RESUMO

Metal sulfides in waste rocks and tailings are susceptible to serious soil and water contamination due to the generation of acid mine drainage (AMD) during stockpiling. The hydrometallurgical process is one of the most essential heavy metal remediation technologies through harmless disposal and resource utilization of the waste sulfides. However, atmospheric hydrometallurgy of sulfides still faces great challenges due to low leaching efficiency and high cost. In this work, we proposed a cooperative leaching system (Fe2(SO4)3-O3) and investigated the oxidative dissolution process of sphalerite (ZnS). Under the optimal conditions, the extracted zinc reached 97.8%. Reactive oxygen species (ROS) (·OH, 1O2 and ·O2-) were identified in the radical quenching experiments. The dissolution of sphalerite did not show passivation due to the ozone's capability to oxidize the sulfur in sphalerite to sulfate. In addition, stirring rate, O3 inlet concentration, and Fe2(SO4)3 concentration had a significant effect on the dissolution of sphalerite. Meanwhile, the apparent activation energy was 24.11 kJ/mol based on kinetic fitting, which indicated that the controlling step of the reaction was mainly a diffusion process. This work demonstrated the cooperative effect of sphalerite leaching in the O3-Fe2(SO4)3 system and provided a theoretical reference for efficient and atmospheric dissolution of sphalerite.

RESUMO

The transport and deposition pathways of anthropogenic radionuclides at the global scale, particularly volatile 129I, remain somewhat elusive due to a dearth of comprehensive investigations. To gain a better understanding of the transport dynamics and deposition mechanism of anthropogenic 129I in the terrestrial environment, one hundred surface soil samples collected from northeast China were analyzed for 129I and 127I concentrations in this study. Our findings reveal that 129I/127I atomic ratios in the mid-eastern Inner Mongolia (MIM) were approximately an order of magnitude higher than the rest of the investigated area. This is, besides the global fallout and the long-range transport of 129I released from the European nuclear reprocessing plants via westerly winds, possibly attributed to the dust with high 129I levels from the East Asian arid regions. In addition to the significant dust-induced 129I input, the unique meteorological conditions and topographical features in the MIM synergistically contribute to the pronounced accumulation and deposition of 129I in this region. This study will provide novel insights into the transport and deposition mechanism of anthropogenic radionuclides, which is significant for the assessment of anthropogenic nuclear activities on the environment in the future.

RESUMO

Long-term deposition of atmospheric radioactive iodine-129 (129I) is important for assessing the impact of human nuclear activities (HNAs), but still not well understood in East Asia. In this study, we quantitatively reconstructed the deposition history of airborne 129I using varved sediment from Sihailongwan Maar Lake (SHLW) in northeast China. Our results revealed significant increases in 129I concentrations and 129I/127I atomic ratios since the 1950s, indicating the influence of HNAs on the environment and marking the onset of the Anthropocene. The variation of 129I in the investigated site can be primarily attributed to the global fallout of ANWT as well as nuclear fuel reprocessing in Europe, Russia and the USA. Notably, neither the Chernobyl nor the Fukushima nuclear accidents have had any discernable impact on the SHLW Lake. Over the past 170 years (1846-2021), the reconstructed fluxes indicate a rapid increase in 129I deposition from the early 1950s until the 1970s followed by dramatic changes thereafter. The measured 129I fluxes range between (1.26-349) × 109 atoms m-2 yr-1 in the SHLW Lake, which are consistent with similar latitude zones across East Asia, but differ significantly from those observed in high-elevation glaciers within the Northern Hemisphere due to prevailing atmospheric circulation patterns. The total 129I inventory was calculated to be 11.9 × 1012 atoms m-2, with natural and anthropogenic 129I accounting for 2.86 % and 97.1 %, respectively, suggesting an overwhelming artificial contribution. The reconstructed fluxes and inventory of atmospheric 129I deposition quantitatively distinguish the natural and artificial contributions, and provide a novel insight into the historical environmental impact of HNAs in East Asia and the characteristics of the Anthropocene.

