OBJECTIVES: To explore the risk factors associated with cow's milk protein allergy (CMPA) in infants. METHODS: This study was a multicenter prospective nested case-control study conducted in seven medical centers in Beijing, China. Infants aged 0-12 months were included, with 200 cases of CMPA infants and 799 control infants without CMPA. Univariate and multivariate logistic regression analyses were used to investigate the risk factors for the occurrence of CMPA. RESULTS: Univariate logistic regression analysis showed that preterm birth, low birth weight, birth from the first pregnancy, firstborn, spring birth, summer birth, mixed/artificial feeding, and parental history of allergic diseases were associated with an increased risk of CMPA in infants (P<0.05). Multivariate logistic regression analysis revealed that firstborn (OR=1.89, 95%CI: 1.14-3.13), spring birth (OR=3.42, 95%CI: 1.70-6.58), summer birth (OR=2.29, 95%CI: 1.22-4.27), mixed/artificial feeding (OR=1.57, 95%CI: 1.10-2.26), parental history of allergies (OR=2.13, 95%CI: 1.51-3.02), and both parents having allergies (OR=3.15, 95%CI: 1.78-5.56) were risk factors for CMPA in infants (P<0.05). CONCLUSIONS: Firstborn, spring birth, summer birth, mixed/artificial feeding, and a family history of allergies are associated with an increased risk of CMPA in infants.

Prussian blue analogues (PBAs) are regarded as promising cathode materials for ammonium-ion batteries (AIBs) because of their low cost and superb theoretical capacity. However, its inherently poor conductivity and structural collapse can significantly limit the enhancement of rate property and cycling stability. In this work, Berlin Green (BG) electrode materials with similar wool-like clusters were constructed by direct precipitation method to accelerate the kinetic, which realizes outstanding cycling stability. Berlin Green with the appropriate amount of iron (BG-2) has a fast ion transport channel, enhanced structure stability, highly reversible insertion/extraction of NH4+, and fine electrochemical reaction activity. Benefiting from the unique architecture and component, the BG-2 electrode shows an excellent rate performance with a discharge/charge specific capacity of 60.1/59.3 mAh g-1 at 5 A g-1. Even at 5 A g-1, BG-2 exhibits remarkable cycling stability with an initial discharge capacity of 59.5 mAh g-1 and a capacity retention rate of approximately 76% after 30,000 cycles. The BG-2 reveals exceedingly good electrochemical reversibility during the process of NH4+ (de)insertion. BG materials indicate huge potential as a cathode material for the next generation of high-performance aqueous batteries.

Constitutive activation of Hedgehog (Hh) signaling has been implicated in many cancers including hepatocellular carcinoma (HCC). Among them, the terminal glioma-associated oncogene homolog 1 (Gli1) regulates the expression of critical genes in the Hh pathway. The current study aims to evaluate the anti-HCC effect of the Gli1 inhibitor, GANT61. In vitro analysis including cell counting kit-8 (CCK-8) assay, flow cytometry, and migration and invasion assay were adopted to evaluate the effect of GANT61 on HCC cell lines. In vivo, xenograft studies were also performed to verify the effect of GANT61 on HCC. By CCK-8 assay, we found that GANT61 could significantly reduce the growth of HCC cell lines Huh7 and hemophagocytic lymphohistiocytosis (HLE), and their IC50 concentrations were 4.481 and 6.734 µM, respectively. Flow cytometry shows that GANT61 induced cell cycle arrest in the G2/M phase and accelerated apoptosis of both HLE and Huh7 cells. While migration and invasion assay shows that GANT61 weakens cells' migration and invasion ability. Besides that, GANT61 inhibits the expression of Gli1, FoxM1, CyclinD1, and Bcl-2, upregulates the level of Bax protein, and also reverses the epithelial-mesenchymal transition program by downregulating the expression of Vimentin and N-Cadherin and upregulating the expression of epithelial E-Cadherin expression. Furthermore, GANT61 inhibits the growth of subcutaneous xenografts of Huh7 cells in nude mice. Overall, this study suggests that Gli1 is a potential target for therapy and GANT61 shows promising therapeutic potential for future treatment in HCC.

