BACKGROUND: Non-small cell lung cancer (NSCLC) is one of the predominant malignancies globally. Percutaneous thermal ablation (PTA) has gained widespread use among NSCLC patients, with the potential to elicit immune responses but limited therapeutic efficacies for advanced-stage disease. T-helper type 9 (Th9) cells are a subset of CD4+ effector T cells with robust and persistent anti-tumor effects. This study proposes to develop PTA-Th9 cell integrated therapy as a potential strategy for NSCLC treatment. METHODS: The therapeutic efficacies were measured in mice models with subcutaneously transplanted, recurrence, or lung metastatic tumors. The tumor microenvironments (TMEs) were evaluated by flow cytometry. The cytokine levels were assessed by ELISA. The signaling molecules were determined by quantitative PCR and Western blotting. The translational potential was tested in the humanized NSCLC patient-derived xenograft (PDX) model. RESULTS: We find that PTA combined with adoptive Th9 cell transfer therapy substantially suppresses tumor growth, recurrence, and lung metastasis, ultimately extending the survival of mice with NSCLC grafts, outperforming both PTA and Th9 cell transfer monotherapy. Analysis of TMEs indicates that combinatorial therapy significantly augments tumor-infiltrating Th9 cells, boosts anti-tumor effects of CD8+ T cells, and remodels tumor immunosuppressive microenvironments. Moreover, combinatorial therapy significantly strengthens the regional and circulation immune response of CD8+ T cells in mice with tumor lung metastasis and induces peripheral CD8+ T effector memory cells in mice with tumor recurrence. Mechanically, PTA reinforces the anti-tumor ability of Th9 cells primarily through upregulating interleukin (IL)-1ß and subsequently activating the downstream STAT1/IRF1 pathway, which could be effectively blocked by intercepting IL-1ß signaling. Finally, the enhanced therapeutic effect of combinatorial therapy is validated in humanized NSCLC PDX models. CONCLUSIONS: Collectively, this study demonstrates that combinatorial therapy displays robust and durable anti-tumor efficacy and excellent translational potential, offering excellent prospects for translation and emerging as a promising approach for NSCLC treatment.

BACKGROUND: Miscarriage is a frustrating complication of pregnancy that is common among women of reproductive age. Insufficient decidualization which not only impairs embryo implantation but disturbs fetomaternal immune-tolerance, has been widely regarded as a major cause of miscarriage; however, the underlying mechanisms resulting in decidual impairment are largely unknown. METHODS: With informed consent, decidual tissue from patients with spontaneous abortion or normal pregnant women was collected to detect the expression profile of UCHL1. Human endometrial stromal cells (HESCs) were used to explore the roles of UCHL1 in decidualization and dNK modulation, as well as the mechanisms involved. C57/BL6 female mice (7-10 weeks old) were used to construct pregnancy model or artificially induced decidualization model to evaluate the effect of UCHL1 on mice decidualization and pregnancy outcome. RESULTS: The Ubiquitin C-terminal hydrolase L1 (UCHL1), as a deubiquitinating enzyme, was significantly downregulated in decidua from patients with miscarriage, along with impaired decidualization and decreased dNKs. Blockage of UCHL1 led to insufficient decidualization and resultant decreased expression of cytokines CXCL12, IL-15, TGF-ß which were critical for generation of decidual NK cells (dNKs), whereas UCHL1 overexpression enhanced decidualization accompanied by increase in dNKs. Mechanistically, the promotion of UCHL1 on decidualization was dependent on its deubiquitinating activity, and intervention of UCHL1 inhibited the activation of JAK2/STAT3 signaling pathway, resulting in aberrant decidualization and decreased production of cytokines associated with dNKs modulation. Furthermore, we found that inhibition of UCHL1 also disrupted the decidualization in mice and eventually caused adverse pregnancy outcome. CONCLUSIONS: UCHL1 plays significant roles in decidualization and dNKs modulation during pregnancy in both humans and mice. Its deficiency indicates a poor pregnancy outcome due to defective decidualization, making UCHL1 a potential target for the diagnosis and treatment of miscarriage.