RESUMO

Drugs targeting the DNA damage response (DDR) are widely used in cancer therapy, but resistance to these drugs remains a major clinical challenge. Here, we show that SYCP2, a meiotic protein in the synaptonemal complex, is aberrantly and commonly expressed in breast and ovarian cancers and associated with broad resistance to DDR drugs. Mechanistically, SYCP2 enhances the repair of DNA double-strand breaks (DSBs) through transcription-coupled homologous recombination (TC-HR). SYCP2 promotes R-loop formation at DSBs and facilitates RAD51 recruitment independently of BRCA1. SYCP2 loss impairs RAD51 localization, reduces TC-HR, and renders tumors sensitive to PARP and topoisomerase I (TOP1) inhibitors. Furthermore, our studies of two clinical cohorts find that SYCP2 overexpression correlates with breast cancer resistance to antibody-conjugated TOP1 inhibitor and ovarian cancer resistance to platinum treatment. Collectively, our data suggest that SYCP2 confers cancer cell resistance to DNA-damaging agents by stimulating R-loop-mediated DSB repair, offering opportunities to improve DDR therapy.

RESUMO

Purpose: Apolipoprotein A1 (APOA1) is a potential crucial protein and treatment goal for pathological myopia in humans. This study set out to discover the function of APOA1 in scleral remodeling in myopia and its underlying mechanisms. Methods: A myopic cell model was induced using hypoxia. Following loss- and gain-of function experiments, the expression of the myofibroblast transdifferentiation-related and collagen production-related factors Forkhead box M1 (FOXM1), APOA1, and methyltransferase-like 3 (METTL3) in the myopic cell model was examined by quantitative reverse transcription polymerase chain reaction (RT-qPCR) and western blotting. The proliferation and apoptosis were determined by Cell Counting Kit-8 assay and flow cytometry, respectively. Chromatin immunoprecipitation (ChIP) was employed to examine FOXM1 enrichment in the METTL3 promoter, methylated RNA immunoprecipitation (Me-RIP) to examine the N6-methyladenosine (m6A) modification level of APOA1, and photoactivatable ribonucleoside-enhanced crosslinking and immunoprecipitation (PAR-CLIP) to examine the binding between METTL3 and APOA1. Results: Hypoxia-induced human scleral fibroblasts (HSFs) had high APOA1 and FOXM1 expression and low METTL3 expression. FOXM1 knockdown elevated METTL3 expression and downregulated APOA1 expression. FOXM1 was enriched in METTL3 promoter. APOA1 or FOXM1 knockdown or METTL3 overexpression reversed the hypoxia-induced elevation in vinculin, paxillin, and α-smooth muscle actin (α-SMA) levels and apoptosis and the reduction in collagen, type I, alpha 1 (COL1A1) level and cell proliferation in HSFs. METTL3 or YTH N6-methyladenosine RNA binding protein F2 (YTHDF2) knockdown or APOA1 overexpression reversed the impacts of FOXM1 knockdown on vinculin, paxillin, α-SMA, and COL1A1 expression and cell proliferation and apoptosis. Conclusions: FOXM1 elevated the m6A methylation level of APOA1 by repressing METTL3 transcription and enhanced APOA1 mRNA stability and transcription by reducing the YTHDF2-recognized m6A methylated transcripts.

RESUMO

In recent years, various materials have been used to adsorb and remove perfluoroalkyl compounds from water. However, most of these materials have limited applications due to their high cost, complex synthesis, inadequate selectivity and sensitivity, and, even worse, the possibility of introducing secondary pollution. Here, under mild conditions, we prepared an inexpensive imidazolium chloride and nitrogen-rich polymer (TAGX-Cl) with a high cationic loading rate and a high yield (>82%). The adsorbent exhibits excellent pH tolerance (pH=1-9) and achieves nearly 99.9% removal of nine perfluoroalkyl carboxylic acids (PFCAs) within 120 min. Experimental data and theoretical simulations confirmed that synergistic electrostatic interactions, hydrogen bonds, and P-π interactions control the adsorptive ability of TAGX-Cl. This work provides a practical strategy for PFCAs removal.