Introduction: Neuronal surface antibody syndromes (NSAS) encompass a growing set of autoimmune neurological disorders, with their predominant clinical presentation being autoimmune encephalitis (AE). The most extensively documented form within NSAS is anti-N-methyl-D-aspartate receptor (NMDAR) autoimmunity. In contrast, other NSAS, such as anti-metabotropic glutamate receptor-5 (mGluR5) autoimmunity, are less common and less comprehensively characterized, particularly in pediatric cases. Case description: In this instance, we present the case of a 7-year-old girl who exhibited abnormal behaviors following hematopoietic stem cell transplantation (HSCT). She received a diagnosis of anti-mGluR5 AE, and her Electroencephalogram (EEG) displayed an increased number of generalized slow waves during wakefulness. Treatment involved intravenous administration of gamma globulin and methylprednisolone, followed by oral prednisone tablets. Levetiracetam was introduced as an antiepileptic therapy during the pulse steroid therapy. Notably, the abnormal behaviors exhibited significant improvement after treatment. Conclusions: To the best of our knowledge, this is the first report of rare pediatric NSAS involving anti-mGluR5 AE following HSCT. Enhancing our understanding and characterization of this condition may facilitate its recognition and treatment in children. Serum antibody testing could enable early identification and treatment of anti-mGluR5 AE.

As short cantilever members, corbels are mainly used to transfer eccentric loads to columns. Because of the discontinuity of load and geometric structure, corbels cannot be analyzed and designed using the method based on beam theory. Nine steel-fiber-reinforced high-strength concrete (SFRHSC) corbels were tested. The width of the corbels was 200 mm, the cross-section height of the corbel column was 450 mm, and the cantilever end height was 200 mm. The shear span/depth ratios considered were 0.2, 0.3, and 0.4; the longitudinal reinforcement ratios were 0.55%, 0.75%, and 0.98%; the stirrup reinforcement ratios were 0.39%, 0.52%, and 0.785%; and the steel fiber volume ratios were 0, 0.75%, and 1.5%. According to the test results, this paper discusses the failure process and failure mode of corbel specimens with a small shear span/depth ratio and analyzes the effects of variables such as shear span/depth ratio, longitudinal reinforcement ratio, stirrup reinforcement ratio, and steel fiber volume content on the shear capacity of corbels. The shear capacity of corbels is significantly affected by the shear span/depth ratio, followed by the longitudinal reinforcement ratio and the stirrup reinforcement ratio. Moreover, it is shown that steel fibers have little impact on the failure mode and ultimate load of corbels, but can enhance the crack resistance of corbels. In addition, the bearing capacities of these corbels were calculated by Chinese code GB 50010-2010 and further compared with ACI 318-19 code, EN 1992-1-1:2004 code, and CSA A23.3-19 code, which adopt the strut-and-tie model. The results indicate that the calculation results by the empirical formula in the Chinese code are close to the corresponding test results, while the calculation method based on the strut-and-tie model of a clear mechanical concept yields conservative results, and hence the related parameter values must be further modified.

BACKGROUND: Although some psychological processes, such as stigma and self-efficacy, affect the complicated relationship between social support and depressive symptoms, few studies explored a similar psychological mechanism among individuals with substance use disorders (SUDs). Hence, this research investigates the mediating effects of stigma and the moderating effects of self-efficacy among the psychological mechanism that social support affects depressive symptoms. METHODS: The study included 1040 Chinese participants with SUDs and completed a series of self-report questionnaires. R software was used to organize and clean up data sets and analyze mediation and moderation effects. RESULTS: The result showed that stigma partially mediated depressive symptoms, while self-efficacy moderated this relationship. More specifically, less social support increased depression symptoms by bringing about higher stigma. Besides, subjects with higher self-efficacy are less susceptible to stigma and therefore have mild depressive symptoms. Furthermore, clinical and theoretical implications are discussed in our study. CONCLUSIONS: Chinese SUDs patients' depressive symptoms were indirectly affected by perceived social support via stigma and less affected by stigma with improved self-efficacy. The theoretical and practical implications of these results are discussed.