BACKGROUND: As is known, CD4 cell count is a significant parameter predicting HIV progression, opportunistic infections and death in HIV-infected individuals, as well was an important indicator for initiating antiretroviral therapy (ART). In China's National Free Antiretroviral Treatment Program, people with HIV (PWH) on ART can receive a CD4 count test at least once every six months. Importantly, the baseline CD4 count (before ART initiation) is significantly correlated with ART and even prognosis, but the influence of the peak CD4 cell count on ART and/or clinical outcomes is still unknown. METHODS: A retrospective study was conducted among 7965 PWH who received ART from October 2003 to September 2022 at Yunnan Infectious Disease Hospital. Clinical features and laboratory data were collected and analyzed by Chi-square test, univariate and multivariate Cox regression analysis. After elimination of confounding variables, multivariate Cox regression analysis was performed to identify survival-related factors. RESULTS: Of a total of 7965 PWH in the ART treatment cohort who met the inclusion and exclusion criteria, 7939 were finally included in the subsequent analyses. First, it was found that the proportion of clinical variables, including sex, age distribution, interval from diagnosis to ART initiation, marital status, and others, was significantly different between the living and dead groups (P < 0.05). Impressively, significantly more PWH had the higher level of baseline, peak and recent CD4 cell counts in the living group compared to those in the dead group. Due to multicollinearity effect, after excluding confounders, the following factors were found to be significantly associated with mortality by multivariate Cox regression analysis: (1) male sex (hazard ratio (HR) = 1.268 [1.032, 1.559]; P = 0.024); (2) time from HIV confirmation to ART initiation ≥ 6 months (HR = 1.962 [1.631, 2.360]; P < 0.001); (3) peak CD4 cell count: Peak CD4 < 100cells/µL group (HR = 16.093 [12.041, 21.508]; P < 0.001), 100cells/µL ≤ x < 200cells/µL group (HR = 7.904 [6.148, 10.160]; P < 0.001), 200cells/µL ≤ x < 350cells/µL group (HR = 3.166 [2.519, 3.980]; P < 0.001), 350cells/µL ≤ x < 500cells/µL group (HR = 1.668 [1.291, 2.155]; P < 0.001). CONCLUSION: Interestingly, patients in male, time from HIV confirmation to ART initiation ≥ 6 months, or peak CD4 count < 500 cells/µl had inferior clinical outcomes, in other word, a lower peak CD4 cell count significantly increased the risk of death, and peak CD4 cell was independent in predicting the overall survival of PWH. It is important to promote "early diagnosis and treatment of HIV" and regularly monitor CD4 levels in HIV/AIDS to evaluate the efficacy of ART and immune reconstitution, and optimize the ART regimen in time to further reduce the mortality of PWH.

In this study, two new species, Rhizoplacaadpressa Y. Y. Zhang & Li S. Wang and R.auriculata Y. Y. Zhang, Li S. Wang & Printzen, are described from Southwest China, based on their morphology, phylogeny and chemistry. In phylogeny, the two new species are monophyletic, and sister to each other within Rhizoplacachrysoleuca-complex. Rhizoplacaadpressa is characterized by its placodioid and closely adnate thallus, pale green and heavily pruinose upper surface, narrow (ca. 1 mm) and white free margin on the lower surface of marginal squamules, the absence of a lower cortex, and its basally non-constricted apothecia with orange discs that turn reddish-brown at maturity. Rhizoplacaauriculata is characterized by its squamulose to placodioid thallus, yellowish green and marginally pruinose squamules, wide (1-3 mm) and bluish-black free margin on the lower surface of marginal squamules, the absence of a lower cortex, and its basally constricted apothecia with persistently orange discs. Rhizoplacaadpressa and R.auriculata share the same secondary metabolites of usnic and placodiolic acids.

Detection of cytoplasmic DNA is an essential biological mechanism that elicits IFN-dependent and immune-related responses. A better understanding of the mechanisms regulating cytoplasmic DNA sensing in tumor cells could help identify immunotherapeutic strategies to improve cancer treatment. Here we identified abundant cytoplasmic DNA accumulated in lung squamous cell carcinoma (LUSC) cells. DNA-PK, but not cGAS, functioned as a specific cytoplasmic DNA sensor to activate downstream ZAK/AKT/mTOR signaling, thereby enhancing the viability, motility, and chemoresistance of LUSC cells. DNA-PK-mediated cytoplasmic DNA sensing boosted glycolysis in LUSC cells, and blocking glycolysis abolished the tumor-promoting activity of cytoplasmic DNA. Elevated DNA-PK-mediated cytoplasmic DNA sensing was positively correlated with poor prognosis of human patients with LUSC. Targeting signaling activated by cytoplasmic DNA sensing with the ZAK inhibitor iZAK2 alone or in combination with STING agonist or anti-PD-1 antibody suppressed the tumor growth and improved the survival of mouse lung cancer models and human LUSC patient-derived xenografts model. Overall, these findings established DNA-PK-mediated cytoplasmic DNA sensing as a mechanism that supports LUSC malignancy and highlight the potential of targeting this pathway for treating LUSC. SIGNIFICANCE: DNA-PK is a cytoplasmic DNA sensor that activates ZAK/AKT/mTOR signaling and boosts glycolysis to enhance malignancy and chemoresistance of lung squamous cell carcinoma.