RESUMO

Iodine is a crucial nutrient for public health, and its presence in the terrestrial atmosphere is a key factor in determining the prevalence of iodine deficiency disorders. While oceanic iodine emissions decrease at lower sea surface temperatures, the primary contributors to atmospheric iodine can vary from oceanic sources in the summer to other sources in winter. However, the specific sources and their respective contributions have remained unexplored. Fortunately, the atomic ratio of 129I to 127I significantly differs between nuclear activity and fossil fuels like coal and petroleum, which formed millions to billions of years ago. This distinction makes 129I a valuable tool for identifying iodine sources. In our study, we analyzed iodine isotopes and incorporated additional indicators such as element content in PM2.5 samples. Our findings reveal, for the first time, that in winter inland areas, fuel oil, alongside coal combustion, is a significant source of atmospheric iodine. This research enhances our comprehension of the impact of human activities on iodine levels in the environment. This understanding is crucial not only for addressing iodine deficiency-related health concerns but also for comprehending stratospheric ozone depletion, a phenomenon closely associated with atmospheric iodine.

RESUMO

PURPOSE: Glioblastoma (GBM) is a highly vascularized tumor with few treatment options after disease recurrence. Here, we report the efficacy and safety of anlotinib hydrochloride plus temozolomide in patients with recurrent GBM. PATIENTS AND METHODS: Patients with first definite postsurgical progression of histologically confirmed GBM preceded by standard radiotherapy and temozolomide chemotherapy were eligible for inclusion. All patients received temozolomide (150-200 mg/m2, orally, every day (QD) d1-5/4 wk) and anlotinib (10 mg, orally, QD, d1-14/3 wk) until disease progression or unacceptable toxicity. The primary endpoint was investigator-assessed 6-month progression-free survival (PFS) rate by the Response Assessment in Neuro-Oncology (RANO) criteria. RESULTS: Twenty-one patients were enrolled between May 2020 and July 2021, with a median age of 55 (range 27-68) years old. According to the Response Assessment in Neuro-Oncology (RANO) criteria, tumor response occurred in 17 patients, of which 9 patients had a complete response, and the objective response rate was 81.0% [95% confidence interval (CI), 62.6-99.3]. The disease control rate was 95.2% (95% CI, 76.2-99.9), with three additional patients achieving a stable disease without tumor progression. The median PFS was 7.3 months (95% CI, 4.9-9.7), and the 6-month PFS rate was 61.9% (95% CI, 39.3-84.6). The median overall survival was 16.9 months (95% CI, 7.8-26.0). The most common adverse events were leukocytopenia (66.7%), thrombocytopenia (38.1%), and hypertriglyceridemia (38.1%). Five patients had nine grade 3 adverse events, with a 23.8% incidence rate. Two patients discontinued therapy due to ischemic stroke (grade 3) and wound dehiscence (grade 1), respectively. No grade 4 or treatment-related deaths occurred in this study. CONCLUSIONS: Anlotinib combined with temozolomide is efficacious and tolerated in patients with recurrent GBM.

RESUMO

Microplastics (MPs) are emerging pollutants, and limited research has focused on their exposure to terrestrial wildlife and transport mode on the remote Tibetan Plateau (TP). Therefore, we investigated MPs in the southern soil and feces of Equus kiang (Tibetan wild ass), a species peculiar to the TP, which is known as the "third pole." We found that MP median abundances were 102 and 4.01 particles/g of dry feces and soil, respectively. In both media, the MP morphology mainly comprised ∼50 µm slender fibrous particles. In total, 29 MP types were identified in the feces, compared to 26 types in the soil. Among them, the acrylate copolymer (35.9%) and polyurethane (24.9%) were predominant in the feces, while polyurethane (22.5%) and silicone (20.4%) were predominant in the soil. After ingesting MPs at one location, E. kiang may become a source of MP pollution when moving to other meadows on the TP. The potential MP transport flux of a herd of 20 kiangs has been estimated at 1736 particles m-2 a-1. A unique "source-sink-source" MP transport model comprising an "atmospheric deposition-vegetation-feces-atmosphere" cycle on the TP was observed based on atmospheric transport simulations and terrestrial food chain transfer processes. Owing to human settlements in south and East Asia adjacent to the TP, atmospheric long-distance transmission is an essential route for MPs to enter the TP.