Platanus fruit-like CuCo2S4 microspheres were fabricated by using a facile hydrothermal method followed by a sulfidation process. As a lithium storage material, they deliver an outstanding initial specific capacity of 1119.3 mA h g-1 at 0.05 A g-1 and a high reversibility of 954 mA h g-1 over 200 cycles even at 1 A g-1. In addition, when applied in supercapacitors they display a superb specific capacitance of 824 F g-1 at 1 A g-1, even over 10 000 cycles and they can also maintain 75% retention at 5 A g-1 and exhibit good reversibility. Furthermore, an advanced asymmetric supercapacitor (ASC) exhibits an advantageous energy density of 36.67 W h kg-1 when the power density increases up to 750 W kg-1. Additionally, the assembled device can easily light a 1.5 V bulb for several minutes. The excellent performance of CuCo2S4 is due to the bimetallic synergistic effect and the unique platanus fruit-like microsphere architecture, which can limit the restacking of the structure and provide suitable voids. This excellent performance confirms that platanus fruit-like CuCo2S4 microspheres are a promising electrode material for energy storge. This work will provide a new strategy to prepare high-performance bimetallic sulfide anode materials by a facile method.

BACKGROUND: Interdigestive migrating motor complexes (MMC) produce periodic contractions in the gastrointestinal tract, but the exact mechanism of action still remains unclear. Intramuscular interstitial cells of Cajal (ICC-IM) participate in gastrointestinal hormone and neuromodulation, but the correlation between ICC-IM and MMC is also unclear. We found that xiangbinfang granules (XBF) mediated the phase III contraction of MMC. Here, the effects of XBF on gastric antrum motility in W/W v mice and the effects of ICC-IM on gastric antrum MMC are reported. AIM: To observe the effects of ICC-IM on gastric antrum motility and to establish the mechanism of XBF in promoting gastric antrum motility. METHODS: The density of c-kit-positive ICC myenteric plexus (ICC-MP) and ICC-IM in the antral muscularis of W/W v and wild-type (WT) mice was examined by confocal microscopy. The effects of XBF on gastric antrum slow waves in W/W v and WT mice were recorded by intracellular amplification recording. Micro-strain-gauge force transducers were implanted into the gastric antrum to monitor the MMC and the effect of XBF on gastric antrum motility in conscious W/W v and WT mice. RESULTS: In the gastric antrum of W/W v mice, c-kit immunoreactivity was significantly reduced, and no ICC-IM network was observed. Spontaneous rhythmic slow waves also appeared in the antrum of W/W v mice, but the amplitude of the antrum slow wave decreased significantly in W/W v mice (22.62 ± 2.23 mV vs 2.92 ± 0.52 mV, P < 0.0001). MMCs were found in 7 of the 8 WT mice but no complete MMC cycle was found in W/W v mice. The contractile frequency and amplitude index of the gastric antrum were significantly increased in conscious WT compared to W/W v mice (frequency, 3.53 ± 0.18 cpm vs 1.28 ± 0.12 cpm; amplitude index, 23014.26 ± 1798.65 mV·20 min vs 3782.16 ± 407.13 mV·20 min; P < 0.0001). XBF depolarized smooth muscle cells of the gastric antrum in WT and W/W v mice in a dose-dependent manner. Similarly, the gastric antrum motility in WT mice was significantly increased after treatment with XBF 5 mg (P < 0.05). Atropine (0.1 mg/kg) blocked the enhancement of XBF in WT and W/W v mice completely, while tetrodotoxin (0.05 mg/kg) partially inhibited the enhancement by XBF. CONCLUSION: ICC-IM participates in the regulation of gastric antrum MMC in mice. XBF induces MMC III-like contractions that enhance gastric antrum motility via ICC-IM in mice.

This study aimed to investigate the shear capacity performance for eight deep beams with HTRB600 reinforced high strength concrete under concentrated load to enable a better understanding of the effects of shear span-depth ratio, longitudinal reinforcement ratio, vertical stirrup ratio and in order to improve design procedures. The dimension of eight test specimens is 1600 mm × 200 mm × 600 mm. The effective span to height ratio l0/h is 2.0, the shear span-depth ratio λ is 0.3, 0.6 and 0.9, respectively. In addition, the longitudinal reinforcement ratio ρs is set to 0.67%, 1.05%, 1.27%, and the vertical stirrup ratio is taken to be 0%, 0.25%, 0.33%, 0.5%. Through measuring the strain of steel bar, the strain of concrete and the deflection of mid-span, the characteristics of the full process of shear capacity, the failure mode and the load deflection deformation curve were examined. The test results showed that the failure mode of deep beams with small shear span-depth ratio is diagonal compression failure, which is influenced by the layout and quantity of web reinforcement. The diagonal compression failure could be classified into two forms: crushing-strut and diagonal splitting. With decreasing of shear span-depth ratio and increasing longitudinal reinforcement ratio, the shear capacity of deep beams increases obviously, while the influence of vertical web reinforcement ratio on shear capacity is negligible. Finally, the shear capacity of eight deep beams based on GB 50010-2010 is calculated and compared with the calculation results of ACI 318-14, EN 1992-1-1:2004 and CSA A23.3-04, which are based on strut-and-tie model. The obtained results in this paper show a very good agreement with GB50010-2010 and ACI 318-14, while the results of EN 1992-1-1:2004 and CSA A23.3-04 are approved to be conservative.