Under the high current density, the excessive strong adsorption of H* intermediates and H2 accumulation the catalysts are the major obstacle to the industrial application of hydrogen evolation reaction (HER) catalysts. Herein, through experimental exploration, it is found that the superaerophobic Nitrogen (N)-doped carbon material can promote the rapid release of H2 and provide H* desorption site for the hydrogen spillover process, which makes it have great potential as the catalysts support for hydrogen spillover. Based on this discovery, this work develops the hydrogen spillover catalyst with electron-rich Pt sites loaded on N-doped carbon nanocage (N-CNC) with adjustable work function. Through a series of comprehensive electrochemical tests, the existence of hydrogen spillover effort has been proved. Moreover, the in situ tests showed that pyrrolic-N can activate adjacent carbon sites as the desorption sites for hydrogen spillover. The Pt@N-1-CNC with the minimum work function difference (ΔΦ) between Pt NPs and support shows superior hydrogen evolution performance, only needs overpotential of 12.2 mV to reach current density of 10 mA cm-2 , outstanding turnover frequency (TOF) (44.7 s-1 @100 mV) and superior durability under the 360 h durability tests at current density of 50 mA cm-2 .

Several specimens of Upretia from Southwest China are morphologically and phylogenetically distinct from currently recognized species in the genus. These specimens are here accommodated within a new species, Upretiazeorina Li J. Li & Printzen. It is characterized by an areolate to squamulose thallus with brown to blackish brown upper surface, pruinose, zeorine type apothecia, black discs, narrowly bacilliform conidia, and the production of gyrophoric acid. Two other specimens of Upretia from China are distinct from currently accepted species and tentatively referred to as Upretia sp. 1 and Upretia sp. 2. A key to all known species of Upretia is also provided.

CONTEXT.­: Molecular stratification of endometrial carcinoma provides more accurate prognostic information than traditional clinicopathologic features. However, because next-generation sequencing is typically recommended for polymerase epsilon (POLE) mutation detection, the practical application of a test based on molecular stratification is limited in the clinical setting. OBJECTIVE.­: To evaluate a polymerase chain reaction (PCR)-based assay for POLE mutation detection in endometrial carcinoma. DESIGN.­: We developed a PCR-based technology called Dalton Mutation Identifier Technology (Dalton-MIT) that targets 9 mutation sites within POLE exons. Endometrial carcinoma specimens from 613 patients were tested for POLE mutations. Correlations between POLE mutations with patient clinicopathologic characteristics and prognosis were analyzed. RESULTS.­: PCR detection data showed that the incidence rate of POLE mutation was 11.4% (70 of 613). Patients with POLE mutations presented better clinicopathologic characteristics and prognosis than those with non-POLE mutations. Comparison between Dalton-MIT and next-generation sequencing in 59.5% (365 of 613) of specimens showed that the sensitivity of Dalton-MIT for detecting POLE pathogenic mutations was 100%, the specificity was 99.3%, the Youden index was .993, and the κ value was .981 (P < .001). CONCLUSIONS.­: Our data demonstrate that POLE mutation detection by Dalton-MIT correlates with next-generation sequencing. This suggests that Dalton-MIT represents a promising alternative assay for detecting POLE mutations and will facilitate the wider application of molecular stratification tools for endometrial carcinoma in the clinic.

Two new multispored species from China, Lecanoraanhuiensis Li J. Li & Printzen, sp. nov. and Lecanorapseudojaponica Li J. Li & Printzen, sp. nov. are described and illustrated here, based on morphological, chemical and molecular evidence. Lecanoraanhuiensis is characterised by an epruinose, yellowish-brown to deep brown apothecial disc, an epihymenium with fine crystals, an amphithecium with small crystals, 16-spored asci and the presence of zeorin, in addition to atranorin. Lecanorapseudojaponica is characterised by an epruinose, red-brown apothecial disc, an epihymenium without crystals, an amphithecium with small crystals, 8 or 16- spored asci and the presence of zeorin and the stictic acid complex, in addition to atranorin. Phylogenetic reconstructions, based on mtSSU, nrITS and nrLSU suggest that these two species are members of the Lecanorasubfusca group. They are compared with morphologically similar and phylogenetically related species, based on a nrITS dataset. Phylogenetic results show that the multispored taxa of Lecanora are polyphyletic. The number of ascospores per ascus appears to be a taxonomic character of minor importance. Detailed descriptions, discussions and figures for the two new species from China and a key for the multispored species of Lecanora worldwide are provided.