RESUMO

Although the incorporation of 2D perovskite into 3D perovskite can greatly enhance intrinsic stability, power conversion efficiency (PCE) of 2D/3D perovskite is still inferior to its 3D counterpart due to poor carrier transport kinetics resulted from the quantum and dielectric confinement of 2D component. To overcome this issue, the electron acceptor molecule 1,2,4,5-tetracyanobenzene (TCNB) was introduced to trigger intermolecular π-π interaction in 2D perovskite along with the electronic doping of 2D/3D perovskite to improve charge transfer efficiency. By virtue of high electron affinity, TCNB can undergo electron transfer reaction and subsequently establish π-π interaction with 1-naphthalenemethylammonium (NMA) cations, greatly strengthening lattice rigidity and reducing exciton binding energy. Transmission electron microscopy results demonstrate that 2D phases are mainly distributed at grain boundaries, reducing defect density and weakening nonradiative recombination. Meanwhile, the p-type doping of perovskite by TCNB optimizes energy level alignment at perovskite/hole transport layer interface. Consequently, PCE of champion device is significantly boosted to 24.01 %. The unencapsulated device retains an initial efficiency close to 94 % after exposure to ambient environment for over 1000 h. This work paves a novel path for designing new mixed-dimensional perovskite solar cells with high PCE and superior stability.

RESUMO

Background: Spinal schwannomas (SSs) are benign tumors affecting the nerve sheath, accounting for 25% of spinal nerve root tumors. Surgery represents the mainstay of treatment for SS patients. Following surgery, approximately 30% of patients experienced developed new or worsening neurological deterioration, which probably represented an inevitable complication of nerve sheath tumor surgery. The objective of this study was to identify the rates of new or worsening neurological deterioration in our center and accurately predict the neurological outcomes of patients with SS by developing a new scoring model. Methods: A total of 203 patients were retrospectively enrolled at our center. Risk factors associated with postoperative neurological deterioration were identified by multivariate logistic regression analysis. ß-coefficients for independent risk factors were used to define a numerical score to generate a scoring model. The validation cohort at our center was used to verify the accuracy and reliability of the scoring model. Receiver operating characteristic (ROC) curve analysis was used to evaluate the performance of the scoring model. Results: In this study, five measured variables were selected for the scoring model: duration of preoperative symptoms (1 point), radiating pain (2 points), tumor size (2 points), tumor site (1 point), and dumbbell tumor (1 point). The scoring model divided the spinal schwannoma patients into three categories: low risk (0-2 points), intermediate risk (3-5 points), and high risk (6-7 points), with predicted risks of neurological deterioration of 8.7%, 36%, and 87.5%, respectively. And the validation cohort confirmed the model with the predicted risks of 8.6%, 46.4%, and 66.6%, respectively. Conclusion: The new scoring model might intuitively and individually predict the risk of neurological deterioration and may aid individualized treatment decision-making for SS patients.

RESUMO

PURPOSE: To assess morphologic characteristics of retinal arterial macroaneurysm (RAM) and their vascular changes using optical coherence tomography angiography (OCTA). METHODS: This observational study included 31 eyes of 29 participants diagnosed with RAM based on fundus fluorescein angiography in Tianjin Medical University Eye Hospital. Multimodal imaging modalities, including fundus photography, fluorescein angiography, and OCTA, were used to examine RAMs. The demographic and clinical characteristics of the RAMs were recorded. RESULTS: Depending on the fundus fluorescein angiography examination, 40 cases of RAM were confirmed in 29 patients. Twenty-three patients were female (79%), and six patients were male (21%). Two patients had binocular RAM, and four eyes had more than one RAMs. Relying on the OCTA technology, RAMs have four different vascular morphology types (i.e., distended, meshed, malformed, and occult types). In the distended type, round or encircled thrombi caused asymmetrical or symmetrical distention of retinal arteriolar, leading to separate true lumen and false thrombus lumen in RAM. In the meshed type, the meshed or dendritic vascular network around the RAM was likely to be the neovascularization due to the ischemia and hypoxia of the arteriolar wall. Finally, in the malformed and occult type, the RAM usually regressed, and the retinal arterioles were remodeled to distorted or normal arterioles accompanied by capillary degradation. CONCLUSION: Relying on the OCTA technology, we found that the RAMs have four different types of vascular morphology. Each group of RAM has different vascular features. The application of OCTA in patients with RAM furthers our understanding of the vasculature of RAMs.