Several additives, including inorganic (NaCl) and organic salts (derivatives of benzoate), were added into aqueous solutions of a gemini cationic surfactant, 2-hydroxypropyl-1,3-bis (myristyldimethylammonium chloride) (abbreviated as 14-3(OH)-14(2Cl)). The mixed systems were investigated using rheological measurement, cryo-TEM and 1H NMR analysis. The results showed that addition of salts induced rich aggregate morphologies in the 14-3(OH)-14(2Cl)/salt systems. The influence of an inorganic salt on the viscoelasticity of 14-3(OH)-14(2Cl) solutions is much weaker than that of organic salts. Furthermore, the ability of three organic salts in enhancing the viscoelasticity of 14-3(OH)-14(2Cl) solutions is in the order sodium m-hydroxybenzoate > sodium o-hydroxybenzoate > sodium p-hydroxybenzoate. The different roles of these organic salt isomers arise from the different types of hydrogen bonding formed between 14-3(OH)-14(2Cl) and the organic counter ions.

Phoebe chekiangensis, as one original species of 'Nanmu with golden tint', harbors limited natural distribution regions, which is recognized as a vulnerable species in China under nationwide protection. Under the background of global climate change, it is of scientific significance to study the community structure and diversity of the natural populations of P. chekiangensis. In this study, community structure, species diversity and the phenotypic variation leaf traits in the communities of thirteen residue natural populations were analyzed. A total of 235 vascular species from 162 genera and 87 families were found in 16 plots within 13 natural populations, including 221 seed plants from 151 genera and 79 families. The species diversity in tree layer of community in Kaihua, Zhejiang and Linan, Zhejiang was significantly lower than that of other communities due to severe disturbance, while intermediate disturbance promoted the species diversity of shrub layer in community in Jianning, Fujian, and slight disturbance was beneficial to natural regeneration of P. chekiangensis. Foliage phenotype was important for species identification within Phoebe genus. There existed substantial variation in foliage phenotypic diversity among and within populations. The mean coefficient of variation was 17.2%, ranging from 10.4% to 27.5%. The variation was greater among populations (53.6%) than within populations (17.0%). Mean phenotypic differentiation coefficient was 75.1% among populations, indicating that the variance among populations was the main source for the phenotypic variation of the species. Results from the cluster analysis indicated that the 13 natural populations were divided into two distinct groups based on the Euclidean distance (10 cm), with stochastic variation.

As a major family of plant-specific transcription factors, SQUAMOSA PROMOTER BINDING PROTEIN-LIKE (SPL) genes play vital regulatory roles in plant growth, development and stress responses. In this study, 18 SPL genes were identified and cloned from Betula luminifera. Two zinc finger-like structures and a nuclear location signal (NLS) segments were existed in the SBP domains of all BlSPLs. Phylogenetic analysis showed that these genes were clustered into nine groups (group I-IX). The intron/exon structure and motif composition were highly conserved within the same group. 12 of the 18 BlSPLs were experimentally verified as the targets of miR156, and two cleavage sites were detected in these miR156-targeted BlSPL genes. Many putative cis-elements, associated with light, stresses and phytohormones response, were identified in the promoter regions of BlSPLs, suggesting that BlSPL genes are probably involved in important physiological processes and developmental events. Tissue-specific expression analysis showed that miR156-targeted BlSPLs exhibited a more differential expression pattern, while most miR156-nontargeted BlSPLs tended to be constitutively expressed, suggesting the distinct roles of miR156-targeted and nontargeted BlSPLs in development and growth of B. luminifera. Further expression analysis revealed that miR156-targeted BlSPLs were dramatically up-regulated with age, whereas mature BlmiR156 level was apparently declined with age, indicating that miR156/SPL module plays important roles in vegetative phase change of B. luminifera. Moreover, yeast two-hybrid assay indicated that several miR156-targeted and nontargeted BlSPLs could interact with two DELLA proteins (BlRGA and BlRGL), which suggests that certain BlSPLs take part in the GA regulated processes through protein interaction with DELLA proteins. All these results provide an important basis for further exploring the biological functions of BlSPLs in B. luminifera.

