed their affiliation to Halorubrum, Natrinema, and Haloarcula genera. Among them, three PHB-producing strains, CEJ34-14, CEJ5-14, and CEJ48-10, related to Halorubrum chaoviator, Natrinema pallidum, and Haloarcula tradensis were found to be the best ones reaching values of 9.25, 7.11, and 1.42% of cell dry weight (CDW), respectively. Our findings highlighted that Halorubrum, Natrinema, and Haloarcula genera were promising candidates for PHB production using soluble starch as a carbon source under high salinity (250 g L-1 NaCl).

We investigated the efficiency of a benthic diatom-associated bacteria in removing benzo(a)pyrene (BaP) and fluoranthene (Flt). The diatom, isolated from a PAH-contaminated sediment of the Bizerte Lagoon (Tunisia), was exposed in axenic and non-axenic cultures to PAHs over 7 days. The diversity of the associated bacteria, both attached (AB) and free-living bacteria (FB), was analyzed by the 16S rRNA amplicon sequencing. The diatom, which maintained continuous growth under PAH treatments, was able to accumulate BaP and Flt, with different efficiencies between axenic and non-axenic cultures. Biodegradation, which constituted the main process for PAH elimination, was enhanced in the presence of bacteria, indicating the co-metabolic synergy of microalgae and associated bacteria in removing BaP and Flt. Diatom and bacteria showed different capacities in the degradation of BaP and Flt. Nitzschia sp. harbored bacterial communities with a distinct composition between attached and free-living bacteria. The AB fraction exhibited higher diversity and abundance relative to FB, while the FB fraction contained genera with the known ability of PAH degradation, such as Marivita, Erythrobacter, and Alcaligenes. Moreover, strains of Staphylococcus and Micrococcus, isolated from the FB community, showed the capacity to grow in the presence of crude oil. These results suggest that a "benthic Nitzschia sp.-associated hydrocarbon-degrading bacteria" consortium can be applied in the bioremediation of PAH-contaminated sites.

Both industrial effluent discharge and the resuspension of contaminated marine sediments are important sources of trace metals in seawater which potentially affect marine ecosystems. The aim of this study was to evaluate the impact of the industrial wastewaters having acidic pH (2-3) and containing trace metals on microbial diversity in the coastal ecosystem of the Gulf of Gabès (Tunisia, southern Mediterranean Sea) subjected to resuspension events of marine sediments. Four trace elements (As, Cd, U, and V) were monitored during 10-day sediment resuspension experiments. The highest enrichment in the seawater dissolved phase was observed for Cd followed by U, V, and As. Cd remobilization was improved by indigenous microbial community, while U release was mainly abiotic. Acidic effluent addition impacted both trace metal distribution and microbial diversity, particularly that of the abundant phylum Bacteroidetes. Members of the order Saprospirales were enriched from sediment in natural seawater (initial pH > 8), while the family Flavobacteriaceae was favored by acidified seawater (initial pH < 8). Some Flavobacteriaceae members were identified as dominant species in both initial sediment and experiments with acidic wastewater, in which their relative abundance increased with increasing dissolved Cd levels. It could be therefore possible to consider them as bioindicators of metal pollution and/or acidification in marine ecosystems.

Particulate-bound aliphatic and polycyclic aromatic hydrocarbons (AHs and PAHs) were investigated in the surface waters of the Gulf of Gabès (Tunisia, southern Mediterranean Sea). Samples were collected off the Sfax and Gabès-Ghannouch coasts. Concentrations in total resolved n-alkanes ranged from 0.03 to 3.2 µg L-1, and concentrations in total parents + alkylated PAHs ranged from bdl to 108.6 ng L-1. The highest concentrations were recorded in the southern Sfax. AHs were mainly of biogenic origin with odd n-alkane predominance, although an anthropogenic contribution was also detected. The PAH molecular patterns revealed a mixed origin with the presence of low molecular weight and alkylated compounds, characteristic of uncombusted oil-derived products, and the presence of high molecular weight compounds, typical of combustion residues. Rainfall events induced an increase in PAH concentrations by a factor 1.5-23.5. The particle-water partition coefficients (Koc) suggest that the partitioning of PAHs between the particulate and dissolved phases is driven by hydrophobicity and organic matter composition.

We investigated the origin and distribution of aliphatic and polycyclic aromatic hydrocarbons (AHs and PAHs) and organic matter (OM) in surficial sediments of the Sfax-Kerkennah channel in the Gulf of Gabès (Tunisia, Southern Mediterranean Sea). TOC, AH and PAH concentrations ranged 2.3-11.7%, 8-174µgg-1sed.dw and 175-10,769ngg-1sed.dw, respectively. The lowest concentrations were recorded in the channel (medium sand sediment) and the highest ones in the Sfax harbor (very fine sand sediment). AHs, PAHs and TOC were not correlated for most of the stations. TOC/N and δ13C values revealed a mixed origin of OM with both marine and terrestrial sources. Hydrocarbon molecular composition highlighted the dominance of petrogenic AHs and the presence of both petrogenic and pyrogenic PAHs, associated with petroleum products and combustion processes. This work underscores the complex distribution patterns and the multiple sources of OM and hydrocarbons in this highly anthropogenized coastal environment.

A full-scale study has been conducted to assess the bioaugmentation efficiency of trickling filter process to treat petrochemical wastewater from a lubricant industry recycling waste oils. During 45 weeks, the organic loading rate (OLR) in the trickling filter was increased stepwise from 0.9 to 4 kg of chemical oxygen demand (COD)/(m3·day) at the end of the upgrading period as the flow rate (FR) reached the value of 30 m3/day. The removal, obtained in terms of percentage, for COD ranged from 60 to 84.5 and greater than 98 for total n-alkane (TNA), while those of total kjeldahl nitrogen (TKN) and total phosphor (TP) were about 32 and 55, respectively. The analytical profile index (API) of trickling biofilm has confirmed that 5 strains are closely related to Acinobacter junii, Stenotrophomonas maltophilia, Vibrio vulnificus, Vibrio metschnikovi, Pseudomona slulzeri and Trichosporon spp2.

The Semi-enclosed Mediterranean Sea records various signals of high anthropic pressures from surrounding countries and the industrialized European countries. This is particularly true for oil pollution. Although accounting for 1% of the world's ocean surface, it receives about 25% of the petroleum inputs to the ocean. To achieve a global budget we need to collect information from different parts of the Mediterranean. Particularly, we focus in this paper on the Southern Mediterranean, where data are presently very scarce. In this context, the University of Sfax has undertaken an estimation of hydrocarbon pollution along the coasts of Sfax and Gabès Gulf. Non-aromatic hydrocarbons were analysed in 8 surface sediments by FT/IR and GC/MS. Non-aromatic hydrocarbon concentrations vary in the range 310-1406 microg g(-1) sediments dry weight, which is high, compared to other Mediterranean sites. GC/MS data indicate a large group of unresolved compounds suggesting a petroleum contamination, confirmed by the identification of hopanes with predominant C29 and C30alpha,beta compounds and steranes with predominance of C27 over C28) and C29 compounds.