RESUMO

The immune microenvironment of glioma attributes to the initiation and development of glioma; however, the underlying mechanisms of tumor microenvironment formation have not been fully understood. In this study, we revealed that Zinc Finger Protein 395 (ZNF395), a member of the Kruppel C2H2-type zinc-finger protein family and also known as a common transcription factor, was aberrantly overexpressed in glioma and positively associated with the poor clinicopathological features and the prognosis of patients with glioma based on the analyses of TCGA, CGGA and other datasets. Further in vitro experimental data demonstrated that the upregulation of ZNF395 promoted the proliferation of glioma cells. In addition, functional enrichment analysis showed that ZNF395 was involved in immune processes and correlated with macrophage infiltration and polarization. Moreover, C-C Motif Chemokine Ligand 20 (CCL20), one of the ZNF395 co-expressed genes, was validated as the downstream factor under the transcriptional regulation of ZNF395. Importantly, cell co-culture experiments confirmed that ZNF395 upregulated both the intracellular and secreted CCL20 level of glioma cells and induced M2 macrophage polarization which is known to promote the malignant progression of glioma. Taken together, our findings suggested that ZNF395 might play an essential role in glioma development, and inhibition of ZNF395 might be a plausible strategy for glioma therapy.

RESUMO

Background: Intravenous thrombolysis (IVT) is a standard procedure for the treatment of patients with acute ischemic stroke (AIS). Improving the therapeutic efficacy of IVT is an important task for neurologists. The aim of this study was to evaluate the efficacy and safety of early low-dose tirofiban treatment in AIS patients with early neurological deterioration (END) after IVT. Methods: In this prospective and randomized pilot study, 73 AIS patients with END were recruited from a local hospital in China. Of these, 14 patients were treated with regular antiplatelet agents (aspirin plus clopidogrel) and 59 patients were treated with tirofiban within 24 h of IVT, followed by regular antiplatelet therapy. Neurological deficits and functional recovery were assessed with NIHSS and modified Rankin Scale (mRS) at 7 and 90 days. During the 90-day follow-up period, both hemorrhagic (e.g., intracerebral hemorrhage) and non-hemorrhagic (e.g., pneumonia) events were recorded. Results: Treatment with tirofiban compared with regular antiplatelet therapy: (1) improved functional recovery of AIS patients to mRS (≤2) at both 7 and 90 days (odds ratios [ORs], 1.37 and 1.64; 95% confidence interval [CI], 1.16-1.61 and 1.26-2.12; P = 0.008 and < 0.001, respectively), and (2) reduced NIHSS scores from 11.14 ± 2.38 to 5.95 ± 3.48 at day 7 (P < 0.001) and from 8.14 ± 2.74 to 4.08 ± 3.50 at day 90 (P < 0.001). Tirofiban treatment did not increase the risk of hemorrhagic complications. Multivariate regression analysis showed that tirofiban treatment independently predicted a favorable functional outcome (P ≤ 0.001). Conclusion: Early treatment with low-dose tirofiban in AIS patients with neurologic deterioration after IVT potentially improved functional recovery and attenuated neurologic deficits as early as 7 days and did not increase the risk of various hemorrhagic complications. However, the therapeutic efficacy of tirofiban treatment in END patients needs to be determined by future randomized clinical trials with a large study population. RESUMO

Anaerobic ammonia oxidation (Anammox) is a highly productive research area in municipal wastewater treatment. A novel combined process driven by partial nitrification/anammox (PN/A) and partial denitrification/anammox (PD/A) was established in this paper using a sequencing batch reactor (SBR) and two up-flow sludge beds (USBs). Municipal wastewater after carbon removal pretreatment in SBR entered PN/A-USB. PN/A process was initiated and enhanced by optimizing the intermittent aeration mode under low dissolved oxygen (DO). After enhancing and stabilizing the PD/A process, PN/A effluent entered the PD/A-USB along with raw municipal wastewater at a ratio of 4:1 and the combined system was established. Through this, this study achieved a nitrogen removal efficiency (NRE) of 84.9 % from municipal wastewater at an ultra-low total hydraulic residence time (HRT) of 3.9 h. Candidatus Brocadia (1.8 % in PN/A, 1.0 % in PD/A) was the only functional anammox bacterium in the combined process.