Nitrogen, an essential macronutrient for the growth and development of plants, affects above- ground biomass accumulation dramatically. Thus, it is very important to reveal the molecular mechanisms of how plants resist or adapt to low nitrogen availability. The NAC1(NAM, ATAF, CUC 1) gene, located in the upstream regulatory network, has been reported to resist low nitrogen by regulating expression of key downstream genes and thus root growth in (Populus tremula × alba).In this study, we detected the responses of miR164 and its target gene NAC1 under nitrate-starvation condition using the Betula luminifera somaclones G49-3 as material. The NAC1 gene which contains 1497 bp sequence, encodes 358 amino acids and contains a highly conserved NAM domain at N terminal was cloned by the RACE method. The NAC1 was then validated to be the target gene of miR164 via 5'-RACE, and the cleavage site was between the 10(th) and 11(th) base. The expression patterns of miR164 and its target gene NAC1 were further detected under nitrate-starvation condition through qRT-PCR analysis. The results showed that miR164 expression was repressed by nitrate-starvation at the beginning of the treatment (4 d) and then ascended. However, the expression pattern of miR164 in roots was different from that in shoots and leaves. Moreover, the expression levels of target gene NAC1 and miR164 were negatively correlated. The expression level of miR164 in root was increased while that of NAC1 was decreased under Re treatment, which indicated that miR164 and its target gene NAC1 play a regulatory role in response to low nitrate availability. The findings of our study may help elucidate the molecular mechanisms by which miR164 regulates target gene NAC1 at post-transcriptional level, and provide valuable information for further study of the regulatory roles of miR164-NAC1 under nitrate-starvation condition.

Chrysin (5,7-dihydroxylflavone, Chry) is a natural flavonoid extracted from plants and propolis. In this work, a novel chrysin-organotin (Chry-Sn) compound with enhanced anticancer activities was synthesized by the reaction of chrysin and triphenyltin chloride, and its potential anticancer effects against cancer cells were measured using various methods. Sulforhodamine B (SRB) results showed that chrysin and Chry-Sn had significant inhibition effects on the proliferation of MCF-7, A549 and HeLa human cancer cell lines in a dose- and time- dependent manner. These results suggested that Chry-Sn possessed enhanced anticancer effects. Hoechst 33258 staining and acridine orange staining results showed apoptosis and nuclei fragments significantly increased after being treated with chrysin and Chry-Sn respectively. Moreover, chrysin and Chry-Sn significantly increased ROS levels in MCF-7 cells. Western blot results showed that chrysin and Chry-Sn activated caspase 3 and induced autophagy by increasing LC3-II level. All results showed collectively that Chry-Sn could be a more promising drug than chrysin in anticancer treatment.

To study the characteristics of adsorption and desorption of Scutellaria baicalensis pieces. On the basic thermodynamic theory, thestatic method was adopted to obtain S. baicalensis pieces' isothermal adsorption and desorption data at 25, 35, 45 ℃, with the water activity between 0.10 and 0.85. Eight moisture models were selected to fit the data and then evaluated to determine the thermodynamic properties of S. baicalensis pieces. The results show that, among the eight adsorption models, Peleg fit the best, in which absolutely-safe and the relatively-safe moisture contents of S. baicalensis pieces were around 9.22% and 13.51% respectively; the net equivalent heat adsorption and desorption and the differential entropy of S. baicalensis pieces were closely related to moisture content, and decrease within crease of water content; when drying the pieces, 12.0% of moisture content can be taken as the drying end; both of adsorption and desorption processes were driven by enthalpy and non-spontaneous. The study on S. baicalensis pieces' isothermal adsorption and desorption rules can help advance studies on adsorption and desorption rules of Chinese herbal pieces, and play a good guiding role in optimizing storage conditions and drying process of Chinese herbal pieces.