Bird songs are considered a sexually selected trait that can reflect the individual health of males as well as the vitality of potential mates and their competitors. Vocalization features should, therefore, be related to parasite load and body condition. Here, we performed a thorough acoustic analysis of the display calls of 9-month-old sub-adults and 18-month-old adults of pasture-raised red junglefowls Gallus gallus. We investigated whether the calls of pasture-raised red junglefowls can indicate body size and body condition, in addition to the influence of parasitic infection intensity on the expression of sexual traits. We found that frequency-related syllable parameters were significantly positively correlated with wing length in adults, whereas body weight was independent of both the frequency and temporal parameters of calls. In addition, we correlated parasitic load with the expression of sexually selected traits and discovered a positive association between the intensity of parasite infection and wing length in sub-adults, independent of vocal parameters. Overall, the results suggest that the vocalization of red junglefowls may convey reliable body size information, which will facilitate further studies of different vocal parameters in the transmission of bird vocalizations.

Aerogels, as three-dimensional porous materials, have attracted much attention in almost every field owing to their unique structural properties. Designing high-entropy alloy aerogels (HEAAs) to quinary and above remains an enormous challenge due to the different reduction potentials and nucleation/growth kinetics of different constituent metals. Herein, a novel and universal chelating co-reduction strategy to prepare HEAAs at room temperature in the water phase is proposed. The addition of chelators (ethylenediaminetetraacetic acid tetrasodium salt, sodium citrate, salicylic acid, and 4,4'-bipyridine) with a certain strong coordination capacity can adjust the reduction potential of different metal components, which is the key to synthesize single-phase solid solution alloys successfully. The optimized AgRuPdAuPt HEAA can be an excellent electrocatalyst for hydrogen evolution reaction (HER) with an ultrasmall overpotential of 22 mV at 10 mA cm-2 and excellent stability for 24 h in an alkaline solution. In situ Raman spectroscopy unveils the enhanced hydrogen evolution reaction mechanism of HEAAs. Overall, this work provides a novel chelating co-reduction strategy for the facile and versatile synthesis and design of advanced HEAAs and broadens the development and utilization of multi-elemental alloy electrocatalysts.

Chitosan bead grafted by third-generation dendrimers (CB-G3) with a diameter of 1.40 mm was synthesized to investigate their performance in recovering Ag(I), Cu(II), and Pb(II) ions in aqueous media. The prepared adsorbents were characterized by XRD, FT-IR, elemental analysis, TGA, and SEM, and the effects of pH, contact time, concentration, and temperature were examined. The results showed that the adsorbents were successfully fabricated. The optimum pH value was 5, and the increased generation number contributed to adsorption capacity improvement, indicating that electrostatic interactions between amine groups and metal ions are the governing mechanism of adsorption by the CB-G3. The kinetics, isotherms, and thermodynamics of Ag(I), Cu(II), and Pb(II) adsorption onto the CB-G3 were investigated. The adsorption processes can be described using pseudo-second-order kinetic and Langmuir models. The maximum monolayer adsorption capacities were 105.62, 88.82, and 97.87 mg·g-1 for Ag(I), Cu(II), and Pb(II) at 30 °C within 210 min, respectively. Electrostatic interactions and hydrogen bonds are the main mechanisms between metal ions and N atoms. Therefore, the CB-G3 is a promising candidate for Ag(I), Cu(II), and Pb(II) adsorption owing to its splendid ability in easy separation, good adsorptivity, and reusability for efficiently adsorbing Ag(I), Cu(II), and Pb(II) ions.

We present a novel series of neutral photo-acid generators (PAGs) based on carbazole derivatives. A photo-induced 6π-electrocyclization reaction of carbazole derivatives triggers the subsequent release of halogen acids. With UV irradiation, PAGs spontaneously release acid molecules quantitatively forming polyaromatic compounds. To our knowledge, it is considered the highest quantum yield (over 85%) among Brønsted PAGs.