RESUMO

Anthropogenic 129I has been dispersed all over the world and could be utilized as an oceanographic tracer based on its conservative nature in the ocean. The first datasets of 129I and 127I were obtained by analysis of seawater of 36 water columns collected in the western South China Sea during August-September 2018. The measured 129I concentrations decreased with depth from (0.93-1.61) × 107 atoms/L in the upper 200 m to (0.04-0.14) × 107 atoms/L at 1500 m, indicating a clear anthropogenic source in the upper layer, mainly originated from the global fallout. The riverine input of the deposited 129I on the catchment area of the Mekong River is an important source besides the direct deposition in the seas. The water mass with high 129I from the Mekong River water moves to the east at 11°N by the North Nansha Current in the surface layer (2-25 m). The exponentially decreasing 129I level with depth indicates that the vertical dispersion of 129I from the upper to the lower layer was mainly through slow diffusion, and the deep water at more than 1500 m was not significantly contaminated by the upper layer water at least in the past 70 years.

RESUMO

The aromatic structure and the rich nitrogen content of polymers based on covalent triazine-based frameworks (CTF) and their unique hydrophilic-lipophilic-balanced adsorption properties make them promising candidates for an adsorbent that can be used for sample pretreatment. Herein, a new covalent triazine-based framework (CTF-DBF) synthesized by a Friedel−Crafts reaction was used for the determination of the content of nucleotides in commercial infant formula. It was shown that the synthetic materials had an amorphous microporous structure, a BET surface area of up to 595.59 m2/g, and 0.39 nm and 0.54 nm micropores. The versatile adsorption properties of this material were evaluated by quantum chemistry theory calculations and batch adsorption experiments using five nucleotides as probes. The quantum chemistry results demonstrated that CTF-DBF can participate in multiple interactions with nucleotides. All the analyses performed present good linearity with R2 > 0.9993. The detection limits of targets ranged from 0.3 to 0.5 mg/kg, the spiked recoveries were between 85.8 and 105.3% and the relative standard deviations (RSD, n = 6) were between 1.1 and 4.5%. All these results suggest that this versatile CTF-DBF has great potential for sample pretreatment.

RESUMO

Atmospheric iodine cycling is of significance in climate change and environmental and health impacts. To better explore speciation transformation of atmospheric stable and radioactive iodine, an ultra-sensitive analytical method was established for determination of 129I and 127I in particulate, gaseous inorganic, and gaseous organic species, which was conducted with a self-designed cascade sampling apparatus, followed by their separation with a pyrolysis system and accelerator mass spectrometry and ICP-MS measurements. Combustion protocols for three sampling matrices and NaOH concentration for iodine trapping were optimized to achieve a safe analytical procedure with a high chemical yield of iodine. Based on the lowest concentrations of 129I and 127I, a suitable activated carbon product for adsorption of gaseous organic iodine was carefully selected. The detection limits of the three species were 0.30-2.21 ng m-3 for 127I and 0.05-0.22 × 105 atoms m-3 for 129I. This newly established method was successfully applied to analyze the levels and species of 129I and 127I in ambinet air from Xi'an, China, from May to August, 2020. Gaseous organic iodine was found to be the dominant species of 127I and 129I, accounting for about half of total iodine, and gaseous inorganic iodine and particulate iodine accounted for one-quarter each during the whole sampling period. Speciation variation of 129I and 127I indicates that speciation transformation apparently occurred at the turn of spring and summer, mainly between particulate and gaseous organic iodine. This study has implications on delicate tracing of the atmospheric behavior of iodine with long-lived anthropogenic 129I.
