RESUMO

Acute pancreatitis (AP) is an acute inflammatory disorder characterized by acinar cell death and inflammation. Multiple factors cause hyperglycemia after AP. Macrophage polarization is involved in tissue injury and repair, and is regulated by Notch signaling during certain inflammatory diseases. The present study explores the relationship among hyperglycemia, macrophage polarization, and Notch signaling during AP and the related mechanisms. A cerulein-induced AP model was established in FVB/N mice, and AP with hyperglycemia was initiated by injection of 50% concentration glucose. Tissue damage, Notch activity, and macrophage polarization were assessed in pancreatic tissues. The role of Notch signaling in macrophage polarization during AP was also assessed in vitro by co-culturing primary macrophages and pancreatic acinar cells, and establishing a lipopolysaccharide (LPS)-induced inflammatory model in RAW264.7 cells. Pancreatic acinar cells were damaged and proinflammatory factor levels were increased in pancreatic tissues during AP. The hyperglycemic conditions aggravated pancreatic injury, increased macrophage infiltration, promoted macrophage polarization towards an M1 phenotype, and led to excessive up-regulation of Notch activity. Inhibition of Notch signaling by DAPT or Notch1 knockdown decreased the proportion of M1 macrophages and reduced the production of proinflammatory factors, thus mitigating pancreatic injury. These findings suggest that hyperglycemia induces excessive Notch signaling after AP and further aggravates AP by promoting pancreatic macrophage polarization towards the M1 phenotype. The Notch signaling pathway is a potential target for the prevention and treatment of AP.

Introduction: DOK is a new type of regulatory protein family that participates in the regulation of tumor cell growth. However, most of the studies are conducted in cell lines, and systematic studies have not been conducted in human tumors. Objectives: We conducted a comprehensive analysis of DOK based on its expression profile and its relationship with patient survival, immune infiltration, tumor microenvironment, and drug sensitivity. Methods: We used the TCGA database to analyze the correlation between DOK family gene expression and prognosis and clinical stage. The protein expression of DOK in tumor tissues was analyzed by immunohistochemistry. Use the cBioPortal database to analyze the alteration frequency in DOK family genes in human tumors. In addition, we used ESTIMATE algorithm and TIMER website to analyze the correlation between DOK family genes and tumor immunity. Finally, we further analyzed the relationship between DOK family genes and tumor stemness and the sensitivity of cancer cells to chemotherapy. Results: We conducted a comprehensive analysis of DOK family genes based on its expression profile and its relationship with patient survival. We also confirmed this conclusion by immunohistochemistry. The expression of DOK family genes is related to OS, clinical stage, tumor mutation, methylation, CNV, and SNV. DOK family genes are significantly associated with poor prognosis of UVM. DOK1-DOK3 has obvious correlation with tumor immunity. DOK2 can increase the sensitivity of chemotherapy drugs, while DOK4 reduces the sensitivity of multiple chemotherapy drugs. In addition, the expression level of DOK family genes is significantly correlated with the activity of cancer marker-related pathways. Conclusions: DOK plays a role of tumor suppressor gene or tumor-promoting gene in different tumors. However, DOK family genes play a role in promoting cancer in UVM. DOK family genes are significantly associated with drug sensitivity.

Some individuals develop prediabetes and/or diabetes following acute pancreatitis (AP). AP-induced beta-cell injury and the limited regenerative capacity of beta cells might account for pancreatic endocrine insufficiency. Previously, we found that only a few pancreatic cytokeratin 5 positive (Krt5+) cells differentiated into beta cells in the murine AP model, which was insufficient to maintain glucose homeostasis. Notch signaling determines pancreatic progenitor differentiation in pancreas development. This study aimed to examine whether Notch signaling inhibition could promote pancreatic Krt5+ cell differentiation into beta cells and improve glucose homeostasis following AP. Pancreatic tissues from patients with acute necrotizing pancreatitis (ANP) were used to evaluate beta-cell injury, Krt5+ cell activation and differentiation, and Notch activity. The murine AP model was induced by cerulein, and the effect of Notch inhibition on Krt5+ cell differentiation was evaluated both in vivo and in vitro. The results demonstrated beta-cell loss in ANP patients and AP mice. Krt5+ cells were activated in ANP pancreases along with persistently elevated Notch activity, which resulted in the formation of massive duct-like structures. AP mice that received Notch inhibitor showed that impaired glucose tolerance was reversed 7 and 15 days following AP, and increased numbers of newborn small islets due to increased differentiation of Krt5+ cells to beta cells to some extent. In addition, Krt5+ cells isolated from AP mice showed increased differentiation to beta cells by Notch inhibition. Collectively, these findings suggest that beta-cell loss contributes to pancreatic endocrine insufficiency following AP, and inhibition of Notch activity promotes pancreatic Krt5+ cell differentiation to beta cells and improves glucose homeostasis. The findings from this study may shed light on the potential treatment of prediabetes/diabetes following AP.

Plenoptic cameras are increasingly gaining attention in various fields due to their ability to capture both spatial and angular information of light rays. Accurate geometric calibration can lay a solid foundation for the applications that use the plenoptic camera. In this paper, to the best of our knowledge, we first introduce an accurate corner detection method based on a novel selection and refinement strategy. The detected-corner candidates on raw images are selected by a random sample consensus (RANSAC)-based algorithm and optimized by the photometric similarity, as well as the sub-pixel refinement. In addition, a robust and accurate stepwise calibration method is proposed based on separated intrinsic parameters, including parameters related to the pinhole model and those unique to the plenoptic camera. Experiments on both simulated and real data demonstrate that our method outperforms the state-of-the-art methods and is able to support a more accurate calibration of plenoptic cameras.