In this paper, a novel grafted zeolite/Fe3O4/chitosan (ZMC-MAH-TEPA) adsorbent was greenly synthesized and evaluated for the removal of Cr(VI) and Ag(I) in single and bi-component solutions. The characterization data of XRF, XRD, FT-IR, VSM, TGA, BET and SEM showed the successful fabrication of the adsorbent with abundant -NH2 and -NH- as well as great recovery properties (11.70 emu/g). The effects of experimental parameters, including pH value, initial concentration, temperature, time, and coexisting ions on single and bi-component adsorption, were investigated. The results demonstrated that the adsorption capacities increased with the enhanced shaking time and concentration until equilibrium was reached. The optimum pH value was 3 for Cr(VI) adsorption and 5 for Ag(I) adsorption. The maximum adsorption capacities for Cr(VI) and Ag(I) ions, obtained by the Langmuir model, were 50.75 and 70.12 mg·g-1 in single metal solutions and 45.45 and 58.94 mg·g-1 in bi-component metal solutions (Cr(VI)/Ag(I) = 1:1), respectively. Additionally, the adsorption mechanism of Cr(VI) and Ag(I) was explained in terms of electrostatic interaction and hydrogen bonding between metal ions and -NH2 and -NH- in ZMC-MAH-TEPA. Taken together, this study provides a clean approach to synthesizing chitosan-based adsorbents for the efficient removal of Cr(VI) and Ag(I) ions.

Intermolecular interactions, including attractive and repulsive interactions, play a vital role in manipulating functionalization of the materials from micro to macro dimensions. Despite great success in generation of ultralong organic phosphorescence (UOP) by suppressing non-radiative transitions through attractive interactions recently, there is still no consideration of repulsive interactions on UOP. Herein, we proposed a feasible approach by introducing carboxyl groups into organic phosphors, enabling formation of the intense repulsive interactions between the isolated molecules and the matrix in rigid environment. Our experimental results show a phosphor with a record lifetime and quantum efficiency up to 3.16 s and 50.0% simultaneously in film under ambient conditions. Considering the multiple functions of the flexible films, the potential applications in anti-counterfeiting, afterglow display and visual frequency indicators were demonstrated. This finding not only outlines a fundamental principle to achieve bright organic phosphorescence in film, but also expands the potential applications of UOP materials.

Robust species delimitations provide a foundation for investigating speciation, phylogeography, and conservation. Here we attempted to elucidate species boundaries in the cosmopolitan lichen-forming fungal taxon Lecanora polytropa. This nominal taxon is morphologically variable, with distinct populations occurring on all seven continents. To delimit candidate species, we compiled ITS sequence data from populations worldwide. For a subset of the samples, we also generated alignments for 1209 single-copy nuclear genes and an alignment spanning most of the mitochondrial genome to assess concordance among the ITS, nuclear, and mitochondrial inferences. Species partitions were empirically delimited from the ITS alignment using ASAP and bPTP. We also inferred a phylogeny for the L. polytropa clade using a four-marker dataset. ASAP species delimitations revealed up to 103 species in the L. polytropa clade, with 75 corresponding to the nominal taxon L. polytropa. Inferences from phylogenomic alignments generally supported that these represent evolutionarily independent lineages or species. Less than 10% of the candidate species were comprised of specimens from multiple continents. High levels of candidate species were recovered at local scales but generally with limited overlap across regions. Lecanora polytropa likely ranks as one of the largest species complexes of lichen-forming fungi known to date.

RESUMO

Based on the concept of responsible holistic nursing care, a whole-process dual-tutor nursing practice model is established and its application effects are explored. This paper firstly reviews the research progress of nursing workload prediction methods at home and abroad, in order to provide a reference for clinical nursing workers in China to choose a scientific, reasonable, and easy-to-use nursing workload prediction method. It is proposed to construct a nursing education management model based on small data to provide ideas and references for nursing education management to effectively predict the evolutionary trend of students' behaviour and improve the level of accurate services. The experimental group adopted a dual-tutor responsibility system for the whole-process nursing practice model, including a complete three-level supervision system: a dual-tutor teaching system, a PDCA responsibility system for continuous improvement, and a multichannel teacher-student interaction platform; the control group adopted the traditional nursing practice model.

The species Immersariacupreoatra has been included in Bellemerea. This caused us to reconsider the relationships between Bellemerea and the lecanorine species of Immersaria and to question the monophyly of Immersaria. Amongst 25 genera of the family Lecideaceae, most have lecideine apothecia, the exceptions being Bellemerea and Koerberiella, which have lecanorine apothecia. According to previous classifications, Immersaria included species with both lecanorine and lecideine apothecia. A five-loci phylogenetic tree (nrITS, nrLSU, RPB1, RPB2, and mtSSU) for Lecideaceae showed that Immersaria was split into two clades: firstly, all the lecideine apotheciate species and secondly, all the lecanorine apotheciate species. The latter clade was closely related to the remaining lecanorine apotheciate genera: Bellemerea and Koerberiella. Therefore, the genus concept of Immersaria is revised accordingly and a new lecanorine genus Lecaimmeria is proposed. Furthermore, four new species for Immersaria and seven new species and three new combinations for the new genus Lecaimmeria are proposed. Keys to Immersaria and the new genus Lecaimmeria are provided.