A series of unexpected thermo-responsive phenomena were discovered in an aqueous solution of the cationic gemini surfactant, 2-hydroxypropyl-1,3-bis(alkyldimethylammonium chloride) (n-3(OH)-n(2Cl), n = 14, 16), in the presence of an inorganic salt. The viscosity change trend for the 14-3(OH)-14(2Cl) system was investigated in the 20-40 °C temperature range. As the temperature increased, the viscosity of the solution first decreased to a minimum point corresponding to 27 °C, and then increased until a maximum was reached, after which the viscosity decreased again. In the 16-3(OH)-16(2Cl) system, the gelling temperature (T(gel)) and viscosity changes upon heating were similar to those in the 14-3(OH)-14(2Cl) system above 27 °C. The reversible conversion of elastic hydrogel to wormlike micelles in the aqueous solution of the 16-3(OH)-16(2Cl) system in the presence of an inorganic salt was observed at relatively low temperatures. Various techniques were used to study and verify the phase-transition processes in these systems, including rheological measurements, cryogenic transmission electron microscopy (cryo-TEM), electric conductivity, and differential scanning calorimetry. The abovementioned phenomena were explained by the formation and destruction of intermolecular hydrogen bonds, and the transition mechanisms of the aggregates were analyzed accordingly.

BACKGROUND: Cyclooxygenase 1 (COX-1), COX-2, and HO-1 are involved in the process of aspirin's effect. The genetic susceptibility of these enzymes to aspirin resistance (AR) is unclear. METHODS: A total of 431 patients took aspirin. Using arachidonic acid-induced light transmittance aggregation combined with adenosine diphosphate-induced light transmittance aggregation, 36 participants served for AR, 164 participants for semi-AR, and 231 participants for aspirin sensitivity (AS). The AR with 9 single-nucleotide polymorphism in COX-1, COX-2, and HO-1 genes was investigated. RESULTS: COX-1 rs1330344 (-1676A>G) is associated with AR. G-Allele carriers significantly increased the risk of AR. For patients with AS as control, P is .02 (odds ratio [OR] = 1.77, confidence interval [CI]: 1.07-2.92). For patients with semi-AR as control, P is .05. HO-1 rs2071746 (-413A>T) is associated with AR. T-Allele carriers significantly increased the risk of AR. For patients with AS as control, P is .04 (OR = 1.70, CI: 1.02-2.79). For patients with semi-AR as control, P is .05 (OR = 1.68, CI: 1.00-2.80). CONCLUSION: rs2071746 in HO-1 gene, rs1330344 in COX-1 gene contribute to AR.

OBJECTIVE: To determine whether apoptosis is induced in rat hepatic stellate cells (HSCs) in response to activation of the hepatocyte growth factor (HGF) by hepatocyte growth factor activator (HGFA) by using a co-culture system of bone marrow mesenchymal stem cells (BMSCs) and HSCs. METHODS: In this study, cells were divided into the following five groups: HSC control group: HSCs co-cultured with fibroblast cells; HSCs blank group: HSCs cultured alone; BMSCs blank group: BMSCs cultured alone; Experimental group: BMSCs + HSCs; HGFA intervention group: HSCs treated with 70 ng/mL of HGFA. The culture systems were established in culture plates with transwell inserts, and cells were assessed at 24, 48, and 72 h of growth. Dynamic changes in cell morphology were observed under an inverted phase contrast microscope. The surface markers of BMSCs and the apoptosis rate of HSCs were detected by Annexin-V-FITC/propidium iodide (PI). Expression of a-smooth muscle actin (SMA) in HSCs was evaluated by immunohistochemistry. The presence of activated HGF (HGF-a chain) was determined by immunofluorescent staining. HSC proliferation was measured by MTT assay, and the concentrations of HGF and HGFA were quantified by enzyme-linked immunosorbent assay (ELISA). RESULTS: MTT results indicated that treatment with HGF alone had no effect on HSC proliferation rate (vs. HSC blank group, P more than 0.05), but that 24 h treatment with HGFA significantly inhibited the proliferation rate (0.26 ± 0.00 vs. blank group: 0.13 ± 0.04, P = 0.02); moreover, this effect was concentration-dependent. Expression of HGF-a was lower in the experimental group than in the HGFA intervention group at 72 h (37.24 ± 1.03 vs. 40.44 ± 0.77, P = 0.04), and both of these groups had higher expression than the control group at all time points examined (P less than 0.05). The apoptosis rate was consistently higher in the experimental group than in the HGFA intervention group, but most robustly at 72 h (40.77 ± 1.16% vs. 33.35 ± 2.04%, P = 0.00); moreover, the apoptosis rate was significantly higher than that in the control group at all time points examined (P less than 0.01). The concentration of HGF in the experimental group and the HGFA intervention group showed a time-dependent reduction, and was consistently lower than that in the HSCs control group (P less than 0.05). Finally, the concentration of HGFA was higher in the experimental group than in the blank group at all time points examined (P less than 0.05). CONCLUSION: The BMSC-HSC co-culture system can promote secretion of HGFA from HSCs and HGF activation, thereby inducing apoptosis of HSCs.