OBJECTIVE: Diabetes is common in COVID-19 patients and associated with unfavorable outcomes. We aimed to describe the characteristics and outcomes and to analyze the risk factors for in-hospital mortality of COVID-19 patients with diabetes. RESEARCH DESIGN AND METHODS: This two-center retrospective study was performed at two tertiary hospitals in Wuhan, China. Confirmed COVID-19 patients with diabetes (N = 153) who were discharged or died from 1 January 2020 to 8 March 2020 were identified. One sex- and age-matched COVID-19 patient without diabetes was randomly selected for each patient with diabetes. Demographic, clinical, and laboratory data were abstracted. Cox proportional hazards regression analyses were performed to identify the risk factors associated with the mortality in these patients. RESULTS: Of 1,561 COVID-19 patients, 153 (9.8%) had diabetes, with a median age of 64.0 (interquartile range 56.0-72.0) years. A higher proportion of intensive care unit admission (17.6% vs. 7.8%, P = 0.01) and more fatal cases (20.3% vs. 10.5%, P = 0.017) were identified in COVID-19 patients with diabetes than in the matched patients. Multivariable Cox regression analyses of these 306 patients showed that hypertension (hazard ratio [HR] 2.50, 95% CI 1.30-4.78), cardiovascular disease (HR 2.24, 95% CI 1.19-4.23), and chronic pulmonary disease (HR 2.51, 95% CI 1.07-5.90) were independently associated with in-hospital death. Diabetes (HR 1.58, 95% CI 0.84-2.99) was not statistically significantly associated with in-hospital death after adjustment. Among patients with diabetes, nonsurvivors were older (76.0 vs. 63.0 years), most were male (71.0% vs. 29.0%), and they were more likely to have underlying hypertension (83.9% vs. 50.0%) and cardiovascular disease (45.2% vs. 14.8%) (all P values <0.05). Age ≥70 years (HR 2.39, 95% CI 1.03-5.56) and hypertension (HR 3.10, 95% CI 1.14-8.44) were independent risk factors for in-hospital death of patients with diabetes. CONCLUSIONS: COVID-19 patients with diabetes had worse outcomes compared with the sex- and age-matched patients without diabetes. Older age and comorbid hypertension independently contributed to in-hospital death of patients with diabetes.

Skylight polarization is used for navigation by some birds and insects. Skylight polarization also has potential for human navigation applications. Its advantages include relative immunity from interference and the absence of error accumulation over time. However, there are presently few examples of practical applications for polarization navigation technology. The main reason is its weak robustness during cloudy weather conditions. In this paper, the real-time measurement of the sky light polarization pattern across the sky has been achieved with a wide field of view camera. The images were processed under a new reference coordinate system to clearly display the symmetrical distribution of angle of polarization with respect to the solar meridian. A new algorithm for the extraction of the image axis of symmetry is proposed, in which the real-time azimuth angle between the camera and the solar meridian is accurately calculated. Our experimental results under different weather conditions show that polarization navigation has high accuracy, is strongly robust, and performs well during fog and haze, clouds, and strong sunlight.

The sky light polarization navigator has many advantages, such as low cost, no decrease in accuracy with continuous operation, etc. However, current celestial polarization measurement methods often suffer from low performance when the sky is covered by clouds, which reduce the accuracy of navigation. In this paper we introduce a new method and structure based on a handheld light field camera and a radial polarizer, composed of an S-wave plate and a linear polarizer, to detect the sky light polarization pattern across a wide field of view in a single snapshot. Each micro-subimage has a special intensity distribution. After extracting the texture feature of these subimages, stable distribution information of the angle of polarization under a cloudy sky can be obtained. Our experimental results match well with the predicted properties of the theory. Because the polarization pattern is obtained through image processing, rather than traditional methods based on mathematical computation, this method is less sensitive to errors of pixel gray value and thus has better anti-interference performance.

Stable information of a sky light polarization pattern can be used for navigation with various advantages such as better performance of anti-interference, no "error cumulative effect," and so on. But the existing method of sky light polarization measurement is weak in real-time performance or with a complex system. Inspired by the navigational capability of a Cataglyphis with its compound eyes, we introduce a new approach to acquire the all-sky image under different polarization directions with one camera and without a rotating polarizer, so as to detect the polarization pattern across the full sky in a single snapshot. Our system is based on a handheld light field camera with a wide-angle lens and a triplet linear polarizer placed over its aperture stop. Experimental results agree with the theoretical predictions. Not only real-time detection but simple and costless architecture demonstrates the superiority of the approach proposed in this paper.

Screening of zearalenone (ZEN)-producing species and monitoring of ZEN in feed were performed by using anti-zearalenone monoclonal antibodies. ELISA recoveries of ZEN from corn distillers dried grains with solubles (DDGS) feed, corn feed, rice bran, soybean meal, wheat bran and rapeseed dregs were between 78.6% and 88.6%. ZEN recovery from culture media was 96.3% at the spiked level of 500 µg/kg. Eighty-three samples of DDGS feed, corn feed and other fee ingredients were collected from 11 provinces of China and analysed for ZEN. Average ZEN levels were 563.4 µg/kg for DDGS feed, 393.1 µg/kg for corn feed and 65.7 µg/kg for other feed ingredients. Eighteen Fusarium species such as Fusarium graminearum which could produce ZEN were isolated from corn feed and other feed ingredients.
