RESUMO

Oxidation of C-B bonds is extensively used in organic synthesis, materials science, and chemically biology. However, these oxidations are usually limited to the oxidation of C(sp3 )-B and C(sp2 )-B bonds. The C(sp)-B bond oxidation is rarely developed. Herein we present a novel strategy for the preparation of γ-lactones via the oxidation of enynyl boronates. This process successively involves the C(sp)-B bond oxidation, the epoxidation of C-C double bond and the lactonization. This protocol provided various γ-lactones and unsaturated butenolides efficiently that are prevalent in numerous nature products and bioactive molecules. Most importantly, asymmetric oxidative lactonization of enynyl boronates was also achieved, providing chiral γ-lactones in high enantioselectivities and diastereoselectivities. The versatile transformations and ubiquity of γ-lactones shed light on the importance of this strategy in the construction and late-stage functionalization of complex molecules.

To ensure food safety and prevent the toxic effects of paclobutrazol (PBZ) and carbofuran (CAR) on humans, a sensitive and rapid method for the detection of PBZ and CAR in fruits and vegetables is required. Herein, a highly sensitive PBZ monoclonal antibody (PBZ mAb) and CAR monoclonal antibody (CAR mAb) with half-inhibitory concentrations (IC50) at 0.77 and 0.82 ng mL-1 were prepared, respectively. We proposed a novel dual-color immunochromatographic assay (ICA) with two test lines (T1 and T2) and an independent control line (C) based on chrysanthemum-like Au@Polydopamine (AuNC@PDA) and colloidal gold (AuNPs) for the simultaneous and sensitive detection of PBZ and CAR with naked-eye detection limits of 10 and 5 µg kg-1, respectively. The limits of detection (LOD) for PBZ and CAR were 0.117 and 0.087 µg kg-1 in orange, 0.109 and 0.056 µg kg-1 in grape, and 0.131 and 0.094 µg kg-1 in cabbage mustard, respectively. The average recoveries of PBZ and CAR in orange, grape, and cabbage mustard were 97.86-102.83%, with coefficients of variation from 8.94 to 11.05. The detection results of this method for 30 samples (orange, grapes, and cabbage mustard) agreed well with those of liquid chromatography-tandem mass spectrometry. The novel dual-color ICA was sensitive, rapid, and accurate for the simultaneous detection of PBZ and CAR in real samples.

We present herein a novel strategy for the preparation of ketones from aldehydes and allylic boronic esters. This reaction involves the allylation of aldehydes with allylic boronic esters and the Rh-catalyzed chain-walking of homoallylic alcohols. The key to this successful development is the protodeboronation of alkenyl borylether intermediate via a tetravalent borate anion species in the presence of KHF2 and MeOH. This approach features mild reaction conditions, broad substrate scope, and excellent functional group tolerance. Mechanistic studies also supported that the tandem allylation and chain-walking process were involved.

RESUMO

We describe a general and efficient protocol for the synthesis of organophosphine compounds from phenols and phosphines (R2PH) via a metal-free C-O bond cleavage and C-P bond formation process. This approach exhibits broad substrate scope and excellent functional group tolerance. The synthetic utilities of this protocol were demonstrated by the synthesis of chiral ligands via the various transformations of cyano groups and their applications in asymmetric catalysis.

Muscle atrophy arises because of many chronic illnesses, as well as from prolonged glucocorticoid treatment and nutrient deprivation. We previously demonstrated that the USP19 deubiquitinating enzyme plays an important role in chronic glucocorticoid- and denervation-induced muscle wasting. However, the mechanisms by which USP19 exerts its effects remain unknown. To explore this further, we fasted mice for 48 hours to try to identify early differences in the response of wild-type and USP19 knockout (KO) mice that could yield insights into the mechanisms of USP19 action. USP19 KO mice manifested less myofiber atrophy in response to fasting due to increased rates of protein synthesis. Insulin signaling was enhanced in the KO mice, as revealed by lower circulating insulin levels, increased insulin-stimulated glucose disposal and phosphorylation of Akt and S6K in muscle, and improved overall glucose tolerance. Glucocorticoid signaling, which is essential in many conditions of atrophy, was decreased in KO muscle, as revealed by decreased expression of glucocorticoid receptor (GR) target genes upon both fasting and glucocorticoid treatment. This decreased GR signaling was associated with lower GR protein levels in the USP19 KO muscle. Restoring the GR levels in USP19-deficient muscle was sufficient to abolish the protection from myofiber atrophy. Expression of GR target genes also correlated with that of USP19 in human muscle samples. Thus, USP19 modulates GR levels and in so doing may modulate both insulin and glucocorticoid signaling, two critical pathways that control protein turnover in muscle and overall glucose homeostasis.

Diet induced obese (DIO) mice can be stratified according to their weight gain in response to high fat diet as low responders (LDR) and high responders (HDR). This allows the study of ß-cell failure and the transitions to prediabetes (LDR) and early diabetes (HDR). C57BL/6N mice were fed for 8 weeks with a normal chow diet (ND) or a high fat diet and stratified as LDR and HDR. Freshly isolated islets from ND, LDR and HDR mice were studied ex-vivo for mitochondrial metabolism, AMPK activity and signalling, the expression and activity of key enzymes of energy metabolism, cholesterol synthesis, and mRNA profiling. Severely compromised glucose-induced insulin secretion in HDR islets, as compared to ND and LDR islets, was associated with suppressed AMP-kinase activity. HDR islets also showed reduced acetyl-CoA carboxylase activity and enhanced activity of 3-hydroxy-3-methylglutaryl-CoA reductase, which led respectively to elevated fatty acid oxidation and increased cholesterol biosynthesis. HDR islets also displayed mitochondrial membrane hyperpolarization and reduced ATP turnover in the presence of elevated glucose. Expression of protein kinase Cε, which reduces both lipolysis and production of signals for insulin secretion, was elevated in DIO islets. Genes whose expression increased or decreased by more than 1.2-fold were minor between LDR and ND islets (17 differentially expressed), but were prominent between HDR and ND islets (1508 differentially expressed). In HDR islets, particularly affected genes were related to cell cycle and proliferation, AMPK signaling, mitochondrial metabolism and cholesterol metabolism. In conclusion, chronically reduced AMPK activity, mitochondrial dysfunction, elevated cholesterol biosynthesis in islets, and substantial alterations in gene expression accompany ß-cell failure in HDR islets. The ß-cell compensation process in the prediabetic state (LDR) is largely independent of transcriptional adaptive changes, whereas the transition to early diabetes (HDR) is associated with major alterations in gene expression.