Methylenetetrahydrofolate reductase (MTHFR) is an essential enzyme involved in folate metabolism; a single nucleotide polymorphism (SNP) C677T has been reported to be linked with altered incidences of several diseases. We here conducted a meta-analysis of 15 published epidemiological studies with a total of 7306 cases and 8062 controls to evaluate its association with prostate cancer risk with overall and subgroup analyses. No statistical relationship was found overall with any genetic model (TT vs. CC: OR = 0.80, 95%CI = [0.62, 1.04], P = 0.094; CT vs. CC: OR = 0.97, 95%CI = [0.84; 1.12], P = 0.667; Dominant: OR = 0.94, 95%CI = [0.82; 1.07], P = 0.343; Recessive: OR = 0.81, 95%CI = [0.64; 1.04], P = 0.104), but after the exclusion of several studies, we could observe the homozygote TT to confer less susceptibility to prostate cancer in carriers; moreover, different effects of the polymorphism on prostate cancer risk was detected from subgroup analysis stratified by participants' residential region: significant reduced prostate cancer risk was found to be associated with the polymorphism from Asian studies (TT vs. CC: OR = 0.47, 95%CI = [0.33; 0.67], P< 0.001; CT vs. CC: OR = 0.73, 95%CI = [0.60; 0.90], P = 0.002; Dominant: OR = 0.67, 95%CI = [0.56; 0.82], P< 0.001; Recessive: OR = 0.55, 95%CI = [0.40; 0.76], P< 0.001) while studies from Europe indicated a slight increased risk under dominant model with marginal significance (OR = 1.14, 95%CI = [0.99; 1.30], P = 0.064). Moreover, the protective effect of the polymorphism against prostate cancer was also shown by studies performed in yellow Asians (TT vs. CC: OR = 0.48, 95%CI = [0.31; 0.75], P = 0.001; CT vs. CC: OR = 0.68, 95%CI = [0.51; 0.90], P = 0.006; Dominant: OR = 0.63, 95%CI = [0.48; 0.82], P < 0.001; Recessive: OR = 0.57, 95%CI = [0.39; 0.84], P = 0.004). We propose that these phenomena should be viewed with the consideration of folate metabolism profile and different gene background as well as living habits of different populations, and more relevant studies should be conducted to confirm our hypothesis and provide a comprehensive and clear picture concerning this topic.

In this study we investigated the potential effects of Angelica sinensis on the growth and metastasis in human lung adenocarcinoma A549 cells. In vitro the Cck-8 assays showed that Angelica sinensis had weak antiproliferative effect on A549 cells only at high concentration. The cell adhesion assay showed that Angelica sinensis decreased the adhesive ability of A549 cells in a dose- and time-dependent manner. Transwell invasion and migration assay showed that Angelica sinensis reduced the invasive and migratory abilities of A549 cells in a dose-dependent manner. In vivo the animal experiments showed that Angelica sinensis suppressed lung metastasis of nude mice at high concentration. Then, we attempted to clarify the mechanisms of anti-metastatic activities of Angelica sinensis. The results showed Angelica sinensis inhibited the enzymatic activity of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9), it involved the down-regulation of the expressions of MMP-2 and MMP-9 at both the protein and mRNA levels, which may be associated with Angelica sinensis suppressing the expression of TGF-ß1. It also involved the increase of the tissue inhibitors of metalloproteinases TIMP-2, but TIMP-1 decreased upon incubation of A549 cells with Angelica sinensis. The results suggest that Angelica sinensis might exert anti-growth and anti-metastasis activity against lung cancer cells through the decrease of MMP-2, MMP-9, TGF-ß1 and TIMP-1 and increase of TIMP-2.
